Abstract:A fundamental understanding of the recent experiments of magnetic separation of amino acids using gold/iron oxide composite nanoparticles is provided for the first time by theoretically studying the interaction of gold-coated iron oxide cluster with sulfur-containing amino acids: cysteine, methionine, and taurine. We find, in agreement with experiments, that the interaction of cysteine is stronger than methionine, the interaction energies being 0.738 and 0.712 eV respectively. These energies are intermediate between van der Waals and covalent bonding and are ideal for effective binding and release in magnetic separation. On the other hand, we find that taurine, which is also a sulfur-containing amino acid, cannot bind to gold-coated iron oxide cluster. This selectivity can be used for the separation of taurine from the other two sulfur-containing amino acids. The present study provides insight on the mechanisms responsible for functionalizing iron oxide nanoparticles and may have far-reaching implications in biomedical research.

Abstract:Nowadays, proteins separation has attracted great attention in proteomics research. Because the proteins separation is helpful for making an early diagnosis of many diseases. Magnetic nanoparticles are an interesting and useful functional material, and have attracted extensive research interest during the past decades. Because of the excellent properties such as easy surface functionalization, tunable biocompatibility, high saturation magnetization etc, magnetic microspheres have been widely used in isolation of proteins/peptides. Notably, with the rapid development of surface decoration strategies, more and more functional magnetic adsorbents have been designed and fabricated to meet the growing demands of biological separation. In this review, we have collected recent information about magnetic adsorbents applications in selective separation of proteins/peptides. Furthermore, we present a comprehensive prospects and challenges in the field of protein separation relying on magnetic nanoparticles.

Abstract:In this work, magnetic graphene double-sided mesoporous nanocomposites (mag-graphene@mSiO₂) were synthesized by coating a layer of mesoporous silica materials on each side of magnetic grapheme. The surfactant (CTAB) mediated sol-gel coating was performed using tetraethyl orthosilicate as the silica source. The as-made magnetic graphene double-sided mesoporous silica composites were treated with high-temperature calcination to remove the hydroxyl on the surface. The novel double-sided materials possess high surface area (167.8 cm²/g) and large pore volume (0.2 cm³/g). The highly open pore structure presents uniform pore size (3.2 nm) and structural stability. The hydrophobic interior pore walls could ensure an efficient adsorption of target molecules through hydrophobic-hydrophobic interaction. At the same time, the magnetic Fe₃O₄ particles on both sides of the materials could simplify the process of enrichment, which plays an important role in the treatment of complex biological samples. The magnetic graphene double-sided nanocomposites were successfully applied to size-selective and specific enrichment of peptides in standard peptide mixtures, protein digest solutions, and human urine samples. Finally, the novel material was applied to selective enrichment of endogenous peptides in mouse brain tissue. The enriched endogenous peptides were then analyzed by LC-MS/MS, and 409 endogenous peptides were detected and identified. The results demonstrate that the as-made mag-graphene@mSiO₂ have powerful potential for peptidome research.
One-step synthesis of magnetic graphene oxide nanocomposite and its application in magnetic solid phase extraction of heavy metal ions from biological samples

Abstract:A novel magnetic graphene oxide nanocomposite was synthesized by one-step coprecipitation method and characterized by transmission electron microscopy, X-ray photoelectron spectroscopy and vibrating sample magnetometer. The nanocomposite beard many intriguing properties, including chemical stability, high adsorption capacity, and superparamagnetic. These properties evoked great interest and desire of its exploration in magnetic solid-phase extraction of heavy metal ions from complex samples. Several parameters effecting the analytical performance, such as the sample pH, amounts of adsorbent, sample volumes, elution volumes, and coexisting ions, had been investigated in detail. The adsorbed metal ions were easy eluted by controlling the pH condition and the materials could be reused more than 20 times. Under the optimized conditions, the limits of detection were 0.016, 0.046, 0.395, 0.038, 0.157 μg L(-1) for Co(2+), Ni(2+), Cu(2+), Cd(2+), and Pb(2+), respectively. The intra-day relative standard deviations (n=5) were in the range of 1.8-5.5% at 10 μg L(-1). The proposed method was successfully applied to biological sample analysis and got excellent recoveries in the range of 81-113% even the matrix was complex.

Abstract:In this article, we report the synthesis of bifunctional Au-Fe(3)O(4) nanoparticles that are formed by chemical bond linkage. Due to the introduction of Au nanoparticles, the resulting bifunctional Au-Fe(3)O(4) nanoparticles can be easily modified with other functional molecules to realize various nanobiotechnological separations and detections. Here, as an example, we demonstrate that as-prepared Au-Fe(3)O(4) nanoparticles can be modified with nitrilotriacetic acid molecules through Au-S interaction and used to separate proteins simply with the assistance of a magnet. Bradford protein assay and sodium dodecyl sulfate-polyacrylamide gel electrophoresis were performed to examine the validity of the separation procedure, and the phosphate determination method suggested that the as-separated protein maintained catalytic activity. This result shows the efficiency of such a material in protein separation and suggests that its use can be extended to magnetic separation of other biosubstances. Moreover, this synthetic strategy paves the way for facile preparation of diverse bifunctional and even multifunctional nanomaterials.

Abstract:A facile approach to prepare water-dispersible super-paramagnetic iron oxide nanoparticles (MNPs) is reported. The homogeneous MNPs prepared in organic phase are functionalized with an aqueous solution of dopamine at room temperature. The obtained dopamine modified MNPs (MNP-DA) exhibiting excellent super-paramagnetic property can bind various molecules, including NTA and proteins under mild conditions. To demonstrate the feasibility for biological application, the MNP-DA was used for rapid and efficient magnetic separation of bovine albumin (BSA) carrying fluorescent Au nanoclusters from fluorescein. Furthermore, the MNP-DA was functionalized with Na, Na-bis (carboxy methyl)-L-lysine hydrate (NTA) to reversibly immobilize and release histidine-tagged proteins. The present study demonstrates a general and facile route to the efficient modification of magnetic nanoparticles with various functional materials and paves the way for a wide range of biomedical applications of MNPs.

Abstract:The graphene oxide (GO)–Fe3O4 and the reduced graphene oxide (rGO)–Fe3O4 composites with good water dispersibility, high affinity and rapid magnetic response have been prepared via a facile grafting method. They can be used for the effective immobilization of proteins and the enrichment of peptides, respectively. By taking advantage of the high loading capacity and the abundant hydrophilic groups of the GO–Fe3O4 composites, they can be applied to immobilize proteins. The amount of loading of protein (BSA) on GO–Fe3O4 is as high as 294.54 mg g−1. The rGO–Fe3O4 composites can be used to enrich the low-concentration peptides conveniently due to their high surface areas, special structures and strong magnetism. Nineteen target peptides with the sequence coverage of 21% can be enriched and detected from the highly diluted digest of BSA (5 fmol μL−1). These results reveal that the prepared GO/rGO–Fe3O4 composites have potential application as a carrier for biomolecule immobilization, enrichment, and separation.
Hydrophilic Polydopamine-Coated Magnetic Graphene Nanocomposites for Highly Efficient Tryptic Immobilization

Abstract:In this work, polydopamine-coated magnetic graphene (MG@PDA) nanocomposites were synthesized by a facile method. Trypsin was then directly immobilized on the surface of the nanocomposites through simple PDA chemistry with no need for introducing any other coupling groups. The as-made MG@PDA nanocomposites inherit not only the large surface area of graphene which makes them capable of immobilizing high amount of trypsin (up to 0.175 mg/mg), but also the good hydrophilicity of PDA which greatly improves their biocompatibility. Moreover, the strong magnetic responsibility makes them easy to be separated from the digested peptide solution when applying a magnetic field. The feasibility of the trypsin-immobilized MG@PDA (MG@PDA-trypsin) nanocomposites for protein digestion was investigated and the results indicated their high digestion efficiency in a short digestion time (10 min). In addition, the reusability and stability of the MG@PDA-trypsin nanocomposites were also tested in our work. To further confirm the efficiency of MG@PDA-trypsin nanocomposites for proteome analysis, they were applied to digest proteins extracted from skimmed milk, followed by nano RPLC-ESI-MS/MS analysis, and a total of 321 proteins were identified, much more than those obtained by 16-h in-solution digestion (264 proteins), indicating the great potential of MG@PDA-trypsin nanocomposites as the supports for high-throughput proteome study.
Facile Fabrication of Superparamagnetic Graphene/Polyaniline/Fe3o4 Nanocomposites for Fast Magnetic Separation and Efficient Removal of Dye

Abstract:Using graphene as adsorbent for removal of pollutants from polluted water is commonly recognized to be costly because the graphene is usually produced by a very complex process. Herein, a simple and eco-friendly method was employed to fabricate efficient superparamagnetic graphene/polyaniline/Fe3O4 nanocomposites for removal of dyes. The exfoliation of graphite as nanosheets and the functionalization of nanosheets with polyaniline and Fe3O4 nanoparticles were simultaneously achieved via a one-pot reaction process combining the intercalation polymerization of aniline and the co-precipitation of the residual Fe3+ and the generated Fe2+. The obtained graphene/polyaniline/Fe3O4 nanocomposites exhibited excellent adsorption performance for Congo red, even in the presence of Brilliant green. The adsorption kinetics and adsorption isotherms were well fitted with pseudo second-order kinetic model and Langmuir isotherm model, respectively. In a word, this method is simple and industrially feasible, which provides a new approach to fabricate highly efficient graphene-based adsorbents on large scale for removal of dyes. In addition, it also can be used to exfoliate other two-dimensional materials, such as boron nitride, carbon nitride and MoS2 for a range of possible applications.

Abstract:Capturing glycopeptides selectively and efficiently from mixed biological samples has always been critical for comprehensive and in-depth glycoproteomics analysis, but the lack of materials with superior capture capacity and high specificity still makes it a challenge. In this work, we introduce a way first to synthesize a novel boronic-acid-functionalized magnetic graphene@phenolic-formaldehyde resin multilayer composites via a facile process. The as-prepared composites gathered excellent characters of large specific surface area and strong magnetic responsiveness of magnetic graphene, biocompatibility of resin, and enhanced affinity properties of boronic acid. Facile Synthesis of Aptamer‐Functionalized Polydopamine‐Coated Magnetic Graphene Oxide Nanocomposites for Highly Efficient Purification of His‐Tagged Proteins

Abstract:Recombinant proteins hold significant importance in numerous disciplines. As the demand for expressing and purifying these proteins grows, the scientific community is in dire need of a simple yet versatile methodology that can efficiently purify these proteins. Aptamers as synthetic nucleic acid–based ligands with high affinity have shown promise in this regard, as they can capture targets through molecular recognition. In this study, novel aptamer‐functionalized polydopamine‐coated magnetic graphene oxide nanocomposites were facilely prepared, achieving an impressive average aptamer coverage density (45 nmol/mg). These nanocomposites exhibited a uniform structure and robust magnetic responsiveness. The findings indicated that they possess several advantages, such as rapid adsorption, substantial capacity (171.4 mg/g), and excellent reusability. Notably, due to the inherent properties of nucleic acids, the immobilized aptamer‐magnetic beads can be utilized repeatedly with high purification efficiency. Finally, the nanocomposites were further employed to purify His‐tagged proteins from actual samples. Remarkably, they were able to selectively and efficiently isolate His‐tagged retinoid X receptor alpha protein from complex Escherichia coli lysate. The purified His‐tagged retinoid X receptor alpha protein was analyzed using matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry. This confirmed the efficacy of developed nanocomposites, reinforcing their vast potential for purification of His‐tagged recombinant proteins.

Abstract:Graphene oxide (GO) is a chemical compound with a form similar to graphene that consists of one-atom-thick two-dimensional layers of sp2-bonded carbon. Graphene oxide exhibits high hydrophilicity and dispersibility. Thus, it is difficult to be separated from aqueous solutions. Therefore, functionalization with magnetic nanoparticles is performed in order to prepare a magnetic GO nanocomposite that combines the sufficient adsorption capacity of graphene oxide and the convenience of magnetic separation. Moreover, the magnetic material can be further functionalized with different groups to prevent aggregation and extends its potential application. Until today, a plethora of magnetic GO hybrid materials have been synthesized and successfully employed for the magnetic solid-phase extraction of organic compounds from environmental, agricultural, biological, and food samples. The developed GO nanocomposites exhibit satisfactory stability in aqueous solutions, as well as sufficient surface area. Thus, they are considered as an alternative to conventional sorbents by enriching the analytical toolbox for the analysis of trace organic compounds.

Abstract:In recent years, graphene-based magnetic composites have attracted tremendous research interest owing to its exceptional properties, such as huge surface area, large delocalized π-electron system, strong magnetic responsiveness, and excellent mechanical/thermal stability. These promising properties together with the ease of processibility and functionalization render graphene-based magnetic composites to be ideal adsorbents in magnetic solid-phase extraction. In this review, we outline the state of the art on the preparation approaches for different graphene-based magnetic composites and its application as adsorbents in preconcentrating organic compounds, biological macromolecules, and metal ions. In addition, future research directions of this type of magnetic materials are identified as well.
Geldanaycin-encapsulated magnetic nanoparticle for isolation of myosin in proteomics

Abstract:The grafting of a drug molecule, i.e., geldanamycin (GA) onto polyethyleneimine (PEI)-coated magnetic nanoparticle produces a novel composite, GA@Fe 3 O 4 –NH 2 . The composite is confirmed by characterizations with FT-IR, Raman, SEM, EDS, VSM and TEM. Due to the high binding-affinity of GA with myosin heavy chain (MYH), GA@Fe 3 O 4 –NH 2 exhibits excellent adsorption performance towards myosin. Consequently, a solid-phase extraction procedure is established for highly efficient and selective separation of myosin from pig heart extract. At pH 6.0, an adsorption efficiency of 97.1 % is achieved for treating 100 μg mL −1 myosin (0.1 mL) with 0.1 mg GA@Fe 3 O 4 –NH 2 as adsorbent. The adsorption behavior of myosin onto GA@Fe 3 O 4 –NH 2 fits Langmuir model, corresponding to a theoretical adsorption capacity of 518.1 mg g −1 . The adsorbed myosin can be readily recycled by the SDS solution (1 %, m/m) with an elution efficiency of 91.8 %. According to circular dichroism spectroscopy, the conformational changes of myosin during adsorption and elution are reversible. For practical application, myosin is successfully isolated from the pig left ventricular protein extract with GA@Fe 3 O 4 –NH 2 , and SDS-PAGE and LC-MS/MS showed that myosin had high purity and a total of 716 proteins could be identified. Significantly, Geldamycin-encapsulated magnetic nanoparticle for the separation of myosin well exploits the potential of the nanomaterials modified by drug molecules in the separation and purification of target proteins.

Abstract:Ultrafine cobalt ferrite (CoFe2O4) nanoparticles with narrow size distributions and regular morphologies were prepared through an improved procedure based on the high-temperature diol reduction method. By replacing the original reducing agent like expensive dihydric alcohol, oleylamine was applied as both stabilizer and reducing agent, which benefit for large scale synthesis of magnetic CoFe2O4 nanoparticles. To produce protein-separation agent with both high specific surface area and magnet content, inverse microemulsion system and substitution reaction were employed to render the produced CoFe2O4 nanoparticles with dense silica bilayer and iminodiacetic acid (IDA)-like structure modification. After the chelation with Cu(2+), the metal-immobilized CoFe2O4 nanoparticles can specifically absorb bovine hemoglobin (BHb) with maximum adsorption capacity as qm=1812.3mg/g. Compared with the reported Cu(2+)-immobilized magnetic nanoparticles, the as-prepared CoFe2O4@Si-IDA-Cu(2+) nanoparticles exhibited excellent adsorption capacity and were applied as high efficient protein separation agent in a real complex biological fluid like bovine blood.
