Abstract:Biodegradable metals have attracted considerable attentions in recent years. Besides the early launched biodegradable Mg and Fe metals, Zn, an essential element with osteogenic potential of human body, is regarded and studied as a new kind of potential biodegradable metal quite recently. Unfortunately, pure Zn is soft, brittle and has low mechanical strength in the practice, which needs further improvement in order to meet the clinical requirements. On the other hand, the widely used industrial Zn-based alloys usually contain biotoxic elements (for instance, ZA series contain toxic Al elements up to 40 wt.%), which subsequently bring up biosafety concerns. In the present work, novel Zn-1X binary alloys, with the addition of nutrition elements Mg, Ca and Sr were designed (cast, rolled and extruded Zn-1Mg, Zn-1Ca and Zn-1Sr). Their microstructure and mechanical property, degradation and in vitro and in vivo biocompatibility were studied systematically. The results demonstrated that the Zn-1X (Mg, Ca and Sr) alloys have profoundly modified the mechanical properties and biocompatibility of pure Zn. Zn-1X (Mg, Ca and Sr) alloys showed great potential for use in a new generation of biodegradable implants, opening up a new avenue in the area of biodegradable metals.

Abstract:Apart from the industrial and automotive applications, Zn and Zn-based alloys are considered as a new kind of potential biodegradable material quite recently. However, one drawback of pure Zn as potential biodegradable metal lies in that pure Zn has quite low strength and plasticity. In the present study, three important IIA essential nutrient elements Mg, Ca and Sr and hot-rolling and hot-extrusion thermal deformations have been applied to overcome the drawback of pure Zn and benefit the biocompatibility of Zn-based potential implants. The microstructure, mechanical properties, corrosion behavior, hemocompatibility, in vitro cytocompatibility were studied systematically to investigate their feasibility as bioabsorbable implants. The results showed that the mechanical properties of the ternary Zn-1Mg-1Ca, Zn-1Mg-1Sr and Zn-1Ca-1Sr alloys are much higher than that of pure Zn, owing to both the alloying effects and thermal deformation effects. In vitro hemolytic rate test and cell viability test indicated that the addition of the IIA nutrient alloying elements Mg, Ca and Sr into Zn can benefit their hemocompatibility and cytocompatibility, which would further guarantee the biosafety of these new kind of biodegradable Zn-based implants for future clinical applications. Abstract:Zn, a promising biodegradable material, possesses excellent biocompatibility and biodegradability, however its low strength and hardness largely limit its application in biodegradable implants. The addition of alloying elements would generally be suggested as an effective method for improving the mechanical properties. In the current study, alloying with a minor amount of Ca or Sr in Zn–1.5Mg alloy, the ternary alloys were composed of the matrix Zn and precipitated phase (Mg2Zn11 and CaZn13 for Zn–1.5Mg–0.1Ca alloy, Mg2Zn11 and SrZn13 for Zn–1.5Mg–0.1Sr alloy, respectively). Besides, the grain size of ternary alloys became more homogeneous and smaller than that of Zn–1.5Mg alloy. Meanwhile, the effect of alloying elements on the mechanical properties and corrosion behavior of Zn–1.5Mg alloy were analyzed. The results showed that the ternary alloys exhibited much higher yield strength (YS), ultimate tensile strength (UTS) and elongation than those of Zn–1.5Mg alloy. The measured corrosion rates of the ternary alloys were slightly increased due to galvanic corrosion reaction. Besides, a model of the corrosion mechanism in the simulated body fluid was discussed here based on the results of the studied alloys.

Abstract:Magnesium (Mg) and its alloys have attracted attention as potential biodegradable materials in orthopedics due to their mechanical and physical properties, which are compatible with those of human bone. However, the effect of the mismatch between the rapid material degradation and fracture healing caused by the adverse effect of hydrogen (H2), which is generated during degradation, on surrounding bone tissue has severely restricted the application of Mg and its alloys. Thus, the development of new Mg alloys to achieve ideal degradation rates, H2 evolution and mechanical properties is necessary. Herein, a novel Mg-1Zn-1Sn-xSr (x = 0, 0.2, 0.4, and 0.6 wt%) quaternary alloy was developed, and the microstructure, mechanical properties, corrosion behavior and biocompatibility in vitro/vivo were investigated. The results demonstrated that a minor amount of strontium (Sr) (0.2 wt %) enhanced the corrosion resistance and mechanical properties of Mg-1Zn-1Sn alloy through grain refinement and second phase strengthening. Simultaneously, due to the high hydrogen overpotential of tin (Sn), the H2 release of the alloys was significantly reduced. Furthermore, Sr-containing Mg-1Zn-1Sn-based alloys significantly enhanced the viability, adhesion and spreading of MC3T3-E1 cells in vitro due to their unique biological activity and the ability to spontaneously form a network structure layer with micro/nanotopography. A low corrosion rate and improved biocompatibility were also maintained in a rat subcutaneous implantation model. However, excessive Sr (>0.2 wt %) led to a microgalvanic reaction and accelerated corrosion and H2 evolution. Considering the corrosion resistance, H2 evolution, mechanical properties and biocompatibility in vitro and in vivo, Mg-1Zn-1Sn-0.2Sr alloy has tremendous potential for clinical applications.

Abstract:Abstract Magnesium-based biodegradable metals (BMs) as bone implants have better mechanical properties than biodegradable polymers, yet their strength is roughly less than 350 MPa. In this work, binary Zn alloys with alloying elements Mg, Ca, Sr, Li, Mn, Fe, Cu, and Ag respectively, are screened systemically by in vitro and in vivo studies. Li exhibits the most effective strengthening role in Zn, followed by Mg. Alloying leads to accelerated degradation, but adequate mechanical integrity can be expected for Zn alloys when considering bone fracture healing. Adding elements Mg, Ca, Sr and Li into Zn can improve the cytocompatibility, osteogenesis, and osseointegration. Further optimization of the ternary Zn-Li alloy system results in Zn-0.8Li-0.4Mg alloy with the ultimate tensile strength 646.69 ± 12.79 MPa and Zn-0.8Li-0.8Mn alloy with elongation 103.27 ± 20%. In summary, biocompatible Zn-based BMs with strength close to pure Ti are promising candidates in orthopedics for load-bearing applications.

Abstract:In the present study, the microstructure, mechanical properties, corrosion behavior, wettability, haemocompatibility, and cytocompatibility of the as-cast and as-rolled biodegradable Zn-1Mg-0.1Ca and Zn-1Mg-0.5Ca have been systematically investigated to evaluate their feasibility as potential biodegradable materials. The results demonstrated that the Zn-1Mg-0.1Ca have significantly improved mechanical properties, with the yield strength (YS), ultimate tensile strength (UTS), and elongation of as-rolled Zn-1Mg-0.1Ca are (209.04 ± 28.31) MPa, (331.51 ± 40.06) MPa, and (35.43 ± 3.53)%, respectively. Wettability test results demonstrated that the Zn-1Mg-0.1Ca and Zn-1Mg-0.5Ca have hydrophilic surfaces that can enhance cell responses and tissue-implant interactions. The haemocompatibility evaluation showed that the hemolysis ratio of Zn-1Mg-0.1Ca have a low hemolysis ratio of 0.6%; the platelets remain sphere morphology and are not activated. High cell viability indicates the cytocompatibility of the as-rolled Zn-1Mg-0.1Ca alloy. The Zn-1Mg-0.1Ca alloy can be considered as new suitable biodegradable Zn-based alloys for further biomedical applications.

Abstract:The microstructure, mechanical properties, in vitro degradation behavior and hemocompatibility of novel Zn–1Mg–0.1Sr and Zn–1Mg–0.5Sr (wt%) ternary alloys were evaluated with pure Zn as control. The results indicated that Zn–Mg–Sr alloys exhibited much higher yield strength (YS), ultimate tensile strength (UTS), hardness and corrosion rate than those of pure Zn. But their elongation and hemolysis rates were reduced. Furthermore, the hot-rolled Zn–1Mg–0.1Sr alloy presented the superior mechanics performance (196.84±13.20MPa, 300.08±6.09MPa, 22.49±2.52%, 104.31±10.18 for YS, UTS, Elongation and hardness, respectively), appropriate corrosion rate (0.15±0.05mm/year) and excellent hemocompatibility (hematolysis rate of 1.10±0.2% and no signs of thrombogenicity), showing a preferable candidate as the biodegradable implant material.

Abstract:Zn-3Cu-xMg (x=0, 0.1, 0.5 and 1.0wt.%) alloys were developed as potential biodegradable metallic materials in this study. The mechanical properties, corrosion behavior and in vitro cytocompatibility of Zn-3Cu-xMg alloys were studied systematically to evaluate the feasibility as biodegradable implant materials. The secondary phase in as-cast and as-extruded Zn-3Cu alloy was CuZn5 phase. Mg2Zn11 phase newly formed and precipitated by Mg addition. The volume fraction of Mg2Zn11 phase increased gradually with increasing Mg concentration. As a result, yield strength was improved from 213.7 to 426.7MPa and increased by 99.7% while elongation decreased from 47.1% to 0.9%. Besides, biocompatibility was improved apparently and in vitro corrosion rates increased from 11.4 to 43.2μmyear−1, which is more suitable for clinic application. The present research indicated that the newly developed alloys could be promising candidates for biomedical use due to the proper mechanical properties, degradation rate and acceptable biocompatibility.

Abstract:Zinc (Zn), one of the promising candidates for biodegradable implant materials, has excellent biocompatibility and biodegradability. In this study, as-cast Zn1FexMg (x <= 1.5 wt %) alloys were prepared to systematically explore the effects of magnesium (Mg) alloying on their microstructures, mechanical properties, and biodegradability. The microstructure of Zn1FexMg alloy consisted of Zn matrix, Zn + Mg2Zn11 eutectic structure, and FeZn13 phase. The addition of Mg not only promoted grain refinement of the alloy, but also improved its mechanical properties. The results of immersion tests showed that the addition of Mg accelerated microcell corrosion between different phases, and the modeling of the corrosion mechanism of alloys in simulated body fluid (SBF) solution was discussed to describe the interaction between different phases in the corrosion process. Zn1Fe1Mg possessed superior comprehensive mechanical properties and appropriate corrosion rate, and the values for hardness, tensile strength, yield strength, elongation, and corrosion rate were 105 HB, 157 MPa, 146 MPa, 2.3%, and 0.027 mm/a, respectively, thus revealing that Zn1Fe1Mg is a preferred candidate for biodegradable implant material.
Development and characterization of ZnxCuyTizMo alloys for biomedical applications: A high-throughput gradient continuous casting approach

Abstract:The limited mechanical properties of pure Zn, such as its low strength and ductility, hinder its application as a material for biodegradable implants. Addressing this challenge, the current study focuses on the development of biodegradable Zn-based alloys, employing innovative alloy design and processing strategies. Here, alloys with compositions ranging from 0.02 to 0.10 weight percent (wt%) Cu, 1.22 to 1.80 wt% Ti, and 0.04 to 0.06 wt% Mo were produced utilizing a high-throughput gradient continuous casting process. This study highlights three specific alloys: Zn1.82Cu0.10Ti0.05Mo (HR8), Zn0.08Cu1.86Ti0Mo (HR7), and Zn1.26Cu0.13Ti0.06Mo (HR6), which were extensively evaluated for their microstructure, mechanical properties, electrochemical performance, potential as bioimplants, and cytotoxicity. These alloys were found to exhibit enhanced mechanical strength, optimal degradation rates, and superior biocompatibility, evidenced by in-vivo experiments with SD rats, positioning them as promising candidates for medical implants. This research not only introduces a significant advancement in biodegradable alloy development but also proposes an efficient method for their production, marking a pivotal step forward in biomedical engineering. STATEMENT OF SIGNIFICANCE: The limited mechanical properties of pure Zn have hindered its application in biodegradable implants. Our research primarily focuses on the alloy design and process strategies of biodegradable Zn-based alloys. We explore the ZnCuxTixMox alloys. This study introduces a high-throughput experimental approach for efficient screening of multi-component alloy systems with optimal properties. The ZnCuxTixMox alloys were designed and processed through gradient continuous casting, followed by homogenization and hot rolling. Our findings indicate that the Zn1.82Cu0.10Ti0.05Mo alloy demonstrates superior tensile, mechanical, and corrosion properties post hot rolling. The study suggests that Zn0.13Cu1.26Ti0.06Mo, Zn0.08Cu1.86Ti0Mo, and Zn1.82Cu0.10Ti0.05Mo alloys hold significant potential as biodegradable materials.

Abstract:Zn has garnered increasing attention in the realm of biodegradable materials due to its moderate degradation rate and no harmful gas released during degradation. However, the poor mechanical properties of Zn necessitate improvement by alloying and deformation processing. Currently, the primary studied Zn-based alloy systems include Zn-Mg, Zn-Li, Zn-Cu, Zn-Mn, Zn-Ag, and Zn-Ca alloys. Among these alloy systems, Mg is an essential element for the human body with advantages of low cost and good ability to enhance mechanical properties of Zn. Consequently, biodegradable Zn-Mg alloy exhibits fascinating application potential. This paper provides a comprehensive overview of research progress on Zn-Mg alloys by summarizing the effects of composition and deformation processing on degradation properties, biocompatibility, and mechanical properties. Additionally, this paper highlights existing challenges and hopes to offer new perspectives and ideas for researchers engaged in the study of degradable materials.

Abstract:The development of biodegradable Zn alloys with comprehensive performance for biomedical applications is still challenging. Hence, the nanocrystalline Zn–Cu–Sr–Li alloys were prepared by alloying design and multi-pass high-speed hot rolling. The Zn–3Cu-0.2Sr-0.4Li alloy achieved nanoscale microstructures, resulting in high strength and an ideal corrosion rate that is more suitable for tissue repair cycles. Scanning Kelvin probe force microscopy analysis revealed the potential from high to low was ranked as: ε phase > SrZn 13 phase > Zn grains > β phase. The nanoscale Zn grains and β particles formed numerous microcells, enhancing the microgalvanic corrosion effect. The Sr ions could be released more efficiently during the degradation in Li-containing alloys, which endowed the alloys with outstanding biocompatibility. Besides, the synergistic release of Zn ions and Cu ions during alloy degradation endowed the alloy with excellent antimicrobial ability against Staphylococcus aureus (inhibition zone diameter ≈ 7.5 mm). The antimicrobial ability of Zn–3Cu-0.2Sr-0.4Li alloy was superior among the reported biodegradable Zn alloys. These results demonstrated nanocrystalline Zn–Cu–Sr–Li alloys have comprehensive performance and broad application prospects in biodegradable bone implants, providing the experimental and theoretical basis for future research in the biomedical field.

Abstract:Biodegradable Zn-based alloys have excellent mechanical properties, a good corrosion rate, and favorable biocompatibility, which have broad application prospects. In this study, new biodegradable Zn-0.5Fe-xLi (x = 0, 0.2, 0.5, 0.8 wt%) alloys were successively prepared by melting and hot extrusion, and the effects of Li content on the microstructure, mechanical properties, corrosion behavior, and biocompatibility of the alloys were investigated. The formation of FeZn13 and eutectic LiZn4 phases in the Zn-0.5Fe-xLi alloys and the grain size of the Zn-0.5Fe-0.5Li alloys significantly reduced with an increase in Li content. The synergistic effect of solid solution and grain refinement of Li significantly improved the strength of the Zn-0.5Fe-0.5Li alloy and ensured the ductility of the alloy, which resulted in its optimum overall performance. The yield strength was 353.6 ± 6.8 MPa, the ultimate tensile strength was 441.9 ± 2.6 MPa, and the elongation was 26.8 ± 0.8%. In addition, the Zn-0.5Fe-0.5Li alloy showed excellent corrosion properties. The electrochemical corrosion rate was 0.735 mm/year, and the surface corrosion degradation morphology was uniform after immersion. The Zn-0.5Fe-0.5Li alloy also demonstrated the most favorable in vitro cytocompatibility in cellular experiments. Overall, the extruded Zn-0.5Fe-0.5Li alloy is a promising biodegradable implant material with excellent mechanical properties, a suitable corrosion rate, and good cytocompatibility.

Abstract:Zn as a novel biodegradable metal holds great potential in bioresorbable implant application since it possesses excellent biocompatibility and more corrosion resistant than Mg. In the present study, the effect of the micro-alloying element Mn on the Zn–Mg alloy was studied with mechanical properties, in vitro degradation behaviors and hemocompatibility being evaluated in comparison with pure Zn as control. The experimental Zn–Mg–Mn alloys were composed of the matrix Zn and secondary phase (MgZn2). Meanwhile, Zn–Mg–Mn alloys ingots exhibited much higher yield strength (YS), ultimate tensile strength (UTS) and hardness compared to pure Zn. But their elongation was reduced. The results of immersion in Hank's solution for 30days revealed that the sequence of corrosion rates from high to low was: as-cast Zn–1Mg–0.1Mn alloy>as-cast Zn–1.5Mg–0.1Mn alloy>pure Zn. The consequences of electrochemical tests indicated that the calculated corrosion rates followed the ranking order: as-cast Zn–1Mg–0.1Mn alloy>as-cast Zn–1.5Mg–0.1Mn alloy>pure Zn. The results of hemolysis rate and platelet adhesion implied that the studied alloys had good blood compatibility. Furthermore, after hot-rolling, the YS, UTS, elongation and hardness of Zn–1Mg–0.1Mn alloys were further improved and possessed the superior mechanics performance (YS 195.02MPa, UTS 299.04MPa, Elongation 26.07%, Hardness 107.82 Hv), appropriate corrosion rate (Vcorr 0.25mm/year) and excellent hemocompatibility (hemolysis rate of 1.10% and no signs of thrombogenicity), showing the preferable candidate as a biodegradable implant material.

Abstract:The effects of Al addition on the microstructure and mechanical properties of Mg-Zn-Sn-Mn-Ca alloys are studied in this paper. It was found that the Mg-6Sn-4Zn-1Mn-0.2Ca-xAl (ZTM641-0.2Ca-xAl, x = 0, 0.5, 1, 2 wt.%; hereafter, all compositions are in weight percent unless stated otherwise) alloys have α-Mg, Mg2Sn, Mg7Zn3, MgZn, α-Mn, CaMgSn, AlMn, Mg32(Al,Zn)49 phases. The grain is also refined when the Al element is added, and the angular-block AlMn phases are formed in the alloys. For the ZTM641-0.2Ca-xAl alloy, the higher Al content is beneficial to elongation, and the double-aged ZTM641-0.2Ca-2Al alloy has the highest elongation, which is 13.2%. The higher Al content enhances the high-temperature strength for the as-extruded ZTM641-0.2Ca alloy; overall, the as-extruded ZTM641-0.2Ca-2Al alloy has the best performance; that is, the tensile strength and yield strength of the ZTM641-0.2Ca-2Al alloy are 159 MPa and 132 MPa at 150 °C, and 103 MPa and 90 MPa at 200 °C, respectively.

Abstract:In recent years, Zn-based materials provide a new option as biodegradable metals for orthopedic applications. To improve the low strength and brittle nature of pure Zn, small amounts of alloying element Mn (0.1, 0.4 and 0.8 wt.%) were added into Zn to fabricate binary Zn-Mn alloys. An extremely high elongation (83.96 +/- 2.36%) was achieved in the resulting Zn-0.8 wt.%Mn alloy. Moreover, Zn-Mn alloys displayed significantly improved cytocompatibility as compared to pure Zn, according to cell proliferation and morphology analyses. More importantly, a significantly improved osteogenic activity was verified after adding Mn regarding ALP activity and osteogenic expression. Furthermore, Zn-0.8 wt.%Mn alloy scaffolds were implanted into the rat femoral condyle for repairing bone defects with pure Ti as control. Enhanced osteogenic activities were confirmed for Zn-0.8Mn alloy in contrast to pure Ti based on Micro-CT and histological results, and favorable in vivo biosafety of Zn-0.8Mn alloy was verified by H&E staining and blood tests. The exceptional mechanical performance and favorable osteogenic capability render Zn-Mn alloy a promising candidate material in the treatment of bone defects or fracture repair. Statement of Significance The element Mn, on the one hand, as an essential trace element in the human body, promotes cell proliferation, adhesion, spreading, and regulates bone metabolism; on the other hand, it could significantly improve the ductility of Zn alloys. Here, we systematically reported the biocompatibility and biofunctionality of binary biodegradable Zn-Mn alloys in the bone environment. The Zn-Mn alloys promoted MC3T3-E1 cell proliferation, adhesion, spreading, and osteogenic differentiation in vitro. Furthermore, a rat femoral condyle defect model was established; porous Zn-Mn alloy scaffolds were manufactured to repair the bone defects. Significant bone regenerations, considerable bone ingrowth, and desirable biosafety were confirmed in vivo. Abstract:In this work, three widely used commercial Zn alloys（ZA4-1, ZA4-3, ZA6-1） were purchased and prepared by hot extrusion at 200 °C. The microstructure, mechanical properties, corrosion behaviors,biocompatibility and hemocompatibility of Zn alloys were studied with pure Zn as control. Commercial Zn alloys demonstrated increased strength and superb elongation compared with pure Zn. Accelerated corrosion rates and uniform corrosion morphologies were observed in terms of commercial Zn alloys due to galvanic effects between Zn matrix and α-Al phases. 100% extracts of ZA4-1 and ZA6-1 alloys showed mild cytotoxicity while 50% extracts of all samples displayed good biocompatibility. Retardant cell cycle and inhibited stress fibers expression were observed induced by high concentration of Zn 2+ releasing during corrosion. The hemolysis ratios of Zn alloys were lower than 1% while the adhered platelets showed slightly activated morphologies. In general, commercial Zn alloys possess promising mechanical properties, appropriate corrosion rates, significantly improved biocompatibility and good hemocompatibility in comparison to pure Zn. It is feasible to develop biodegradable metals based on commercial Zn alloys.
