Structure and Catalytic Activity of Hemoglobin Assembled with Layered Double Hydroxide Nanosheets by Coprecipitation Using a T-shaped Microreactor

Abstract:In this study, the direct assembling of hemoglobin (Hb) with layered double hydroxide nanosheets (Hb-LDHNS) has been achieved by using a T-shaped microreactor, and the structure and bioactivity of the immobilized protein have been investigated. The XRD data show that Hb is immobilized on LDHNS surface through ultrafine assembling and electrostatic interactions rather than interaclation. The SEM and TEM images display that the size of LDHNS in biohybrid are ranged from 10 to 40 nm with a thickness of less than 5 nm. The spectroscopic results exhibit that the immobilized Hb molecules strongly interact with LDHNS matrixes, however they still maintain the native structure in biohybrids. The biocatalytic performance of the obtained Hb-LDHNS is also evaluated by employing the oxidation of o-phenylenediamine. Results reveal that the reduction of bioactivity resulted from the ultrafine assembling is more than the decrease derived from the electrostatic adsorption at 25 degrees C. After heated to 90 degrees C, LDHNS demonstrate the superior protection role on bioactivity than large sized LDH plate. The immobilized Hb in LDHNS can retain their bioactivity up to 30.98-73.50%, far higher than that in LDH flakes obtained by traditional coprecipitation or its free form. The resulting biocomposites can be stored for several months at 4 degrees C with losing insignificant bioactivity. (C) 2016 Elsevier B.V. All rights reserved.
