Abstract:Low temperature is adverse to anaerobic ammonium oxidation (anammox) reaction while proper Fe addition can enhance anammox performance. Therefore, batch assays were conducted to investigate the potential effects of magnetite (100 μm, 20 μm and 200 nm) on anammox performance which were achieved from the reactor operated at 10–25 °C. After 3 runs, the results indicated that nano-scale magnetite improved the nitrogen elimination significantly. The specific anammox activity (SAA) of the group with nano-magnetite amendments was greater than the other groups after 3 runs (13.5, 12.9, 14.3, 15.4 and 15.7 mgTN/(gVSS·h)), reaching 18.0 mgTN/(gVSS·h). The distribution of magnetite in the granules were then analyzed using X-ray diffraction (XRD) and energy dispersive X-ray spectroscopy (EDS). The results indicated that nano-magnetite was more feasible to attached to the surface of the granules which might accelerate the release of Fe(II) or Fe(III) to enhance anammox performance.

Magnetite addition reduces nitrite requirement for efficient anaerobic ammonium oxidation by facilitating mutualism of ANAMMOX and FEAMMOX bacteria

Abstract:Partial nitrification (PN) is crucial for anaerobic ammonium oxidation (ANAMMOX), but faces challenges such as high energy demands and process control. Recent research has highlighted additives like magnetite as potential alternatives to conventional electron acceptors (O₂ and NO₂-) for enhancing ammonium (NH4+) oxidation with lower energy consumption. This study investigated the effect of adding 50 mg/L of magnetite to ANAMMOX reactors, resulting in improved nitrogen (N) removal efficiency. The magnetite-added ANAMMOX (M-ANA) reactor yielded N removal efficiencies of 71 %, 66 %, and 57 % for NH4+:NO2- molar ratios of 1:1.3, 1:0.8, and 1:0.5, respectively. The M-ANA reactor operated under a 0.5 mol lower NO2- concentration achieved similar performance to the control ANAMMOX (C-ANA) reactor operated with a theoretical amount of NO2-. Moreover, the M-ANA reactor showed the potential to remove NH4+ by 56 % without any NO2- supplementation. Metagenomic analysis showed that the addition of magnetite significantly improved the relative abundance of microorganisms involved in the FEAMMOX reaction, such as Fimbriimonas ginsengisoli and Pseudomonas stutzeri. It also facilitated positive mutualism between ANAMMOX and FEAMMOX reactions. In addition, M-ANA granules exhibited a dense and compact structure compared with C-ANA, and the presence of magnetite facilitated the formation of resilient granules. Notably, the useful protein (Heme C) concentration and specific microbial activity in the M-ANA reactor were 1.3 and 2.2 times higher than those in the C-ANA reactor. Overall, the results demonstrate that an appropriate amount of magnetite can enhance the N removal efficiency while reducing the energy input requirements and associated carbon emissions. These findings can guide the future development of carbon- and energy-neutral N removal processes.
Effect of Ferrous Ion on the Performance of an Anammox Reactor

Abstract:The effect of ferrous ion on the performance of an anaerobic ammonium oxidation (anammox) reactor was investigated. The substrate removal rate of the reactor was boosted by addition of ferrous ion within 205 d. The maximum NH+4-N and NO-2-N removal rate with 0.075 mmol·L-1 Fe2+ was 1.8-fold and 1.6-fold of the control (Fe2+ concentration = 0.03 mmol·L-1). Microbial growth,denoted by production of nitrate and volatile solids (VS),was also enhanced. The biomass increment at 0.075 mmol·L-1 Fe2+ was 1.36-fold over the control,and the VS of the test reactor was 2.15-fold of the control. The anammox bacteria varied in cellular structure under iron-replete conditions (0.075 mmol·L-1). An unknown grey area appeared between the light area and the dark area,which was unprecedented in anammox bacteria. Based on this research,anammox bacteria have a large demand for iron and the former medium for the anammox bacteria was iron limited.
The role of magnetite (Fe3O4) particles for enhancing the performance and granulation of anammox

Abstract:In this study, two lab-scale sequencing batch reactors each with an effective volume of 2.3 L were operated as C-AMX (no carrier addition) and M-AMX (magnetite carrier added) for 147 days with synthetic wastewater at an NLR range of 0.19-0.47 kgN/m3/d. The long-term effect of magnetite on the granulation and performance of anammox bacteria in terms of nitrogen removal and other essential parameters were confirmed. In phase I (1-24 days), M-AMX took approximately 12 days to obtain a nitrogen removal rate (NRR) above 80 % of the initial input nitrogen. Although free nitrous acid inhibited the reactor at a high concentration at the onset of phase III, the NRR of M-AMX recovered about 3.7 times faster than that of C-AMX. In addition, it was confirmed that the M-AMX granules had a dense and compact structure compared to C-AMX, and the presence of the carrier promoted the development of these resilient granules. While the measured microbial stress gradually increased in C-AMX reactor, a vice versa was observed in the M-AMX reactor as granulation proceeded. Compared to other alternative iron-based carrier particles, the stable crystal structure of magnetite as a carrier created a mechanism where filamentous bacteria groups were repelled from the granulation hence the microbial stress in the M-AMX in the final phase was 61.54 % lower than that in the C-AMX. The iron rich environment created by the magnetite addition led to Ignavibacteria, (a Feammox bacteria) increasing significantly in the M-AMX bioreactor.

Abstract:Magnetite has been widely reported to accelerate methane production in anaerobic digestion. However, the effects of magnetite on the process of methane consumption, specifically nitrate-dependent anaerobic oxidation of methane (AOM), and its underlying mechanisms remain poorly understood. In this study, the addition of magnetite significantly improved the removal of nitrate in an anaerobic membrane biofilm reactor (AnMBfR) (25.2 ± 2.5 mg NO 3− -N/L/d) compared to that in an AnMBfR without magnetite (8.7 ± 4.2 mg NO 3− -N/L/d). Metatranscriptomic analysis showed that genes encoding type IV pilus assembly proteins were highly expressed in nitrate/nitrite-reducing bacteria in the biofilms of the two AnMBfRs. Specifically, the transcript abundance of these genes was observed to be higher in the presence of magnetite (gene expression levels, log 2 FPKM = 13.08) compared to the absence of magnetite (12.04). Biofilms with magnetite exhibited a linear increase in electron transfer resistance with increasing temperature, and the decrease in pH significantly increased their conductivity, indicating a metallic-like conductivity. Contrarily, an increase in temperature or decrease in pH did not influence the resistance or conductivity of biofilms without magnetite. Some of these cytochrome c (CYC)-like proteins were electrically active in the electrochemical Fourier transform infrared spectra and were closely associated with the outer-membrane multi-heme c-type cytochromes (OMCs). The intensities of the OMC-like bands in the surface-enhanced resonance Raman spectra with biofilms in the presence of magnetite were higher than those without magnetite. This study suggests that magnetite promotes the nitrate-dependent AOM process because of the formation of methanotrophic consortia based on direct interspecies electron transfer (DIET).

Abstract:In this study, the effect of Fe3+ on the start-up of Anammox process was investigated. Four EGSB reactors were operated with the addition of 0 (R1), 0.04 (R2), 0.08 (R3) and 0.14 (R4) mmol/L Fe3+, respectively. The results showed that Fe3+ remarkably improved the nitrogen loading rate (NLR) and operation efficiency of the reactor. After 180 days, the influent NH4+-N concentration in the four reactors was 201.4, 301.8, 343.2, 380.2 mg N/L, and the NLR was 589.3, 877.6, 993.0, 1105.8 mg N/(L.d), respectively. And the nitrogen removal rate (NRR) in R2, R3 and R4 was respectively 1.54, 1.73 and 1.94 times of that in R1. High throughput sequencing revealed that Fe3+ could promote the enrichment of Anammox bacteria Candidatus Brocadia. Moreover, the analysis by qPCR indicated that the abundance of Anammox 16S rRNA gene and the functional gene hzsB increased, which showed a positive correlation with the concentration of Fe3+.

Abstract:Nitrite-dependent anaerobic methane oxidation (n-DAMO) is a novel denitrification process that simultaneously further removes and utilizes methane from anaerobic effluent from wastewater treatment plants. However, the metabolic activity of n-DAMO bacteria is relative low for practical application. In this study, conductive magnetite was added into lab-scale sequencing batch reactor inoculated with n-DAMO bacteria to study the influence on n-DAMO process. With magnetite amendment, the nitrogen removal rate could reach 34.9 mg N·L −1 d −1 , nearly 2.5 times more than that of control group. Magnetite significantly facilitated the interspecies electron transfer and built electrically connected community with high capacitance. Enzymatic activities of electron transport chain were significantly elevated. Functional gene expression and enzyme activities associated with nitrogen and methane metabolism had been highly up-regulated. These results not only propose a useful strategy in n-DAMO application but also provide insights into the stimulating mechanism of magnetite in n-DAMO process.

Long-term and multiscale assessment of methanogenesis enhancement mechanisms in magnetite nanoparticle-mediated anaerobic digestion reactor

Abstract:Magnetite nanoparticles (Fe3O4-NPs) have been demonstrated to be involved in direct interspecies electron transfer between syntrophic bacteria, yet a comprehensive assessment of the ability of Fe3O4-NPs to cope with process instability and volatile fatty acids (VFAs) accumulation in scaled-up anaerobic reactors is still lacking. Here, we investigated the start-up characteristics of an expanded granular sludge bed (EGSB) with Fe3O4-NPs as an adjuvant at high organic loading rate (OLR). The results showed that the methane production rate of R1 (with Fe3O4-NPs) was approximately 1.65 folds of R0 (control), and effluent COD removal efficiency was maintained at approximately 98.32% upon 20 kg COD/(m3·d) OLR. The components of volatile fatty acids are acetate and propionate, and the rapid scavenging of propionate accumulation was the difference between R1 and the control. The INT-ETS activity of R1 was consistently higher than that of R0 and R2, and the electron transfer efficiencies increased by 68.78% and 131.44%, respectively. Meanwhile, the CV curve analysis showed that the current of R1 was 40% higher than R3 (temporary addition of Fe3O4-NPs), indicating that multiple electron transfer modes might coexist. High-throughput analysis further revealed that it was difficult to reverse the progressive deterioration of system performance with increasing OLR by simply reconfiguring bacterial community structure and abundance, demonstrating that the Fe3O4-NPs-mediated DIET pathway is a prerequisite for establishing multiple electron transfer systems. This study provides a long-term and multi-scale assessment of the gaining effect of Fe3O4-NPs in anaerobic digestion scale-up devices, and provides technical support for their practical engineering applications.
Enhanced Anammox Consortium Activity for Nitrogen Removal: Impacts of Static Magnetic Field

Abstract:Anaerobic ammonium oxidation (anammox) process has been becoming a promising technology for the removal of nitrogenous contaminants from wastewater. In short-term batch tests, we observed the anaerobic ammonium oxidizing activity of anammox consortium increased as the magnetic field varied in the range of 16.8-95.0mT. A maximum 50% increase was obtained at the value of 75.0mT. In order to study long-term effect of magnetic field on anammox consortium, an anammox reactor with magnetic field of 60.0mT was operated in laboratory-scale. The results demonstrated that a significant 30% increase in maximum nitrogen removal rate and an approximate 1/4 saving in cultivation time were achieved by using the magnetic system. Microbiological composition analysis showed that bacterial diversity in the reactor decreased under magnetic-exposed condition. Nevertheless, some strains belonging to Planctomycetales were highly enriched. These findings indicated that the magnetic field was useful and reliable for fast start-up of anammox process since it was proved as a simple and convenient approach to enhance anaerobic ammonium oxidizing activity.
Enhancing mainstream anammox process by adding Fe3O4 nanoparticles

Abstract:In order to further promote the application of mainstream anammox, magnetic field enhancing technology was applied to improve the anammox start-up and operation performance under the conditions of low temperature and substrates concentrations. Two UASBs were adopted as anammox reactors and run in parallel. The controlling group and the magnetic field enhancing group (with 2.4 g/L Fe3O4 NPs) were recorded as UASBC and UASBM. The results showed that UASBC and UASBM successfully took 37 days and 33 days to start mainstream anammox with TNRE of 90.03% and 85.05%, respectively. The maximum TNLR of UASBC and UASBM reached 0.54 mg N(m3·d)−1 and 0.82 mg N(m3·d)−1 with the maximum TNRE of 86.41% and 85.50% during 121 days operation. The results showed that magnetic filed could change morphological characteristics of AnAOB and strengthen sludge granulation by adjusting the composition of EPS. Microbial high-throughput sequencing analysis results showed the phylum of Planctomycetes dominated in UASBC and UASBM with the relative abundance of 26.41% and 32.68% on 121 d. Candidatus Kuenenia was cultured as the dominant bacteria, and it was speculated that magnetic filed had the ability to promoting the enrichment of Candidatus Kuenenia. These findings were interesting and meaningful, and could provide new ideas for long-term application of mainstream anammox.

Abstract:Two up-flow anaerobic sludge blanket (UASB) reactors (RM with 10 g Fe L-1 magnetite, RB without magnetite) feeding with synthetic wastewater were built to investigate the effect of magnetite addition on anaerobic digestion (AD) performance during start-up and starvation recovery stages. With the magnetite addition, the COD removal efficiency and biogas production during the two stages were enhanced, and the recovery time of RM was shortened by about 50%. The reduced synthesis of riboflavin and heme c along with enhanced sludge conductivity of RM indicated that magnetite could replace their roles for efficient extracellular electron transfer (EET), which favors the growth of anaerobes. Microbial community analysis showed that potential syntrophic partners like Syntrophaceae and Methanothrix were enriched in RM during the recovery stage, and the performance was improved with quick responsiveness. Results demonstrated that addition of conductive materials like magnetite could improve the stability and restorability of AD process efficiently.
Insights into feasibility and microbial characterizations on simultaneous elimination of dissolved methane from anaerobic effluents and nitrate/nitrite reduction in a conventional anoxic reactor with magnetite

Abstract:Discovery of nitrate/nitrite-dependent anaerobic methane oxidation (DAMO) challenges the conventional biological treatment processes, since it provides a possibility of simultaneously mitigating dissolved methane emissions from anaerobic effluents and reducing additional carbon sources for denitrification. Due to the slow growth of specialized DAMO microbes, this possibility has been just practiced with biofilms in membrane biofilm reactors or granular sludge in membrane bioreactors. In this study, simultaneous elimination of dissolved methane from anaerobic effluents and nitrate/nitrite reduction was achieved in a conventional anoxic reactor with magnetite. Calculations of electron flow balance showed that, with magnetite the eliminated dissolved methane was almost entirely used for nitrate/nitrite reduction, while without magnetite approximately 52 % of eliminated dissolved methane was converted to unknown organics. Metagenomic sequencing showed that, when dissolved methane served as an electron donor, the abundance of genes for reverse methanogenesis and denitrification dramatically increased, indicating that anaerobic oxidation of methane (AOM) coupled to nitrate/nitrite reduction occurred. Magnetite increased the abundance of genes encoding the key enzymes involved in whole reverse methanogenesis and Nir and Nor involved in denitrification, compared to that without magnetite. Analysis of microbial communities showed that, AOM coupled to nitrate/nitrite reduction was proceeded by syntrophic consortia comprised of methane oxidizers, Methanolinea and Methanobacterium, and nitrate/nitrite reducers, Armatimonadetes_gp5 and Thauera. With magnetite syntrophic consortia exchanged electrons more effectively than that without magnetite, further supporting the microbial growth.
[Effect of Sequential Biocatalyst Addition on Anammox Process].

Abstract:Anaerobic ammonium oxidation (Anammox) process is a high-rate nitrogen removal technology that has been applied in sludge dewatering effluents treatment with nitrogen removal rate as high as 9.5 kg/(m x d). However, due to the slow growth rate of the autotrophic Anammox bacteria and the susceptivity to environmental conditions, the start-up of Anammox process is very long; the operation is unstable; and the nitrogen removal from organic-containing and/or toxicant-containing ammonium-rich wastewaters using Anammox process becomes difficult. Thus, the application of this high-rate process is significantly limited. In this paper, a newly-developed Anammox process with sequential biocatalyst (Anammox biomass) addition was established based on the procedure in fermentation engineering. We introduced the Anammox process with sequential biocatalyst addition on start-up, stable operation and the treatment of organic-containing and toxicant-containing ammonium-rich wastewaters. Results show that supplementing high-activity Anammox biomass into reactors will increase the amount of as well as the ratio of Anammox bacteria. Thus, the innovative Anammox process with sequential biocatalyst addition not only accelerates the start-up course, but also enhances the stability of Anammox process. Furthermore, it overcomes the drawbacks of wastewaters containing high organic content and toxic substances. Therefore, the application of Anammox process may be further enlarged.
Magnetite Enhancing Sludge Anaerobic Fermentation to Improve Wastewater Biological Nitrogen Removal: Pilot-scale Verification

Abstract:Alkaline anaerobic fermentation for acids production has been considered as an effective method to recover resources from waste activated sludge, and magnetite could improve the quality of fermentation liquid. Here, we have constructed a pilot-scale sludge alkaline anaerobic fermentation process enhanced by magnetite to produce short chain fatty acids (SCFAs), and used them as external carbon sources to improve the biological nitrogen removal of municipal sewage. Results showed that the addition of magnetite could significantly increase the production of SCFAs. The average concentration of SCFAs in fermentation liquid reached 3718.6 ± 101.5 mg COD/L, and the average concentration of acetic acid reached 2368.8 ± 132.1 mg COD/L. The fermentation liquid was used in the mainstream A2O process, and its TN removal efficiency increased from 48.0% ± 5.4% to 62.2% ± 6.6%. The main reason was that the fermentation liquid is conducive to the succession of sludge microbial community the denitrification process, increasing the abundance of denitrification functional bacteria and realizing the enhancement of denitrification process. Besides, magnetite can promote the activity of related enzymes to enhance the biological nitrogen removal. Finally, the economic analysis showed that magnetite-enhanced sludge anaerobic fermentation was economically and technically feasible to promote the biological nitrogen removal of municipal sewage.

Abstract:Magnetic nanoparticles (NPs) have been widely applied in environmental remediation, biomass immobilization and wastewater treatment, but their potential impact on anaerobic ammonium oxidation (anammox) biomass remains unknown. In this study, the short-term and long-term impacts of maghemite NPs (MHNPs) on the flocculent sludge wasted from a high-rate anammox reactor were investigated. Batch assays showed that the presence of MHNPs up to 200 mg L-1 did not affect anammox activity, reactive oxygen species production, or cell membrane integrity. Moreover, long-term addition of 1-200 mg L-1 MHNPs had no adverse effects on reactor performance. Notably, the specific anammox activity, the abundance of hydrazine synthase structural genes and the content of extracellular polymeric substance were increased with elevated MHNP concentrations. Meanwhile, the community structure was shifted to higher abundance of Candidatus Kuenenia indicated by high-throughput sequencing. Therefore, MHNPs could be applied to enhance anammox flocculent sludge due to their favorable biocompatibility.
Studies on Start-Up Technique of Anammox Bioreactor

Abstract:The feasibility of start-up of anammox bioreactor, using autotropic denitrifying biofilm was studied. The anammox bioreactor could be started up successfully within 110d, adopting the method of cultivating firstly autotrophic denitrifying biofilm, then start-up of the bioreactor. The volumetric total nitrogen loading was 0.145kg/(m3.d), and the removal rate of NH4+-N and NO2--N was 98.59% and 99.08%, respectively. At the initial period of start-up, the effluent pH value of the bioreactor was lower than the influent pH value, and tended to be unanimous with the advance of the start-up process. At steady operation, the liquid pH value in the bioreactor was alkaline, the ratio of removed NH4+-N, NO2--N and produced NO3--N was 1:1.01:0.19. In the start-up process, the ratio of removed NH4+-N, NO2--N and produced NO3--N as well as the change of pH value and biofilm color could indicate the start-up course of the bioreactor.
Effect of Ferric Iron on Enrichment Cultivation Process of Anammox Sludge

Abstract:Effect of ferric iron on enrichment cultivation process of anammox sludge was investigated through control trial,moreover,qPCR analysis and high-throughput 16S rRNA sequencing were conducted to make the mechanism analysis.The results showed that nitrogen load rate (NRR) and operational performance were significantly enhanced with ferric iron addition in influent.Besides,qPCR analysis revealed that functional genes relevant to denitrification were dramatically decreased and abundance of anammox 16S rRNA and gene hzsB encoding hydrazine synthase were enriched.It was noteworthy that high abundance of anammox 16S rRNA and gene hzsB was achieved in reactors with ferric iron addition.High-throughput 16S rRNA sequencing indicated that Candidate Brocadia,a typical anaerobic ammonium oxidation bacteria (AAOB) were enriched remarkably in reactors with ferric iron addition.

Abstract:Nitrate is a common pollutant in the aquatic environment. Denitrification and dissimilatory nitrate reduction to ammonium (DNRA) are the main reduction processes of nitrate. In the relatively closed sediment environment, the competitive interaction of these two nitrate reduction determines whether the ecosystem removes or retains nitrogen. In the process of NO3--N bioreduction, Magnetite, which is a common mineral present in soil and other sediments can play a crucial role. However, it is still not clear whether magnetite promotes or inhibits NO3--N bioreduction. In this paper, the effect of magnetite on NO3--N bioreduction was studied by batch experiments. The results show that magnetite can increase the NO3--N reduction rate by 1.48 %, and can inhibit the DNRA process at the beginning of the reaction and then promote the DNRA process. Magnetite changed the microbial community structure in our experiment systems. The relative abundance of Sphingomonas, which mainly exists in a high carbon and low nitrogen environment, increased under sufficient carbon source conditions. The relative abundance of Fe-oxidizing and NO3--N reducing bacteria, such as Flavobacterium, increased in the absence of carbon sources but in the presence of magnetite. In addition, magnetite can significantly increase activity of the microbial electron transport system (ETS). the added microbial electronic activity of magnetite increased nearly two-fold under the same experiment conditions. The acid produced by the metabolisms of Pseudomonas and Acinetobacter further promotes the dissolution of magnetite, thus increasing the concentration of Fe (II) in the system, which is beneficial to autotrophic denitrifying bacteria and promote the reduction of NO3--N. These findings can enhance our understanding of the interaction mechanism between iron minerals and nitrate reducing bacteria during nitrate reduction under natural conditions.

Abstract:Direct interspecies electron transfer (DIET) has been shown to be more efficient than conditional mediated interspecies electron transfer via hydrogen/formate. However, the effective enrichment of functional organisms and the development of DIET to improve the anaerobic digestion performance of sludge remain a challenge. This study aimed to develop a magnet-Fe3O4 composite structure capable of enriching functional microorganisms without the need to continuously add conductive materials. The composition of the microbial community and its corresponding functionalities were compared using metagenomic analysis in response to the addition of free Fe3O4 and magnet-Fe3O4 composite groups. The magnet-Fe3O4 composite structure reduced the lag phase by 51.2% and enhanced the maximum methane production rate by 50.1% compared with those of the free Fe3O4 group. Electrochemically active bacteria such as Dissulfurimicrobium and Geobacter were enriched in the magnet-Fe3O4 biofilm, along with archaea Methanosaeta. The presence of DIET was proved by the enriched CO2 reduction pathway assigned to Methanosaeta and a larger quantity of type IV pili genes possessed by electrochemically active bacteria in the magnet-Fe3O4 biofilm. These results provide a promising strategy for enriching functional microorganisms and stably improving methanogenesis efficiency in anaerobic sludge digestion systems.

Abstract:Nitrite-dependent anaerobic methane-oxidizing (n-damo) process has a promising prospect in anaerobic wastewater treatment, utilizing methane as the sole electron source to remove nitrite. However, the metabolic activity of n-damo bacteria is too low for practical application. This study aimed to stimulate n-damo process by introducing conductive nano-magnetite and/or electron shuttle anthraquinone-2,6-disulfonate (AQDS), and also set a comparative treatment of adding insulated ferrihydrite. The results showed that the nitrite reduction rate was enhanced the most significantly in treatment with nano-magnetite, approximately 1.6 times higher than that of the control without any supplement. While ferrihydrite application showed an adverse effect on n-damo process. The well-known aerobic methane oxidizer Methylomonas spp. was found to be enriched under n-damo condition with the supplementation of nano-magnetite and/or AQDS, but abundance of n-damo bacteria did not exhibit significant increase. It was hypothesized that Methylomonas spp. could be survived under anaerobic n-damo condition using oxygen produced by n-damo bacteria for the self-growth, and the nitrite reduction could be promoted through the enhancement of microbial interspecies electron transfer triggered by the introduction of conductive materials. It opens a new direction for the stimulation of n-damo activity, which needs more evidences to verify the hypothetic mechanism.
