Starting with direct trifluoromethylation, new synthons and new reactions, the research progress of the synthesis of trifluoromethylated quaternary carbon centers is reviewed.

Abstract:Due to the uniqueness of fluorine atom and C—F bond,difluoromethylene has special properties.As a bioisostere of an oxygen or a carbonyl group,it plays an important role in medicines,pesticides and materials.Difluoromethyl heteroaryl sulfones,represented by 2-PySO2CF2H (Hu reagent),have been developed recently as difluoromethylation reagents,and widely recognized by synthetic chemists for their ease of preparation,good functional group tolerance and universal applicability to a wide range of carbonyl compounds.Through different types of reactions such as nucleophilic substitution,nucleophilic addition,Julia-Kocienski olefination reaction,and radical-mediated difunctionalization,they introduced difluoromethyl,difluoromethylene,gem-difluoroalkene and other fluorine-containing groups into aldehydes,ketones,and heterocyclic compounds.For the first time,the synthesis of fluorine-containing organic compounds involved in various difluoromethyl heteroaryl sulfones in the past decade is reviewed from the perspective of reaction types and their application studies.

Abstract:As one kind of tetrahydrophthalimide herbicides with high efficiency to control the weed in corn and soybean fields,flumiclora-pentyl was firstly synthesized by Sumitomo Chemical.Based on the traditional synthesis process,using p-fluorophenol as a starting material,a new synthesis process of flumiclora-pentyl was developed with several steps such as chlorination,etherification,nitrification,reduction and condensation.The yield of the product was 33%,and the purity was higher than 95%.The structures of the product and the intermediate were confirmed by Elementary Analysis,1H-NMR,MS and IR methods.

Abstract:The demand for effective fire extinguishants has spurred investigation into novel perfluorinated molecules due to their exceptional properties, including high thermal stability, low toxicity, and eco-friendliness. This study presents a computational framework for designing and tailoring perfluorinated compounds as potential alternatives to traditional extinguishing agents. The research centers on elucidating the synthesis, molecular architecture, and characterization of these compounds to enhance their fire suppression capabilities while mitigating environmental impact. By employing computational methods, molecular modeling, and advanced spectroscopic techniques, the structural intricacies and potential applications of perfluorinated compounds in fire suppression are investigated. Additionally, future research directions aimed at addressing challenges and advancing the development of environmentally sustainable fire extinguishants are discussed. This manuscript contributes to the ongoing efforts to replace conventional extinguishing agents with safer and more environmentally friendly alternatives.

Abstract:Fluorine-containing polyester (FPE) was synthesized by the reaction of phthalic anhydride, epichlorohydrin, and 2,2,3,3-tetrafluoropropyl glycidyl ether (TFGE) at the catalysis of tetraethylammonium bromide using 1,4-butanediol as the initiator. The structure of FPE was characterized by 1H NMR and 19F NMR, and the result showed that the fluorine in TFGE was introduced into FPE. Fluorine content in FPE was determined by oxygen-flask combustion and fluoride selective electrode. It was found that fluorine content in FPEs increased with the increase of TFGE content. The wettability of water and oil on the cured coating based on FPE was investigated by contact angle meter. Contact angles of water and diiodomethane on the cured coatings first increased with the increase of the fluorine content in FPEs and then remained constant. The X-ray photoelectron spectroscopic analysis showed that the F/C molar ratio on the coatings surface was much higher than the overall F/C molar ratio, which indicated that fluoroalkyl groups in FPEs had enriched on the coatings surface. This review summarises the confluence of these two important moities and provides a comprehensive account of the methods for synthesising and applying fluorine-, polyfluoroalkyl- and polyfluoroaryl-substituted 1,2,3-triazoles. Focus has been devoted to recent examples and applications where the properties of both the 1,2,3-triazole and organofluorine substituent are proposed to be important in enhancing the compounds functionality.

Abstract:Fluorinated molecules have a wide range of applications and are used as medicines, agrochemicals and refrigerants and in smartphone liquid crystal displays, photovoltaic solar cells, Teflon tapes and the coatings of textiles and buildings. Fluorination and fluoroalkylation — incorporation of a trifluoromethyl, difluoromethyl or monofluoromethyl group — are the major strategies used for the construction of carbon–fluorine bonds and fluorinated carbon–carbon bonds, respectively. The past two decades have witnessed a rapid growth in fluorination and fluoroalkylation methods thanks to the development of new reagents and catalysts. This Primer aims to provide an overview of state-of-the-art strategies in fluorination, trifluoromethylation, difluoromethylation and monofluoromethylation, with an emphasis on using C–H functionalization, although other strategies for fluorination and fluoroalkylation are also discussed. Further landmark achievements are expected in the fields of fluorination and fluoroalkylation as organofluorine compounds are used increasingly in everyday applications.

Abstract:Pentafluorosulfanyl (SF 5 )‐containing compounds and corresponding analogs are a highly valuable class of fluorine‐containing building blocks owing to their unique properties. The reason for that is the set of peculiar and tremendously beneficial characteristics they can impart on molecules once introduced onto them. Despite this, their application in distinct scientific fields remains modest, given the extremely harsh reaction conditions needed to access such compounds. The recent synthetic approaches via S–F, and C–SF 5 bond formation as well as the use of SF 5 ‐containing building blocks embody a "stairway‐to‐heaven" loophole in the synthesis of otherwise‐inaccessible chemical scaffolds only a few years ago. Herein, we report and evaluate the properties of the SF 5 group and analogs, by summarizing synthetic methodologies available to access them as well as following applications in material science and medicinal chemistry since 2015.

Abstract:The special properties of fluorine atoms and fluorine-containing groups have led to an increasing number of applications for fluorine-containing organic compounds, which are also extremely widely used in the field of new drug development. Unfortunately, naturally fluorinated organics are rare in nature, so the selective introduction of fluorine atoms or fluorine-containing groups into organic molecules is very important for pharmaceutical/synthetic chemists. N-2,2,2-trifluoroethylisatin ketimines have received the attention of many chemists since they were first developed as fluorine-containing synthons in 2015. This paper reviews the organic synthesis reactions in which trifluoroethyl isatin ketimine has been involved in recent years, focusing on the types of reactions and the stereoselectivity of products, and also provides a prospect of its application in this field.

Abstract:Organofluorine compounds have greatly benefited the pharmaceutical, agrochemical, and materials sectors. However, they are plagued by concerns associated with PFAS. Additionally, the widespread use of the trifluoromethyl group is facing imminent regulatory scrutiny. Defluorinative functionalization, which converts the trifluoromethyl to the difluoromethyl motifs, represents the most efficient synthetic strategy. However, general methods for robust C(sp3)–F bond transformations remain elusive due to challenges in selectivity and functional group tolerance. Here, we present a method for C(sp3)–F bond defluorinative functionalization of the trifluoromethyl group via difluoromethyl anion in flow. This new approach tames the reactive difluoromethyl anion, enabling diverse functional group transformations. Our methodology offers a versatile platform for drug and agrochemical discovery, overcoming the limitations associated with fluorinated motifs.

Abstract:The selective C‐F bonds functionalization in available trifluoromethyl compounds is an economic route for obtaining high‐value organic fluorine compounds. However, due to the high C‐F bonds dissociation energy in trifluoromethyl group, the strength of the bond decreases continuously during defluorination, posing a challenge in modifying such molecules without over‐conversion. Over the past few years, there has been significant progress in developing efficient methods for constructing difluoromethylene unit, using approaches such as radical chemistry, photochemistry, electrochemistry, and organometallic chemistry. These methods involve difluorocarbon radicals, difluorocarbon cations, difluorocarbon anions, and trifluoromethyl carbenes as intermediates. In this review, we summarized the recent five years of research achievements in this field for selective single C‐F bond cleavage in trifluoromethyl aromatic and carbonyl compounds to gain diverse difluoromethylene‐containing organic molecules via different intermediate strategies.
