Abstract:Three distinct geminivirus species Tobacco curly shoot virus (TbCSV), Tobacco leaf curl Yunnan virus (TbLCYNV) and Tomato yellow leaf curl China virus (TYLCCNV) belonging to the genus Begomovirus have previously been found associated with tobacco leaf curl disease (TLCD) in Yunnan province, China.For determination of their distribution, 109 samples were collected from symptom expressing tobacco plants in fields located in four districts of Yunnan province in the period from 2000 to 2005. Primer pairs specific to each of the three viruses were used to determine their occurrence. PCR detections revealed that all the three viruses occurred in Baoshan district while only TYLCCNV was found in the other three districts (Dali, Wenshan and Honghe ). Mixed infections comprising TbCSV + TbLCYNV, TbCSV + TYLCCNV and TbCSV +TbLYNV + TYLCCNV were found in Baoshan district. Comparisons and phylogenetic analysis of the partial sequences obtained from a total of 39 isolates showed that they clustered into different groups on the basis of virus species and geographical location. The epidemiology of TLCD and the development of strategies for effective disease management were discussed.

Abstract:Viral diseases are becoming an important problem in Amorphophallus production due to the propagation of seed corms and their trade across regions. In this study, combined-High-Throughput Sequencing, RT-PCR, electron microscopy, and mechanical inoculation were used to analyze virus-like infected Amorphophallus samples in Yunnan province to investigate the distribution, molecular characterization, and diversity and evolution of Amorphophallus-infecting viruses including three isolates of dasheen mosaic virus and three orthotospoviruses: mulberry vein banding associated virus (MVBaV), tomato zonate spot virus (TZSV) and impatiens necrotic spot virus (INSV). The results showed that DsMV is the dominant virus infecting Amorphophallus, mixed infections with DsMV and MVBaV to Amorphophallus were quite common in Yunnan province, China. This is the first report on infection of Amorphophallus with MVBaV, TZSV, and impatiens necrotic spot virus (INSV) in China. This work will help to develop an effective integrated management strategy to control the spread of Amorphophallus viral diseases.

Abstract:This paper briefly summarized the research progress of carnation virus disease in Kunming. By identified procedure, we affirmed there are carnation mottle virus，carnation vein mottle virus，carnation latent virus，carnation etched ring virus and carnation necrotic fleck virus in Kunming area. Among these viruses, carnation mottle virus is the superior virus. Meanwhile, this paper also put forward the strategy of carnation virus disease's integrated management in order to supply in the producing.
Detection and Diagnosis of Viruses Infecting Lily

Abstract:CMV and several filamentous viruses were detected in samples of lily from Lishui and Hangzhou by electron microscopy and enzyme-linked immunosorbent assay (ELISA). Host range tests suggested that these viruses had narrowly host ranges, they couldn't infect 6 families 12 tested plants including Nicotiana glutinosa,N. tobacum,Cucumis sativus, Chenopodium quinoa, C. amaranticolor. CMV was detected in 26 out of 64 samples by ELISA, so the infection rate of CMV was 40.6%. Filamentous virus particles were examined in 58 out of 75 samples by electron microscopy, so the infection rate of filamentous virus was 77.3%. Pinwheel inclusions were observed in the cytoplasm of the leaf cells in 27 out of 40 samples infected by filamentous virus, which indicated the rate of potyvirus was 67.5%. Electron microscopic examination on the morphology and pathology of filamentous viruses suggested that they belonged to three genera: Potyvirus, Carlaviru and Potexvirus.

Abstract:The current occurrence and damage of bacterial fruit blotch on cucurbits caused by Acidovorax citrulli in Shandong province were summarized. From these, the cause analysis of the disease were discussed. The results showed that one of the chief factors was the pathogen carried by the seeds and seedlings. Based on these, the integrated management for the disease was proposed.

Abstract:Ageratum conyzoides is a common weed in agricultural regions in Asia. A. conyzoides plants exhibiting yellow vein symptoms were collected from Yunnan and Guangxi provinces of China. Polymerase chain reaction detection and sequence analysis showed that samples collected from Yunnan were mainly infected by Tobacco curly shoot virus (TbCSV) associated with Ageratum yellow vein China betasatellite (AYVCNB), while samples from Guangxi were mostly infected by Papaya leaf curl China virus (PaLCuCNV) and AYVCNB, or by Ageratum yellow vein China virus (AYVCNV) and AYVCNB, with a few exhibiting dual infections by PaLCuCNV, AYVCNV, and AYVCNB. Agrobacterium-mediated inoculation of infectious clones showed that both TbCSV and AYVCNB or PaLCuCNV and AYVCNB produced typical yellow vein symptoms in A. conyzoides. Consequently, Ageratum yellow vein diseases in Yunnan and Guangxi provinces were caused by TbCSV/AYVCNB, PaLCuCNV/AYVCNB, or AYVCNV/ AYVCNB. The implications of these results in relation to the prevalence of begomoviruses in cultivated plants are discussed.
Molecular identification of viral pathogens in Crocus sativus

Abstract:Objective: To investigate the viral pathogens in cultivated Crocus sativus. Methods: Viral pathogen identification was carried out by the observation of virus particle morphology and cytopathology as well as the detection of DAS-ELISA, RT-PCR, and sequencing. Results: Linear virus particles of 600-900 nm in length were observed in C. sativus by negative staining under transmission electron microscope (TEM). Bundles of linear virus particles, cylindrical inclusion bodies of subdivision II, and amorphous inclusion bodies were observed in the cells of C. sativus under TEM after ultrathin-section. These observations resembled the cytopathology of infectious Bean yellow mosaic virus (BYMV). Positive result of DAS-ELISA was obtained from the leaves of C. sativus by using monoclonal antiserum against BYMV capsid protein. Positive result of RT-PCR induced by the Potyvirus specific primers (Sprimer and M4) was also obtained. Sequencing after RT-PCR revealed that the viral sequence in this diseased C. sativus had a homology of 99% with the BYMV sequence. Conclusion: The pathogenic virus of this C. sativus disease is identified as BYMV.

Abstract:Cucumber mosaic virus(CMV),which is transmitted by over sixty species of aphids and infects over 1000 species of plants ,has been processing a gradual severity in the past fifteen years. It is now the severest crop virus in China. Climate warming has played an important role on this phenomenon. Relationship between the rampancy of cucumber mosaic virus and the climate waring was analyzed, and countermeasure of disease-management which depends on agricultural and biological control measures was put forward.

Abstract:The Rice black-streaked dwarf viral disease(RBSDV),transmitted by the small planthopper Laodelphax striatellus Fallen,is one of the most destructive diseases in rice.In order to control this disease,a provincial collaborative research was conducted across the Zhejiang province,China,during 2000 to 2005.The epidemiology,causes of resurgence and damage-yield loss relationship were understood and were applied to design the field sampling and forecast methods,and to determine the economic thresholds of the disease through control of the insect vector.A provincial disease alert system including the GPS,GIS and RS approaches,and an estimation model for virus-infestation in rice were established and were used for long-term forecast of the disease.A new management strategy was proposed which integrated the extension of RBSDV disease-and vector-resistant varieties,improvement of seedling nursery,healthy cultivation of diseased hill gap-filling by removing tillers from nearby healthy hills,and control of insect vectors in the seedlings stage to protect rice plants from the viral disease in field stages.An integrated RBSDV disease management technical system was therefore shaped for the Yangtze Delta region,and applied in large scale,effectively reduced the disease occurrence and damage,achieved significant economic and social effects.

Abstract:The earliest HIV-1 epidemic in China started in Yunnan province, which continues to fall down into the most severely affected area. Multiple HIV-1 subtypes have been detected in Yunnan. There are some evidences supporting the hypothesis that the virus disseminating widely into many other regions of China may originate from Yunnan. Here we review the HIV-1 epidemic in Yunnan, hoping to provide the basic information for the prevention, control of HIV-1 infection not only in Yunnan but also in China, and for the reasonable designation of HIV-1vaccines.

Abstract:Investigation on Verticillium wilt of Erigeron breviscapus,including the pathogens,factors and incidences were conducted in main planting areas in different seasons from 2002 to 2005.The results showed Verticillium albo-atrum was the pathogen of E.breviscapus wilt.The incidence of the wilt was 10%-15%,sometimes exceeding 20%.Improper irrigation or rainy seasons were direct reasons causing the wilt to occur on a large scale.

Abstract:Morning glory (Ipomoea indica) plants showing yellow mosaic and vein yellowing symptoms were collected from Jiangsu and Yunnan province of China. The amplified DNA-A components of two isolates named JS1 and Y338 were comprised of 2827 and 2801 nucleotides, respectively, in length with genome organization similar to that of begomoviruses. Complete nucleotide sequence revealed the presence of two ORFs (AV1 and AV2) in the virion sense and four ORFs (AC1, AC2, AC3 and AC4) in the complementary sense. Comparison analysis showed that DNA-A sequence of JS1 isolate was closely related to that of sweet potato leaf curl virus (SPLCV) from United States with nucleotide sequence identity of 97.0% and DNA-A of Y338 showed highest sequence identity at 97.8% with an isolate of SPLCV from China. Phylogenetic analysis showed that JS1 clustered together with SPLCV-US and Y338 showed close relationship with SPLCV-CN. Sequence analysis indicated that isolates JS1 and Y338 were strains of SPLCV.

Abstract:Tomato yellow leaf curl virus (TYLCV), belonging to the genus Begomovirus in the family Geminiviridae , is the most damaging virus for global tomato production. This virus has become one of the most studied plant viral pathogens because of its huge economic importance. Since it was firstly identified in Shanghai, China, in 2006, it has been spread to most parts of the country. The widespread occurrence, rapid spread to new regions, and enormous damage to tomato production, makes it an important agricultural pathogen in China. TYLCV has been characterized extensively at the molecular level. This review is focused on the occurrence and distribution of all TYLCV isolates in China, providing valuable information for further epidemiological studies. In addition, management strategies for TYLCV are also proposed, with the ultimate goal to prevent and control the further occurrence of this viral disease.

Abstract:Objective To study the Echinococcus granulosus infection in Yunnan province and draw up the prevention and control measures. Method Brightness mode of ultrasonic examination, questionnaire and the serum sample analysis by ELISA were used to study the Echinococcus granulosus infection.Result The infection rate was 13.52% and did not show any significant difference from that (14.11%)of the first investigation of important human parasite disease in China. Conclusion The result indicates that Yunnan is a low-epidemic area for echinococcosis.

Abstract:Virus is known as one of the smallest creatures in nature,its morphology and size can only be observed and identified by electron microscopy enlarging it by tens of thousands or even hundreds of thousands of times. The virus is strictly parasitic so that its growth and reproduction depend on the host cell. Plant viruses are an important part of the virus family. The plant virus disease,which affects the growth and development of plants and decreases the yield and quality,is increasingly serious year by year. Due to the numerous varieties of viruses and complex damage mechanisms,it is difficult to prevent and control the viral diseases. This article reviews the major methods of detecting plant virus as well as the technologies of virus-elimination and its application in recent years. Further,the article discusses the issues which need to be paid attention to in the virus-elimination. Finally,the article forecasts the future research direction for providing references and theoretical basis for further study.
Molecular identification of geminiviruses on tomato and morning glory in Hunan Province

Abstract:Geminiviruses including a group of single-stranded circular DNA viruses occurred worldwide.Six plant samples collected from Hunan Province with symptoms of suspicious geminivirus infection were examined in this study.After conducting rolling circle amplification (RCA),the RCA products were digested by restriction endonuclease.Sequencing analysis revealed that virus isolates from tomato and morning glory shared 99％ of homology with Tomato yellow leaf curl virus (TYLCV) and Sweet potato leaf curl virus (SPLCV),respectively.Thcse results indicated that tomato and morning glory plants were infected by monopartite geminivirus,and this is the first report of geminivirus infection in Hunan Province.
