Abstract:From alkylidenecyclopropanes (MCPs), 4-alkylidene-5,6-dihydro-4H-pyrrolo-[1,2-c][1,2,3]triazoles 6 were prepared in moderate yields through diiodogenation, Cu(I)-catalyzed 1,3-dipolar cycloaddition and subsequent intramolecular Heck reaction.

Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:A series of novel 1,3,5-triarylpyrazoles were synthesized from flavanones, arylhydrazines, and trimethyl phosphate in an one-pot procedure. Facile reaction process, easy after-reaction workshop, and good yields are the distinct characteristics of the developed protocol. The target compounds were characterized by element analysis, infrared ray (IR), 1H NMR spectra, and electrospray ionization-mass spectrometry. The structure of representative compound (C23H20N2O3, Mr = 372.42) was further confirmed by X-ray diffraction. It crystallizes in monoclinic, space group P 21/c, a = 8.9720(5), b = 24.5523(13), c = 8.9687(6) angstrom, = 90.0000, = 102.6417(17), = 90.0000 degrees, V = 1927.76(20) angstrom 3, Z = 4, (MoK) = 0.086, F(000) = 784, Dc = 1.283 g/cm3, the final R = 0.0349 and wR = 0.0844 for 1668 observed reflections (I > 2 sigma(I))

Abstract:A series of non-annulated products: trifluoromethylpyrazolone-tethered trisubstituted methane derivatives were synthesized in high yields in acetonitrile solvent catalyzed by NH4OAc via one-pot, three-component reactions of 2-hydroxy-1,4-naphthoquinone, aromatic aldehydes and 1-aryl-3-trifluoromethyl-5-pyrazolone. Treatment of trifluoromethylpyrazolone-tethered trisubstituted methane derivatives with the dehydrating agent SOCl2/Pyridine in acetonitrile afforded the corresponding annulated fused polyheterocyclic products trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives. Furthermore, an efficient, straightforward protocol has been developed for the synthesis of trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives at medium yields via one-pot, two-step, three-component reactions. Meanwhile, the structures of typical products were further confirmed by XRD-analysis.

Abstract:The formal [3+2] cycloaddition of 1,3,5-triaryl-1,3,5-triazinanes with donor-acceptor cyclopropanes has been found to provide pyrrolidines in good to excellent yields under mild reaction conditions. Abstract:8-Bromo-7-chloro[1,2,4]triazolo[4,3-c]pyrimidines were prepared by bromine-mediated oxidative cyclization of the aldehyde-derived hydrazones. The structure of one such product was unambiguously confirmed by single-crystal X-ray analysis. While the C-5 unsubstituted 1,2,4-triazolo[4,3-c]pyrimidine chemotype is extremely susceptible to ring isomerization at ambient conditions, the C-5-substituted analogues were found to be quite stable, permitting isolation in pure form. Nevertheless, they can still be converted into the 1,2,4-triazolo[1,5-c]pyrimidines by base-or acid-promoted Dimroth rearrangement. The presence of halogen functionalities on the pyrimidine nucleus renders the products useful as versatile synthetic intermediates for the easy diversification, as evidenced by palladium-catalyzed Kumada cross-couplings and Buchwald-Hartwig amination, as well as direct aromatic substitution.

Abstract:A novel approach to chemoselective synthesis of biologically important CF3-subsituted pyrazolines was developed via a Lewis base catalyzed intermolecular triazene cycloaddition reaction of an array of terminal/internal alkenes with CF3CHN2. This strategy features a catalytic amount of 1,8-diazabicyclo[5.4.0]undec-7-ene, high yields (up to 95 %), wide substrate scope and excellent functional group tolerance (54 examples). Importantly, we preformed scaffold diversification of a panel of known pharmaceuticals, natural products, and bioactive heterocycles to generate the corresponding pyrazoline derivatives with potential broad bioactivities for further development.

Abstract:Incorporation of a trifluoromethyl group with 1,2,3-triazoles motifs was described. We explored a click reaction approach for regioselective synthesis of 1-susbstituted-4-trifluoromethyl-1,2,3-triazoles in which 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) reacts with commercial 2-bromo-3,3,3-trifluoropropene (BTP) to form 3,3,3-trifloropropyne (TFP) in situ. Arising from merits associated with the availability and stability of BTP, and the high efficiencies of CuI/1,10-Phenanthroline (Phen)-catalyzed cycloaddition reactions of azides with alkynes, this readily performed click process takes place to form the target 1,2,3-triazoles in high yields, and with a wide azide substrate scope. The potential value of this protocol was demonstrated by its application to a gram-scale reaction.

Abstract:An efficient and general method for the synthesis of 3-trifluoromethyl-4 trifluoroacetyl pyrazoles from the cycloaddition of sydnones with 1,1,1,5,5,5-hexafluoropentane-2,4-dione has been established. Using ZnI2/bpy as a catalyst in THF, this protocol proved to be general to prepare a variety of 3-trifluoromethyl-4 trifluoroacetyll pyrazoles in good yields. Abstract:An efficient and highly regioselective synthesis of 1-allenyl-5-aryl-1,2,3-triazoles from 1-sulfonyl-4-aryl-1,2,3-triazoles is disclosed. 1-Sulfonyl-4-aryl-1,2,3-triazoles reacted with propargylic alcohols in the presence of trifluoroboron etherate to furnish 1-allenyl-5-aryl-1,2,3-triazoles. When secondary alcohols were used as substrate, the outcome of the reaction was 1-alkyl-5-aryl-1,2,3-triazoles.

Abstract:An efficient synthesis of 1,3-diaryl-4-halo-1H-pyrazoles was achieved. The synthesis involves the [3 + 2] dipolar cycloaddition of 3-arylsydnones and 2-aryl-1,1-dihalo-1-alkenes. The process proceeds smoothly in moderate to excellent yields. 1,3-Diaryl-4-halo-1H-pyrazoles are found to be important intermediates that can easily be converted into 1,2,5-triaryl-substituted pyrazoles via Pd-catalyzed C-H bond activation.
