Abstract:A high-throughput semi-automated procedure for simultaneously stability evaluation of multiple compounds in plasma using direct single column high-performance liquid chromatography (HPLC) combined with tandem mass spectrometry (MS/MS) was developed to eliminate the laborious procedures that are traditionally used for stability studies. Untreated human, monkey, mouse and rat plasma samples containing ten drug components were directly injected into a mixed-functional column that provided both protein removal and chromatographic functionality. Ten test compounds were simultaneously assayed using a tandem mass spectrometer in the positive ion mode using multiple reaction monitoring (MRM). Plasma samples containing ten test compounds were placed in a thermostatic autosampler and then sequentially monitored in one analytical procedure. The time between each injection was set about 7 minutes. The peak responses of the test compounds in individual plasma samples were repeatedly determined every 28 minutes. Drug stability in plasma was indicated by the change of the mass chromatographic peak areas for the test compounds and was observed to be a function of animal species, incubation time and incubation temperature. The potential for matrix ionization suppression on the direct single column HPLC-MS/MS system was also investigated using the post-column infusion technique. The proposed cassette assay procedure provides an analytical throughput ten times greater than the single component approach for the evaluation of drug stability in plasma without compromising data quality.

Abstract:A simple procedure for the measurement of stability of drug candidates in plasma was developed to eliminate the traditional labor-intensive and time-consuming sample preparation procedures that are typically used for these studies. The procedure makes use of a thermostatic autosampler as an incubator combined with the direct plasma injection method based on high-performance liquid chromatography (HPLC) coupled to tandem mass spectrometry (MS–MS). Untreated human, monkey, mouse and rat plasma containing the test compound was directly injected into a mixed-function column for on-line protein removal and chromatography. The test compound and its biotransformation product were separated via HPLC and monitored using the tandem mass spectrometer. The need for adequate chromatographic separation of the test compound (M) from its carboxylic acid metabolite (M+1) is demonstrated. Plasma samples from four different species at specified incubation temperatures were sequentially assayed in one analytical procedure. The injection-to-injection time was about 6 min. The peak responses of the test compound in individual plasma samples were repeatedly determined every 24 min. The retention times and peak shape of all analytes were found to be consistent throughout the experiments. The stability of the test compound in plasma was found to be a function of animal species, incubation time and temperature. The test compound was rapidly degraded in rat plasma at 37°C, but it could be stabilized by adding sodium thiosulfate.

Abstract:An ultrafast bioanalytical method using monolithic column high-performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS) was evaluated for the simultaneous determination of a drug discovery compound and its metabolite in plasma. Baseline separation of the two compounds was achieved with run times of 24 or 30 s under isocratic or gradient conditions, respectively. The monolithic column HPLC/MS/MS system offers shorter chromatographic run times by increasing flow rate without sacrificing separation power for the drug candidate and its biotransformation product (metabolite). In this work, the necessity for adequate chromatographic resolution was demonstrated because the quantitative determination of the drug-related metabolism product was otherwise hampered by interference from the dosed drug compound. The chromatographic performance of a monolithic silica rod column as a function of HPLC flow rates was investigated with a mixture of the drug component and its synthetic metabolite. The assay reliability of the monolithic column HPLC/MS/MS system was checked for matrix ionization suppression using the post-column infusion technique. The proposed methods were successfully applied to the analysis of study rat plasma samples for the simultaneous quantitation of both the dosed drug and its metabolite. The analytical results obtained by the proposed monolithic column methods and the 'standard' silica particle-packed HPLC column method were in good agreement, within 10% error. Abstract:A simple, easy to use and efficient method was described for simultaneous determination of ten cardiovascular drugs with a broad range of physicochemical properties in rat plasma via online solid phase extraction (online SPE) and high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Following a simple centrifugation step, a 10 mL aliquot of the plasma sample was injected directly onto the HPLC system. The LiChrospher (R) RP-18 ADS (25 mm x 4 mm, 25 mu m, Merck) cartridge was washed with 10 mM ammonium acetate buffer (pH 9.5) for 1 min, after which time the analytes were removed by back-flushing directly onto the analytical column (Acclaim 120 C18 column, 150 mm x 4.6 mm, 5 mu m) with gradient elution using acetonitrile-10 mM ammonium acetate buffer (pH 3.5) as mobile phase. The flow rate through both columns was 1 mL min(-1), and the analytes were quantified using a triple-quadrupole tandem mass spectrometer in multiple-reaction monitoring mode. Linear calibration curves were obtained over the range of 0.2-100 ng mL(-1), and the method limits ranged from 0.2 ng mL(-1) to 1 ng mL(-1) which is sensitive enough for clinical drug monitoring. The intra-and inter-day precisions were in the range of 0.20-2.32%, and the accuracies were between 93.33% and 114.60%. Excellent recoveries from plasma were achieved with a range from 83.52% to 107.38%. The procedure was easier to execute and required less sample handling than methods previously described in the literature. This easy to use and high-throughput method with direct injection of plasma samples for the analysis of multiple cardiovascular drugs may provide a practical solution for tailoring drug dosage in a rational manner to rapidly achieve optimal efficacy and safety of medication.

Abstract:A bioanalytical method has been developed and validated for quantitation of a beta-lactam drug candidate and its open-ring biotransformation product utilizing high-flow high-performance liquid chromatography (HPLC) for on-line purification of plasma samples and electrospray tandem mass spectrometry for detection and quantitation. The HPLC system used two columns: an Oasis column (1 x 50 mm, 30 microm) as the on-line extraction column and a conventional C18 column (2 x 50 mm, 5 microm) as the analytical column. Each plasma standard or quality control (QC) sample (50 microL) was mixed with 50 microL of a working solution of the internal standard in aqueous 0.5 M ammonium acetate (pH 4.0). Portions (10 microL) of these samples were then injected into an Oasis column with a mobile phase consisting of 100% aqueous 1 mM formic acid at a high flow rate (4.0 mL/min), with the effluent from the Oasis column directed to waste and not to the mass spectrometer. After the purification step, the Oasis column effluent was directed to the analytical column and the mass spectrometer and the analytes were eluted with methanol/aqueous 1 mM formic acid (70:30) at a flow rate of 1.0 mL/min. The total analysis time was 1.6 min per sample. The standard curve range was 0.980 to 250 ng/mL. The accuracy, inter-day precision and intra-day precision were within 10% for both compounds.
Direct stability characterization of aconite alkaloids in different media by autosampler-mediated incubation-online solid phase extraction-LC-MS/MS

Abstract:Because degradation might occur at any step of conventional sample preparation protocols, it is of great importance to carry out direct analysis for stability characterization of drug candidates in vitro. Herein, an automated system for directly monitoring the stability of drug candidates in different media was configured by hyphenating autosampler-mediated incubation, online solid phase extraction, and high performance liquid chromatography-tandem mass spectrometry (AMI-SPE-LC-MS/MS). The thermostatic autosampler served as an incubator, and an SPE column was employed to online capture analytes after direct injection of incubated matrices. To achieve sensitive detection, a predictive multiple reaction monitoring approach was introduced to monitor the degradation products. As a proof-of-concept, this system was applied for direct stability characterization of six aconite alkaloids, including aconitine (AC), mesaconitine (MA), hypaconitine (HA), benzoylmesaconine (BMA), benzoylaconine (BAC), and benzoylhypaconine (BHA), which are the primary effective as well as toxic components in aconite-derived herbal medicines. Extensive hydrolysis to generate two degradation products was observed for AC, MA, or HA in phosphate buffer saline (PBS, pH 7.4), PBS fortified with fresh rat plasma and PBS spiked denatured rat plasma, whereas BMA, BAC, and BHA remained intact in PBS, PBS spiked rat plasma, methanol, acetonitrile (ACN), and 50% aqueous ACN. Therefore, care should be taken to maintain the stability of aconite alkaloids, in particular in alkaline media, during metabolic and pharmacokinetic evaluations. More importantly, the developed AMI-SPE-LC-MS/MS system provides a reliable tool for directly and veritably monitoring the in vitro stability of drug candidates.

Abstract:Shuang-Huang-Lian powder injection (SHLPI) is a natural drug injection made of honeysuckle, scutellaria baicalensis and forsythia suspensa. It has the characteristics of complex chemical composition and difficult metabolism research in vivo. LC-MS platform has been proven to be an important analytical technology in plasma metabolomics. Unfortunately, the lack of an effective sample preparation strategy before analysis often significantly impacts experimental results. In this work, twenty-one extraction protocols including eight protein precipitation (PPT), eight liquid-liquid extractions (LLE), four solid-phase extractions (SPE), and one ultrafiltration (U) were simultaneously evaluated using plasma metabolism of SHLPI in vivo. In addition, a strategy of “feature ion extraction of the multi-component metabolic platform of traditional Chinese medicine” (FMM strategy) was proposed for the in-depth characterization of metabolites after intravenous injection of SHLPI in rats. The results showed that the LLE-3 protocol (Pentanol:Tetrahydrofuran:H2O, 1:4:35, v:v:v) was the most effective strategy in the in vivo metabolic detection of SHLPI. Furthermore, we used the FMM strategy to elaborate the in vivo metabolic pathways of six representative substances in SHLPI components. This research was completed by ion migration quadrupole time of flight mass spectrometer combined with ultra high performance liquid chromatography (UPLC/VionTM-IMS-QTof-MS) and UNIFITM metabolic platform. The results showed that 114 metabolites were identified or preliminarily identified in rat plasma. This work provides relevant data and information for further research on the pharmacodynamic substances and in vivo mechanisms of SHLPI. Meanwhile, it also proves that LLE-3 and FMM strategies could achieve the in-depth characterization of complex natural drug metabolites related to Shuang-Huang-Lian in vivo.

High-speed gradient parallel liquid chromatography/tandem mass spectrometry with fully automated sample preparation for bioanalysis: 30 seconds per sample from plasma

Abstract:In this work, a high-throughput and high-performance bioanalytical system is described that is capable of extracting and analyzing 1152 plasma samples within 10 hours. A Zymark track robot system interfaced with a Tecan Genesis liquid handler was used for simultaneous solid-phase extraction of four 96-well plates in a fully automated fashion. The extracted plasma samples were injected onto four parallel monolithic columns for separation via a four-injector autosampler. The use of monolithic columns allowed for fast and well-resolved separations at a considerably higher flow rate without generating significant column backpressure. This resulted in a total chromatographic run cycle time of 2 min on each 4.6 x 100 mm column using gradient elution. The effluent from the four columns was directed to a triple quadrupole mass spectrometer equipped with an indexed four-probe electrospray ionization source (Micromass MUX interface). Hence, sample extraction, separation, and detection were all performed in a four-channel parallel format that resulted in an overall throughput of about 30 s per sample from plasma. The performance of this system was evaluated by extracting and by analyzing twelve 96-well plates (1152) of human plasma samples spiked with oxazepam at different concentrations. The relative standard deviation (RSD) of analyte sensitivity (slope of calibration curve) across the four channels and across the 12 plates was 5.2 and 6.8%, respectively. An average extraction recovery of 77.6% with a RSD of 7.7% and an average matrix effect of 0.95 with a RSD of 5.2% were achieved using these generic extraction and separation conditions. The good separation efficiency provided by this system allowed for rapid method development of an assay quantifying the drug candidate and its close structural analog metabolite. The method was cross-validated with a conventional liquid chromatography/tandem mass spectrometry (LC/MS/MS) assay.
Stability-indicating high-performance thin-layer chromatography method development and validation for dapagliflozin propanediol monohydrate, vildagliptin, and metformin hydrochloride in active pharmaceutical ingredient and its tablet dosage form

Abstract:A simple, precise, and accurate high-performance thin-layer chromatography (HPTLC) method was developed and authenticated to perform simultaneous estimation of dapagliflozin propanediol monohydrate (DAPA), vildagliptin (VILDA), and metformin (MET) in active pharmaceutical ingredient (API) and tablet dosage form. It was found that there is not any reported stability-indicating HPTLC method for the simultaneous determination of DAPA, VILDA and MET; hence, this method was developed. Merck HPTLC silica gel 60 F 254 plates were selected as the stationary phase. Acetonitrile, formic acid, and water (9:1:0.5, V/V ) were the chromatographic solvents used. A densitometric study was conducted at the wavelength of 215 nm. For DAPA, VILDA, and MET, the concentrations used to conduct the study were 0.2‒0.6 μg/band, 2‒6 μg/band, and 10‒30 μg/band, respectively. The peaks of DAPA, VILDA, and MET have retardation factors ( R F ) of 0.70, 0.45, and 0.34, respectively. The regression coefficients ( R 2 ) of DAPA, VILDA, and MET were 0.9955, 0.9958, and 0.9972, respectively. The limit of detection values of DAPA, VILDA, and MET were 0.0018 μg/band, 0.0094 μg/band, and 0.1434 μg/band. The limit of quantification of DAPA, VILDA, and MET were 0.0056 μg/band, 0.0285 μg/band, and 0.4346 μg/band, respectively. With respect to International Council for Harmonisation (ICH) prerequisites, the accuracy, precision, and robustness outcomes were within suitable limits. The outcomes of stability data revealed that the degradation by acid, base, hydrogen peroxide, and water produced distinct peaks of DAPA, VILDA, and MET, along with a few other peaks, having different R F values. The forced degradation study for DAPA, VILDA, and MET was performed. DAPA was found to be stable in acidic, oxidative, and neutral conditions. Under acidic, basic, and oxidative conditions, VILDA was observed to be stable. The proposed technique passes all the ICH criteria; hence, it is an accurate, easy, suitable, and stable method for the analysis of DAPA, VILDA, and MET in combined dosage form.

An Integrated Hepatocyte Stability Assay for Simultaneous Metabolic Stability Assessment and Metabolite Profiling

Abstract:The determination of metabolic stability is critical for drug discovery programs, allowing for the optimization of chemical entities and compound prioritization. As such, it is common to perform high-volume in vitro metabolic stability experiments early in the lead optimization process to understand metabolic liabilities. Additional metabolite identification experiments are subsequently performed for a more comprehensive understanding of the metabolic clearance routes to aid medicinal chemists in the structural design of compounds. Collectively, these experiments require extensive sample preparation and a substantial amount of time and resources. To overcome the challenges, a high-throughput integrated assay for simultaneous hepatocyte metabolic stability assessment and metabolite profiling was developed. This assay platform consists of four parts: 1) an automated liquid-handling system for sample preparation and incubation, 2) a liquid chromatography and high-resolution mass spectrometry-based system to simultaneously monitor the parent compound depletion and metabolite formation, 3) an automated data analysis and report system for hepatic clearance assessment; and 4) streamlined autobatch processing for software-based metabolite profiling. The assay platform was evaluated using eight control compounds with various metabolic rates and biotransformation routes in hepatocytes across three species. Multiple sample preparation and data analysis steps were evaluated and validated for accuracy, repeatability, and metabolite coverage. The combined utility of an automated liquid-handling instrument, a high-resolution mass spectrometer, and multiple streamlined data processing software improves the process of these highly demanding screening assays and allows for simultaneous determination of metabolic stability and metabolite profiles for more efficient lead optimization during early drug discovery. SIGNIFICANCE STATEMENT: Metabolic stability assessment and metabolite profiling are pivotal in drug discovery to fully comprehend metabolic liabilities for chemical entity optimization and lead selection. Process of these assays can be repetitive and resource demanding. Here, we developed an integrated hepatocyte stability assay that combines automation, high-resolution mass spectrometers, and batch-processing software to improve and combine the workflow of these assays. The integrated approach allows simultaneous metabolic stability assessment and metabolite profiling, significantly accelerating screening and lead optimization in a resource-effective manner.

Abstract:A high-throughput method for rapid screening of active ingredients in drugs has been developed with mass spectrometry coupled to a low-temperature plasma (LTP) probe ion source. Without sample preparation or pretreatment, the active ingredients of 11 types of commercial pharmaceuticals, including hormones, antipyretic analgesics, cardiovascular, digestant, neuro-psychotherapeutic, diuretic, antithyroid, sulfa anti-inflammatory, antiparastic, sedative-hypnotics, and antibacterial, were directly desorbed/ionized and detected by a linear ion trap mass spectrometry (MS). The structures of these ingredients were elucidated by tandem MS. The analysis of 18 methyltestosterone tablets could be accomplished within 1.9 min, which allows fast detection with a speed of approximate 600 samples within 1 h. This work demonstrated that LTP probe ion source combined with MS is a high-throughput method for screening of pharmaceuticals and potentially applied to on-line quality control in pharmaceutical industry. Abstract:Direct mass spectrometry (MS) analysis of biofluids with simple procedures represents a key step in the translation of MS techniques to clinical and point-of-care applications. The current study reports the development of a single-step method using slug-flow microextraction and nano-electrospray ionization for MS analysis of organic compounds in blood and urine. High sensitivity and quantitation precision have been achieved in the analysis of therapeutic and illicit drugs in 5 mL samples. Real-time chemical derivatization has been incorporated for analyzing anabolic steroids. The monitoring of enzymatic functions has also been demonstrated with cholinesterase in wet blood. The reported study encourages the future development of disposable cartridges, which function with simple operation to replace the traditional complex laboratory procedures for MS analysis of biological samples.

Abstract:Highly polar metabolites are abundant in human plasma. Analysis of these compounds by standard reverse-phase chromatography is difficult. Highly polar metabolites were extracted by a two-step method. They were analyzed by ultra-performance hydrophilic interaction chromatography (HILIC) coupled to quadrupole-time of flight mass spectrometer detection technology. The middle and low polar fraction were also analyzed using a standard C18 column. Data were extracted by Markerlynx, and analyzed by multivariate statistical methods to discriminate human plasma samples by sex and age. The results not only showed good classification of samples by sex and age, but also revealed new potential biomarkers not revealed by standard RPLC/MS analysis. This study showed that HILIC is an effective technique to analyze highly polar metabolites for metabonomic studies.

Abstract:Antimicrobial resistance (AMR) is a major threat to global health due to the wide use of antibacterial drugs. Multiple studies show that the pharmacokinetic/pharmacodynamic (PK/PD) studies of antibiotics are an approach to prevent/delay AMR. The pharmacokinetic parameters of antibiotics are the basis of PK/PD studies, and therapeutic drug monitoring (TDM) is the key method to obtain pharmacokinetic information. We developed an ultra-performance liquid chromatography–tandem mass spectrometry to determine 18 antibacterial drugs (piperacillin, cefazolin, cefuroxime, cefoperazone, ceftriaxone, cefepime, aztreonam, meropenem, imipenem, levofloxacin, moxifloxacin, azithromycin, clindamycin, tigecycline, linezolid, vancomycin, voriconazole and caspofungin) in human plasma for practical clinical usage. Samples were prepared using protein precipitation with methanol. Chromatographic separation was accomplished in 6 min on a BEH C 18 column (2.1 × 100 mm, 1.7 μm) using a gradient elution of acetonitrile and 0.1% formic acid in water at a flow rate of 0.3 ml/min. The electrospray ionization source interface was operated in the positive and negative ionization modes. Inter- and intra-day precision, accuracy, recovery, matrix effect, and stability were validated according to the Food and Drug Administration guidance. The correlation coefficients of calibration curves were all greater than 0.99. The accuracies of the 18 antibacterial drugs ranged from 89.1% to 112.4%. The intra-day precision of the analytes ranged from 1.4% to 9.3% and the inter-day precision from 2.1% to 7.2%. The matrix effects ranged from 93.1% to 105.8% and the extraction recoveries ranged between 90.1% and 109.2%. The stabilities of the 18 antibacterial drugs in plasma were evaluated by analyzing three different concentrations following storage at three storage conditions. All samples displayed variations less than 15.0%. The validated method was successfully applied to routine clinical TDM for 231 samples.

Simultaneous Determination of Contents of Two 10-Hydroxycamptothecins in Human Plasma by HPLC Method

Abstract:Objective:To establish a HPLC method for simultaneous determination of the lactone form(L-HCPT) and carboxylate form of 10-hydroxycamptothecin(C-HCPT) in human plasma and determine the stability of HCPT.Methods:The HPLC-fluorometric method was used with the excitation wavelength at 363 nm and the emission wavelength at 530 nm.The samples were injected into the Lichrospher C18 column(250 mm×4.6 mm,5 μm) with acetonitrile-0.075 mol/L ammonium acetate buffer solution(32∶68,pH 6.4)as the mobile phase at the flow rate of 1.0 mL/min.HCPT content was quantified by the internal standard method.Results:L-HCPT and C-HCPT could be separated completely by this method.The detection limit was 5 ng/mL.The linearity ranges of L-HCPT and C-HCPT were 16.99-2 175.00 ng/mL and 14.94-1 912.50 ng/mL,respectively.The relative recovery rates of CHCPT and L-HCPT were 96.79%-119.64% and 100.23%-112.93%,respectively.The intra-and inter-day precisions were both less than 10%.Ninty-three point thirty-four percentage to 97.18% of C-HCPT transformed to L-HCPT after acidified by 1 mol/L hydrochloric acid.The HCPT in plasma was stable at-20 ℃ for 28 days.Conclusion:This method is specific,sensitive and reliable for the determination of HCPT in human plasma and pharmacokinetic study.
Determination Of Diethylstilbestrol In Human Plasma With Measurement Uncertainty Estimation By Liquid Chromatography-Tandem Mass Spectrometry

Abstract:A rapid and sensitive liquid chromatography/tandem mass spectrometry (LC-MS/MS) using an electrospray ionization (ESI) method was developed and validated to determine the concentration of diethylstilbestrol in human plasma. Samples were extracted with methyl tert-butyl ether after alkalified by sodium tetraborate solution, using 3,5-dihydroxy-4-isopropyl toluylene as the internal standard (IS). Separation was performed on a Restek C-18 column (2.1mmx150mm, 5.0 mu m) guarded by a Hypersil Gold pre-column (2.1mmx10mm, 3 mu m). The mobile phase consisted of acetonitrile-water (85:15, containing 0.2mmol/L ammonium acetate) at a flow rate of 0.3mL/min. Detection was performed using negative multiple reaction monitoring (MRM) mode. The mass transitions monitored were [M-H](-)m/z267.2237.2 and [M-H](-)m/z253.2211.0 for diethylstilbestrol and IS, respectively. The method was fully validated using total error theory, which is based on -expectation tolerance intervals and include trueness and intermediate precision. The method was found to be accurate over a concentration range of 0.1-50ng/mL. The main advantages of this method were the high sensitivity (a lower limit of quantification, LLOQ, of 0.1ng/mL) and the short analysis time (total run time 5min per sample). The measurement uncertainty based on -expectation tolerance intervals was assessed at each concentration level of the validation standards. The method is specific, sensitive, and accuracy, and it is suitable for plasma concentration determination of diethylstilbestrol.
Development and Validation of the Stability indicating Assay Methodology Employing LC-MS/MS for Concurrent Quantification of Dapagliflozin Propanediol Monohydrate and Metformin Hydrochloride: Probable Degradants based on Mass Spectra

Abstract:For the concurrent measurement of dapagliflozin propanediol monohydrate (DAPA) and metformin hydrochloride (MET) in combined dosage form, a quick, accurate, specific and easy liquid chromatography tandem mass spectrometry (LC-MS/MS) approach was created. The method was performed on a column C8 RRHD Eclipse (150 × 4.60 mm, 5 μm). In 5 mM ammonium acetate buffer pH-4.0, methanol and acetonitrile were the components of the mobile phase in the ratios of 30:65:05, respectively. The effluent was detected at 227 nm at a 0.4 mL/min flow rate. The observed retention times for DAPA and MET were 7.297 and 3.230 minutes, respectively. The drug was stressed by being exposed to acid and alkali hydrolysis. From the mass spectra, it was found that two degradant peaks were observed in the standard mixture and the sample during alkali stress condition and probable degradants formed. The developed approach was validated in accordance with ICH guidelines. With correlation coefficients of 0.9969 for DAPA and 0.9975 for MET, it was discovered that the standard curve was linear over the range of 60 to 140 and 300 to 700 μg/mL for DAPA and MET, respectively. The limit of detection (LoD) was 2.959 and 8.893 μg/mL for DAPA and MET, respectively. The limit of quantification (LoQ) was 8.967 and 26.949 μg/mL for DAPA and MET, respectively. The %recovery was determined in between 98 to 102%. The precision was within the limit (Relative Standard Deviation (RSD) <2%). The proposed stability indicates that LC-MS/MS method can be successfully utilized for simultaneous estimation of DAPA and MET in combined dosage form without any prior separation of individual drugs and no interference was found due to degradant formed during stress condition.

Abstract:Abstract A stability-indicating hybrid micelle liquid chromatography accompanied by UV detection was developed for the simultaneous analysis of either paracetamol (PCA) or pseudoephedrine hydrochloride (PSU) with their synthetic impurities. Mixture I contains PCA with p -amino phenol and p -nitro phenol, while mixture II involves the estimation of PSU with benzaldehyde and benzoic acid. Both mixtures were separated using a C18 column that was thermostatically maintained at 40°C and operating under a flow rate of 1.5 mL/min, applying UV detection at 240 nm for mixture I and 220 nm for mixture II. In both cases, the mobile phase consisted of 0.1 M sodium dodecyl sulfate, acetonitrile, and triethylamine (90:10:0.3, v/v/v) and adjusted to pH 4 (mixture I) or pH 3.7 (mixture II) using 2.0 M O -phosphoric acid. The proposed method was validated and successfully applied to assay different pharmaceuticals containing PCA or PSU. Moreover, the stability-indicating nature of the proposed method was proved through applying photolytic degradation procedures for PCA.

Abstract:The pharmaceutical industry is rapidly advancing toward new drug modalities, necessitating the development of advanced analytical strategies for effective, meaningful, and reliable assays. Hydrophilic Interaction Chromatography (HILIC) is a powerful technique for the analysis of polar analytes. Despite being a well-established technique, HILIC method development can be laborious owing to the multiple factors that affect the separation mechanism, such as the selection of stationary phase chemistry, mobile phase eluents, and optimization of column equilibration time. Herein, we introduce a new automated multicolumn and multi-eluent screening workflow that streamlines the development of new HILIC assays, circumventing the existing tedious ‘hit-or-miss’ approach. A total of 12 complementary columns packed with sub-2 µm fully porous and 2.7 µm superficially porous particles operated on readily available ultra-high pressure liquid chromatography (UHPLC) instrumentation across a diverse set of commercially available polar stationary phases were investigated. Different mobile phases with pH ranging from pH 3 to 9 were evaluated using different organic modifier. The gradient and column re-equilibration were judiciously set to ensure a reliable assay screening framework yielding straightforward separation conditions for subsequent optimization and method deployment in fast-paced laboratory settings. This UHPLC screening system is coupled with a diode array and charged aerosol detectors (DAD and CAD) to ensure versatile detection for a variety of compounds. This fast-screening platform lays the foundation for a convenient generic workflow, accelerating the pace of HILIC method development and transfer across both academic and industrial sectors.

Abstract:In mass spectrometry (MS), nonvolatile salts contaminate the transmission system and cause ion suppression, hampering MS analysis. When MS is combined with liquid chromatography (LC) that uses a salty mobile phase, the problems become more intractable due to long analysis time. Here, a novel heat-assisted dual neutral spray ionization (HADSI) method was developed, which projected sample solution spray and solvent spray onto a heated plate to achieve online desalting and high ionization. The experimental parameters of HADSI were optimized, which indicated that the plate temperature was crucial for ionization and desalination. Eight drug compounds dissolved in various commonly used buffers were directly analyzed using HADSI-MS, even though the concentration of PBS buffer reached 500 mmol/L. The established method showed considerable sensitivity in the positive ion mode with the limits of detection at the level of nmol/L, and good linearity ( > 0.99) was achieved for all the analyzed compounds. The repeatability and intra- and interday precisions of the method were evaluated, demonstrating the feasibility and reliability of the analysis of salty samples by HADSI-MS. Further, the method was demonstrated to tolerate the long-time analysis of high-salt LC eluates and the device was easy to maintain. Finally, a crude roxithromycin product was separated by LC and then analyzed by HADSI-MS, and seven unknown impurities and nine known impurities were successfully detected. Our results indicated that HADSI-MS may have potential applications in academic and industrial fields.
