Modulated Current Based Scanning Ion Conductance Microscopy (SICM) with High Frequency Vibrating Probe for Cell Imaging

Abstract:Scanning ion conductance microscopy (SICM) has been becoming a powerful tool for imaging and characterizing biological samples in recent years. However, the regulating speed of probes in a majority of the existing modulated current based SICM systems is limited by the resonant frequency of Z-direction piezoelectric ceramic with long travel. In this study, a modulated current based SICM system with a high frequency vibrating probe was constructed to improve the regulating speed of the probe. The material for each component of the designed probe was selected using finite element simulation. A multi-objective optimization method was utilized to determine the key dimensions of each component. A microstructured sample was imaged using the modulated current based hopping mode with the designed high frequency vibrating probe, and the imaging result was compared to that of using the conventional hopping mode. Experimental results indicated that the designed high frequency vibrating probe has more robust feedback control over the probe position, thus effectively improving the imaging quality of the conventional SICM systems with relatively slow regulating speed of the probes. The imaging results of living cells further verify the feasibility of the newly built SICM system.
