Abstract:Due to the uniqueness of fluorine atom and C—F bond,difluoromethylene has special properties.As a bioisostere of an oxygen or a carbonyl group,it plays an important role in medicines,pesticides and materials.Difluoromethyl heteroaryl sulfones,represented by 2-PySO2CF2H (Hu reagent),have been developed recently as difluoromethylation reagents,and widely recognized by synthetic chemists for their ease of preparation,good functional group tolerance and universal applicability to a wide range of carbonyl compounds.Through different types of reactions such as nucleophilic substitution,nucleophilic addition,Julia-Kocienski olefination reaction,and radical-mediated difunctionalization,they introduced difluoromethyl,difluoromethylene,gem-difluoroalkene and other fluorine-containing groups into aldehydes,ketones,and heterocyclic compounds.For the first time,the synthesis of fluorine-containing organic compounds involved in various difluoromethyl heteroaryl sulfones in the past decade is reviewed from the perspective of reaction types and their application studies.

The high reactivity of reagent 6 allowed to highly selectively access either C‐monofluoromethylated or O ‐monofluoromethylated β‐ketoesters in high yields by the use of different bases. In addition, reactions of reagent 3 with a variety of nucleophiles including phenols, carboxylic acids, thiophenols or heteroaryl nucleophiles occurred in full conversion within 10 min at room temperature and the scopes for these reactions were reported in detail.

Abstract:The Lewis acidity of metal-organic frameworks (MOFs) has attracted much research interest in recent years. We report here the development of two quantitative methods for determining the Lewis acidity of MOFs-based on electron paramagnetic resonance (EPR) spectroscopy of MOF-bound superoxide (O2•-) and fluorescence spectroscopy of MOF-bound N-methylacridone (NMA)-and a simple strategy that significantly enhances MOF Lewis acidity through ligand perfluorination. Two new perfluorinated MOFs, Zr6-fBDC and Zr6-fBPDC, where H2fBDC is 2,3,5,6-tetrafluoro-1,4-benzenedicarboxylic acid and H2fBPDC is 2,2',3,3',5,5',6,6'-octafluoro-4,4'-biphenyldicarboxylic acid, were shown to be significantly more Lewis acidic than nonsubstituted UiO-66 and UiO-67 as well as the nitrated MOFs Zr6-BDC-NO2 and Zr6-BPDC-(NO2)2. Zr6-fBDC was shown to be a highly active single-site solid Lewis acid catalyst for Diels-Alder and arene C-H iodination reactions. Thus, this work establishes the important role of ligand perfluorination in enhancing MOF Lewis acidity and the potential of designing highly Lewis acidic MOFs for fine chemical synthesis.

Abstract:AbstractFluorine incorporation into organic molecules is expected to lower the pKa of neighboring functionality via its strong electron‐withdrawing effect, and this strategy has been widely exploited in diverse disciplines. Herein, we report a striking anomalous α‐fluorine substitution effect on the α‐Csp3‐H acidity. We have experimentally measured the pKa values of a series of popular nucleophilic monofluoromethylating reagents α‐fluoro(phenylsulfonyl)methane derivatives as well as their C‐H analogues by Bordwell's overlapping indicator method in dimethyl sulfoxide solution. Contrary to expectations, we found that α‐fluorine substituent does not generally enhance but rather weaken the α‐Csp3‐H acidity of most (phenylsulfonyl)methane derivatives. DFT computations reproduce and provide insight into the anomalous α‐fluorine effect. A correlation was identified between the C‐H pKa of (phenylsulfonyl)methane derivatives and Mayr's nucleophilicity parameter (N) of the corresponding carbanions.

Abstract:Abstract Organofluorine compounds have shown their great value in many aspects. Moreover, allenes are also a class of important compounds. Fluorinated or fluoroalkylated allenes might provide an option as candidates for drug and material developments, as allenes allow a great number of valuable transformations. Herein, we report a metal-free synthesis of difluoromethylated allenes via regioselective trifunctionalization of 1,3-enynes. This method proceeds through double C–F bond formation with concomitant introduction of an amino group to the allene. Synthetic applications are conducted and preliminary mechanistic studies suggest that a two-step pathway is involved. DFT calculations revealed an unusual dibenzenesulfonimide-assisted fluorination/fluoroamination with NFSI. In addition, kinetic reaction study revealed the induction period of both major and side products to support the proposed reaction mechanism. This work offers a convenient approach for the synthesis of a range of difluoromethylated allenes and is also a rare example of trifunctionalization of 1,3-enynes.

Abstract:Amides are ubiquitous in physical and life sciences. Given the significant abundance of arenes, dearomative aminocarbonylation of arenes would lead to a large and underexplored chemical space for amide discovery. However, such reactions are challenging due to the high degree of resonance stabilization and selectivity issues. Herein, we disclose an unprecedented dearomative trifluoromethylative aminocarbonylation of arenes via bifunctional coordination to chromium, providing a modular platform for the construction of amides possessing trifluoromethyl (CF3) groups and three-dimensional rings. Its versatility further enabled a switchable difluoromethylation or trifluoromethylation aminocarbonylation of arene C-H bonds. A possible mechanism was proposed based on control experiments. Finally, the synthetic utility was well demonstrated by diverse applications in the total synthesis of CF3-functionalized amide-type drugs, including praziquantel, nateglinide, maraviroc and alloyohimbane.

Abstract:The internal C-C double bond in 3-aryl-1,2-allenes was highly regioselectively fluorohydroxylated to afford 2-fluoroalken-3-ols in 37-88% yields by using Selectfluor as the electrophilic reagent. The regioselectivity may be determined by the electronic effect, while the reactivity may be controlled by the stabilization effect of the aryl group in the allylic cationic intermediates.

Abstract:Although defluorinative functionalization of trifluoromethylarenes has been studied by several research groups, different kinds of catalysts are requested accordingly based on specific substrate families to obtain corresponding RCF2Ar compounds. Herein, we report a general photocatalytic approach for selective hydrodefluorination (HDF) and defluoroalkylation (DFA) of ArCF3. Using electronically-different arenethiolates as photosensitizer, 88 gem-difluoromethyl compounds were synthesized from a broad range of electronically-variable trifluoromethylarenes, including natural products and bioactive molecules. Our mechanistic studies uncovered photo-induced Electron Donor-Acceptor (EDA) complexation followed by the silane-assisted C-F bond activation.

Abstract:Recent advances in photoinduced trifluoromethylation and difluoroalkylation under photocatalysis are summarized. Most of the photoredox reactions proceed efficiently under mild conditions with simple operation. Various fluorinated reagents are developed and applied in different transformations. Usually, the photocatalyst in a photocatalytic cycle is initially stimulated to the excited state in the presence of visible light, which then provides an electron to the fluorinated reagent to release a CF3 radical or CF2 radical. The subsequent radical addition to an unsaturated bond with further transformations would produce the corresponding fluorinated products. Alkenes, alkynes, isocyanides, and arenes are frequently employed as substrates for access to fluorochemicals. The advantages of trifluoromethylation and difluoroalkylation under photocatalysis including mild reaction conditions, good functional group tolerance, and experimental ease will make the approaches attractive for further applications.

Abstract:Trifluoroethyl group served as the bioisosteres of ethyl or ethoxy group widely exists in the fields of pharmaceuticals and agrochemicals. Developing novel strategies of introducing trifluoroethyl group into various skeletons has become a hot topic in organic synthetic chemistry. In this review, the trifluoroethylation reaction of various partners such as alkynes, alkenes, functionalized arenes, amines and so on were summarized. This review was categorized into four parts according to the bond formation: C(sp)-CH2CF3, C(sp2)-CH2CF3, C(sp3)-CH2CF3, and X (X = N, O, S, Se)-CH2CF3. Some representative examples along with reaction mechanisms were also discussed.

Abstract:Reaction of 4-aryl-1,1,1-trifluorobut-3-en-2-ols [CF3-allyl alcohols, ArCH═CHCH(OH)CF3] with arenes under activation with anhydrous FeCl3 or FSO3H was studied. We found that the transformation led to trifluoromethylated alkenes [Ar(Ar')CHCH═CHCF3] or 1-trifluoromethylated indanes (CF3-indanes). The formation of these two types of reaction products strongly depends on the nucleophilicity of the starting arene and the electrophilicity of cationic intermediates generated from CF3-allyl alcohols under reaction conditions. Benzene, anisole, veratrole, and ortho-xylene lead exclusively to CF3-alkenes with an E-configuration. More π-donating polymethylated arenes (pseudocumene, mesitylene) afford only CF3-indanes with a predominantly cis-orientation of substituents at positions 1 and 3 of the indane ring. Meta- and para-xylenes show an intermediate behavior; they may form both CF3-alkenes and/or CF3-indanes. The mechanisms of the investigated transformations are discussed.

Abstract:Dearomative trifluoromethylation of arenes provides an ideal method for constructing CF3‐containing three‐dimensional architectures which are of increasing interest in drug discovery. However, such transformation is rare and challenging because of the inherently low reactivity of arene π‐systems and selectivity issues. Herein, we disclose a general dearomative 1,2‐hydrotrifluoromethylation of (hetero)arenes utilizing activation by temporary π‐complexation to chromium, thus affording convenient access to 1,3‐cyclohexadienes bearing the privileged CF3 group. The versatility of this strategy further enables a dearomative 1,2‐deuterotrifluoromethylation with high deuterated ratios. The related dearomative 1,2‐(deutero)hydroperfluoroethylation reactions were conducted well for a range of chromium‐bound (hetero)arenes. Finally, a reasonable mechanism was proposed based on the intermediate capture and control experiments.

Abstract:A general and convenient method for the direct transition-metal-free intermolecular chloro- difluoromethylthiolation of a variety of unactivated alkenes and alkynes has been described. This methodology exhibits good functional group tolerance and operationally simple. Mechanistic studies indicated that this is the electrophilic substitution mechanism, and the strong electrophilic difluoromethylthiolating reagent HCF 2 SCl was generated in situ under the standard reaction conditions. The developed protocol fulfills the requirements of sustainable and green chemistry.

Abstract:Sulfenyl fluorides are organic compounds of sulfur in formal oxidation state +2 with the formula R–S–F. Although the chloride, bromide, and iodide analogues have been extensively described in the literature, organic sulfenyl fluorides remain essentially unstudied. These structures have been implicated as putative intermediates in established processes to access polyfluorinated sulfur species; however, definitive and direct evidence of their existence has not been achieved, nor has a systematic understanding of their reactivity. Here, we report the synthesis, isolation, and spectroscopic characterization of several arenesulfenyl fluorides, including structural analysis of 2,4-dinitrobenzenesulfenyl fluoride by single-crystal X-ray diffraction. The functional group undergoes direct, efficient, and highly regioselective anti-addition to alkenes and alkynes, as well as insertion by carbenes. The resulting α- or β-fluoro thioether adducts can be readily transformed into useful fluorinated motifs, for example by modification of the sulfur groups (to sulfonamides or sulfonyl fluorides), by sulfur elimination (to generate formal C–H fluorination products), or by Julia–Kocienski olefination (to form vinyl fluorides). Thus, we establish that sulfenyl fluorides are unexpectedly accessible and stable compounds. Further, they serve as versatile reagents for the production of fluorinated organic compounds.
