Abstract:We report a MgF2-Au-MgF2-polydopamine based surface plasmon resonance (SPR) sensor and its biomedical applications. The symmetrical MgF2-Au-MgF2 geometry induced a minimum loss of surface plasmons leading to, so far, the best refractive index resolution of 2.2 x 10(-8) refractive index units (RIU) in a glucose solution protocol. Meanwhile, a high detection performance could be maintained in the sensed medium with a refractive index ranging from 1.03 to 1.33 RIU. In order to solve the problem of surface functionalization on MgF2, which was thought to be inert, we managed to form adherent polydopamine films onto the MgF2-Au-MgF2 surface by dopamine self-polymerization, and found that secondary reactions could be used to create a variety of ad-layers for further biological uses. Finally, a model experiment on a chip detecting cellular mediators in human aqueous humor demonstrated the biomedical application of this system.
A polydopamine-modified optical fiber SPR biosensor using electroless-plated gold films for immunoassays

Abstract:A sensitive and stable electroless-plated gold film for the preparation of an optical fiber surface plasmon resonance (SPR) sensor is presented in this work, together with a facile antibody immobilization method. Gold nanoparticles were uniformly adsorbed onto the surface of an optical fiber forming a film with a thickness of approximately 56.3nm. The sensor had a high sensitivity with 2054nm/RIU and 3980nm/RIU in the refractive index ranges of 1.333–1.359 and 1.359–1.386, respectively. An SPR biosensor was developed based on polydopamine-modified gold film (PDA-Au), which was fabricated by a simple and quick spontaneous polymerization of dopamine (DA) on the gold film. When goat anti-human IgG antibodies were immobilized, the PDA-Au surface had a larger resonant wavelength shift of 66.21nm compared with the traditional 11-mercaptoundecanoic acid-modified gold film (MUA-Au) surface. In addition, the PDA-Au surface enabled the sensitive and selective determination of human IgG down to a concentration of 2μgmL−1 with a high sensitivity of 0.41nm per μgmL−1. The PDA-Au surface exhibited an approximately four fold higher sensitivity and an about seven fold lower LOD than the MUA-Au surface to human IgG.
Polydopamine-Assisted Fabrication of Stable Silver Nanoparticles on Optical Fiber for Enhanced Plasmonic Sensing

Abstract:We present a facile and effective method for fabrication of the localized surface plasmon resonance (LSPR) optical fiber sensor assisted by two polydopamine (PDA) layers with enhanced plasmonic sensing performance. The first PDA layer was self-polymerized onto the bare optical fiber to provide the catechol groups for the reduction from Ag+ to Ag-o through chelating and redox activity. As the reduction of Ag+ proceeds, Ag nanoparticles (NPs) were grown in-situ on the PDA layer with uniform distribution. The second PDA layer was applied to prevent Ag NPs from oxidating and achieve an improvement of LSPR signal. The PDA/Ag/PDA-based optical fiber sensor has an enhanced LSPR sensitivity of 961 nm/RIU and excellent oxidation resistance. The stable PDA/Ag/PDA-based LSPR sensor with high optical performance is very promising for future application in optical sensing field.

Abstract:Immunoassay can be divided into two aspects, one is immobilization of antibodies for the efficient detection of corresponding antigens, and the other is immobilization of antigens to search for antibodies which work against them. In this paper, we demonstrated these two aspects of immunoassay by using the disposable fiber optic biosensors based on surface plasmon resonance (SPR) through surface decoration with half-antibody fragments, which has been scarcely ever reported to the best of our knowledge. We first fabricated the fiber optic SPR biosensor which consists of one gold film coated single-mode fiber sandwiched by two multimode fibers. Then, we decorated the fiber optic SPR biosensors with antibody fragments and antigen fragments, respectively, and compared the specific detection performances of these two kinds of sensors. After surface decoration with half-antibody fragments, the antigen-decorated fiber probe has a demonstrated sensitivity and limit of detection of 0.9771nm/(μg/mL) and 0.1 μg/mL, respectively, which improves by 10 times compared with the performance of the antibody-decorated fiber probe. Additionally, the selective detection results indicate that our proposed biosensor can be employed as a reliable antigen detector or an effective antibody filter. Our proposed sensor has the advantages of miniaturization, low cost, simple usage, label-free detection, high efficiency and sensitivity, and can effectively avoid cross-contamination caused by reuse. Given the reliable and clean detection method for immunoassay, our work should open a new window for the utilization of miniaturized fiber optic sensors in biochemical sensing.

Abstract:An all-optical fiber multi-layer surface plasmon resonance (SPR) biosensor based on a sandwich structure of polydopamine-MoSe2@Au nanoparticles-polydopamine (PDA-MoSe2@AuNPs-PDA) was designed for the detection of specific immunoreactions. By optimizing the multi-layer structure and the ratio of MoSe2: AuNPs, a sensitivity of 5117.59 nm/RIU has been obtained, which is more than double that of the only Au-filmed optical fiber SPR sensor. A large surface area was produced by integrating the MoSe2 primitive unit cell and the AuNPs into a hybrid plasmonic nanostructure of MoSe2@AuNPs, leading to optical fiber SPR signal amplification. The nanostructure of MoSe2@AuNPs was surrounded by the PDA layer to guarantee the efficient immobilization of the protein molecules on the optical fiber by strong covalent bond. This biosensor achieved a detection limit of 54.05 ng/mL for detecting the goat-anti-rabbit IgG, which demonstrated enhancements of 12.1%, 23.3% and 184.6% in comparison with three reported SPR biosensors decorated with PDA-AuNPs-PDA, PDA and Cysteamine-MoSe2@AuNPs-Cysteamine nanostructure, respectively. This biosensor achieved favorable selectivity and outstanding sensitivity compared with the reported SPR immuno-sensors, which will provide a miniaturized, rapid-response and label-free optical fiber bio-sensing platform for clinical diagnosis in the future.

Abstract:A new type of human immunoglobulin G (IgG) sensors based on the surface plasmon resonance (SPR) in the low refractive index (RI) plastic optical fiber (POF) and an antibody immobilization method is presented. A 50-nm-thick gold film was formed on the polished D-shaped fiber surface by magnetron sputtering. The RI response of the POF sensor is 30 049.61 nm/RIU, which is 26.5 times higher than that of single mode fiber (SMF) SPR sensors. The proposed SPR biosensor can be developed by simple and rapid modification of the gold film with 11-mercapto undecanoic acid (MUA). Upon immobilization of the goat anti-human IgG antibody, the resonance wavelength shifts by 11.2 nm. The sensor can be used to specifically detect and quantify the human IgG at concentrations down to 245.4 ng/mL with the sensitivity of 1.327 7 nm per μg/mL, which offers an enhancement of 12.5-fold compared to that of the conventional SMF based SPR sensors. The proposed device may find the potential applications in the case of use at the point of care.

Abstract:A novel in situ electrochemical surface plasmon resonance (EC-SPR) immunosensor is presented in this paper. The EC-SPR measurement can be used to in situ monitor the polymer formation, probe immobilization, antigen–antibody interaction and protein immunosensing process. A sandwich immunosensor based on permeable polypyrrole propylic acid (PPA) film is constructed using mouse IgG as a model analyte. The results show that the introduction of capture antibody conjugated enzyme not only enhances the current responses but also increases the SPR angle shift. The calibration curves of electrochemical (EC) and surface plasmon resonance (SPR) measurement exhibit a similar dependence on the bulk concentration of antigen. An approximate linear relationship can be obtained by plotting the data in semi-logarithmic reference frame. Compared with SPR, EC shows higher sensitivity with prolonged time. The in situ EC-SPR immunosensor described herein could have important potentials for diagnostics and medicine applications.

Abstract:This article provides a novel method of constructing an optical fiber localized surface plasmon resonance (LSPR) biosensor. A gold nanoparticle (NP) assembled film as the sensing layer was built on the polyelectrolyte (PE) multilayer modified sidewall of an unclad optical fiber. By using a trilayer PE structure, we obtained a monodisperse gold NP assembled film. The preparation procedure for this LSPR sensor is simple and time saving. The optical fiber LSPR sensor has higher sensitivity and outstanding reproducibility. The higher anti-interference ability for response to an antibody makes it a promising method in application as a portable immuno-sensor.

Abstract:In producing high-performance optical biosensors, the selected coupling agent and its fixation mode play an essential role as one of the decisive conditions for antibody incubation. In this work, we designed optical fiber biosensors by electrochemical polymerization to enable low detection limit (LOD) immunoassay. Based on the optical fiber lossy mode resonance (OF-LMR) achieved by In 2 O 3 -SnO 2 -90/10 wt% (ITO), we have simultaneously implemented the electropolymerized dopamine (ePDA) film on the ITO-coated fiber via the electrochemical method, utilizing the excellent electrical conductivity of ITO. After that, the immunoglobulin G (IgG) antibody layer was immobilized on the entire sensing region with the assistance of the ePDA film. The results of immunoassay were analyzed by recording the shift of the LMR resonance wavelength to verify the sensor performance. The LOD was evaluated as the lowest concentration of human IgG detected by the OF-LMR sensor, which was confirmed to be 4.20 ng·mL −1 . Furthermore, the sensor achieved selective detection for specific antigens and exhibited a good recovery capability in chicken serum samples. The developed scheme provides a feasible opportunity to enhance the intersection of electrochemistry and optics subjects and also offers a new promising solution to achieve the immunoassay.

Polymer-Templated Gold Nanoparticles on Optical Fibers for Enhanced-Sensitivity Localized Surface Plasmon Resonance Biosensors

Abstract:Dense arrays of well-dispersed gold nanoparticles (AuNPs) on optical fibers are shown to bridge the gap in sensitivity and sensing performance between localized surface plasmon resonance (LSPR) and classical SPR sensing. A simple self-assembly method relying on a poly(styrene-b-4-vinylpyridine) (PS-b-P4VP) block copolymer brush layer was used to immobilize AuNPs of different diameters from 10 to 92 nm on optical fibers. In comparison with standard AuNP deposition methods using (3-aminopropyl)trimethoxysilane (APTMS) and polyelectrolytes, the sensitivity with the PS-b-P4VP templating method was found to be 3-fold better, a consequence of the smaller gap between particles and the presence of fewer AuNP aggregates. Hence, the sensitivity of the LSPR sensor for IgG was comparable to a classical SPR, also on optical fibers, and about 68% of that for a prism-based wavelength-interrogation SPR instrument. The reproducibility and the detection limit of the LSPR sensor were about the same as the SPR sensor. The enhanced performance of the LSPR sensors using the PS-b-P4VP block copolymer fabrication method paves the way for use of these LSPR biosensors in a smaller and more cost-effective platform.

Abstract:Optical fiber surface plasmon resonance (SPR) sensors are widely used in concentration measurement. Conventional approaches usually exploit wet electroless plating to deposit silver film on the surface of the core of optical fiber to produce surface plasmon resonance effect. The inability to accurately control and measure the thickness of metal growth is a drawback of chemical methods to prepare optical fiber SPR sensors. Here, we propose an indirect method to measure the thickness of metal film and design an experiment to control the thickness of metal film by using gradient concentration solution. We also discuss the design and fabrication of a multimode optical fiber SPR urea biosensor. In addition, we build a reflective fiber optic sensor system. We test the performance of different thicknesses of the sensor probe in solution. The best sensitivity is about 31.7 nm/M from 0 to 1 M.
Highly Reproducible Fiber Optic Surface Plasmon Resonance Biosensors Modified by CS2 for Disposable Immunoassays

Abstract:To achieve fiber optic surface plasmon resonance (SPR) biosensors for disposable immunoassays, the following activities are required: first, to manufacture highly consistent fiber optic SPR sensors; second, to develop highly reproducible and reliable surface modification methods for the sensors; third, to establish methods of analysis and evaluation to determine the capability and reliability of biomolecular detection. In this work, we prepared microprobe-type fiber optic SPR sensors and selected highly consistent sensors according to the Pearson correlation coefficient (PCC) model. To obtain reliable surface modification, carbon disulfide (CS2) was introduced as a crosslinker to modify anti-human IgG on the surface of fiber optic SPR biosensors for the first time. After systematic improvement and optimization, the CS2-based biosensors obtained a higher human IgG sensitivity and lower human IgG detection limit than conventional (11-MUA)-based biosensors. The PCC model was employed to evaluate the consistency of the surface-functionalized biosensors, and experimental results showed that the CS2-based biosensors exhibited reliable consistency, while the (11-MUA)-based biosensors did not. Finally, the proposed CS2-based biosensors have high selectivity and work well in serum. The surface modification method and reliable and consistency evaluation model for the sensors reported in this paper can contribute to the development of disposable fiber optic SPR biosensors.

Abstract:Surface Plasmon Resonance (SPR) is a powerful technique for directly sensing in biological studies, chemical detection and environmental pollution monitoring. In this paper, we present polymer optical fiber application in SPR sensors, including wavelength interrogation SPR sensor and surface enhanced Raman scattering (SERS) probe. Polymer optical fiber (POF) is used to effectively couple the incident light from light source, and collect reflective light into spectrometer. It also makes the sensing system more flexible and reliable. Gold nanorods with 30 nm long axis and average aspect ratios 3:1 are synthesized using seed-mediated growth method. Combining Au nanorods with POF, we can easily realize fiber SERS sensor. Long-period fiber gratings are fabricated on single mode polymer optical fiber with 120 um period and 50% duty cycle. The polarization characteristic of this kind of birefringent grating is studied. Theoretical analysis shows this type grating has two separated polarization peaks, and it will be advantageous in SPR sensing applications.

Abstract:We developed an all optic-fiber waveguide-coupled surface plasmon resonance (SPR) sensor using zirconium disulfide (ZrS 2 ) and poly-dopamine (PDA) as the dielectric layer and biological cross-linker, respectively. This sensor can be employed to monitor the entire process of the C-reactive protein (CRP) sensing, including antibody modification and antigen detection. The design and the optimization of the optical fiber waveguide-coupled SPR sensor were realized, based on the transfer matrix method and first-principles calculations. The sensor was fabricated and characterized according to the optimized parameters. The experimental setup was implemented to measure the entire process of antibody modification and antigen detection for CRP with the detection limit of 3.21 pmol·mL −1 , and the specificity tests were also carried out.

Abstract:Fiber optic-based biosensors with surface plasmon resonance (SPR) technology are advanced label-free optical biosensing methods. They have brought tremendous progress in the sensing of various chemical and biological species. This review summarizes four sensing configurations (prism, grating, waveguide, and fiber optic) with two ways, attenuated total reflection (ATR) and diffraction, to excite the surface plasmons. Meanwhile, the designs of different probes (U-bent, tapered, and other probes) are also described. Finally, four major types of biosensors, immunosensor, DNA biosensor, enzyme biosensor, and living cell biosensor, are discussed in detail for their sensing principles and applications. Future prospects of fiber optic-based SPR sensor technology are discussed.

Abstract:In recent years, surface plasmon resonance devices (SPR, or named plamonics) have attracted much more attention because of their great prospects in breaking through the optical diffraction limit and developing new photons and sensing devices. At the same time, the combination of SPR and optical fiber promotes the development of the compact micro-probes with high-performance and the integration of fiber and planar waveguide. Different from the long-range SPR of planar metal nano-films, the local-SPR (LSPR) effect can be excited by incident light on the surface of nano-scaled metal particles, resulting in local enhanced light field, i.e., optical hot spot. Metal nano-particles-modified optical fiber LSPR sensor has high sensitivity and compact structure, which can realize the real-time monitoring of physical parameters, environmental parameters (temperature, humidity), and biochemical molecules (pH value, gas-liquid concentration, protein molecules, viruses). In this paper, both fabrication and application of the metal nano-particles modified optical fiber LSPR sensor probe are reviewed, and its future development is predicted.

Abstract:Localized surface plasmon resonance (LSPR) and lossy mode resonance (LMR) are excited simultaneously by the gold nanoparticle (AuNP) arrays and the top-coated polyelectrolyte layer on optical fibers. The well-dispersed and dense AuNP arrays ordered by the diblock copolymer-template have LSPR sensing characteristics, whereas the layer-by-layer coated polyelectrolyte film carries LMR-based sensitivity enhancement. Both the experimental and simulation results illustrated that the thickness of the polyelectrolyte layer is the key factor affecting the optical properties of the proposed polyelectrolyte-enhanced LSPR sensor, and the sensor with a thicker polyelectrolyte layer presents a higher refractive index (RI) sensitivity because the lossy mode of the polyelectrolyte layer enhances the plasmonic resonance of AuNP arrays. The optimal sensor, which self-assembles with 50 nm AuNPs and 20 PAH/PSS bilayers, shows higher sensitivity and stability, with an RI sensitivity of 1312 nm/RIU and a detection limit for human IgG of 32.1 μg/mL. In addition, we observed an abnormally enhanced peak with an invariable wavelength, which has great potential to eliminate interference as a self-reference signal.

Abstract:In this work, surface plasmon resonance (SPR) was used to study protein immobilization on poly(pyrrole-co-pyrrole propylic acid) (PPy/PPa) for immunosensing applications. SPR was employed to in situ monitor the electropolymerization process and to control thickness of the PPy/PPa copolymer film. Goat IgG as a model protein was covalently immobilized on the carboxyl-containing film through EDC/NHS as the coupling reagents. The effect of pyrrole propylic acid (Pa) proportion in the deposition solution on the protein immobilization capability was systemically investigated. The immobilization efficiency was demonstrated by a label-free SPR immunosensor. The heterogeneous kinetics of the immune reaction was discussed. This work could provide a facile method to immobilize proteins on an electrode surface by electropolymerized copolymer, and renders a universal approach to in situ study the protein immobilization process and sensing kinetics for scientific insights of the heteroimmunosensing scheme particularly in surface chemistry and molecular biology for further improvement of immunosensors.
Direct Electronical Readout of Surface Plasmon Resonance Biosensor Enabled by On-Fiber Graphene/Pmma Photodetector

Abstract:Surface plasmon resonance (SPR) optical fiber sensors are appealing for biomolecular detection due to their inherent characteristics such as flexibility, real-time performance, and high sensitivity. Concurrently, incorporating SPR sensors into wearable devices has emerged as a significant strategy. However, the majority of traditional SPR optical fiber sensors utilize spectrometers for optical readout, which leads to a relatively bulky overall size of the sensing system. Herein, we present the first optical fiber device capable of conducting sensitive SPR measurements and providing direct electronical readout. This has been achieved by integrating a hyperbolic-metamaterial SPR (HMM-SPR) sensor with an on-fiber graphene/PMMA photodetector (oFGPD). The HMM, composed of three pairs of Au/ZrO2, has been employed to develop highly sensitive SPR sensors. The oFGPD, which was constructed by transferring a single layer of graphene onto a tapered fiber region and subsequently covering it with a PMMA protecting film, achieved a high responsivity of 3.42×106 A W−1 (at 14.07 pW) and a rapid response time of approximately 90 ms at 1550 nm. More significantly, we have incorporated an SPR sensor based on a side-polished fiber (SPF) into the oFGPD, enabling an electronical readout technique for environmental refractive index (RI) SPR signals in a broad potential spectral range, from visible to near-infrared, all within a more compact device. This integration has been successfully validated in the detection of urea and glucose concentrations in artificial perspiration. This approach provides a novel direction for SPR sensor detection and establishes a solid foundation for their application in wearable technology.
