Abstract:Two strains (designated F2, F6) which produce excellent flocculating substance was isolated from soil samples and identified as Rhizobium radiobacter and Bacillus sphaeicus. The compound bioflocculant (CBF) with higher flocculating activity was obtained by co culturing F2 and F6. The isolates F2 and F6 produced the flocculating substance during the late logarithmic growth phase and the early stationary growth phase. The flocculation activated substances of CBF distributed mainly in upper phase and was mainly composed of polysaccharide (90.6%) and protein (9.3%). The compound bioflocculant is thermo-stable and 75% of its activity remains after heating at 100 degrees C for 30 min. The flocculation efficiency of this bioflocculant increased with the pH within a range of pH 7-10 and was 86%-94%. The flocculation activated substances could be recovered from the supernatant of the fermented medium by cold acetone precipitation and purified by treating with Savage reagent. The experimental result showed that the optimum extracting conditions were that concentrating multiple is 1/2, acetone. consistency is 80%, acetone quantity is 1:1, precipitating time is 24h, and pH is 7, the rate of extraction for the polysaccharides was 55.5%; the optimum purifying conditions were that Vsample:Vsevag is 2:1, Vchloroform:Vbutanol is 5:2, purifying time is 90 min, the rate of eliminating for the protein was 92%. After the preliminary extraction and purification, CBF remained high flocculating activity.
