Abstract:Both clenbuterol (CLB) and ractopamine (RAC) are beta-adrenergic agonists. After long-term excessive intake, there will be adverse reactions such as headache, chest tightness, limb numbness, and serious lifethreatening. Simultaneous detection of CLB and RAC in related samples is of great importance for human health. In this work, we outline a microfluidics-based indirect competitive immunoassay (MICI) system that can sensitively detect residual CLB and RAC in pork, swine blood and swine urine. The rapid detection of multiple samples can be achieved in one chip, which greatly improves the detection efficiency. This method has good stability and reproducibility and the microfluidic chips are easy to manufacture. The linear ranges for CLB and RAC detection by MICI are 0.1-2.5 ng/mL and 0.1-5 ng/mL, and the limits of detection (LODs) are 0.094 ng/mL and 0.091 ng/mL, respectively. This straightforward and portable immunoassay system provides a good platform for rapid detection of harmful substances in food samples. Abstract:Phenylethanolamine A (PA) is a new kind of beta-agonist, which was illegally used as a feed additive for growth promotion in China. In this study, a novel immunochromatographic assay (ICA) based on surface-enhanced Raman scattering (SERS) for the ultrasensitive and quantitative detection of phenylethanolamine A is presented. The principle of this new ICA is similar to that based on colloidal gold particles, but using Au(MBA)@Ag-Ab [e.g., polyclonal antibody of PA labeled Au@Ag core-shell nanoparticles (NPs) sandwiched with a Raman reporter (4-mercaptobenzoic acid, MBA)] as a probe. After ICA procedures, the specific Raman scattering intensity of MBA on the test line was measured for quantitative detection of PA. This assay was completed within 15 min. The IC50 and limit of detection (LOD) values of the ICA for PA detection were 0.06 ng mL(-1) and 0.32 pg mL(-1), respectively, which were 1-3 orders of magnitude lower than those obtained by other immunoassays, indicating the ultrasensitivity of this ICA. There was no cross-reactivity (CR) of the assay with another three beta-agonists (ractopamine, clenbuterol, and salbutamol), suggesting high specificity of the SERS-based ICA. A spiking experiment revealed that the recoveries of PA from pig urine samples were in range of 99.9- 101.2% with relative standard deviations (RSDs) of 3.6-5.8%. The results demonstrated that this SERS-based ICA was able to quantitatively detect PA in urine samples with high sensitivity, specificity, precision, and accuracy and might be a powerful method for the analysis of other target analytes in the food area.

Abstract:A nitrogen-doped Fe-MOF shows a high specific surface area and excellent electrical conductivity after high temperature carbonization. A novel electrochemical sensor based on a N@Fe-MOF@C loaded dual-template molecularly imprinted polymer (DTMIP) modified glassy carbon electrode (GCE) was proposed for the rapid and ultra-sensitive simultaneous detection of clenbuterol hydrochloride (CLB) and ractopamine (RAC). N@Fe-MOF@C combined with a MIP significantly enhanced the electrical signal. Cyclic voltammetry (CV) was used to detect CLB and RAC. The electrochemical polymerization was conducted with O-phenylenediamine as the functional monomer and CLB and RAC as template molecules. The factors affecting the sensor response were optimized. Under the optimal experimental conditions, the CV current response showed a linear range of 10-12-8 × 10-9 M for both CLB and RAC, and the detection limit (LOD) for both CLB and RAC was 3.03 × 10-13 M (S/N = 3). This electrochemical sensing system has high sensitivity, selectivity, excellent reproducibility, repeatability and stability. The recoveries of the actual samples (97.4%-101.2%) and reasonable relative standard deviations (RSDs) (1.06%-3.17%) indicate the practicability of the sensor system. The system has high application value in the rapid detection of CLB and RAC in clenbuterol hydrochloride tablets, human urine and raw pork.
An Automated Immunoassay Based on Magnetic Microspheres for the Determination of Clenbuterol in Food

Abstract:An automated magnetic immunoassay for the determination of clenbuterol in food is reported. Magnetic microspheres were employed as the solid phase for binding anti-clenbuterol antibodies. Fe3O4 microspheres (506.9nm) were synthesized from FeCl3 and ethylene glycol. The magnetic saturation value was 72.4emu g(-1) for the microspheres. The magnetic microspheres were subsequently surface modified using tetraethyl orthosilicate. Carboxylate groups were produced on the surface of the microspheres and the anti-clenbuterol antibodies were covalently attached using 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide and N-hydroxysuccinimide. The incubation, washing, and detection procedures were automated. The limits of detection and quantification were 0.02ng mL(-1) and 0.1ng mL(-1), respectively. Recoveries of clenbuterol in sausage, pork, and beef were 93.5% to 110.0%, 98.9- 109%, and 94.0-101.9%, respectively. This approach allowed analysis in 25min compared to 50min for a commercial method while reducing the reagents required.
Detection of Ractopamine and Clenbuterol Hydrochloride Residues in Pork Using Surface Enhanced Raman Spectroscopy

Abstract:Surface enhanced Raman spectroscopy (SERS) coupled with chemometric methods, such as adaptive iteratively reweighted penalized least squares (AIR-PLS), wavelet transform, and least squares support vector machine (LSSVM), was investigated to realize the rapid detection and identifi cation of ractopamine (RAC) and clenbuterol hydrochloride (CL) residues in pork. First-level wavelet detail signal intensities at 1168 cm –1 were used to establish a standard curve of the RAC residues in pork, and the linear regression equation and the correlation coefficient were y = –4. 3683x – 11.059 and –0.9726. Second-level wavelet detail signal intensities at 1258 cm –1 were used to establish a standard curve of the CL residues in pork, and the linear regression equation and the correlation coeffi cient were y=33.595x + 36.538 and 0.9842. The second-level wavelet detail signals of the SERS spectra were selected as the inputs of the LSSVM classifi cation model for the identifi cation of the RAC and CL residues in pork, with a total accuracy rate reaching 100%. The experimental results demonstrate that the proposed method based on SERS is a good detection scheme for the rapid detection and identifi cation of RAC and CL residues in pork.

Abstract:Direct and sensitive detection of multiple illegal additives in complex food samples is still a challenge in on-site detection. In this study, an ultrasensitive immunochromatographic assay (ICA) using magnetic Fe3O4@Au nanotags as a capture/detection difunctional tool was developed for the direct detection of β2-adrenoceptor agonists in real samples. The Fe3O4@Au tag is composed of a large magnetic core (~160 nm), a rough Au nanoshell, dense surface-modified Raman molecules, and antibodies, which cannot only effectively enrich targets from complex solutions to reduce the matrix effects of food samples and improve detection sensitivity, but also provide strong colorimetric/surface-enhanced Raman scattering (SERS) dual signals for ICA testing. The dual readout signals of the proposed ICA can meet the detection requirements in different environments. Specifically, the colorimetric signal allows for rapid visual detection of the analyte, and the SERS signal is used for the sensitive and quantitative detection modes. The proposed dual-signal ICA can achieve the simultaneous determination of two illegal additives, namely, clenbuterol hydrochloride and ractopamine. The detection limits for the two targets via colorimetric and SERS signals were down to ng mL−1 and pg mL−1 levels, respectively. Moreover, the proposed assay has demonstrated high accuracy and stability in real food samples.
Simultaneous Determination of Four Β2-Agonist Residues in Pig Feed and Urine by Capillary Electrophoresis with Field Amplified Sample Injection and Electrochemiluminescent Detection

Abstract:A novel method for simultaneous determination of clenbuterol (CLB), terbutaline (TER), salbutamol (SAL), and procaterol (PRO) was proposed based on capillary electrophoretic (CE) separation and electrochemiluminescent (ECL) detection. Matrix solid phase dispersion (MSPD) and ion exchange coupled with mesoporous nanocarbon adsorption were adopted to pretreat solid and aqueous samples, respectively. The sensitivity was enhanced by about 9-fold due to the online stacking via field amplified sample injection. The conditions for CE separation and ECL detection were investigated in detail. The four beta(2)-agonists were baseline separated under the optimum conditions within 10 min. The linear ranges were 0.25-10.00 mu mol L-1 with detection limits (S/N = 3) of 0.081, 0.076 and 0.034 mu mol L-1 for CLB, TER and SAL, respectively, and 0.5-20.00 mu mol L-1 for PRO with a detection limit of 0.12 mu mol L-1. The developed method was successfully applied to determine the four b2-agonists in real samples, offering average recoveries ranging from 88.9% to 107.1% for pig feed, and from 86.7% to 109.1% for pig urine.
Rapid Detection of Clenbuterol Residues in Pork Using Enhanced Raman Spectroscopy

Abstract:Clenbuterol (CB) is a synthetic β-receptor agonist which can be used to improve carcass leanness in swine, but its residues in pork also pose health risks. In this report, surface-enhanced Raman scattering (SERS) technology was used to achieve rapid detection and identification of clenbuterol hydrochloride (CB) residues. First, the effects of several different organic solvents on the extraction efficiency were compared, and it was found that clenbuterol in pork had a better enhancement effect using ethyl acetate as an extraction agent. Then, SERS signals of clenbuterol in different solvents were compared, and it was found that clenbuterol had a better enhancement effect in an aqueous solution. Therefore, water was chosen as the solvent for clenbuterol detection. Next, enhancement effect was compared using different concentration of sodium chloride solution as the aggregating compound. Finally, pork samples with different clenbuterol content (1, 3, 5, 7, 9, and 10 μg/g) were prepared for quantitative analysis. The SERS spectra of samples were collected with 0.5 mol/L of NaCl solution as aggregating compound and gold colloid as an enhanced substrate. Multiple scattering correction (MSC) and automatic Whittaker filter (AWF) were used for preprocessing, and the fluorescence background contained in the original Raman spectra was removed. A unary linear regression model was established between SERS intensity at 1472 cm-1 and clenbuterol content in pork samples. The model had a better linear relationship with a correlation coefficient R2 of 0.99 and a root mean square error of 0.263 μg/g. This method can be used for rapid screening of pork containing clenbuterol in the market.

Abstract:Salbutamol (SAL), a kind of β-agonist which can enhance the lean meat-to-fat ratio, has been inhibited as an additive used in animal feeds for livestock production in many countries due to its harmful effect to the consumers. In this study, an ultrasensitive and specific competitive immunochromatographic test (ICT) integrated with surface-enhanced Raman scattering (SERS) for the detection of SAL was described. The immunoprobe was prepared by immobilizing polyclonal antibody against SAL on the surface of Au@Ag nanoparticles in which the Raman reporter (4-mercaptobenzoic acid, MBA) had been sandwiched. After ICT procedures, the specific SERS signals generated from MBA on the test line of the ICT strip were measured for the quantitative determination of SAL. The assay was completed in 15 min. The IC50 and the limit of detection (LOD) values of the assay for SAL were 0.028 ng mL−1 and 3.0 pg mL−1, respectively. There was no cross-reactivity (CR) of the assay with other three β-agonists (clenbuterol, phenylethanolamine A, and ractopamine), showing high specificity of the assay. Spiking experiments indicated that the average recoveries (n = 3) of SAL from swine feed, meat, and urine samples were in ranges of 98.4–105.2 % with the relative standard deviations (RSDs) of 1.7–7.8 %. The results demonstrated that the proposed ICT was a feasible method for ultrasensitive and specific detection SAL in swine feed, meat and urine samples, and could be extended for the detection of other target analytes.

Abstract:Clenbuterol (CL), as a feed additive, has been banned in many countries due to its potential threat to human health. In detection of CL, a fast, low‐cost technique with high accuracy and specificity would be ideal for its administrative on‐field inspections. Among the attempts to pursue a reliable detection tool of CL, a technique that combines surface enhanced Raman spectroscopy (SERS) and immunoassay, is close to meet the requirements as above. However, multiple steps of interactions between CL analyte, antibody, and antigen are involved in this method, and under conventional setup, the operation of SERS/immunoassay were unwieldy. In this paper, to facilitate a more manageable sample manipulation for SERS‐immunoassay measurement, a 3D paper chip was suggested. A switch‐on‐chip multilayered (abbreviated as SoCM‐) microfluidic paper‐based analysis device (μPad) was fabricated to provide operators with manual switches on the interactions between different microfluids. Besides, on a detection slip we made on the main body of our SoCM‐μPad, antigen was anchored in pattern. With this architecture, multistep interactions between the CL analyte in swine hair extract and the SERS probe‐modified antibody and antigen, were managed for on‐chip SERS‐immunoassay detection. This would be very attractive for fast, cheap, accurate, and on‐site specific detection of CL from real samples.
Simultaneous Detection of Ractopamine, Clenbuterol and Salbutamol by Suspension Array Technology

Abstract:A multi-residue analytical method was established for the simultaneous detection of ractopamine (RAC), clenbuterol(CL) and salbutamol(SAL) by suspension array technology. Based on the principle of indirect competitive enzyme linked immunosorbent assay, three veterinary antigens coupled with different fluorescent microspheres as probes were prepared and the phycoerythrin (PE)-labeled secondary antibodies were used as reporter signal. Standard curves of three veterinary drugs were separately established under optimum conditions. According to the specific recognition experiment, the standard curves for simultaneous detection of those three veterinary drugs were established. Good linearity was obtained in the concentration range of 1 - 500 mu g/L, 0. 1 - 500 mu g/L, 1 - 100 mu g/L, and the detection limits were 0. 68, 0. 095 and 0. 884 mu g/L for RAC, CL, SAL, respectively, with correlation coefficients (R-2) > 0. 99 for all three drugs. The cross. reactivity of other structural analogues was lower than 1. 5%. Good recoveries were found at four spiked levels for both single and mixed standards in real samples.

Abstract:Lateral flow immunoassay (LFIA) has emerged as an effective technique in the field of food safety and environmental monitoring. However, sensitive and quantitative detection is still challenging for LFIAs in complex environments. In this work, a dual-model colorimetric/SERS lateral flow immunoassay for ultrasensitive determination of clenbuterol was constructed based on a metallic core-shell Au/Au nanostar acting as a multifunction tag. Raman reporter molecules are located between the core (AuNP) and shell (Au nanostar) to form a sandwich structure, which contributes to eliminate the environmental interference and improve the detection stability. In addition, the Au/Au nanostar provides a much higher Raman enhancement due to the presence of sharp tips and larger surface roughness in comparison with gold nanoparticles (AuNPs). Thus, on the basis of the antibody-antigen interaction, the dual-model immunoassay can produce strong colorimetric and surface-enhanced Raman spectroscopy (SERS) signals for highly sensitive detection of the target analyte, clenbuterol. Under optimal conditions, clenbuterol could be detected by the colorimetric model with a visual detection limit of 5 ng/mL. Meanwhile, the SERS signal of the Au/Au nanostar was accumulated on the test line for the SERS model detection with a quantitative detection limit as low as 0.05 ng/mL, which is at least 200-fold lower than that of the traditional AuNPs-based immunoassay. Furthermore, recovery rates of the proposed method in food samples were 86-110%. This dual-model immunoassay provides an effective tool for antibiotic residues analysis and demonstrates a broad potential for future applications in food safety monitoring.
Fe3O4@Au@Ag Nanoparticles As Surface-Enhanced Raman Spectroscopy Substrates for Sensitive Detection of Clenbuterol Hydrochloride in Pork with the Use of Aptamer Binding

Abstract:Clenbuteml hydrochloride (CLB), a kind of p-agonist, has been illegally used as a feed additive for growth promotion in the food industry in many countries. Herein, surface-enhanced Raman scattering (SERS) combined with aptamer recognition was used for the ultrasensitive and quantitative detection of CLB. Fe3O4@Au@Ag nanoparticles displaying strong SERS enhancement were synthesized and used as the active substrate. Aptamer against CLB was immobilized on the surface of the substrate to form the capture probe. The complementary DNA (cDNA) was linked to gold nanoparticles (AuNPs) carrying the Raman reporter 4-mercaptobenzoic acid (4-MBA) to form the signal probe. The detection of CLB was carried out by competitive binding between free CLB and cDNA linked on the signal probe with the aptamer immobilized on the capture probe. The SERS intensity was significantly improved by the coupling effect between the Fe3O4@Au@AgNPs and AuNPs, resulting in high sensitivity for the quantification assays with a limit of detection of 0.003 ng mL(-1). The recoveries of the CLB from spiked pork samples were 90.7-108.0% and the coefficients of variation (CV%) were 2.2-3.1%, demonstrating the high precision and accuracy of the assay.

Abstract:A surface-enhanced Raman scattering (SERS)/electrochemical dual-signal readout immunosensor was developed for simultaneous detection of β-adrenergic agonists salbutamol (SAL), ractopamine (RAC) and phenylethanolamine A (PA). The highly-ordered gold/silver bimetallic cavity array (BMCA) was prepared by electrodepositing Au/Ag nanoparticles to the interstice of highly ordered close-packed polystyrene templates. After electrochemical and SERS characterization, the BMCA was used as the substrate for constructing SERS/electrochemical dual-signal readout immunosensor. 3,3′,5,5′-tetramethylbenzidine (TMB), methylene blue (MB) and Nile blue (NB) were selected as the dual-signal reporters, and hybridization chain reaction (HCR) was used as the signal amplifier. The immunoprobe was prepared by absorption of the antibody (Ab) and constructing HCR system embedded with electro/SERS reporter on Au nanoparticles (NPs). After competitive immuno-reaction between coating antigen and analyte for limited Ab on immunoprobe, the SERS/electrochemical dual-signals on BMCA were measured for quantitatively detecting SAL, RAC and PA simultaneously. SAL, RAC and PA were detected in concentration range of 1 pg mL −1 to 100 ng mL −1 with LOD of 0.8, 0.4, and 1.3 pg mL −1 , respectively. The applicability of the proposed immunosensor in spiked pork liver samples was verified by the recovery of 95.0%–108.5% with RSD of 6.9%–10.7%. It was proven that the immunosensor was able to detect multiple β-adrenergic agonists with high sensitivity, specificity, accuracy and precision. The immunosensor can be used as a platform for the determination of other small molecular compounds in biological, food and environmental analytical fields.

Raman Enhancement Effects of Gold Nanoparticles with Different Particle Sizes on Clenbuterol and Ractopamine

Abstract:Clenbuterol and ractopamine can be used to improve carcass leanness in swine, but their residues in pork also pose health risks. This study used surface-enhanced Raman scatter (SERS) technology to establish MLR and PLSR prediction models for clenbuterol and ractopamine, respectively, to achieve rapid detection and identification of ractopamine (RAC) and clenbuterol hydrochloride (CL) residues. In this report, the detection of clenbuterol and ractopamine was carried out using three gold colloids with different particle sizes as enhanced substrates. It was found that the larger particle size of gold nanoparticles was, the better enhancement effect was, which indicated that gold nanoparticles with larger particle sizes were more conducive to the adsorption of RAC and CL. Then, the SERS spectra of 0.1-10 mu g/mL clenbuterol and ractopamine solution were collected by using NaCl aqueous solution of 1 mol/L as aggregating compound and gold colloid A as enhanced substrate. Automatic Whittaker Filter (AWF) was used for preprocessing to reduce fluorescence background contained in the original Raman spectra collected. Finally, models were established between SERS intensity at each characteristic peak and the concentration of clenbuterol and ractopamine, respectively. The results show that the PLSR model for clenbuterol using six peaks was stable. The correlation coefficient R2 and the root mean square error of correction set were 0.99 and 0.025 mu g/mL, respectively. The correlation coefficient R2 and the root mean square error of prediction set were 0.99 and 0.026 mu g/mL, respectively. The MLR model for ractopamine using six peaks had a higher correlation coefficient and a lower root mean square error (RMSEC) than other models. The correlation coeffi-cient R2 and the root mean square error of correction set were 0.99 and 0.043 mu g/mL, respectively. The corre-lation coefficient R2 and the root mean square error of prediction set were 0.99 and 0.05 mu g/mL, respectively. The detection limit (LOD) value of this method for the detection of clenbuterol and ractopamine was 18 ng/mL and 24 ng/mL, respectively. This study is helpful for further development of a simple SERS detection method of clenbuterol and ractopamine based on gold colloid.

Abstract:Rapid, sensitive, and accurate detection of adrenoceptor agonists is a significant research topic in the fields of food safety and public health. Immunoassays are among the most widely used methods for detecting adrenoceptor agonists. In recent years, surface-enhanced Raman spectroscopy combined with immunoassay (SERS-IA) has become an effective technique for improving detection sensitivity. This review focuses on the innovation of Raman reporter molecules and substrate materials for the SERS-IA of adrenoceptor agonists. In addition, it also investigates the challenges involved in potentially applying SERS-IA in the detection of adrenoceptor agonists. Overall, this review provides insight into the design and application of SERS-IA for the detection of adrenoceptor agonists, which is critical for animal-derived food safety and public health.

Abstract:In this paper, two immunoassay methods based on SERS are developed for multiplex analysis, both of which stemmed from the concept of forming a sandwich structure 'capture antibody substrate/antigen/Raman-reporter-labeled immuno-nanoparticles'. They are two-molecule labeled one-nanoparticle and one-molecule labeled two-nanoparticle methods. In both the methods, two different antibodies covalently bound to a solid substrate can specifically capture two different antigens from a sample. The captured antigens in turn bind selectively to their corresponding antibodies immobilized on Raman-reporter-labeled nanoparticles. Multianalyte immunoassay is successfully demonstrated by the detection of characteristic Raman bands of the probe molecules only when the antigen and antibody are matched. Spatial Scattering Raman Spectral Characteristics of Clenbuterol

Abstract:Problems with “clenbuterol”, a synthetic β2-adrenergic stimulant that includes clenbuterol, ractopamine, and albuterol, have been increasing in recent years. This article focuses on the detection of albuterol, chemically called 3,6-dihydroxy1-methyl-5-oxo-3,5-dihydro-2H-indolium betaine. It is difficult to directly measure the content of clenbuterol in pork due to its messy and uneven composition, which requires a destructive pre-treatment process that takes time and effort. In this study, a method for subsurface food inspection was presented based on a newly developed line-scan spatially scattering Raman spectroscopy technique. A spatially offset Raman spectroscopy system based on line-scan Raman chemical imaging system was built, which was used to collect spatially offset Raman spectra from the samples. Under the condition of one CCD exposure, the spatially offset Raman spectroscopy system can collect a series of Raman spectra simultaneously in a narrow space interval and a wide offset range. Through data analysis, the rare clenbuterol species in the sample can be determined by the position of characteristic peaks. The system was used to collect salbutamol Raman signal, and the results showed that the characteristic peaks of salbutamol were consistent with the standard characteristic peaks, so it was possible to use the system to detect lean meat in meat.
