Abstract:This study explored a new rosuvastatin calcium- and heparin-loaded poly(l-lactide- co-caprolactone) (PLCL) scaffold for covered stents for treating aneurysms. The mechanism of rosuvastatin-induced endothelialization via vascular endothelial growth factor (VEGF)-A elevation was further explored. Rosu50, Rosu75, Rosu100, and phosphate-buffered saline (PBS) nanofibrous scaffolds were fabricated by coaxial electrospinning and observed by electron microscopy. Anticoagulation and pro-endothelialization properties were tested. Sixteen rabbits were selected for an in vivo assay and underwent microsurgery to establish a carotid aneurysm model. The animals were treated with covered stents and followed for 4 months using digital subtraction angiography (DSA), electron microscopy, and histology. Rosuvastatin-treated human umbilical vein endothelial cell (HUVEC) viability, function, and VEGF-A modulation were further studied to elucidate the pro-endothelialization mechanism of rosuvastatin. Our study demonstrates that rosuvastatin and heparin can be incorporated into PLCL nanofibers via electrospinning. Rosu100 nanofiber scaffolds exhibited significant anticoagulation properties. The viability of HUVECs transferred to Rosu100 nanofiber scaffolds was increased significantly. In vivo, DSA revealed that the Rosu100 group had better outcomes than the PBS group. In addition, the Rosu100 stents induced more integrated endothelialization. Further study demonstrated that rosuvastatin promoted HUVEC viability and function in vitro. The effects of rosuvastatin may be attributed to an elevation in VEGF-A. We demonstrated that rosuvastatin- and heparin-loaded PLCL-covered stents show favorable anticoagulation and pro-endothelialization properties in vitro and in vivo in a rabbit aneurysm model. VEGF-A elevation played a crucial role in rosuvastatin-promoted endothelialization. This work provides an additional option for treating cerebral aneurysms with covered stents.

Abstract:Restenosis caused by thrombopoiesis is one of the biggest hinders of endovascular stent-graft used in small-diameter vessels. Rapid endothelialization of the lumen of stent is a promising approach to prevent thrombosis. In this study, we aimed at loading heparin, a potent anticoagulants, and vascular endothelial growth factor (VEGF) into the core of poly(Llactide-co-caprolactone) nanofiber via emulsion electrospinning. The nanofiber was covered on the stent and applied in the treatment of vascular diseases such as aneurysm. The morphologies of the emulsion electrospun nanofibers and core shell structure were observed by scanning electron microscope and laser scanning confocal microscope. The release profiles of heparin and VEGF, degradation rate of nanofiber mats and cell proliferation in vitro were investigated. It was found that the release of both heparin and VEGF from the nanofiber lasted for more than 30 days without serious initial burst release. The degradation rate of nanofiber mats containing heparin and VEGF was faster than that of pure PLCL nanofiber mats. Moreover, the released VEGF could promote the proliferation of Pig iliac endothelial cells (PIECs) cultured on the nanofiber mat, which was of great benefit to stent endothelialization. The results of digital subtraction angiography (DSA) follow-up indicated the aneurysm was obliterated by separating the aneurysm dome from the blood circulation and the parent artery kept long-term patency. Results of the study demonstrated that the heparin and VEGF loaded nanofiber could provide an approach to fabricate covered stent-graft with properties of anticoagulation and induction of rapid endothelialization.
Heparin- and Rosuvastatin- loaded Poly(L-lactide-co-caprolactone) Nanofiber Aneurysm Covered Stent Inhibits Inflammatory Smooth Muscle Cell in Reducing in-stent Stenosis and Thrombosis

Abstract:Abstract Background: Endovascular covered-stent has unique advantages in treating complex intracranial aneurysms. However, in-stent stenosis and late thrombosis have become the main factors affecting the efficacy of covered-stent treatment. Smooth-muscle-cell phenotypic modulation plays an important role in late in-stent stenosis and thrombosis. Thus, covered stents loaded with drugs that can inhibit smooth-muscle-cell phenotypic modulation may lower the incidence of long-term complications. Methods: Nanofiber covered stents were prepared using coaxial electrospinning. A rabbit-carotid-artery aneurysm model was established and treated with covered stents. Angiography and histology were performed to evaluate the therapeutic efficacy and incidence rate of in-stent stenosis and thrombosis. Phenotype, function, and inflammatory factors of smooth-muscle cells were studied to explore the mechanism of rosuvastatin in smooth-muscle cells. Result: Heparin–rosuvastatin-loaded nanofiber scaffolds inhibited the proliferation of synthetic smooth-muscle cells. Heparin–rosuvastatin-loaded nanofiber covered stent effectively treated aneurysms without showing any notable in-stent stenosis. In vitro experiments showed that rosuvastatin could inhibit the smooth muscle cell phenotypic modulation of platelet-derived growth factor-BB induction. The inflammatory cytokines secretion and cell viability were inhibited after rosuvastatin treatment.Conclusion: Rosuvastatin could inhibit the abnormal proliferation of synthetic smooth-muscle cells. Heparin–rosuvastatin-loaded covered stents could reduce the incidence of stenosis and late thrombosis and improve the healing rates of stents used for aneurysm treatment.

Abstract:The metallic stents covered with heparin loaded poly(l-lactide-co-caprolactone) nanofibers via emulsion electrospinning have been fabricated as a novel covered stent. The morphology and inner-structure of core–shell nanofibers were respectively ion observed by scanning electron microscopy and transmission electron microscopy. The distribution of heparin aqueous solution and chemical component in nanofibers was separately determined by fluorescence microscopy and Fourier transform infrared spectrum. The results showed that the nanofibrous matrix successfully encapsulated with heparin would not rupture with the expansion of metallic stent, which could effectively separate the aneurysm dome with bloodstream in the rabbit model. The aneurysm was immediately obliterated after the stenting and angiogram at 14 days follow-up showed that the aneurysm was still obliterated. No obvious stenosis and intima hyperplasia in parent artery were found. Therefore, this work provides a promising approach to fabricate covered stent for aneurysm treatment.
Atorvastatin Calcium and Poly(L-lactide-co-caprolactone) Core-Shell Nanofiber Covered Stent Separating Aneurysm and Promoting Endothelialization

Abstract:Aneurysmal subarachnoid hemorrhage caused by intracranial aneurysm is one of the common cerebrovascular diseases can lead to hemorrhagic stroke, brain damage and death. The morphology and inner-structure of the core-shell nanofibers were respectively observed by scanning electron microscopy and transmission electron microscopy. The release of atorvastatin calcium from the nanofiber lasted for more than 10 weeks without serious initial burst release. The nanofiber films could keep complete morphology after degraded for 3 months. The study demonstrated that atorvastatin calcium promoted the synthesis of nitric oxide (NO) in HUVECs and further the proliferation of HUVECs in vitro. Animal studies showed that PLCL-Atv covered stent could separate the aneurysm dome from the blood circulation and obliterate aneurysm. Moreover, the release of Atv could promote the proliferation of HUVECs on nanofiber films and induction of rapid endothelialization.

Abstract:Aneurysmal subarachnoid hemorrhage is a common complication caused by an intracranial aneurysm that can lead to hemorrhagic stroke, brain damage, and death. Knowing this clinical situation, the purpose of this study was to develop a controlled-release stent covered with a core-shell nanofiber mesh, fabricated by emulsion electrospinning, for the treatment of aneurysms. By encapsulating atorvastatin calcium (AtvCa) in the inner of poly (L-lactide-co-caprolactone) (PLCL) nanofibers, the release period of AtvCa was effectively extended. The morphology and inner structure of the core-shell nanofibers were observed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM), respectively. The release of AtvCa from the nanofiber system continued for more than ten weeks without a significant initial burst release. The nanofiber mesh structure degraded gradually but maintained its fiber morphology before neovascularization. The results of this study further elucidated the reendothelialization mechanism of AtvCa by analyzing the nitric oxide (NO) expression from seeded HUVECs. The in vivo studies demonstrated that the PLCL-AtvCa covered stents were capable of separating the aneurysm dome from the blood circulation, leading to the abolishment of the aneurysm. Moreover, the AtvCa controlled release promoted the in vitro proliferation of HUVECs on the nanofiber meshes, and the PLCL-AtvCa covered stents induced in vivo neovascularization. Statement of Significance Intracranial aneurysms are pathological dilatations of blood vessels that have developed an abnormally weak wall structure, thus prone to rupture. Covered stents had been demonstrated to be a method for the treatment of intracranial aneurysm. We prepared a controlled-release stent covered with a core-shell nanofiber mesh, fabricated by emulsion electrospinning, which encapsulated atorvastatin calcium in the inner portion of nanofibers. The results of this study further elucidated the reendothelialization mechanism of AtvCa by analyzing the nitric oxide (NO) expression from seeded HUVECs. The generated AtvCa-load covered stents separated the aneurysm dome from the blood circulation, and keep long-term patency of the parent artery. Abstract:Emulsion electrospinning is a convenient and promising method for incorporating proteins and drugs into nanofiber scaffolds. The aim of this study was to fabricate a nanofiber scaffold for anticoagulation and rapid endothelialization. For this purpose, we encapsulated heparin and vascular endothelial growth factor (VEGF) into the core of poly(L-lactic acid-co-epsilon-caprolactone) (P(LLA-CL)) core-shell nanofibers via emulsion electrospinning. The fiber morphology, core-shell structure and hydrophilicity of the nanofiber mats were analyzed by scanning electron microscopy, transmission electron microscopy and water contact angle. The blood compatibility was measured by hemolysis and anticoagulation testing. A CCK-8 assay was performed to study the promotion of endothelial progenitor cell (EPC) growth and was complemented by immunofluorescent staining and SEM. Our study demonstrates that heparin and VEGF can be incorporated into P(LLA-CL) nanofibers via emulsion. The released heparin performed well as an anticoagulant, and the released VEGF promoted EPC growth on the fiber scaffolds. These results imply that electrospun P(LLA-CL) nanofibers containing heparin and VEGF have great potential in the development of vascular grafts in cases where antithrombogenicity and accelerated endothelialization are desirable. Improved Hydrophilicity of Poly-L-Lactic Acid Blending with Nanoparticulate Amorphous Calcium Phosphate Encourages Its Application in Vascular Stents

Abstract:Hydrophobicity of the biodegradable stent surface, which leads to the formation of an incompatible interface with blood and cells, is a crucial cause of late stent thrombosis. The co-formulation of nanoparticulate amorphous calcium phosphate (ACP) with poly-L-lactic acid (PLLA) was investigated to address this issue. The hydrophilicity was investigated by calculating the static contact angle of ultrapure water, which confirmed that static contact angle of PLLA/ACP was significantly less than that of PLLA. In a hemolysis test, both polymers presented good hemolytic properties. In a platelet adhesion test, fewer activated platelets were found on the than on the PLLA/ACP film compared with PLLA film. Cytocompatibility studies indicated that more cell mulitiplication occurred, forming a confluent ECV304 cell layer, and that the pseudopodia of the cells stretched and were thus more fully developed on PLLA/ACP films compared with those on PLLA films, according to microscopic observations. Furthermore, the effects of the two polymers on the release of proinflammatory cytokines (IL-6 and TNF-alpha) from a human monocyte cell line (THP-1) were evaluated by ELISA. PLLA/ACP elicited a lower release of IL-6 and TNF-alpha from THP-1 than did PLLA. No systemic biochemical toxicity was found in either PLLA or PLLA/ACP in a rabbit iliac artery stenting model. The improved hydrophilicity promised that PLLA/ACP would be useful for fully biodegradable stent applications.

Abstract:Biodegradable polymers used as vascular stent coatings and stent platforms encounter a major challenge: biocompatibility in vivo, which plays an important role in in-stent restenosis (ISR). Co-formulating amorphous calcium phosphate (ACP) into poly(lactic-co-glycolic acid) (PLGA) or poly-L-lactic acid (PLLA) was investigated to address the issue. For stent coating applications, metal stents were coated with polyethylene-co-vinyl acetate/poly-n-butyl methacrylate (PEVA/PBMA), PLGA or PLGA/ACP composites, and implanted into rat aortas for one and three months. Comparing with both PEVA/PBMA and PLGA groups after one month, the results showed that stents coated with PLGA/ACP had significantly reduced restenosis (PLGA/ACP vs. PEVA/PBMA vs. PLGA: 21.24 +/- 2.59% vs. 27.54 +/- 1.19% vs. 32.12 +/- 3.93%, P < 0.05), reduced inflammation (1.25 +/- 0.35 vs. 1.77 +/- 0.38 vs. 2.30 +/- 0.21, P < 0.05) and increased speed of re-endothelialization (1.78 +/- 0.46 vs. 1.17 +/- 0.18 vs. 1.20 +/- 0.18, P < 0.05). After three months, the PLGA/ACP group still displayed lower inflammation score (1.33 +/- 0.33 vs. 2.27 +/- 0.55, P < 0.05) and higher endothelial scores (2.33 +/- 0.33 vs. 1.20 +/- 0.18, P < 0.05) as compared with the PEVA/PBMA group. Moreover, for stent platform applications, PLLA/ACP stent tube significantly reduced the inflammatory cells infiltration in the vessel walls of rabbit iliac arteries relative to their PLLA cohort (NF-kappaB-positive cells: 23.31 +/- 2.33/mm2 vs. 9.34 +/- 1.35/mm2, P < 0.05). No systemic biochemical or pathological evidence of toxicity was found in either PLGA/ACP or PLLA/ACP. The co-formulation of ACP into PLGA and PLLA resulted in improved biocompatibility without systemic toxicity.

Abstract:In-stent restenosis caused by tumor ingrowth increases the risk of secondary surgery for patients with abdominal aortic aneurysms (AAA) because conventional vascular stent grafts suffer from mechanical fatigue, thrombosis, and endothelial hyperplasia. For that, we report a woven vascular stent-graft with robust mechanical properties, biocompatibility, and drug delivery functions to inhibit thrombosis and the growth of AAA. Paclitaxel (PTX)/metformin (MET)-loaded silk fibroin (SF) microspheres were self-assembly synthesized by emulsification-precipitation technology and layer-by-layer coated on the surface of a woven stent via electrostatic bonding. The woven vascular stent-graft before and after coating drug-loaded membranes were characterized and analyzed systematically. The results show that small-sized drug-loaded microspheres increased the specific surface area and promoted the dissolution/release of drugs. The stent-grafts with drug-loaded membranes exhibited a slow drug-release profile more for than 70 h and low water permeability at 158.33 ± 17.56 mL/cm2·min. The combination of PTX and MET inhibited the growth of human umbilical vein endothelial cells. Therefore, it was possible to generate dual-drug-loaded woven vascular stent-grafts to achieve the more effective treatment of AAA.

Abstract:With the increase of vascular diseases in recent years, it is of importance to develop an anti-occlusion stent graft, which can meet the requirements of transplants for a long term. In this paper, we describe a silk fibroin (SF)/heparin-functionalized bifurcated stent graft (BSG) using textile forming technology. The BSGs were prototyped based on seamless weaving technology, and the surface was modified with SF-loaded heparin under steam/air treatment to improve their patency. The physical properties such as thickness, water permeability, contact angle, mechanical properties, and in vitro drug release and coagulation time of the BSGs were examined. The results showed that heparin modification can improve its coagulation time, and the water permeability resistance of the BSGs reached 1.154 ± 0.854 mL/(cm2×min), while their thicknesses were just 0.085 ± 0.004 mm. The heparin release of the BSGs showed that the release time was prolonged upon steam treatment by means of the increase in the β-sheet structure and crystallinity of SF. The viability and attachment of human vascular smooth muscle (HVSM) cells cultured in the release of modified BSGs demonstrated that the modified BSGs could significantly inhibit the proliferation of HVSM cells. The heparin-functionalized BSG with satisfactory thickness, water permeability resistance and anti-occlusion function, which has potential applications in the treatment of vascular diseases.

Abstract:Small-diameter vascular grafts (inner diameter < 6 mm) are useful in treating cardiovascular diseases. The off-the-shelf small-diameter vascular grafts for clinical applications remain a great limitation owing to their thrombogenicity or intimal hyperplasia. Herein, bilayer anticoagulant hydrogel tubes with poly(ε-caprolactone) (PCL) sheaths are prepared by freeze-thawing and electrospinning, which contain nanofibrillated cellulose (NFC)/poly(vinyl alcohol) (PVA)-heparin/poly-L-lysine nanoparticles tube as an inner layer and PCL sheath as an outer layer. The structure, anticoagulant property, and biocompatibility of the inner layer are studied. The effects of thickness of the outer layer on perfusion performance and mechanical property of hydrogel tubes with PCL sheaths (PCL-NFC/PVA-NPs tubes) are investigated. The effect of compliance of PCL-NFC/PVA-NPs tubes on their blood flow is studied by numerical simulation. The tissue compatibility and the patency of PCL-NFC/PVA-NPs tubes are evaluated by implantation in subcutaneous tissue of rats and carotid artery of rabbits. PCL-NFC/PVA-NPs tubes have prominent anticoagulation, sufficient burst pressure and good compliance similar to native arteries. PCL-NFC/PVA-NPs tubes facilitate infiltration of host cells and achieve active proliferation of recruited cells, which will be a promising candidate for small-diameter vascular grafts.

Abstract:Aim: Poly-l-lactic acid (PLLA) based bioabsorbable stents with or without amorphous calcium phosphate (ACP) were implanted and compared the inflammation in coronary arteries. Methods: 6 PLLA and 6 PLLA/ACP based paclitaxel-eluting stents were randomly implanted into the coronary arteries of 12 healthy mini-pigs. The segments with stent were used to evaluate inflammation score and endothelialization score by hematoxylin-eosin staining. Results: At the 28th day after stent implantation, no in-stent restenosis or stent thrombosis was found in both PLLA and PLLA/ACP group. Histological analysis indicated that the inflammation score in PLLA/ACP group was less than that of in PLLA group (2.20 +/- 0.42 vs. 2.80 +/- 0.48, P < 0.05). Consist with that, the expression of NF-kappa B was lower in PLLA/ACP group. The results from immunohistochemistry showed that the expressions of endothelial nitric oxide synthase (eNOS) and CD 31 in PLLA/ACP group were dramatically higher than those in PLLA group respectively. The serum levels of vascular endothelial growth factor (VEGF) and nitric oxide (NO) in PLLA/ACP group were significantly higher than those in PLLA group respectively (509.86 +/- 49.37 pg/ml vs. 322.04 +/- 35.16 pg/ml and 139.46 +/- 7.52 mu mol/L vs. 29.55 +/- 16.55 mu mol/L, P < 0.05). Conclusion: The application of ACP helps to reduce the inflammation caused by PLLA, and is also helpful in endothelial formation and function for PLLA-based bioabsorbable stent.
Heparin-immobilized polymers as non-inflammatory and non-thrombogenic coating materials for arsenic trioxide eluting stents.

Abstract:We have synthesized heparin-immobilized copolymers of l-lactide (LA) and 5-methyl-5-benzyloxycarbonate-1,3-dioxan-2-one (MBC) as non-inflammatory and non-thrombogenic biodegradable coating materials. These copolymers were used in fabricating arsenic trioxide (As2O3)-eluting stents to reduce the late-stage adverse events, such as thrombosis, localized hypersensitivity and inflammation, that occur when applying stents to treat coronary artery diseases. Heparinized copolymers effectively reduced platelet adhesion and protein adsorption while increasing the plasma recalcification time and thromboplastin time in vitro. Histological analysis of the polymer-coated stents in a porcine coronary artery injury model indicated that one heparinized copolymer (Hep-Co90, LA:MBC=90:10), with the highest LA content of 90% and the lowest degradation rate, induced the least foreign body reactions and inflammation, which were as small as those induced by bare metal stents. Consequently, Hep-Co90 was used as the stent coating material for local As2O3 delivery. Histomorphometric evaluations suggested no significant difference between bare metal stents and As2O3-eluting stents at 1 and 3months post-implantation. At 6months, the lumen area in the porcine coronary arteries treated with As2O3-eluting stents is 32.4% higher than those treated with bare metal stents while the neointimal area, neointimal thickness and stenosis rate decreased by 25.8%, 32.5% and 31.2%, respectively. The As2O3-eluting stent using Hep-Co90 as the drug carrier and stent coating material presented in this study represents a novel promising device in preventing in-stent restenosis.

Abstract:Abdominal aortic aneurysm (AAA) is a common vascular disease that has attracted widespread attention due to its characteristic of being asymptomatic and high mortality rate. In this study, we present a novel woven vascular stent-graft, which can improve thrombus formation resulting from excessive smooth muscle cell proliferation following blood vessel stent implantation. To achieve this, we adopted the orthogonal design method to optimize the production process parameters and then coated the surface of the woven stent with chitosan/heparin using a layer-by-layer self-assembly technique based on the electrostatic binding principle, and the performance of the stent before and after modification was comprehensively evaluated. The results demonstrated that the coating had no impact on the mechanical properties, wall thickness, and cross-sectional water permeability of the membranes. Remarkably, the release rate of heparin reached as high as 90.88% at 216 h, effectively inhibiting the growth of smooth muscle cells. In conclusion, the new woven polyester vascular stent-graft provides a potential application prospect for the treatment of AAA.

Abstract:Drug-eluting stents have been widely implanted to prevent neointimal hyperplasia associated with bare metal stents. Conventional polymers and anti-proliferative drugs suffer from stent thrombosis due to the non-selective nature of the drugs and hypersensitivity to polymer degradation products. Alternatively, various herbal anti-proliferative agents are sought, of which biochanin A (an isoflavone phytoestrogen) was known to have anti-proliferative and vasculoprotective action. PLA-PEG diblock copolymer was tagged with heparin, whose degradation releases heparin locally and prevents thrombosis. To get a controlled drug release, biochanin A was loaded in layered double hydroxide nanoparticles (LDH), which are further encapsulated in a heparin-tagged PLA-PEG copolymer. LDH nanoparticles are synthesized by a co-precipitation process; in situ as well as ex situ loading of biochanin A were done. PLA-PEG-heparin copolymer was synthesized by esterification reaction, and the drug-loaded nanoparticles are coated. The formulation was characterized by FTIR, XRD, DSC, DLS, and TEM. In vitro drug release studies, protein adhesion, wettability, hemocompatibility, and degradation studies were performed. The drug release was modeled by mathematical models to further emphasize the mechanism of drug release. The developed drug-eluting stent coating is non-thrombogenic, and it offers close to zero-order release for 40 days, with complete polymer degradation in 14 weeks.

Abstract:In the treatment of coronary artery disease (CAD), the use of stent implantation often leads to clinical complications such as restenosis, delayed endothelial healing, and thrombosis. Here, we develop a double drug sustained-release coating for the stent surface by grafting heparin/NONOate nanoparticles (Hep/NONOates). The Hep/NONOates and surface modification of the stent were characterized by X-ray photoelectron spectroscopy, attenuated total reflection Fourier-transform infrared spectroscopy, static water contact angle, and scanning electron microscopy (SEM), and the release behaviors of the anticoagulant, heparin (Hep) and the bioactive molecule, nitric oxide (NO) were studied. Furthermore, the blood compatibility and cytotoxicity of the modified stent were evaluated by whole blood adhesion and platelet adhesion tests, hemolysis assay, morphological changes of red blood cells, plasma recalcification time assay, in vitro coagulation time tests, and MTT assay. Finally, the results of a rabbit carotid artery stent implantation experiment showed that the double drug sustained-release coating for the stent can accelerate regeneration of endothelial cells and keep good anticoagulant activity. This study can provide new design ideas based on nanotechnology for improving the safety and effectiveness of drug-eluting stents.
Effect of Sustained Heparin Release from PCL/chitosan Hybrid Small-Diameter Vascular Grafts on Anti-Thrombogenic Property and Endothelialization.

Abstract:Thrombus formation and subsequent occlusion are the main reasons for the failure of small-diameter vascular grafts. In this study, a hybrid small-diameter vascular graft was developed from synthetic polymer poly(ε-caprolactone) (PCL) and natural polymer chitosan (CS) by the co-electrospinning technique. Heparin was immobilized on the grafts through ionic bonding between heparin and CS fibers. The immobilization was relatively stable, and heparin could continuously release from the grafts for more than 1month. Heparin functionalization evidently improved the hemocompatibility of the PCL/CS vascular grafts, which was illustrated by the reduced platelet adhesion and prolonged coagulation time (activated partial thromboplastin time, prothrombin time and thromboplastin time) as shown in the human plasma assay, and was further confirmed by the ex vivo arteriovenous shunt experiment. In vitro cell proliferation assay showed that heparin can promote the growth of human umbilical vein endothelial cells, while moderately inhibiting the proliferation of vascular smooth muscle cells, a main factor for neointimal hyperplasia. Implantation in rat abdominal aorta was performed for 1month. Results indicate that sustained release of heparin provided optimal anti-thrombogenic effect by reducing thrombus formation and maintaining the patency. Furthermore, heparin functionalization also enhanced in situ endothelialization, thereby preventing the occurrence of restenosis. In conclusion, it provides a facile and useful technique for the development of heparinized medical devices, including vascular grafts.
Nanofibrous Vascular Scaffold Prepared from Miscible Polymer Blend with Heparin/stromal Cell-Derived Factor-1 Alpha for Enhancing Anticoagulation and Endothelialization

Abstract:There is an urgent clinical demand to develop a functional small diameter vascular graft with long-term patency, which mainly relies on the anticoagulation and endothelialization of the vascular graft. In addition, improved degradation of vascular graft provides more space for cell infiltration and facilitates remodeling of blood vessel. In this study, an elastic and biomimetic nanofibrous vascular scaffold with improved degradability was prepared by adding poly(lactic-co-glycolic acid) (PLGA) into the poly(L-lactic acid) (PLLA)/poly(L-lactide-co-ε-caprolactone) (PLCL) blend using thermally induced phase separation (TIPS) technique. The incorporation of PLGA also improved the hydrophilicity of the composite scaffold. Then the vascular scaffold was surface modified by combination of heparin and stromal cell-derived factor-1 alpha (SDF-1α). The results of whole blood clotting kinetics and plasma recalcification profiles indicated that heparinized modification significantly enhanced the anticoagulation of vascular scaffold. In vitro cell culture assays demonstrated the immobilized SDF-1α facilitated recruitment of endothelial progenitor cells (EPCs), migration and proliferation of mature endothelial cells, human umbilical vein endothelial cells (HUVECs), and expression of VE-cadherin/CD144 and endothelial nitric oxide synthase (eNOS) genes in HUVECs, thereby accelerating the endothelialization of the modified vascular scaffold. Besides, the nanofibrous vascular scaffold modified with heparin and SDF-1α inhibited the proliferation of human vascular smooth muscle cells (HVSMCs). Therefore, the phase separated nanofibrous vascular scaffold modified with heparin and SDF-1α shows the promising in vitro performance as a functional small diameter vascular graft.

Abstract:Although poly(l-lactic acid) (PLLA) is the widely used material for bioresorbable stents, publications on PLLA braided stents are still lacking. In this paper, the PLLA monofilaments were prepared via melt spinning and solid-state drawing. The total draw ratios are from 2.8 to 30.8. The properties of the monofilaments, such as crystallinity, elastic modulus, elongation at break, were characterized. Due to the relatively good mechanical properties, PLLA monofilaments with draw ratio of 16.8 were used to braid carotid stents. The curve of the radial force of the braided PLLA stent almost overlaps with that of a Carotid Wallstent with similar parameters, which indicates that the braided PLLA stent can provide adequate support to the carotid lesion.
