Abstract:OBJECTIVE To develop a sensitive and specific reverse-phase high-performance liquid chromatography(RP-HPLC) method to determine propofol in follicular fluid.METHODS Thymol was added into follicular fluid samples as the internal standard,and the samples were then precipitated with methanol.After centrifugation at 20 000 r·min-1 for 15 min,20 μL of supernatant was directly injected into an Agilent Extend-C18 column(4.6 mm×250 mm,5 μm).The mobile phase was composed of methanol-water(77∶23).Analysis was run at a flow rate of 1.0 mL ·min-1.Fluorescence detection was applied with excitation and emission wavelengths of 276 and 310 nm,respectively.RESULTS The calibration curves had good linearity in the range of 0.108-5.4 mg·L-1.The linear regression equation was A=0.207 4ρ-0.001 2(r=0.999 3).The limits of detection and quantity proved to be 54 and 108 μg·L-1,respectively.The method recoveries for low,intermediate and high concentrations were 117.9%,101.7% and 99.4%,respectively.The RSDs of inter-day assay were 4.10%,1.28% and 0.20%(n=5),and intra-day assay were 4.34%,2.33% and 0.61%(n=5),respectively.CONCLUSION The method is accurate and convenient.It can be used to determine propofol for studying its clinical pharmacokinetics.
Simultaneous Determination of Phthalates and Parabens in Cosmetic Products by Gas Chromatography/Mass Spectrometry Coupled with Solid Phase Extraction

Abstract:Studies have been carried out on the simultaneous determination of 8 phthalates, i. e. di-ethyl phthalate (DEP) , di-propyl phthalate (DPP) , di-isobutyl phthalate (DIBP) , dibutyl phthalate (DBP) , benzyl butyl phthalate ( BBP) , di-cyclohexyl phthalate (DCHP) , di-(2-ethylhexyl) phthalate (DEHP), di-octyl phthalate (DOP) and 4 parabens, i. e. methylparaben (MPB), ethylparaben (EPB), propyl paraben (PPB), and butyl paraben (BPB) by gas chromatography in combination with mass spectrometry (GC/MS) in electron ionisation mode (EI) with selected-ion monitoring (SIM) acquisition method. The phthalates and parabens in 15 cosmetic products, including hair sprays, perfumes, deodorants, cream, lotion, etc. were determined. The determination of the samples were performed after sonication-assisted extraction with methanol, cleaned up with an LC-C18 column (3 mL) and analyzed by GC/MS method. The base peak (m/z 149) of the phthalates and the base peak (m/z 121) of the parabens were selected for the screening studies. The characteristic ions, m/z 121, 149, 177, 222 for DEP; m/z 149, 191, 209 for DPP; m/z 57, 149, 223 for DIBP; m/z 104, 149 for DBP; m/z 91, 132, 149, 206 for BBP; m/z 55, 149, 167 for DCHP; m/z 113, 149, 167, 279 for DEHP; m/z 149, 279 for DOP; m/z 65, 93, 121, 152 for MPB; m/z 93, 121, 138, 166 for EPB; m/z 93, 121, 138, 180 for PPB; and m/z 93, 121, 138, 194 for BPB were chosen for quantitative studies. These techniques are capable to detect phthalates and parabens at the level of 0. 1 -5. 0 microg/kg. Overall recoveries were 80% - 100% with relative standard deviations (RSDs) less than 10%. Only one of the 15 examined samples was free from phthalates and parabens. The rest 14 samples were found to contain at least 3 or more of these phthalates and/or parabens. The predominant phthalates detected in the studied samples were MPB, PPB, DPP, DCHP and DEHP. The residue levels were at 1. 42 -4 278 mg/kg.

Abstract:Objective To determination of propofol in biological samples by GC/MS.Methods The propofol in biological samples was extracted with cyclohexane and detected by GC/MS.The value of m/z of 163,178 for propofol and 150,135 for thymol(internal standard) were adopted in the MS determination.The method specificity,linear range,quantitative line and the detection limit,recovery and precision were examined.Results The retention time of propofol which was well separated from the internal standard thymol was 8.17min.The good linear range of propofol had been obtained(r>0.992) in blood,kidney and liver samples,and the following detection limit was 0.05μg/mL,0.1μg/g,0.05μg/g.The method recoveries were between 90% and 110% and the intraday and interday relative standard deviations were less than 7%.Conclusion This method is simple,sensitive and rapid.Its accuracy,precision and recovery can meet the determination of propofol concentration in biological samples,and it is suitable for analysis of propofol in biological samples.
Fast Determination of Propofol in Human Plasma Using C18-Functionalized Magnetic Nanomaterials Followed by Supercritical Fluid Chromatography

Abstract:Novel C18-functionalized magnetic nanomaterials; i.e., C18@poly-styrene-divinylbenzene-glycidyl methacrylate-Fe3O4 (C18@PS-DVB-GMA-Fe3O4) have been synthesized by using N, N-dimethyloctadecylamine as modifying agent, which could be beneficial to remove the blood phospholipids. The C18@PS-DVB-GMA-Fe3O4 nanoparticles have been used and evaluated in the Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) procedure for human plasma prior to the analysis of propofol by supercritical fluid chromatography (SFC). In the QuEChERS procedure, human plasma samples are directly mixed with extraction solvent and C18@PS-DVB-GMA-Fe3O4 nanoparticles, and the extraction and cleanup procedures have been accomplished simultaneously. The SFC separation was performed with a C18 column (Thermo Scientific™ Acclaim™ 120, 250 × 4. 6 mm, 5 μm) within 5 min, using thymol as the internal standard. Supercritical carbon dioxide was used as the mobile phase with methanol as the cosolvent at the flow rate of 1.0 mL/min. The column temperature was 38°C, and detection wavelength was 275 nm. A good linearity was obtained among the propofol concentration range of 0.5-10 mg/L (R2 = 0.9997) with the limit of detection of 0.17 mg/L. Recoveries were in the range of 76.5-91.9%, with RSD less than 7.9%. These results suggested that method is convenient, rapid with high accuracy and little matrix effect, and suitable for rapid determination of propofol plasma concentration.
Physicochemical Characterization and Pharmacological Evaluation of Novel Propofol Micelles with Low-Lipid and Low-Free Propofol

Abstract:We developed safe and stable mixed polymeric micelles with low lipids and free propofol for intravenous administration, to overcome the biological barrier of the reticuloendothelial system (RES), reduce pain upon injection, and complications of marketed propofol formulation. The propofol-mixed micelles were composed of distearoyl-phosphatidylethanolamine-methoxy-poly (ethylene glycol 2000) (DSPE mPEG2k) and Solutol HS 15 and were optimized using Box Behnken design (BBD). The optimized formulation was evaluated for globule size, zeta potential, loading content, encapsulation efficiency, pain on injection, histological evaluation, hemolysis test, in vivo anesthetic action, and pharmacokinetics, in comparison to the commercialized emulsion Diprivan. The optimized micelle formulation displayed homogenous particle sizes, and the free drug concentration in the micelles was 60.9% lower than that of Diprivan. The paw-lick study demonstrated that propofol-mixed micelles significantly reduced pain symptoms. The anesthetic action of the mixed micelles were similar with the Diprivan. Therefore, we conclude that the novel propofol-mixed micelle reduces injection-site pain and the risk of hyperlipidemia due to the low content of free propofol and low-lipid constituent. It may be a more promising clinical alternative for anesthetic.

Abstract:The aim of the present study was to develop an oil-free o/w microemulsion, composed of pluronic F68, propylene glycol and saline, which solubilized poorly soluble anesthetic drug propofol for intravenous administration. The ternary diagram was constructed to identify the regions of microemulsions, and the optimal composition of microemulsion was determined by in vitro evaluation such as globule size upon dilution and rheology. The droplet size of the diluent emulsion corresponding to oil-in-water type ranged from 200 to 300nm in diameter. Stability analysis of the microemulsions indicated that they were stable upon storage for at least 6 months. Hemolysis percent of propofol microemulsions was lower than that of commercial lipid emulsion (CLE) at 4h. Acute toxicity test showed that median lethal dose of propofol microemulsion was the same as that of CLE. No significant difference in time for unconsciousness and recovery of righting reflex was observed between the prepared microemulsions and CLE. In conclusion, microemulsion would be a promising intravenous delivery system for propofol.

Abstract:To develop a new high-performance liquid chromatographic (HLPC) method for determination of propofol in human serum.Human serum samples were precipitated with 20% perchloric acid and centrifuged to obtain 50 microl of the supernatant for analysis by HPLC coupled with fluorescence detection. The analysis was performed with a C(18) reversed-phase column using a acetonitrile-water (90:10) phase delivered at 1.0 ml/min, with the excitation wavelength of 276 nm and emission wavelength of 310 nm.The calibration curves were linear (r=0.997 5) within the concentration range of 0.05-10 microg/ml, the limit of propofol quantification was 50 ng/ml and the intra- and inter-day precisions were between -/+15%.The method is accurate, sensitive and simple for propofol determination in clinical anesthesia.

Abstract:Rapid and quantitative determination of blood propofol concentration is important for anesthesiologists to accurately control intraoperative propofol dose, timely monitor physiological statuses of patients and greatly improve the safety of surgery. Herein, a dopant-assisted negative photoionization ion mobility spectrometer with the optimized ionization region structure and the three-way inlet design was developed, increasing the generation ratio of the reactant ions O2-, and improving the ionization efficiency of propofol molecules. Besides, the addition of methanol-anisole solution during injection promoted the neutral desorption of propofol in blood, further improving the detection sensitivity by an order of magnitude, eliminating any sample pretreatment and effectively reducing the single analysis time to less than 1 min compared to the previous article. The dual calibration quantitative method, i.e. the method of calibrating the O2- concentration and the sample concentration changes during the entire process of detecting propofol through the integral value of M·O2- and the maximum signal intensity of O2-, successfully achieved accurate quantification of blood propofol. And the linear calibration curve of propofol was obtained with the range of 0.1 to 15 ng μL-1 and with the limit of detection of 0.03 ng μL-1, which was fulfilled to conduct propofol determination throughout the perioperative period. Finally, this method was applied to clinically measure the blood propofol concentration in patients newly regained consciousness with concentrations ranging from 0.2 ng μL-1 to 3 ng μL-1, and it turned out that the older patient had the lower propofol concentration in blood.

Abstract:Objective To determine the vinpocetine content and its related substances in lyophilized submicroemulsion of vinpocetine for quality control.Methods Separation was carried out on a C18 column(4.6 mm×250 mm,5 μm),using a mobile phase of methanol-0.5% triethylamine(84∶16,pH8)at a flow rate of 1.0 ml/min.The ultraviolet detection wavelength was set at 273 nm.Results Under the established chromatographic conditions,the related substances and vinpocetine were separated completely.The method was validated and stability under various conditions was evaluated.The standard curve was linear within the range of 5-50 μg/ml with a correlation coefficient of 0.9998.The average recovery was 100.79%(RSD=0.67%,n=9)in lyophilized submicroemulsion.Conclusion This method is simple,specific and can be used for quality control of vinpocetine.
Preparation and evaluation of novel mixed micelles as nanocarriers for intravenous delivery of propofol

Abstract:Novel mixed polymeric micelles formed from biocompatible polymers, poly(ethylene glycol)-poly(lactide) (mPEG-PLA) and polyoxyethylene-660-12-hydroxy stearate (Solutol HS15), were fabricated and used as a nanocarrier for solubilizing poorly soluble anesthetic drug propofol. The solubilization of propofol by the mixed micelles was more efficient than those made of mPEG-PLA alone. Micelles with the optimized composition of mPEG-PLA/Solutol HS15/propofol = 10/1/5 by weight had particle size of about 101 nm with narrow distribution (polydispersity index of about 0.12). Stability analysis of the mixed micelles in bovine serum albumin (BSA) solution indicated that the diblock copolymer mPEG efficiently protected the BSA adsorption on the mixed micelles because the hydrophobic groups of the copolymer were efficiently screened by mPEG, and propofol-loaded mixed micelles were stable upon storage for at least 6 months. The content of free propofol in the aqueous phase for mixed micelles was lower by 74% than that for the commercial lipid emulsion. No significant differences in times to unconsciousness and recovery of righting reflex were observed between mixed micelles and commercial lipid formulation. The pharmacological effect may serve as pharmaceutical nanocarriers with improved solubilization capacity for poorly soluble drugs.
Efficacy of High Performance Liquid Chromatography for Simultaneous Determination of Propofol and Remifentanil Concentrations in Human Plasma

Abstract:Objective To evaluate the efficacy of high performance liquid chromatography (HPLC) for simultaneous determination of propofol and remifentanil concentrations in human plasma.Methods Methods Eighteen healthy volunteers of both sexes,aged 18-45 yr,weighing 52-81 kg,were enrolled in the study.Venous blood samples were collected,and the concentrations of propofol and remifentanil in human plasma were detected simultaneously by HPLC.The internal standard was thymol.Potassium dihydrogen phosphate 0.1 mol/L was added to the plasma and then the plasma samples were extracted with extract liquor (ethyl acetate ∶ hexane =4 ∶ 1,V/V).The analytical column was ZORBAX Eclipse XDB-C18 (4.6 mm×250 mm,5 μm).The mobile phase was methano ∶ 0.02 mol/L NaH2PO4 ∶ acetonitrile,the flow rate was 1.0 ml/min,the detection wavelength was 210 nm within 1-7 min,and 266 nm within 7-16 min,and the sample size was 20 μl.Linear regression analysis was performed by using the least-squares method.The specimens of the blood with the final concentration of remifentanil 1.00,5.00 and 20.00 ng/ml and propofol 0.50,2.00 and 10.00 μg/ml were obtained to determine the recovery,precision and stability.Results Linear regression equation of remifentanil was C=12.853 5Ai/As+0.084 8 (R2 =0.999 4),and this system showed a good linear relationship with the concentration of remifentanil ranged 0.5-40.0 ng/ml.Linear regression equation of propofol was C=8.554 3 Ai/As+0.029 1 (R2=0.998 6),and this system showed a good linear relationship with the concentration of propofol ranged 0.2-20.0 μg/ml.For both propofol and remifentanil concentrations,the relative recovery was within the range of 85％-115％,the absolute recovery was larger than 75％,and the relative standard deviation of intra-and inter-day precision and stability was less than 5％.The method was proved to meet the requirements of biological sample analysis.Conclusion For HPLC method established in this trial,the determination is sensitive,reproducible,rapid and simple,and it can be used for simultaneous determination of propofol and remifentanil concentrations in human plasma and for clinical pharmacokinetic research.
GC determination of propylene glycol in rifamycin sodium injection

Abstract:Objective: To establish a GC method for the determination of propylene glycol in rifamycin sodium injection. Methods: The GC method was conducted on a DB- FFAP capillary column. The injector and detector were operated at 230 ℃ and 250 ℃,respectively. The temperature program consisted of an initial temperature of 80℃ held for 3 min,rose to 230 ℃ at a rate of 10 ℃ ·min- 1and held for 3 min. The injection volume was 1 μL.Results: The calibration curve was linear in the range of 0. 02%- 0. 15%( v /v) and the correlation coefficient was 0. 999. The average recoveries of two kinds of samples from two different manufacturers were 98. 8%- 99. 7%and 97. 4%- 104. 3%( n = 3),respectively. Conclusion: The established method is accurate,simple and reproducible. It is suitable to determinate propylene glycol in rifamycin sodium injection and can be effectively used for quality control of the drug.
RP-HPLC Method for Determination of Propofol in Human Serum

Abstract:OBJECTIVE: To develop a RP HPLC method for determining serum concentration of propofol. METHODS: The propofol was separated on a Diamonsil C_(18) column, the mobile phase was a mixture of acetonitrile and water (70:30, containing 0.10% trifluoroacetic acid). The excitation and emission wavelength were 276nm and 310nm, respectively. Sample preparation was simply a deproteinsation process with saturated ammonium sulphate. The propofol was extracted by the acetonitrile, the solvent of internal standard (thymol). RESULTS: A good linear relationship was obtained between 0.25 - 10 mg · L~(-1) (γ=0.9996). The detection limit was 25 Mg·L~(-1) with an injection volume of 20 μ L. The RSD of within-day and between-day were less than 2.5%, the average recovery was 91.2% - 100.9%. CONCLUSION: The proposed method is sensitive, reliable and can be used for pharmacokinetic studies of propofol and monitoring propofol in serum.
Simultaneous Quantitative Analysis of Six Proton-Pump Inhibitors with a Single Marker and Evaluation of Stability of Investigated Drugs in Polypropylene Syringes for Continuous Infusion Use.

Abstract:OBJECTIVE:We developed and validated a simple, convenient and reproducible method for simultaneous estimation of six proton-pump inhibitors (PPIs), omeprazole (OPZ), esomeprazole (EOPZ), lansoprazole (LPZ), pantoprazole (PPZ), rabeprazole (RPZ) and ilaprazole (IPZ) in pharmaceutical dosage forms by a single marker. Meanwhile, the stability of the cited PPIs in 0.9% sodium chloride injection stored in polypropylene syringes up to 48 hours for continuous infusion use was investigated.MATERIALS AND METHODS:The chromatographic separation was achieved on an InterSustain® C18 column (150 × 4.6 mm, 5 μm). The isocratic mobile phase made up of 0.05 M potassium dihydrogen phosphate buffer (pH 4.0): acetonitrile (65:35, v/v) was pumped through the column at a temperature maintained at 30°C and a flow rate of 1.0 mL/min. The relative retention time, UV spectral similarity and relative correction factors between OPZ and the other five PPIs were calculated and investigated using the quantitative analysis of multi-components with a single marker (QAMS) method. The stability study examined physical parameters, pH values and drug concentrations of the PPIs mixtures.RESULTS:Under these conditions, all cited PPIs were separated simultaneously at a retention time of 6.0, 7.3, 7.3, 9.9, 12.5 and 13.9 min for RPZ, OPZ, EOPZ, IPZ, PPZ and LPZ, respectively, with a total run time less than 20.0 min. Comparative analysis results indicated that there were no significant differences observed between the QAMS method and the external standard method. The percentage of initial concentration of each PPI gradually decreased during the storage time.CONCLUSION:The proposed method, which is selective, economical and accurate, was applied successfully for determination of the cited PPIs in their respective pharmaceutical dosage forms. Admixtures of OPZ, EOPZ, PPZ, IPZ in 0.9% sodium chloride injection were stable for 24 hours and LPZ, RPZ in 0.9% sodium chloride injection were stable for 8 hours in polypropylene syringes.
Preparation and Property of Mpeg-Pla/pluronic Mixed Micelles and Their Role in Solubilization of Propofol

Abstract:Novel mixed polymeric micelles formed by biocompatible polymers,mPEG-PLA and Pluronic P105,were fabricated and used as a nanocarrier to solubilize the poorly solublc anesthetic drug propofol.Propofol was added directly to an aqueous solution of mPEG-PLA/Pluronic P105 mixed micelles and stirred into a miccllar solution.The average particle size and size distribution of micelles were evaluated by the dynamic light scattcring technology.Drug loading content,encapsulation cfficiency and free drug concentration were determined by using ultracentrifugation and lyophilization.In vitro release characteristic of propofol formulation was investigated by dialysis method.The physical stability of mixed micelles was also assessed under storagc condition (4 ℃) after six months.Sleep-recovery studies in male Sprague-Dawley rats,at a dose of 10 mg/kg were performed to compare the pharmacodynamic profiles of propofol in mixed micelles with that of commercial lipid emulsion (CLE).The results indicated that solubilization of propofol in the mixed micelles was more efficient than that in mPEG-PLA alone.Micelles with the optimized composition of mPEG-PLA/Pluronic P105/Propofol (10∶4∶5,w/w/w) had particle size of about 90 nm with narrow distribution (polydispersity index of aboul 0.2).The content of free propofol in the aqueous phase of mixed micelles was significantly lower than that in CLE (P＜0.05).There was no remarkable differences for particle size,polydispersity index,and free drug concentration when the mix micelles were stored at 4 ℃ for six months,suggesting that the propofol-loaded mixed micelles were stable for at least six months.The accumulative release of mixed micelles was significantly higher than that of CLE at the corresponding time points,suggesting that quick release rate for mixed micelles might produce favorable pharmacological effect.No significant differences in the unconsciousness time and recovery time of righting reflex were observed between the mixed micelles and CLE (P＞0.05).In conclusion,the mixed micelle of mPEG-PLA and pluronic copolymer may be a promising candidate for intravenous delivery of propofol in clinic.
Analysis of gemcitabine liposome injection by HPLC with evaporative light scattering detection

Abstract:Gemcitabine liposome injection (i.e., stealth liposomes) has facilitated the targeting of gemcitabine for cancer treatment. We systemically reviewed liposome-based drug-delivery systems, which can improve pharmacokinetics, reduce side effects, and potentially increase tumor uptake, for pancreatic cancer therapy. A novel liposomal formulation, which allows for higher tumor-targeting efficiencies and can be used in current clinical trials to treat this challenging disease, has gained great popularity and attention. In this work, a simple, rapid high-performance liquid chromatography (HPLC) method was developed for the simultaneous determination of N-(carbonyl-methoxypolyethylene glycol 2000)-1, 2-distearoyl-sn-glycero-3-phosphoethanolamine sodium salt and neutral colipids cholesterol and hydrogenated soy phosphatidylcholine or distearoyl phosphatidylcholine. Because of the poor ultraviolet absorbance of the lipids, evaporative light-scattering detection (ELSD) was used to monitor the separation. The separation was carried out on a YMC-Pack column (YMC Co., Ltd., Kyoto, Japan). Lipids were eluted using binary linear gradients starting from a mixture of 80% A and 20% B to 100% B in 10 minutes, followed by a 6-minute plateau at 100% B, where A is chloroform/isopropyl alcohol/diethylamine/trifluoroacetic acid (TFA) (50:50:0.01:0.0025) and B is chloroform/isopropyl alcohol/H₂O/diethylamine/TFA (41:50:9:0.01:0.0025). The mobile phase composition was then changed back to initial solvent mixture in 1 minute, and the column was equilibrated for 13 minutes before every subsequent run. Then, 0.025% (v/v) TFA was added into the mobile phase to enhance the retaining of the stealth lipids. This newly developed method enabled direct analysis of liposomes without solvent lipid extraction and was validated to be linear, precise, accurate, specific, and sensitive. The method has been successfully employed in a wide range of lipid-based formulation screening, process development, and stability testing. Further, we describe the simple, rapid HPLC-ELSD method for the simultaneous determination of all the lipids and active pharmaceutical ingredients in various liposome-based drug formulations. The method can quantitate all the lipids of active targeting liposomes, which bond with folic acid.

Abstract:Hyperlipidemia is a disease characterized by abnormal blood lipid levels and is the leading risk factor for cardiovascular disease. 3′,4′-Dimethoxy flavonol-3-O-glucoside (abbreviated DF3G) is a new lipid-lowering drug created as a flavonoid structural analog. The principal metabolite of DF3G in human plasma is the aglycone glucuronide conjugate M2. The purpose of this study is to use liquid chromatography-tandem mass spectrometry to develop and validate a quantitative analysis method for DF3G and its metabolite M2 in human plasma, and to use the method to investigate the pharmacokinetics of DF3G and M2 in a clinical trial. This method employed DF3G-d 6 as the internal standard, and plasma samples were processed by protein precipitation. Isocratic separation could accurately differentiate DF3G, M2, and DF3G-d 6 from endogenous components in the matrix or other components in the samples, and endogenous components in the matrix had little impact on ionization efficiency. Positive electrospray ionization with multiple reaction monitoring (MRM) transitions of m/z 461.2 → 299.0 for DF3G, m/z 475.1 → 299.1 for M2 and m/z 467.1 → 305.1 for DF3G-d 6 was used for quantification. The DF3G and M2 linear range for plasma were in the range of 4.00/4.00 ng/mL to 4000/4000 ng/mL. Both the analytes and the internal standard were stable regardless of whether they were in solution or plasma samples. The accuracy of the average concentration of the quality control samples was within 15% of the theoretical value, and the RSD was less than 15%. The method is rapid, accurate, straightforward, and precise. It is appropriate for the determination of DF3G and M2 concentrations in human plasma and has been successfully applied to determine the pharmacokinetic analysis in phase I clinical trials.

Abstract:A method was established for the simultaneous quantification of nine components of five different structural types in Qingkailing injection. High performance liquid chromatography coupled with a photo diode array detector and an evaporative light scattering detector (HPLC-DAD-ELSD) was employed in the determination. Four monitoring wavelengths of 240, 254, 280 and 330 nm were set to determine nucleosides (uridine and adenosine), iridoid glucoside (geniposide), flavone glycoside (baicalin) and organic acids (chlorogenic acid and caffeic acid) respectively, and a combined evaporative light scattering detector was used to detect three steroid compounds (cholic acid, ursodesoxycholic acid and hyodeoxycholic acid). The proposed method permitted the simultaneous separation and determination of five groups of compounds in Qingkailing injection, and acceptable validation results of the precision, repeatability, stability and accuracy tests were achieved. The method was applied to the analysis of 19 Qingkailing injection samples from three different plants, and the results indicated that the method could be used as a convenient and reliable method in the multi-component determination and quality control of traditional Chinese medicines.
Simultaneous Determination of 20(S)-Protopanaxadiol and Its Three Metabolites in Rat Plasma by LC-MS/MS: Application to Their Pharmacokinetic Studies.

Abstract:The aim of this study was to develop an LC-MS/MS method for simultaneous determination of 20(S) protopanaxadiol (PPD) and its three metabolites, PPD-glucuronide (M1), (20S,24S)-epoxy-dammarane-3,12,25-triol (M2) and (20S,24R)-epoxydammarane-3,12,25-triol (M3), in rat plasma. Precipitation with acetonitrile was employed for sample preparation and chromatographic separations were achieved on a C-18 column. The sample was detected using triple quadrupole tandem mass spectrometer with selected reaction monitoring mode. The monitored precursor-to-product ion transitions were m/z 459.4375.3 for PPD, m/z 635.4113.0 for M1, m/z 477.4441.4 for M2 and M3 and m/z 475.4391.3 for IS. The developed assay was validated according to the guidelines of the US Food and Drug Administration. The calibration curves showed good linearity over the tested concentration ranges (r>0.9993), with the LLOQ being 1ng/mL for all analytes. The intra- and inter-day precisions (RSD) were<9.51% while the accuracy (RE) ranged from -8.91 to 12.84%. The extraction recovery was >80% and no obvious matrix effect was detected. The analytes were stable in rat plasma with the RE ranging from -12.34 to 9.77%. The validated assay has been successfully applied to the pharmacokinetic study of PPD as well as its metabolites in rat plasma. According to the pharmacokinetic parameters, the in vivo exposures of M1, M2 and M3 were 11.91, 47.95 and 22.62% of that of PPD, respectively.

Abstract:A novel simple, specific, sensitive, accurate and precise reversed phase high performance liquid chromatographic method (RP-HPLC/UV) was developed and validated for the simultaneous estimation of Glycopyrronium bromide (GLY), Indacaterol acetate (IND) and Mometasone furoate (MOF) in pure form, in laboratory prepared mixtures and in pharmaceutical dosage form. Experimental design methodology was applied by using Plackett-Burman and face-centered composite designs to achieve the best resolution with minimum experimental trials. The designed model was statistically analyzed, graphically presented by surface plots and the relationships between coefficients of the derived polynomial equations were interpreted. Chromatographic separation was achieved on Inertsil ODS C18 column (250 ×4.6 mm, 5 µm) at ambient temperature using a mobile phase composed of methanol: 0.1% glacial acetic acid (pH4) in a gradient elution at a flow rate 1 mL /min. UV detection was carried out at 233 nm. Response was found to be linear in the concentration range of 20-120 µg /mL with regression coefficient (r2 = 0.999) for GLY, 50-300 µg /mL with regression coefficient (r2 = 0.9995) for IND and 50-300 µg /mL with regression coefficient (r2 = 0.9998) for MOF. The method was validated as per ICH guidelines and satisfactory results were achieved. The method was successfully applied for the analysis of the cited drugs in their fixed dose combination (FDC) pharmaceutical formulation. Statistical comparison between the results obtained by the proposed method and the reference methods for GLY, IND and MOF showed no significant difference. The developed method could be implemented in quality control aspects of the cited drugs. Four green metrics were used to evaluate the new RP-HPLC/UV method's greenness and compare it to other published techniques.
