Abstract:Advanced implants with simultaneous accelerated osteogenic and angiogenic capacities are of great importance for osteointegration. Much attention has been paid to simultaneously enhancing the osteogenesis and angiogenesis by surface decoration of bioactive molecules or ions on biomaterial surface, but the inherent physical cue of material surface down to the atomic-scale features have always been ignored. In this study, we demonstrate that regulation of surface oxygen vacancies defects of rutile nanorods are able to simultaneous accelerate the osteogenesis and angiogenesis. The concentration of surface oxygen vacancies defects of rutile nanorods can be manipulated by simple redox processing. The osteogenic differentiation of mesenchymal stem cells (MSCs), angiogenic differentiation and vessel-like tube structures of human umbilical vein endothelial cells (HUVECs) on oxygen vacancies rich surface are significantly up-regulated. This work therefore emphasizes the critical role of the inherent material atomic-scale features and provides a novel strategy to accelerate the osteogenesis and angiogenesis of Ti-based implant.
Osteogenesis of Rat Mesenchymal Stem Cells and Osteoblastic Cells on Strontium-Doped Nanohydroxyapatite-Coated Titanium Surfaces.

Abstract:PURPOSE:The aim of this study was to compare the promotion of osteogenesis in vitro on three types of titanium surfaces: a strontium-hydroxyapatite (Sr-HA)-coated surface, a nano-HA-coated surface, and an uncoated roughened surface.MATERIALS AND METHODS:Sr-HA and HA were placed on disks with a roughened titanium surface by electrochemical deposition. MC3T3-E1 preosteoblast cells and rat bone mesenchymal stem cells were cultured on the Sr-HA, HA-coated, and uncoated roughened disks, and cell adhesion, proliferation, viability, osteogenic differentiation, and formation of mineralized nodules were measured at various time points.RESULTS:The Sr-HA coating produced by a simple electrochemical deposition treatment evidently enhanced the attachment, spreading, alkaline phosphatase activity, and extracellular matrix calcium mineralization of mouse bone mesenchymal stem cells and MC3T3-E1 cells compared with an untreated roughened titanium surface and a nano-HA-coated surface.CONCLUSION:This study suggests that a Sr-doped nano-HA coating produced through electrochemical deposition improves the osteoconductivity of a microrough titanium surface.
The Characterization and Osteogenic Activity of Nanostructured Strontium-Containing Oxide Layers on Titanium Surfaces

Abstract:Purpose: The aim of this study was to compare strontium-containing implant (Sr-Ti) surfaces with modified pure Ti (Mod Ti) on the promotion of osteogenesis in vitro and in vivo. Materials and Methods: The Sr-Ti surfaces were fabricated by hydrothermal treatment. The Mod Ti surfaces were used as a control. These surfaces were characterized with scanning electron microscopy, x-ray diffractometer, and x-ray photoelectron spectroscopy. The biologic activity was analyzed by initial adhesion, proliferation, alkaline phosphatase (ALP), osteocalcin, and osteogenesis-related genes in vitro. Hematoxylin and eosin (H&E) staining and immunohistochemistry analysis of tissue adjacent to this implant were used to evaluate osteogenic activity in vivo. Results: The results confirmed multiple nanogranules on the Sr-Ti surface composed of well-crystallized TiO2 and SrTiO3 phases. Long-term release of Sr enhanced mesenchymal stem cell (MSC) initial adhesion, the production of ALP, osteocalcin, and upregulated osteogenesis-related genes. For in vivo osteogenic evaluation, mesenchymal-like cells were adhered on these two surfaces under the electron microscope and transitory hematoma in adjacent bone tissue by H&E staining. The Stro-1-labeled MSCs were noted around the Sr-Ti surface in immunohistochemistry analysis. Conclusion: This study suggests that Sr-Ti surfaces with nanostructures could promote the response of MSCs in vitro, and recruit Stro-1 labeled MSCs in vivo.
Influence of Integration of TiO2 Nanorods into Its Nanodot Films on Pre-Osteoblast Cell Responses

Abstract:Design of an implant surface with appropriate nanostructures has been considered to be an effective way to promote osteoblast cell growth and osseointegration. In this work, TiO2 nanorods were integrated into TiO2 nanodot films by hydrothermal growth to engineer a nanostructured surface composed of two nano-structural elements. The surface microstructure of the films depended on the integration degree. Low integration led to a surface nanostructure with exposure of the two nano-structural elements, and high integration resulted in a similar surface to a nanorod film. The nanorod integration had an obvious influence on pre-osteoblast cell responses and accelerated the cellular osteogenesis. Two factors of the integrated nano-structural element and the changed surface topology are believed to be responsible for enhancing cellular responses. The former is considered to play a more significant role because the nanorods tended to adsorb human fibronectin preferentially so as to begin differentiation to osteogenesis earlier. We anticipate this work may assist to gain insight into understanding of the interactions between nanostructured surfaces with cells.
Effect of the Nanostructure of Porous Alumina on Growth Behavior of MG63 Osteoblast-Like Cells.

Abstract:It is well known that cellular responses to materials, in terms of adhesion, migration and proliferation, are highly affected by the surface characteristics of the materials. The investigation of the effect of material surface topography on cell behaviors is of great importance for the development of implanted biomaterials in tissue engineering. Alumina is one of the most popular implant materials used in orthopedics, but few data are available concerning the potential cellular responses of MG63 to nanoporous alumina. The present study investigated the size effect of nanoporous alumina substrates on MG63 cell behaviors in terms of cell viability, expression of integrin β1, alkaline phosphatase (ALP) activity and changes of cell morphology, respectively. Cell viability was measured by means of MTT assay and integrin β1 expression was detected by immunofluorescence staining and real-time PCR. Scanning electron microscopy (SEM) was used to observe cell morphology. Cell function was evaluated by detecting the ALP activity and mineralization. Results showed that cell viability and expression of integrin β1 were decreased with the increasing pore size, however, the increasing pore size of the alumina resulted in elongated cell morphology, enhanced ALP activity and mineralization. This study showed that the surface topography of nanoporous alumina plays an important role in regulating the behaviors of MG63 osteoblast-like cells and porous alumina can be regarded as useful substrate in tissue engineering.
Influence of Rod-Surface Structure on Biological Interactions Between TiO2 Nanorod Films and Proteins/cells

Abstract:We investigated the use of TiO2 nanorod films to construct an instructive microenvironment for regulation of biological activity. Rutile TiO2 nanorod films were fabricated via a hydrothermal method. Heat treatment was used to adjust the atomic arrangement order degree of the nanorod surfaces. The influence of changes in the nanorod surface on the films was characterized. The results show that heated nanorod films were well crystallized and the nanorod surfaces had a regular atomic arrangement. Unheated nanorod films had a higher contact angle and lower sensitivity to UV illumination, and showed good capacity for protein adsorption at an early stage, which may lead to better cytocompatibility. The results provide an insight into construction of an appropriate microenvironment for interaction between biomaterials and cells.

Abstract:Abstract Hierarchical surface structures with micro–nano scale play a crucial role in regulation of cell proliferation and osteogenic differentiation. It has been proven that cells are extremely sensitive to the nanoscaled structure and show multifarious phenotypes. Though a vital function of microstructure on osseointegration has been confirmed, the cell performances response to different microscaled structure is needed to be further dissected and in depth understood. In this work, the ordered micro–nano hierarchical structures with varying micro-scaled pits were precisely fabricated on titanium successfully by the combination of electrochemical, chemical etching and anodization as well. In vitro systematical assessments indicated that the micro–nano multilevel structures on titanium exhibited excellent cells adhesion and spreading ability, as well as steerable proliferation and osteogenic differentiation behaviors. It is shown that smaller micro-pits and lower roughness of the hierarchical structures enabled faster cell propagation. Despite cell growth was delayed on micro–nano titanium with relatively larger cell-match-size micro-pits and roughness, osteogenic-specific genes were significantly elevated. Furthermore, the alkaline phosphatase activity, collagen secretion and extracellular matrix mineralization of MC3T3-E1 on multi-scaled titanium were suppressed by a large margin after adding IWP-2 (an inhibitor of Wnt/β-catenin signal pathway), indicating this pathway played a crucial part in cell osteogenic differentiation modulated by micro–nano structures.

Abstract:Many studies have demonstrated the possibility to regulate cellular behavior by manipulating the specific characteristics of biomaterials including the physical features and chemical properties. To investigate the synergistic effect of chemical factors and surface topography on the growth behavior of mesenchymal stem cells (MSCs), bone morphorgenic protein 2 (BMP2) was immobilized onto porous alumina substrates with different pore sizes. The BMP2-immobilized alumina substrates were characterized with scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). Growth behavior and osteogenic differentiation of MSCs cultured on the different substrates were investigated. Cell adhesion and morphological changes were observed with SEM, and the results showed that the BMP2-immobilized alumina substrate was able to promote adhesion and spreading of MSCs. MTT assay and immunofluorescence staining of integrin β1 revealed that the BMP2-immobilized alumina substrates were favorable for cell growth. To evaluate the differentiation of MSCs, osteoblastic differentiation markers, such as alkaline phosphatase (ALP) activity and mineralization, were investigated. Compared with those of untreated alumina substrates, significantly higher ALP activities and mineralization were detected in cells cultured on BMP2-immobilized alumina substrates. The results suggested that surface functionalization of nanoporous alumina substrates with BMP2 was beneficial for cell growth and osteogenic differentiation. With the approach of immobilizing growth factors onto material substrates, it provided a new insight to exploit novel biofunctional materials for tissue engineering.

Abstract:The topography at the micro-/nanoscale level and bioactive composition of material surfaces have been thought to play vital roles in their interactions with cells. However, it is still a challenge to further modify special topography with biodegradable composition or vice versa. In this study, TiO2 nanorod films covered with microscale-distributed Zn-containing calcium phosphate (Zn-CaP) were prepared, trying to create a micro-/nanoscale topography and Zn2+ release capability. MC3T3-E1 cells cultured on TiO2 nanorod film with sparsely distributed Zn-CaP (TiO2/S-ZCP) had significantly higher biological responses than those on the films with densely distributed Zn-CaP (TiO2/D-ZCP) and fully covered Zn-CaP (F-ZCP). Abstract:Human bone marrow-derived mesenchymal stem cells (hBM-MSCs) are considered a great promise in the repair and regeneration of bone. Considerable efforts have been oriented towards uncovering the best strategy to promote stem cells osteogenic differentiation. In previous studies, hBM-MSCs exposed to physical stimuli such as pulsed electromagnetic fields (PEMFs) or directly seeded on nanostructured titanium surfaces ($TiO_{2}$) were shown to improve their differentiation to osteoblasts in osteogenic condition. In the present study, the effect of a daily PEMF-exposure on osteogenic differentiation of hBM-MSCs seeded onto nanostructured $TiO_{2}$ (with clusters under 100 nm of dimension) was investigated. $TiO_{2}$-seeded cells were exposed to PEMF (magnetic field intensity: 2 mT; intensity of induced electric field: 5 mV; frequency: 75 Hz) and examined in terms of cell physiology modifications and osteogenic differentiation. Results showed that PEMF exposure affected $TiO_{2}$-seeded cells osteogenesis by interfering with selective calcium-related osteogenic pathways, and greatly enhanced hBM-MSCs osteogenic features such as the expression of early/late osteogenic genes and protein production (e.g., ALP, COL-I, osteocalcin and osteopontin) and ALP activity. Finally, PEMF-treated cells resulted to secrete into conditioned media higher amounts of BMP-2, DCN and COL-I than untreated cell cultures. These findings confirm once more the osteoinductive potential of PEMF, suggesting that its combination with $TiO_{2}$ nanostructured surface might be a great option in bone tissue engineering applications.

Abstract:Surface modification of medical implants is considered as an effective method to improve cellular behaviors and the integration of tissues with materials. Titanium (Ti)-based materials with four different micro/nano-structures and compositions were prepared by acid etching, electrochemical anodization and alkali-heat treatment. The surface morphologies and compositions of the different surface-modified Ti materials were characterized by field-emission scanning electron microscopy (FE-SEM), atomic force microscopy (AFM) and X-ray diffraction (XRD). The effects of the micro/nano structured and compositions of the surfaces on cellular responses were investigated in vitro by observing the morphology, adhesion, proliferation and osteogenic differentiation of osteoblasts. To further investigate the underlying mechanisms, an RT-PCR assay was performed to analyze the expression levels of cell adhesion-related genes. Our results indicated that the nanosized structure and anatase composition could promote the adhesion and proliferation of MC3T3-E1 pre-osteoblast, as well as alkaline phosphatase activity and extracellular matrix mineralization via the integrin-FAK signaling pathway. Taken together, our innovation presented in this work demonstrated that the surface nano-structure design and composition of biomedical implants can be modified of for future orthopaedic applications.
Optimizing Stem Cell Functions and Antibacterial Properties of TiO2 Nanotubes Incorporated with ZnO Nanoparticles: Experiments and Modeling

Abstract:To optimize mesenchymal stem cell differentiation and antibacterial properties of titanium (Ti), nano-sized zinc oxide (ZnO) particles with tunable concentrations were incorporated into TiO2 nanotubes (TNTs) using a facile hydrothermal strategy. It is revealed here for the first time that the TNTs incorporated with ZnO nanoparticles exhibited better biocompatibility compared with pure Ti samples (controls) and that the amount of ZnO (tailored by the concentration of Zn(NO3)(2) in the precursor) introduced into TNTs played a crucial role on their osteogenic properties. Not only was the alkaline phosphatase activity improved to about 13.8 U/g protein, but the osterix, collagen-I, and osteocalcin gene expressions was improved from mesenchymal stem cells compared to controls. To further explore the mechanism of TNTs decorated with ZnO on cell functions, a response surface mathematical model was used to optimize the concentration of ZnO incorporation into the Ti nanotubes for stem cell differentiation and antibacterial properties for the first time. Both experimental and modeling results confirmed (R-2 values of 0.8873-0.9138 and 0.9596-0.9941, respectively) that Ti incorporated with appropriate concentrations (with an initial concentration of Zn(NO3)(2) at 0.015 M) of ZnO can provide exceptional osteogenic properties for stem cell differentiation in bone cells with strong antibacterial effects, properties important for improving dental and orthopedic implant efficacy.
Nanostructure Mediated Piezoelectric Effect of Tetragonal BaTiO3 Coatings on Bone Mesenchymal Stem Cell Shape and Osteogenic Differentiation

Abstract:In recent years, porous titanium (Ti) scaffolds with BaTiO3 coatings have been designed to promote bone regeneration. However, the phase transitions of BaTiO3 have been understudied, and their coatings have yielded low effective piezoelectric coefficients (EPCs < 1 pm/V). In addition, piezoelectric nanomaterials bring many advantages in eliciting cell-specific responses. However, no study has attempted to design a nanostructured BaTiO3 coating with high EPCs. Herein, nanoparticulate tetragonal phase BaTiO3 coatings with cube-like nanoparticles but different effective piezoelectric coefficients were fabricated via anodization combining two hydrothermal processes. The effects of nanostructure-mediated piezoelectricity on the spreading, proliferation, and osteogenic differentiation of human jaw bone marrow mesenchymal stem cells (hJBMSCs) were explored. We found that the nanostructured tetragonal BaTiO3 coatings exhibited good biocompatibility and an EPC-dependent inhibitory effect on hJBMSC proliferation. The nanostructured tetragonal BaTiO3 coatings of relatively smaller EPCs (<10 pm/V) exhibited hJBMSC elongation and reorientation, broad lamellipodia extension, strong intercellular connection and osteogenic differentiation enhancement. Overall, the improved hJBMSC characteristics make the nanostructured tetragonal BaTiO3 coatings promising for application on implant surfaces to promote osseointegration.
Effects of the Hierarchical Macro/mesoporous Structure on the Osteoblast-Like Cell Response

Abstract:To improve the success of medical devices, implants with strong surface bioactivity are urgently required. Coatings with a macroporous structure produced by micro-arc oxidation possess advantages, such as strong adhesion to substrate and excellent resistance to wear and corrosion. Mesoporous structures contain pores with sizes of 2-50 nm, which can endow the biomaterials with the ability to enhance osteogenesis and to be loaded with diverse drugs. Thus, in this study, we aimed to evaluate the effects of both macroporous and mesoporous structures using a hierarchical macro/mesoporous structure to modify the titanium implant surface. The behaviors of SaOS-2 human osteosarcoma cells on the macro/mesoporous structure, including initial adhesion, proliferation, alkaline phosphatase (ALP) activity, and collagen secretion, were investigated. Cells that attached on the macro/mesoporous surface showed the highest cell numbers and greatest spreading area after incubation for 1, 2, and 4 h compared with the polished smooth substrate and macroporous surface in the presence of fetal bovine serum (FBS). However, in the absence of FBS, cell adhesion on the polished substrate, macroporous structure, and macro/mesoporous structure did not differ significantly. Cell proliferation on the macroporous and macro/mesoporous surfaces increased compared with that on the smooth substrate surface. Furthermore, ALP activity and collagen secretion were enhanced on the macro/mesoporous structure. Titania nanotubes promote osteogenesis via mediating crosstalk between macrophages and MSCs under oxidative stress

Abstract:Before mesenchymal stem cells (MSCs) adhere to the surface of an implant and differentiate into osteoblasts, monocytes (especially macrophages) arrive at the bone injury site and interact with the implant and subsequent MSCs. In our previous study, large titania nanotubes (TNT110) had been verified to endow superior oxidation resistance to osteoblasts. The early regulation between macrophages and MSCs by surface nanotubes under oxidative stress (OS) was evaluated further in this study. Cellular and molecular results show that TNT110 greatly increased early inflammation of macrophages by activating integrin/FAK-mediated MAPK and NFκB signals and simultaneously promoted their gene expression of SDF1, IL-8, and CCL2 (chemokines) compared with 30 nm nanotubes and titanium substrates. Co-culture results show that more MSCs were recruited by those chemokines and may promote osteogenic self-differentiation, reducing early inflammation of macrophages by accelerating their M1-to-M2 transition in the TNT110 group. All findings reveal that the early cellular behavior of macrophages and MSCs was effectively regulated by TNT110, indicating large nanotubes would be more suitable to prevent oxidative damages.
Effects of the micro-nano surface topography of titanium alloy on the biological responses of osteoblast

Abstract:In clinical applications, osseointegration is essential for the long-term stability of dental implants. Inspired by the hierarchical structure of natural bone, we applied the electrochemical etching (EC) technique to form a micro-nano structure on a titanium alloy (Ti6Al4V) substrate, called EC surface. Sand blasting and acid etching (SLA) and machined (M) methods were employed to generate micro and smooth textures, respectively, as the control groups. The surface topographies of the three substrates were characterized using scanning electron microscopy (SEM). Then, human osteoblast-like cells (MG63) were cultured on substrates, and adhesion, proliferation, morphology, alkaline phosphatase activity (ALP), and gene expression levels of Runt-related transcription factor 2 (RUNX2), osteocalcin (OCN), osteopontin (OPN), and type I collagen (COLIA 1) were analyzed. MG63 cells cultured on the EC Ti alloy substrates displayed better cell adhesion, significant proliferation, and a higher production level of ALP, gene expressions of RUNX2, OCN, OPN and COLIA 1 (p < 0.01 or p < 0.05) compared with those of SLA and M substrates. These results indicate that the micro-nano structure fabricated by electrochemical etching method is beneficial for the biological functions of MG63 cells and may be a promising candidate in dental implants. © 2016 Wiley Periodicals, Inc. Spatial Control of Cell-Nanosurface Interactions by Tantalum Oxide Nanodots for Improved Implant Geometry

Abstract:Nanotopological cues can be exploited to understand the nature of interactions between cells and their microenvironment to generate superior implant geometries. Nanosurface parameters which modulate the cell behavior and characteristics such as focal adhesions, cell morphology are not clearly understood. Here, we studied the role of different nanotopographic dimensions in modulating the cell behavior, characteristics and ultimately the cell fate and accordingly, a methodology to improve implant surface geometry is proposed. Tantalum oxide nanodots of 50, 100nm dot diameter with an inter-dot spacing of 20, 70nm and heights 40, 100nm respectively, were engineered on Silicon substrates. MG63 cells were cultured for 72 hours and the modulation in morphology, focal adhesions, cell extensible area, cell viability, transcription factors and genes responsible for bone protein secretion as a function of the nanodot diameter, inter-dot distance and nanodot height were evaluated. Nanodots of 50nm diameter with a 20nm inter-dot spacing and 40nm height enhanced cell spreading area by 40%, promoted cell viability by 70% and upregulated transcription factors and genes twice as much, as compared to the 100nm nanodots with 70nm inter-dot spacing and 100nm height. Favorable interactions between cells and all dimensions of 50nm nanodot diameter were observed, determined with Scanning electron microscopy and Immunofluorescence staining. Nanodot height played a vital role in controlling the cell fate. Dimensions of nanodot features which triggered a transition in cell characteristics or behavior was also defined through statistical analysis. The findings of this study provide insights in the parameters of nanotopographic features which can vitally control the cell fate and should therefore be taken into account when designing implant geometries.

Abstract:Cell sheet technology is a new strategy in tissue engineering which could be possible to implant into the body without a scaffold. In order to get an integrated cell sheet, a light-induced method via UV365 is used for cell sheet detachment from culture dishes. In this study, we investigated the possibility of cell detachment and growth efficiency on TiO2 nanodot films with RGD immobilization on light-induced cell sheet technology. Mouse calvaria-derived, preosteoblastic (MC3T3-E1) cells were cultured on TiO2 nanodot films with (TR) or without (TN) RGD immobilization. After cells were cultured with or without 5.5mW/cm(2) UV365 illumination, cell morphology, cell viability, osteogenesis related RNA and protein expression, and cell detachment ability were compared, respectively. Light-induced cell detachment was possible when cells were cultured on TR samples. Also, cells cultured on TR samples showed better cell viability, alongside higher protein and RNA expression than on TN samples. This study provides a new biomaterial for light-induced cell/cell sheet harvesting.

Abstract:The topography at the micro/nanoscale level for biomaterial surfaces has been thought to play vital roles in their interactions with cells. However, discovering the interdisciplinary mechanisms underlying how cells respond to micro-nanostructured topography features still remains a challenge. In this work, ∼37 μm 3D printing used titanium microspheres and their further hierarchical micro-nanostructured spheres through hydrothermal treatment were adopted to construct typical model surface topographies to study the preosteoblastic cell responses (adhesion, proliferation, and differentiation). We here demonstrated that not only the hierarchical micro-nanostructured surface topography but also their distribution density played critical role on cell cytocompatibility. The microstructured topography feature surface with middle-density distributed titanium microspheres showed significantly enhanced cell responses, which might be attributed to the better cellular interaction due to the cell aggregates. However, the hierarchical micro-nanostructured topography surface, regardless of the distribution density of titanium microspheres, improved the cell-surface interactions because of the enhanced initial protein adsorption, thereby reducing the cell aggregates and consequently their responses. This work, therefore, provides new insights into the fundamental understanding of cell-material interactions and will have a profound impact on further designing micro-nanostructured topography surfaces to control cell responses.
