Abstract:Objective To explore the expression levels and clinical significance of ABCG2 and OCT4 in gallbladder carcinoma tissues.Methods Adopting real-time PCR to detect the mRNA expression of ABCG2 and OCT4 in fresh gallbladder benign and malignant tissues.Western blot method was used to detect the protein expression of ABCG2 and OCT4 in gallbladder benign and malignant tissues.Immunohistochemical method was used to detect the expression levels of ABCG2 and OCT4 on paraffin-embedded tissues in 40 patients with gallbladder cancer and 8 cases with chronic cholecystitis.Statistical analysis was made for the correlation of the expression levels of ABCG2 and OCT4,and for the association of the expression of ABCG2 and OCT4 with the clinicopathological characteristics.Results ① The mRNA expression of ABCG2 and OCT4 in gallbladder cancer group were(117.9±10.5) and(86.2±13.2),which were obviously higher than those in chronic cholecystitis tissues [(27.1±7.9) and(2.1±1.0)].The difference between the two groups was present significantly(P<0.05).② It was shown with Western blot that the protein expression level of ABCG2 of gallbladder cancer tissues was obviously high,while its level of chronic cholecystitis tissues was weak.Meanwhile the protein expression of OCT4 in chronic cholecystitis tissues could not be detected,but in the tumor tissues the expression level of OCT4 was higher.③ Immunohistochemical results displayed that the positive rates of ABCG2 in gallbladder carcinoma and chronic cholecystitis groups were 60.0% and 37.5% respectively,and the difference between the two groups was present significantly(P<0.05).④ There was no expression of OCT4 in chronic cholecystitis tissues,but in gallbladder carcinoma tissues the positive rate of OCT4 was 45%.⑤ In gallbladder carcinoma tissues,the differences of ABCG2 and OCT4 expression levels were not statistically significant with age and gender(P>0.05),but the expression levels of ABCG2 and OCT4 were related to gallbladder carcinoma invasion,cancerous differentiation degree and TNM staging(P<0.05).⑥ ABCG2 and OCT4 expression levels were positively correlated each other(rs=0.65,P<0.05).Conclusions Compared with chronic cholecystitis tissue,the transcription and translation levels of ABCG2 and OCT4 in gallbladder cancer tissues are over expressed.The expression levels of ABCG2 and OCT4 are associated with gallbladder carcinoma invasion,differentiation and TNM staging,and the relationship between the 2 parmeters is significantly positively correlated,which suggests that ABCG2 and OCT4 become to be important indexes reflecting gallbladder carcinoma biological behavior.
