Abstract:An aluminum chloride promoted Hantzsch reaction of N-tosylhydrazones has been developed. The reaction is general for a wide range of N-tosylhydrazones, and a series of 1,4-dihydropyridines (1,4-DHPs) were prepared in moderate to excellent yields.

Abstract:Aromatization of Hantzsch 1,4-dihydropyridines (1a-n) were promoted by tetrakis-pyridine cobalt (II) dichromate to yield corresponding pyridine derivatives (2a-n) in moderate to high yields (70-87%). The reaction was carried out under mild and convenient conditions.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:A simple and efficient synthesis of Hantzsch 1,4-dihydropyridines from the condensation of benzaldehydes with alkyl acetoacetates/benzoylacetate and aqueous ammonia in the absence of catalyst has been developed by using a continuous flow microreactor. Under optimized condition, various 1,4-dihydropyridines were obtained in 80-93% yield with high atom efficiency. Compared with using batch system, the present smooth procedure greatly accelerated the reaction due to the excellent mixing and mass transfer of reactant in micro flow system.

Abstract:A coupling reaction of N-phenoxyacetamides with N-tosylhydrazones or diazoesters through Rh(III) -catalyzed CH activation is reported. In this reaction, ortho-alkenyl phenols were obtained in good yields and with excellent regio- and stereoselectivity. Rh-carbene migratory insertion is proposed as the key step in the reaction mechanism.

Abstract:An efficient protocol for the Hantzsch synthesis of 2, 3, 5, 6-tetrasubstituted unsymmetrical pyridines has been developed through Fe(OAc)2 promoted condensation of multicomponent carbonyl compounds, in which the C4 of the pyridine rings was derived from DMF and the nitrogen atom resulted from 1, 1dichloro-2 nitroethene (DCNE). Abstract:The difluoromethyl-containing Hantzsch 1,4-dihydropyridines were synthesized in good yields by a one-pot cyclocondensation of ethyl difluoroacetoacetate (EDFAA), a variety of aromatic aldehydes and ammonium acetate under solvent and catalyst free conditions. The comparison of reaction conditions and products was made among the different 1,3-carbonyl substrates (ethyl acetoacetate, ethyl difluoroacetoacetate and ethyl trifluoroacetoacetate) for the Hantzsch reaction.

Abstract:A novel route for tandem C–C/C–N formation, annulation and aromatization of hydrazones with 1,2-dichloroethane to synthesize 1H-pyrazoles has been developed. Furthermore, the 1,2-dichloroethane serves as alkylation reagent in good to excellent yields. This methodology features mild reaction conditions and good functional group tolerance, providing a direct approach for the preparation of 1H-pyrazoles.

Abstract:It has been reported previously that treatment of arylketone-derived arylsulfonylhydrazones with NXS/(nBu)4NX affords exclusively vinyl halides. In contrast, we have found that treatment of arylaldehyde-derived arylsulfonylhydrazones with N-chlorosuccinimide in the presence of potassium hydroxide affords 1,2,4,5-tetrazine derivatives in good to excellent yields. The present reactions are carried out under metal-free and mild reaction conditions.

Abstract:An efficient palladium‐catalyzed olefination of N‐tosylhydrazones as β‐diazo phosphonate precursors with aryl halides has been developed. 2,2‐Disubstituted vinylphosphonates bearing versatile functional groups were easily accessed in moderate to excellent yields. Various aryl halides could be employed as the coupling partners, such as (hetero)arylbromides, aryliodides, and even arylchlorides. Moreover, with similar strategy (2,2‐diphenylvinyl)diarylphosphine oxides can also be attained by the reaction of N‐tosylhydrazones and aryl bromides. This protocol features easily available raw materials, simple reaction conditions, broad substrate scope as well as scale‐up ability. Moreover, the potential application of this product was exemplified by further transformations.

The simultaneous formation of C–C and C–N bonds in a single step is particularly appealing. Abstract:The palladium-catalyzed oxidative borylation reaction of N-tosylhydrazones has been developed. The reaction features mild conditions, broad substrate scope, and good functional group tolerance. It thus represents a highly efficient and practical method for the synthesis of di-, tri-, and tetrasubstituted alkenylboronates from readily available N-tosylhydrazones. One-pot Suzuki coupling and other transformations highlight the synthetic utility of the approach. DFT calculations have revealed that palladium-carbene formation and subsequent boryl migratory insertion are the key steps in the catalytic cycle.
