Abstract:Liriope muscari (Decne.) L. H. Bailey is a well-known traditional Chinese medicine used for treating cough and insomnia. There are few reports on the quality evaluation of this herb partly because the major steroid saponins are not readily identified by UV detectors and are not easily isolated due to the existence of many similar isomers. In this study, a qualitative and quantitative method was developed to analyze the major components in L. muscari (Decne.) L. H. Bailey roots. Sixteen components were deduced and identified primarily by the information obtained from ultra high performance liquid chromatography with ion-trap time-of-flight mass spectrometry. The method demonstrated the desired specificity, linearity, stability, precision, and accuracy for simultaneous determination of 15 constituents (13 steroidal glycosides, 25(R)-ruscogenin, and pentylbenzoate) in 26 samples from different origins. The fingerprint was established, and the evaluation was achieved using similarity analysis and principal component analysis of 15 fingerprint peaks from 26 samples by ultra high performance liquid chromatography. The results from similarity analysis were consistent with those of principal component analysis. All results suggest that the established method could be applied effectively to the determination of multi-ingredients and fingerprint analysis of steroid saponins for quality assessment and control of L. muscari (Decne.) L. H. Bailey.

Abstract:Six new steroidal saponins (1-6) and one known steroidal saponin (7) were obtained from the roots of Ophiopogon japonicus (L. f.) Ker-Gawl. Their structures were determined by the detailed analysis of extensive nuclear magnetic resonance and mass spectroscopic data. The in vitro cytotoxic activities of these compounds against MDA-MB-435, HepG2 and A549 cell lines were also investigated.

Abstract:A new eudesmane sesquiterpene glycoside, ophiopogonoside B (1), along with five known compounds, ophiopogonoside A (2), ruscogenin-1-O-[β-D-glucopyranosyl (1 → 2)]-[β-D-xylopyranosyl (1 → 3)]-β-D-fucopyranoside (3), palmitic acid (4), palmitic acid glyceride (5), and β-sitosterol-D-glucopyranoside (6),was isolated from the tuberous roots of Liriope muscari (Decn.) Bailey (Liliaceae). Their structures were confirmed by 1D and 2D NMR spectroscopy. Among them, compounds 1, 2, 4, and 5 were the first reported from the genus Liriope. Ophiopogonoside B (1) showed moderate inhibitory activity to glycogen phosphorylase a.

Abstract:The screening of several Chinese medicinal herbs for nematocidal properties showed that the ethanol extract of Liriope muscari fibrous roots possessed significant nematocidal activity against the pine wood nematode (Bursaphelenchus xylophilus). From the ethanol extract, a new constituent (1,4-epoxy-cis-eudesm-6-O-β-D-glucopyranoside) and three known glycosides [1β,6α-dihydroxy-cis-eudesm-3-ene-6-O-β-D-glucopyranoside (liriopeoside A), 1β,6β-dihydroxy-cis-eudesm-3-ene-6-O-β-D-glucopyranoside, and 1α,6β-dihydroxy-5,10-bis-epi-eudesm-4(15)-ene-6-O-β D-glucopyranoside] were isolated by bioassay-guided fractionation. The structures were elucidated by 1D and 2D NMR and MS techniques. 1,4-Epoxy-cis-eudesm-6-O-β-D-glucopyranoside possessed moderate nemato-cidal activity against B. xylophilus with a LC(50 )value of 339.76 μg/mL, while liriopeoside A (LC(50) = 82.84 μg/mL) and 1β,6β-dihydroxy-cis-eudesm-3-ene-6-O-β-D-glucopyranoside (LC(50) = 153.39 μg/mL) also exhibited nematocidal activity against B. xylophilus. The crude extract of L. muscari fibrous roots exhibited nematocidal activity against the pine wood nematode with a LC(50) value of 182.56 μg/mL.

Abstract:Two new steroidal glycosides (1 and 2), together with 15 known compounds (3-17) were isolated from the fibrous roots of Ophiopogon japonicas, and three new steroidal glycosides (18-20), together with 14 known compounds (21-34) were isolated from the fibrous roots of Liriope spicata var. prolifera. The structures of the new compounds were elucidated on the basis of extensive one-dimensional (1D)- and 2D-NMR spectroscopic analyses and mass spectrometry. The isolated compounds were evaluated for their anti-inflammatory activity in vitro. Most of these steroidal glycosides showed significant inhibitory activity against neutrophil respiratory burst stimulated by phorbol myristate acetate.

Abstract:Lonicera Linn.is an important genus of the familyCaprifoliaceaecomprising of approximately 200 species, and some species of which have been usually used in traditional Chinese medicine for thousands of years. Some species of this genus can also be used in functional foods, cosmetics and other applications. The saponins, as one of most important bioactive components of theLonicera Linn.genus, have attracted the attention of the scientific community. Thus, a comprehensive and systematic review on saponins from the genus is indispensable. In this review, 87 saponins and sapogenin from the genus ofLonicera Linn., together with their pharmacological activities including hepatoprotective, anti-inflammatory, anti-bacterial, anti-allergic, anti-tumor, and immunomodulatory effects, and hemolytic toxicity were summarized.

Abstract:In a continuation of phytochemical studies on the underground organs of Liriope spicata var. prolifera, four new steroidal glycosides, lirioproliosides A-D, along with two known compounds, 25(S)-ruscogenin 1-O-[alpha-L-rhamnopyranosyl(1-->2)[beta-D-xylopyranosyl (1-->3)]-beta-D-fucopyranoside and ophiopogonin A, were identified. The structures of lirioproliosides A-D were established by a combination of spectroscopic and chemical methods as 25(S)-ruscogenin 1-O-[alpha-L-rhamnopyranosyl(1-->2)][beta-D-xylopyranosyl (1-->3)]-beta-D-fucopyranoside-3-O-alpha-L-rhamnopyranoside, 25(S)-ruscogenin 1-O-[3-O-acetyl-alpha-L-rhamnopyranosyl(1-->2)]-beta-D-fucopyranoside, 25(S)-ruscogenin (1-O-[2-O-acetyl-alpha-L-rhamnopyranosyl (1-->2)]-beta-D-fucopyranoside and ruscogenin (1-O-[2-O-acetyl-alpha-L-rhamnopyranosyl(1-->2)]-beta-D-fucopyranoside, respectively. Among these steroidal glycosides, ophiopogonin A and lirioprolioside B, and lirioproliosides C and D, were isolated as epimeric pairs.

Abstract:Two new steroidal saponins, diosbulbisides D (1) and E (2), along with five known saponins (3-7) were isolated from the rhizomes of Dioscorea bulbifera L. Their structures were elucidated on the basis of spectral data. Compounds 6 and 7, two 3-O-trisaccharides of diosgenin spirostanes, showed moderate cytotoxic activity against human hepatocellular carcinoma cells, with IC(50) values of 3.89 mu M and 7.47 mu M on SMMC7721, and 10.87 mu M and 19.10 mu M on Bel-7402 cell lines, respectively.

Abstract:Henryiosides F and G (1 and 2), two new steroidal saponins along with two known analogues (3 and 4) were obtained from the roots and rhizomes of Maianthemum henryi. Their structures were determined by physicochemical properties and spectroscopic methods including 1D, 2D-NMR, IR and HR-ESI-MS data analysis. Cytotoxic activity in human HepG2 and SW620 tumour cells were evaluated by the MTT method and all of the saponins exhibited cytotoxicity with IC50 values ranging from 15.33 μM to 57.85 μM.

Abstract:Five phenolic compounds, namely N-trans-coumaroyltyramine (1), N-transferuloyltyramine (2), N-trans-feruloyloctopamine (3), 5,7-dihydroxy-8-methoxyflavone (4) and (3S)3,5,4'-trihydroxy-7-methoxy-6-methylhomoisoflavanone (5), were isolated from the fibrous roots of Liriope muscari (Liliaceae). Compounds 2-5 were isolated for the first time from the Liriope genus. Their in vitro antioxidant activities were assessed by the DPPH and ABTS scavenging methods with microplate assays. The structure-activity relationships of compounds 1-3 are discussed.

The invention also provides the application of the rusogenin fucopyranoside C in the preparation of the medicine for resisting tumor metastasis, and more particularly relates to the application in the preparation of the medicine for generating tumour new blood vessels and inhibition of relative tumor metastasis. Being proved by the study of the invention, the rusogenin fucopyranoside C can effectively inhibit the growth, migration and adhesive ability, can inhibit lung metastasis of melanoma B16, and provides new selection in the application of preparing the medicine for resisting tumor metastasis.

Abstract:Nine steroidal components, including a new pregnane glycoside (1), were obtained from the roots and rhizomes of Smilacina henryi. Their structures were determined via extensive spectroscopic data including, IR, HRESIMS and 1D, 2D NMR data analysis. Furthermore, their cytotoxic activities against human HepG2 and SW620 tumor cells were evaluated by the MTT method and compounds 2, 3, 5, 8 and 9 showed moderate activity with IC50 values raging from 18.4 to 86.3 μM.

Their structures were determined by means of extensive spectroscopic analyses (IR, ESIMS, HRESIMS, 1D and 2D NMR). Compounds 1-9 were tested for their cytotoxic activities by MTT assay, and 1, 3, 5 and 6 showed moderate cytotoxic activities against HeLa, SMMC-7721, and HL-60 human tumor cell lines. Abstract:Bulbs ofLilium brownii, commonly known as "Bai‐he" in China, serve both edible and medicinal purposes in clinical practice. In this study, two new isospirostanol‐type saponins were isolated from L. brownii, and their structures were identified by spectroscopic method, and absolute configurations were elucidated by comprehensive analysis of spectral data obtained from combined acid hydrolysis. Two compounds were finally identified as 3‐O‐[α‐L‐rhamnopyranosyl‐(1→2)‐β‐D‐glucopyranoside]‐(22R,25R)‐5α‐spirosolane‐3β‐ol (1) and 3‐O‐{α‐L‐rhamnopyranosyl‐(1→2)‐[β‐D‐glucopyranosyl‐(1→4)]‐β‐D‐glucopyranoside}‐(22R,25R)‐5α‐spirosolane‐3β‐ol (2), respectively. Further, we found that compound 2 significantly suppressed the proliferation of SMMC‐7721 and HepG2 cells with IC50 values of 26.3±1.08 μM and 30.9±1.59 μM, whereas compound 1 didn't inhibit both of the two hepatocellular carcinoma. Subsequently, compound 2 effectively decreased the levels of interleukin‐1β and tumor necrosis factor‐α and the expression of Bcl‐2, and increased the expression of Bax and Caspase‐3 proteins. Which indicated that the anti‐hepatocellular carcinoma effect of compound 2 involves reducing the level of inflammation and inducing apoptosis.

Abstract:Two new triterpenoid saponins, a cycloartane-type triterpenoid saponin (1) and a oleanane-type triterpenoid saponin (2), were isolated from the whole plant of Ligularia przewalskii. Their structures were elucidated based on extensive spectroscopic studies, including IR, UV, HR-ESI-MS, 1D and 2D NMR experiments. (1H-NMR, 13C-NMR, DEPT, 1H-1H COSY, HMBC, HSQC, and NOESY). Their cytotoxic activities were evaluated against six human cancer (HeLa, HepG2, SGC7901, MDA231, HL60, and Lewis) cell lines in vitro using MTT assay. As a result, two triterpenoid saponins showed potent cytotoxic activities against six tumors cell lines and with broad spectrum activities against six human cancer, with IC50 values from 8.40 to 24.39 μM. The cytotoxic activities of 2 are more potent.

Abstract:The present study was designed to investigate the chemical constituents of the roots of Cyathula officinalis. Compounds were isolated by silica gel, Sephadex LH-20, ODS column chromatography, and preparative HPLC. Their structures were determined on the basis of 1D and 2D NMR techniques, mass spectrometry, and chemical methods. One new oleanane-type triterpenoid saponin, 28-O-[α-L-rhamnopyranosyl-(1→3)-β-D-glucuronopyranosyl-(1→3)-β-D-glucopyranosyl] hederagenin (1), was isolated from the roots of Cyathula officinalis. The anti-inflammatory activities of the isolates were evaluated for their inhibitory effects against LPS-induced nitric oxide (NO) production in RAW 264.7 macrophages cells. Compounds 2, 4, and 6 exhibited moderate anti-inflammatory activities.
