Abstract:An effective method for the synthesis of fully substituted 5-sulfonamidoimidazoles by Rh(II)-catalyzed transannulation of 1,2,4-oxadiazole derivatives with N-sulfonyl-1,2,3-triazoles is reported. The reaction works well with both aromatic 1,2,4-oxadiazoles and 1,2,4-oxadiazol-5-ones providing a flexible approach to N-(alkoxy/amino)carbonyl- and N-alkyl-substituted imidazoles. Both the disclosed reactions are completely regioselective and provide the first examples of a carbenoid-mediated transformation of N,N,O-heterocycles.

Abstract:Novel cyclic fluorinated sulfoximines featuring an E-monofluoroalkenyl benzoisothiazole 1-oxide moiety as single diastereomers can be facilely accessed by Rh(III)-catalyzed redox-neutral [4+1] annulation of sulfoximines with α,α-difluoromethylene alkynes. The reaction proceeds with sequential selective cleavage of both a C-H and a C-F bond, thus endowing with high step and atom economy. Through a combined experimental and computational mechanistic study, the origin of annulative chemoselectivity, unconventional E-selectivity, and excellent diastereoselectivity have been revealed accordingly.

Abstract:A facile access to highly fused tetracyclic indeno-1,2-benzothiazines has been established via a Rh(III)-catalyzed C-H bond activation and intramolecular annulation cascade between sulfoximides and all-carbon diazo indandiones. This strategy is characterized by the fact that the diazo coupling partners do not require preactivation, along with its high efficiency, broad substrate generality, and facile transformation. Particularly, the highly conjugated tetracyclic products demonstrate good optical properties and can easily enter cells to emit bright fluorescence for live cell imaging.

Abstract:The present study provided an efficient synthesis pathway to the azepino[5,4,3‐cd]indoles using N‐sulfonyl‐1,2,3‐triazoles and 4‐vinylindoles through a cascade carbene insertion and intramolecular aza‐Michael addition. This method is distinguished by mild reaction conditions, straightforward operational steps, accessible starting materials, and broad substrate compatibility. Furthermore, the versatility of this reaction with various Michael acceptors significantly broadens the scope of potential substrates. The ability to further transform the resulting multifunctional products underscores the promising applications of this approach in indole nitrogen heterocyclic synthesis.

Abstract:A rhodium-catalyzed denitrogenative formal (3 + 3) transannulation of 1,2,3-thiadiazoles with alk-2-enals is achieved, producing 2,3-dihydrothiopyran-4-ones in moderate to excellent yields. An inverse KIE of 0.49 is obtained, suggesting the reversibility of the oxidative addition of thioacyl Rh(i) carbenes to alk-2-enals. The late-stage structural modifications of steroid compounds are realized. Moreover, our studies show that thioacyl carbenes have different reactivities to those of α-oxo and α-imino carbenes, and highlight the importance of heteroatoms in deciding the reactivities of heterovinyl carbenes.

Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:The Rh(II)-catalyzed formal [3+2] and [3+3] cycloadditions of 1-tosyl 1,2,3-triazoles with 2H-azirines have been developed, which enable the efficient synthesis of polysubstituted 3-aminopyrroles and 1,2-dihydropyrazines, respectively. Abstract:An efficient formation of fused tricyclic [1,3]oxazino[3,4‐a]indol‐1‐ones and dihydropyrimido[1,6‐a]indol‐1(2H)‐ones via Rh(III)‐catalyzed [3+3] or [4+2] annulation of N‐methoxy‐1H‐indole‐1‐carboxamides with sulfoxonium ylides has been developed. These selective annulation reactions were carried out by switching the additives and notable features of this protocol were low catalyst loading and a broad substrate scope providing the corresponding products in up to 99% yields.

Abstract:Described here is a selective synthesis of 2,3-dihydropyrrole-fused N-glycosides through rhodium-catalyzed denitrogenative transannulation of N-sulfonyl-1,2,3-triazoles with glycals. A series of pyrroline-fused N-glycosides are afforded in moderate to excellent yields with exclusive regioselectivity and stereoselectivity. Functional application of such a resultant product by oxidative addition and epoxidation is also explored. Abstract:Sulfur-stereogenic containing benzo-fused heterocycles have gained much attention in drug discovery. However, the asymmetric synthesis of these chiral molecules with structural diversity is very challenging. Herein, we report the synthesis of chiral benzothiadiazine-1-oxides with a seven-membered ring via achiral Ru(II)-catalyzed asymmetric [4 + 3] annulation of sulfoximines with α,β-unsaturated ketones assisted by chiral carboxylic acid (CCA). A broad range of chiral benzothiadiazepine-1-oxides bearing various functional groups could be prepared in up to 90% yield with up to >99% ee, expanding the chemical space of chiral sulfoximines. Notably, the oxidative cleavage of the double bonds in the products gave chiral N-benzoyl sulfoximines with a C–S chiral axis.
