Abstract:Direct interspecies electron transfer (DIET) has been shown to be more efficient than conditional mediated interspecies electron transfer via hydrogen/formate. However, the effective enrichment of functional organisms and the development of DIET to improve the anaerobic digestion performance of sludge remain a challenge. This study aimed to develop a magnet-Fe3O4 composite structure capable of enriching functional microorganisms without the need to continuously add conductive materials. The composition of the microbial community and its corresponding functionalities were compared using metagenomic analysis in response to the addition of free Fe3O4 and magnet-Fe3O4 composite groups. The magnet-Fe3O4 composite structure reduced the lag phase by 51.2% and enhanced the maximum methane production rate by 50.1% compared with those of the free Fe3O4 group. Electrochemically active bacteria such as Dissulfurimicrobium and Geobacter were enriched in the magnet-Fe3O4 biofilm, along with archaea Methanosaeta. The presence of DIET was proved by the enriched CO2 reduction pathway assigned to Methanosaeta and a larger quantity of type IV pili genes possessed by electrochemically active bacteria in the magnet-Fe3O4 biofilm. These results provide a promising strategy for enriching functional microorganisms and stably improving methanogenesis efficiency in anaerobic sludge digestion systems.

A Short-Term Stimulation of Ethanol Enhances the Effect of Magnetite on Anaerobic Digestion

Abstract:Conductive iron oxides (CIO) have been proved recently to facilitate the anaerobic microbial syntrophy based on the direct interspecies electron transfer (DIET) in batch experiments. However, the effect of CIO was always insignificant in anaerobic digestion (AD) reactor especially when the DIET-based syntrophic partners were absent. In this study, the effect of magnetite on performance of AD system with sucrose as a sole carbon source was investigated, but limited enhancement was achieved during the first 36-day operation. The short-term effect of ethanol addition was further studied in the magnetite-amended AD reactor, and results showed that the AD reactor with 10gFe/L micro-sized magnetite (R3) achieved higher performance of COD removal and methane proportion compared with the other reactors (R1 without magnetite; R2 with 2gFe/L micro-sized magnetite; R4 with 2gFe/L nano-sized magnetite). Meanwhile, the pyridoxine in extracellular polymeric substances (EPS) and conductivity of anaerobic sludge from R3 increased more significantly than those of the others. Analysis of high-throughput sequencing indicated that the abundance of archaea increased in sludge from R3 and Methanosarcina responsible for DIET was dominant (63.64%). Additionally, the abundance of potential electroactive bacteria Chloroflexi in R3 was 7.57-fold, 3.61-fold and 7.37-fold as that of R1, R2 and R4, respectively. These results demonstrated that the electroactive microbes and methanogens could be enriched efficiently in anaerobic sludge via synergetic effect of magnetite addition and ethanol short-term stimulation.
Direct interspecies electron transfer stimulated by coupling of modified anaerobic granular sludge with microbial electrolysis cell for biogas production enhancement

Abstract:The magnetite nanoparticle-anaerobic granular sludge complex (Fe-AnGS) was formed by a biologically induced mineralization method, then applied in microbial electrolysis cells (MEC) to enhance the direct interspecies electron transfer (DIET) pathway via selective enriched electrogenic microorganisms for biogas production. Results showed with the highest Fe loading amount of 325.03 ± 8.40 mg/gVS, the magnetization and conductivity of Fe-AnGS were improved by 30.94 % and 29.16 %, respectively. The MEC with Fe-AnGS enriched the specific electrogenic microorganisms in the anode and cathode chambers. Simultaneously, genes coding for carbon dioxide reduction [M00567] increased by 29.40–43.54 % and 34.29–43.43 %, respectively. Inoculation of Fe-AnGS in MEC with glucose as substrate enhanced the experimental methane production of the cathode chambers ( EMP C ), and theoretical methane production of the cathode chamber ( TMP CE ) by 8.00–14.10 %, and 11.83–29.30 %, respectively. Consequently, inoculation of Fe-AnGS in MEC could establish and enhance the DIET pathway for methane production.

Abstract:Direct interspecies electron transfer (DIET) is a new mechanism responsible for syntrophic methane production. Supplementation of conductive material in syntrophic communities can facilitate electron transfer and enhance methane production. Ferroferric oxide (Fe3O4) was dosed in an anaerobic sequencing batch reactor (ASBR) fed with tryptone-based synthetic wastewater to examine its effect on the anaerobic treatment process. Long-term dosage of Fe3O4 not only enhanced the maximum methane production rate by 78.3% in a reaction cycle but also improved methane production when hydrogen/carbon dioxide or acetate was used as the substrate. The conductivity of anaerobic sludge, activity of the electron transport chain, and extracellular electron transfer ability were enhanced with the addition of Fe3O4, inducing enhanced system performance for methane production. Proteiniclasticum and Prolixibacter were the enriched acidogens responsible for hydrolysis and acidification. With the addition of Fe3O4, Methanosarcina was the dominant methanogen, and metagenomic analysis further revealed the genes involved in the hydrogenotrophic pathway of methanogenesis. Methanosarcina might be involved in DIET, and Fe3O4 was responsible for its stimulation.

Abstract:Metal oxides are wildly studied to enhance anaerobic digestion and the methanogenic process, which is generally interpreted by increased direct interspecies electron transfer (DIET). Yet microbial mechanisms involved are under debate. Herein, methane production dynamics were analyzed, and acceleration on biogas accumulation was presented. Complementing previous findings, Fe3O4 nanoparticles stimulated bacterial fermentation rather than methanogenesis or syntropy between electro-microorganism and methanogen. More importantly, metagenome-assembled genomes proved that Fe3O4 nanoparticles increased acetogenesis by Parabacteroides chartae, which provided abundant substrates for acetoclastic methanogenesis. Interestingly, the weakly conductive V3O7·H2O nanowires increased potential hydrogen-producing bacteria, Brevundimonas, and electro-microorganisms, Clostridium and Rhodoferax, which is convenient for conducting DIET. Collectively, conductivity may not be a critical factor in mediating DIET and distinct strategies of metal oxides on methane production propose more possibilities, such as fermentation process.

Abstract:Discovery of nitrate/nitrite-dependent anaerobic methane oxidation (DAMO) challenges the conventional biological treatment processes, since it provides a possibility of simultaneously mitigating dissolved methane emissions from anaerobic effluents and reducing additional carbon sources for denitrification. Due to the slow growth of specialized DAMO microbes, this possibility has been just practiced with biofilms in membrane biofilm reactors or granular sludge in membrane bioreactors. In this study, simultaneous elimination of dissolved methane from anaerobic effluents and nitrate/nitrite reduction was achieved in a conventional anoxic reactor with magnetite. Calculations of electron flow balance showed that, with magnetite the eliminated dissolved methane was almost entirely used for nitrate/nitrite reduction, while without magnetite approximately 52 % of eliminated dissolved methane was converted to unknown organics. Metagenomic sequencing showed that, when dissolved methane served as an electron donor, the abundance of genes for reverse methanogenesis and denitrification dramatically increased, indicating that anaerobic oxidation of methane (AOM) coupled to nitrate/nitrite reduction occurred. Magnetite increased the abundance of genes encoding the key enzymes involved in whole reverse methanogenesis and Nir and Nor involved in denitrification, compared to that without magnetite. Analysis of microbial communities showed that, AOM coupled to nitrate/nitrite reduction was proceeded by syntrophic consortia comprised of methane oxidizers, Methanolinea and Methanobacterium, and nitrate/nitrite reducers, Armatimonadetes_gp5 and Thauera. With magnetite syntrophic consortia exchanged electrons more effectively than that without magnetite, further supporting the microbial growth.
Target the Enhancement of DIET-Based CO2-Dependent Methanogenesis Via Synergy of Magnetite Addition and Inorganic Carbon Recirculation

Abstract:In anaerobic systems, CO2-dependent methanogens as terminal electron acceptors determine the feasibility of organic degradation. In this study, targeting the enhancement of CO2-dependent methanogenesis, a strategy coupling the addition of magnetite and inorganic carbon recirculation was proposed. The results showed that the strategy facilitated the enrichment of Methanosaeta and Methanosarcina to the greatest extent (abundance of 42.9 and 38.8%, respectively), and metaproteomic analysis revealed that their enzymes coding for CO2-dependent methanogenesis were maximally expressed, compared with the single strategy. Combined with electrochemical analysis, magnetite was speculated to trigger direct interspecies electron transfer (DIET)-based syntrophy with extracellular redox-active mediators (Cyt C and flavins) in anaerobic sludge. The ability of Methanosaeta and Methanosarcina to accept electrons was significantly improved, as evidenced by the upregulated electron transfer proteins, such as F420H2 dehydrogenase, coenzyme F-420 hydrogenase, and CoB-CoM heterodisulfide reductase. Meanwhile, Methanosaeta and Methanosarcina enhanced HCO3 - takeup into cells via an ABC-type transport system under the condition of inorganic carbon recirculation, thus thermodynamically inducing electron consumption and inorganic carbon reduction. In summary, the coupling strategy had a great synergistic effect on the enhancement of DIET-based CO2-dependent methanogenesis and endowed the up-flow anaerobic sludge bed reactor with superior methanogenic performance for stable anaerobic wastewater treatment.
Residence Time Reduction in Anaerobic Reactors: Investigating the Economic Benefits of Magnetite-Induced Direct Interspecies Electron Transfer Mechanism

Abstract:Existing research on direct interspecies electron transfer (DIET) has predominantly focused on the types and concentrations of conductive materials across diverse anaerobic digestion. However, insufficient understanding of the impact of residence time, a critical economic factor, prompted this investigation. Magnetite, a conductive material, was introduced into the anaerobic digestion of food wastewater, leading to a significant increase in ultimate methane production (Bu) with 25 mM-Fe3O4 (p < 0.05). Despite a subsequent decline in methane production efficiency from 388.9% to 7.1% over the 15- to 65-day anaerobic digestion period, the initial impact of increased methane production due to magnetite addition was evident. Control's maximum methane production rate (Rm) was 27.5 mL/day, reaching its highest point at 37.4 mL/day with 15 mM-Fe3O4, accompanied by a noteworthy 56.6% reduction in the attainment day of Rm (Rm-day), shortened to 8.2 days. Even with 100 mM-Fe3O4, while Bu showed no significant difference, Rm-day exhibited a substantial reduction of 22.8. Despite the lower overall anaerobic digestion efficiency under some magnetite input conditions, this study confirmed a substantial shortening of Rm-day, suggesting that the DIET mechanism induced by conductive materials such as magnetite could reduce the residence time in continuous-type anaerobic reactors, contributing to improved economic feasibility.

Abstract:Increasing studies indicate that magnetite addition could accelerate the methanogenesis via enhancing direct interspecies electron transfer (DIET)-based anaerobic syntrophy. However, magnetite is found to run off in continuous bioreactor, and the effect of magnetite loss on syntrophic aggregates is still underreported. In this study, two EGSB reactors (RM with magnetite-enhanced sludge, and RB as a control) were operated to investigate the magnetite behavior in continuous bioreactor and the corresponding response of syntrophic aggregates. Results showed that magnetite in RM was washed out gradually in form of iron ions, and a slightly acidic niche was supposed to be the major cause. Nevertheless, candidate DIET partners like Geobacter and Methanothrix along with syntrophic volatile fatty acids (VFAs)-degrading microbes were enriched in RM. In addition, the improved redox activity of extracellular polymeric substance (EPS), higher sludge conductivity and electron transport activity suggested that the DIET ability of sludge in RM was still enhanced, which favors the syntrophic metabolism of VFAs. Interestingly, syntrophic partners were loosely combined under the condition of high organic loading rate (OLR) in the presence of magnetite, but with gradual loss of magnetite, dense and active anaerobic granular sludge (AGS) was formed in RM. This study provided a comprehensive understanding of magnetite behavior in continuous bioreactor and the response of syntrophic aggregates. The robust DIET-based syntrophy after magnetite adding could favor the high-efficient anaerobic wastewater treatment and resource recovery in the future, and further investigations on magnetite resupply and the mechanism of magnetite enriching candidate DIET partners are recommended.
Effects of Magnetite (Fe3O4) as an Electrical Conductor of Direct Interspecies Electron Transfer on Methane Production from Food Wastewater in a Plug Flow Reactor

Abstract:This study was conducted in order to examine the impact of magnetite (Fe3O4), a conductive material capable of promoting direct interspecies electron transfer (DIET) among microorganisms, on the efficiency of anaerobic digestion in a plug flow reactor (PFR) using food wastewater (FW) as the substrate. The effects of recovering and replenishing magnetite discharged along with the digestate during continuous operation of the PFR were also evaluated. A PFR with a total volume of 17 L was utilized as the reactor for anaerobic digestion. The inoculum was obtained from Icheon Biogas Research Facility, which operated with a mixture of pig slurry and FW in a 7:3 (w/w) ratio. FW was used as the substrate (volatile solids (VS) content of 85,865 mg-VS/L). The PFR was set for operation at 39 °C, and after a stabilization period of approximately 82 days, the hydraulic retention time (HRT) was set at 40 days. The study was conducted in three stages: stage 1 (83~122 days), stage 2 (123~162 days), and stage 3 (163~202 days). For the maintenance of an organic loading rate of 2.12 kg-VS/m3/d, 0.3 L/d of substrate was added every 24 h, and analysis of an equal amount of discharged digestate was performed. The experimental treatments included a control without the addition of magnetite after the stabilization period, treatment (T1) with addition of magnetite (20 mM in digestate) and subsequent recovery and replenishment of magnetite on the discharge of digestate, and treatment (T2) with addition of magnetite (20 mM) without the replenishment of magnetite. Analytical parameters included the characteristics of the discharged digestate (pH, NH4+-N, chemical oxygen demand (CODCr), total volatile fatty acids (TVFAs), and alkalinity), and methane production (Mp). During the period of operation of the PFR after the stabilization period, no significant differences in pH and NH4+-N, based on the recovery and replenishment of magnetite, were observed, and a stably functioning PFR was observed. However, in stage 2, due to the increased degradation of organic matter caused by DIET, the CODCr of T1 and T2 decreased by 9.42% compared with the control. In stage 3, the magnetite content in the reactor in T2 decreased by a maximum of 9.42% compared to T1. In stage 3, the Mp for T2 was similar to that of the control, with a maximum discharge of magnetite of 3.06%, and the Mp decreased by 5.40% compared to T1. Regarding the ratio of methanogens in the community, the results of an analysis of the digestate from stage 3 showed an increase in the community of acetotrophic methanogens, specifically Methanosarcina. The findings of this study confirm that DIET was effectively promoted by maintaining the concentration of 20 mM magnetite in the PFR while using FW as a substrate.

Abstract:Two up-flow anaerobic sludge blanket (UASB) reactors (RM with 10 g Fe L-1 magnetite, RB without magnetite) feeding with synthetic wastewater were built to investigate the effect of magnetite addition on anaerobic digestion (AD) performance during start-up and starvation recovery stages. With the magnetite addition, the COD removal efficiency and biogas production during the two stages were enhanced, and the recovery time of RM was shortened by about 50%. The reduced synthesis of riboflavin and heme c along with enhanced sludge conductivity of RM indicated that magnetite could replace their roles for efficient extracellular electron transfer (EET), which favors the growth of anaerobes. Microbial community analysis showed that potential syntrophic partners like Syntrophaceae and Methanothrix were enriched in RM during the recovery stage, and the performance was improved with quick responsiveness. Results demonstrated that addition of conductive materials like magnetite could improve the stability and restorability of AD process efficiently.
Enhanced Anaerobic Digestion of Long Chain Fatty Acid by Adding Magnetite and Carbon Nanotubes

Abstract:This study investigated the impact of stimulating direct interspecies electron transfer (DIET), by supplementing nano-sized magnetite (nFe3O4, 0.5 g Fe/g VSS) and carbon nanotubes (CNT, 1 g/L), in anaerobic digestion of oleic acid (OA) at various concentrations (0.10–4.00 g chemical oxygen demand(COD)/L). Both supplementations could enhance CH4 production, and its beneficial impact increased with increased OA concentration. The biggest improvements of 114% and 165% compared to the control were achieved by nFe3O4 and CNT, respectively, at OA of 4 g COD/L. The enhancement can be attributed to the increased sludge conductivity: 7.1 ± 0.5 (control), 12.5 ± 0.8 (nFe3O4-added), and 15.7 ± 1.1 μS/cm (CNT-supplemented). Dissolved iron concentration, released from nFe3O4, seemed to have a negligible role in improving CH4 production. The excretion of electron shuttles, i.e., humic-like substances and protein-like substances, were found to be stimulated by supplementing nFe3O4 and CNT. Microbial diversity was found to be simplified under DIET-stimulating conditions, whereby five genera accounted for 88% of the total sequences in the control, while more than 82% were represented by only two genera (Methanotrix concilli and Methanosarcina flavescens) by supplementing nFe3O4 and CNT. In addition, the abudance of electro-active bacteria such as Syntrophomonas zehnderi was significantly increased from 17% to around 45%.

Abstract:As a potential approach for enhanced energy generation from anaerobic digestion, iron-based conductive nanoparticles have been proposed to enhance the methane production yield and rate. In this study, the impact of two different types of iron nanoparticles, namely the nano-zero-valent-iron particles (NZVIs) and magnetite (Fe3O4) nanoparticles (NPs) was investigated, using batch test under mesophilic conditions (35 °C). Magnetite NPs have been applied in doses of 25, 50 and 80 mg/L, corresponding to 13.1, 26.2 and 41.9 mg magnetite NPs/gTS of substrate, respectively. The results reveal that supplementing anaerobic batches with magnetite NPs at a dose of 25 mg/L induces an insignificant effect on hydrolysis and methane production. However, incubation with 50 and 80 mg/L magnetite NPs have instigated comparable positive impact with hydrolysis percentages reaching approximately 95% compared to 63% attained in control batches, in addition to a 50% enhancement in methane production yield. A biodegradability percentage of 94% was achieved with magnetite NP doses of 50 and 80 mg/L, compared to only 62.7% obtained with control incubation. NZVIs were applied in doses of 20, 40 and 60 mg/L, corresponding to 10.8, 21.5 and 32.2 mg NZVIs/gTS of substrate, respectively. The results have shown that supplementing anaerobic batches with NZVIs revealed insignificant impact, most probably due to the agglomeration of NZVI particles and consequently the reduction in available surface area, making the applied doses insufficient for measurable effect.

Abstract:The effect of magnetite nanoparticles and nanocomposites (magnetite nanoparticles impregnated into graphene oxide) supplement on the recovery of overloaded laboratory batch anaerobic reactors was assessed using two types of starting inoculum: anaerobic granular sludge (GS) and flocculent sludge (FS). Both nanomaterials recovered methane production at a dose of 0.27 g/L within 40 days in GS. Four doses of magnetite nanoparticles from 0.075 to 1 g/L recovered the process in FS systems between 30 and 50 days relaying on the dose. The presence of nanomaterials helped to reverse the effect of volatile fatty acids inhibition and enabled microbial communities to recover but also favoured the development of certain microorganisms over others. In GS reactors, the methanogenic population changed from being mostly acetoclastic (Methanothrix soehngenii) to being dominated by hydrogenotrophic species (Methanobacterium beijingense). Nanomaterial amendment may serve as a preventative measure or provide an effective remedial solution for system recovery following overloading.

Abstract:Conductive materials have been demonstrated to enhance sludge methanogenesis, but few researches have concentrated on the interaction among conductive materials, microorganisms and their immediate living environment. In this study, Fenton iron mud with a high abundance of Fe(III) was recycled and applied in anaerobic reactors to promote anaerobic digestion (AD) process. The results show that the primary content of extracellular polymeric substances (EPS) such as polysaccharides and proteins increased significantly, possibly promoting microbial aggregation. Furthermore, with the increment of redox mediators including humic substances in EPS and Fe(III) introduced by Fenton iron mud, the direct interspecies electron transfer (DIET) between methanogens and interacting bacteria could be accelerated, which enhanced the rate of methanogenesis in anaerobic digestion (35.21 ± 4.53% increase compared to the control). The further analysis of the anaerobic microbial community confirmed the fact that Fenton iron mud enriched functional microorganisms, such as the abundance of CO 2 -reducing (e.g. Chloroflexi) and Fe(III)-reducing bacteria (e.g., Tepidimicrobium ), thereby expediting the electron transfer reaction in the AD process via microbial DIET and dissimilatory iron reduction (DIR). This work will make it possible for using the recycled hazardous material – Fenton iron mud to improve the performance of anaerobic granular sludge during methanogenesis.

Abstract:Although the promotive effect of direct interspecies electron transfer (DIET) on methane production has been well-documented, the practical applicability of DIET in different scenarios have not yet been systematically studied. This study compared the effects of magnetite-mediated DIET with conventional biogas mixing-driven interspecies hydrogen transfer (IHT) on anaerobic digestion (AD) of swine manure (SM). Compared with control, magnetite supplementation, biogas circulation, and their integration enhanced the CH 4 yield by 19.3%, 25.9%, and 26.2%, respectively. Magnetite mainly enriched DIET-related syntrophic bacteria ( Anaerolineae and Synergistia ) and methanogens ( Methanosarcina ) to accelerate acidification and establish DIET, while biogas circulation mainly enriched hydrolytic bacteria ( Clostridia ) and hydrogenotrophic methanogens ( Methanolinea and Methanobacterium ) to promote hydrolysis and accelerate IHT. Coupling magnetite addition with biogas circulation led to the enrichment of the above six microorganisms to different extents. The effectiveness of the strategies for lowering the H 2 pressure followed: magnetite + biogas circulation ≈ biogas circulation > magnetite. Under stress-free environment, the enhancement effect of magnetite-induced DIET was not even as pronounced as biogas circulation—a simple and common mixing strategy in commercial AD plants, and the promotion effect of magnetite was insignificant in the well-mixed digesters. In short, the magnetite-mediated DIET is not always effective in improving AD of SM.

Abstract:In this study, a batch of acid-tolerant inoculation sludge was domesticated as inoculum, and different concentrations of nano-magnetite were added to the propionic acid (PA) anaerobic digestion system to explore its effect on the PA degradation. It was found the PA degradation rates of nano-magnetite groups were 10.96-74.62% higher than the control group without magnetite in the 6th day, especially with the dosage of 100 mg/L. Microbial community analysis showed some potential DIET participants of Chloroflexi, Proteobacteria and Bacteroidetes had the highest abundance, while metabolite analysis showed that nano-magnetite promoted the transformation of malic acid to oxaloacetic acid and the PA disproportionation to butyric acid. It indicated that nano-magnetite could effectively promote the early degradation of PA in the sludge acclimation system by accelerating DIET or PA disproportionation, so as to prevent the PA accumulation, which could provide reference for the acid accumulation regulation of anaerobic digestion system.

Abstract:Magnetite has been proved to facilitate direct interspecies electron transfer (DIET)-based syntrophys and might alleviate inhibitory effects of antibiotics in anaerobic digestion (AD), while feeding ethanol was an effective approach to enrich the DIET partners. However, most of the existing studies were conducted at fixed ethanol concentration, few attentions were paid on the effects of differential ethanol proportion on AD, the underlying roles and mechanisms of ethanol stimulation remains unclear. This study systematically investigated the impact of ethanol stimulation on anaerobic processes treating oxytetracycline (OTC)-contaminated wastewater at varying proportions (20%, 50%, and 80%, based on equivalent COD value). In the presence of magnetite, ethanol stimulation promoted the methane production from 244.9 mL/g COD to a maximum 434.2 mL/g COD, with the most pronounced enhancement observed at high ethanol proportions. In particular, the average methane production obtained at 50% and 80% ethanol was 328.5 and 297.7 mL/g COD, respectively, whereas the enhancement of 20% ethanol stimulation was relatively limited. Concurrently, more stable COD removal and OTC reduction was noted in the existence of both magnetite and high ethanol proportions. Microbial analysis revealed the pivotal roles of Methanosaeta , alongside the predominance of Methanobacterium , in regulating COD conversion and driving methanogenesis through the CO 2 reduction pathway. Notably, high ethanol proportions fostered the enrichment of exoelectrogens (Geobacter , Desulfovibrio) in the magnetite-amended system, accompanied by the up-regulation of genes involved in organic metabolism pathways. Further investigation of functional genes highlighted the prevalence of pilA enrichment in the magnetite-amended system at low ethanol proportions, whereas omcS became more abundant at high ethanol proportions.

Abstract:Anaerobic granular sludge (AGS) is a promising technology for organic wastewater treatment and energy recovery. In this study, three different kinds of Fe and Fe oxides nanoparticles (Fe3O4, Fe2O3 and ZVI) were tried to be incorporated into AGS through direct loading or aided with biofilm disassembly agents of norspermidine and D-tyrosine, which was aimed to enhance methane production capacity of AGS via increasing redox activity of extracellular polymer substance (EPS) and interspecies electron transfer. Despite the loading methods, incorporation of Fe and Fe oxides nanoparticles into AGS increased methane production capacity remarkably, with an enhancement of 36.49-85.17%, 20.37-204.95% and 189.71-243.32%, respectively, for the Fe3O4, Fe2O3 and ZVI loaded AGS. Pretreatment of AGS using biofilm disassembly agents helped to incorporate more Fe and Fe oxides into the inner structure of AGS, which further enhanced methane production capacity by 48.68% and 184.58%, respectively, for the Fe3O4 and Fe2O3 loaded AGS. Loading Fe and Fe oxides into AGS not only introduced exogenous conductive substances and Fe(III)/Fe(II) redox couples into EPS matrix of AGS, but also stimulated the production of redox active components of flavins and c-Cyts. All these factors may contribute to the reduced resistance of EPS, enhanced interspecies electron transfer and methane production capacity of AGS. This study provides a novel strategy and facile method to accelerate interspecies electron transfer and enhance methane production for matured AGS.

Abstract:Nitrite-dependent anaerobic methane oxidation (n-DAMO) is a novel denitrification process that simultaneously further removes and utilizes methane from anaerobic effluent from wastewater treatment plants. However, the metabolic activity of n-DAMO bacteria is relative low for practical application. In this study, conductive magnetite was added into lab-scale sequencing batch reactor inoculated with n-DAMO bacteria to study the influence on n-DAMO process. With magnetite amendment, the nitrogen removal rate could reach 34.9 mg N·L −1 d −1 , nearly 2.5 times more than that of control group. Magnetite significantly facilitated the interspecies electron transfer and built electrically connected community with high capacitance. Enzymatic activities of electron transport chain were significantly elevated. Functional gene expression and enzyme activities associated with nitrogen and methane metabolism had been highly up-regulated. These results not only propose a useful strategy in n-DAMO application but also provide insights into the stimulating mechanism of magnetite in n-DAMO process.
