Abstract:Studies on toxicity and safety evaluation of pesticides to silkworm(Bombyx mori) constitute the basic work for early-warning of pesticides' potential hazard to silkworm raising in sericultural area.This article reviewed the research progresses of this field in the past 20 years.Mainly,it introduced the acute and chronic toxicity of frequently used pesticides in mulberry field and nearby crop field to silkworm and the determination methods of pesticides' residue toxicity,compared the characteristics and applicable scopes of toxicity grading methods such as reference method,toxicity ratio method,and grade setting method,and put forward recommendations for improving performance in such aspects as the coverage of determination targets,the normalization of determination protocols,and the comprehensiveness of evaluation indexes in toxicity and safety evaluation of pesticides to silkworm.Furthermore,a number of suggestions were put forward for establishing a collaborative research network in the field,formulating testing procedures and comprehensive evaluation technological system,and implementing comprehensive hazard-reduce measures to prevent silkworm from being poisoned by pesticides.
Researches and Applications of PCR Assay Technology in Silkworm Pebrine Disease Diagnosis

Abstract:An effective diagnosis technology is crucial to the study and control of silkworm pebrine disease.With the development of PCR technology,it is currently possible to apply this technology in detecting silkworm pebrine disease.This review is given on the researches and applications of PCR assay technology on pebrine disease diagnosis.We discussed potential approaches to improve sensitivity and specificity of the diagnosis based on template DNA extraction and primer design etc.Furthermore,we proposed that silkworm egg production farms take the responsibility to diagnose pebrine disease among the mother moths and the provincial inspection and quarantine authorities assume the responsibility to supervise pebrine disease inspection and quarantine of silkworm egg products,both of which form the important prerequisite to apply such PCR assay technology in practical quality inspection of silkworm eggs production.

Abstract:Sgnificant differences in transovarial transmission ratio of pebrine among commercial silkworm races were observed in differenct production seasons in Zhejiang Province. Resuluts showed that Japanese strains were more susceptible to pebrin disease than the Chinese strains, and high silk yield varieties were more susceptible than that with relatively low yield varieties. Moreover, significant differences of pebrin incidence were also found between eggs produced in different production units and years.
Examination on Clinical Effect of "MieCanYing"

Abstract:Myiasis is one of the serious disease in sericulture, and application of "MieCanYing" is the major preventive method in cocoon production. "MieCanYing" was sampled from the market in the present experiment, its chemical active ingredients were determined and clinical efficacy was tested. The results showed that effective component content of "MieCanYing" from the the market was significantly lower tha standard. Recommended efficacy index is up to 50.83. Half of the recommended dosage will result in significant decrease in disease control; however doubled recommended dose can not significantly increase in clinical effect but will cause decrease of economic characters of silkworm. Although effective control of disease can be achieved by reducing the dilution fator if "MieCanYing" with insufficient active ingredients,was used, obvious side effects could be observed. Development of new disease control technique to replace the "MieCanYing" is an urgent issue due to the devastating effect of organic phosphorus pesticide on the environment and the security needs of sericulture production.

Abstract:Bombyx mori Infectious flacherie virus (BmIFV) is the agent causing flacherie disease of silkworm. It makes serious loss of cocoon production. The paper introduced the history of the survey and described the virus genome organization that encodes corresponding putative proteome. We gave out an overview of BmIFV on replication mechanism, polyprotein cleavage, protein processing, sequence alignment, and epidemiology, Meanwhile. Comparing BmIFV with other picorna(-like) virus, the studies on cell factor involving in virus infection, virus gene function and epidemiological investigation are important.
Studies on Pathology of Silkworm, Bombyx Mori During Nosema Bombycis Infection

Abstract:When Nosema bombycis infected silkworm, the activity of midgut trehalase decreased significantly, which is mainly due to formation of large quantities of schizonts. The content of haemolymph sugar and trehalose, which related to energy metabolism also decreased significantly. The activity of haemolymph trehalase increased slightly in infected silkworm, but less influence to the content of haemolymph sugar. The content of haemolymph protein decreased behind to the substances which related to energy metabolism and the amount of storage protein decreased largely with respect to 30 K protein.

Abstract:The silkworm Bombyx mori is one of the most economically important insects and serves as a model for Lepidoptera insects. We used serial analysis of gene expression (SAGE) to derive profiles of expressed genes during the developmental life cycle of the silkworm and to create a reference for understanding silkworm metamorphosis. We generated four SAGE libraries, one from each of the four developmental stages of the silkworm. In total we obtained 257,964 SAGE tags, of which 39,485 were unique tags. Sorted by copy number, 14.1% of the unique tags were detected at a median to high level (five or more copies), 24.2% at lower levels (two to four copies), and 61.7% as single copies. Using a basic local alignment search tool on the EST database, 35% of the tags matched known silkworm expressed sequence tags. SAGE demonstrated that a number of the genes were up- or down-regulated during the four developmental phases of the egg, larva, pupa, and adult. Furthermore, we found that the generation of longer cDNA fragments from SAGE tags constituted the most efficient method of gene identification, which facilitated the analysis of a large number of unknown genes.

Abstract:Silkworm eggs are the most basic production material for sericultural production.They are also an important carrier of sericultural scientific and technological progress.The stability and development of silkworm eggs industry is an important basis for implementation of sustainable development of sericulture.Through analyzing the history and current status of China's silkworm eggs production's scale and regional distribution,market supply and demand,production technology and efficeincy,management and administrative system in recent 20 years,it was found that there were a series of problems in silkworm eggs production and management including excessive productivity,regional segmentation,small business scale,research and production disconnected,and low privatization degree.It was suggested to reform and improve the management and administrative system so as to increase degrees of marketization,privatization,business scale,integration,and standardization of silkworm eggs industry.

Abstract:The silkworm Bombyx mori is exploited both as a powerful biological model system and also as a tool to convert leaf protein into silk. Silkworm larvae often suffer from viral infections causing heavy losses to the economy of the silk industry. Insects exhibit both humoral and cellular immune responses that are effective against various pathogens like bacteria, fungi, protozoa, etc., but no insect immune response is effective against viral infections. To date, no satisfactory reports are available on antiviral immunity of the silkworm. Some efforts have been made by very few workers to identify and characterize the antiviral proteins in the silkworm. In the present article the mode of viral infection, and the activity of certain antiviral proteins involved in silkworm immunity and also in some other insects are discussed. The investigation will be helpful in understanding the molecular aspects of antiviral immunity, disease control and may form the basis for potential use of silkworm in other fields such as medicine.
Current Research Status and Prospects of Genomes of Insects Important to Agriculture in China

Abstract:Insects are the largest class of all living things with overwhelming diversity in the earth. Many of them are of agricultural importance: serious crop pests or beneficial insects. Modern entomological researches are more and more relying on information obtained from different insect genomes and transcriptomes since Drosophila melanogaster genome being sequenced in 2000. To better understand insect biology and transform our ability to manage insects that threaten our health, food supply, and economic security and to use beneficial insects that are essential to the maintenance and productivity of natural and agricultural ecosystems or provide us with silk, honey, medicine and other insect products, we need to know their genomic and transcriptomic information. Up to date, genomes of more than 50 insect species have been sequenced and analyzed around the world, and genomes of five insects of agricultural importance, including the domestic silkworm (Bombyx mori), the diamondback moth (Plutella xylostella), the oriental migratory locust (Locusta migratoria manilensis), the fig wasp (Ceratosolen solmsi) and the brown planthopper (Nilaparvata lugens), have been analyzed during the past decade in China, and genome sequencing for several agricultural insects including the whitefly (Bemisia tabaci), the oriental leafworm moth (Spodoptera litura), the rice stem borer (Chilo suppressalis), the white backed planthopper (Sogatella furcifera), the small brown planthopper (Laodelphax striatellu) and several parasite wasps, are in progress. Transcriptomes of the whiteflyand brown planthopper were reported in China in 2010, first two insect transciptomes reported in the world. Hundreds of insect transcriptomes have been reported in China since that year. Many important progresses in the functional genomics of the silkworm, the locust and the brown planthopper have been achieved, including the resequencing of 40 varieties of silkworm genomes which revealed domestication events and genes in silkworm, the uncovering of the precise mechanism of phase changes of the migratory locust, and the finding of that two insulin receptors determine alternative wing morphs in planthoppers. Data mining of insect genomes and transcriptomes, together with newly developed targeted genome editing and RNAi technologies, will lead to a revolutionary change in insect pest control and beneficial insect utilization.

Abstract:As a representative species of Lepidoptera, Bombyx mori has been widely studied and applied. However, bacterial infection has always been an important pathogen threatening the growth of silkworms. Bombyx mori can resist various pathogenic bacteria through their own physical barrier and innate immune system. However, compared with other insects, such as Drosophila melanogaster, research on the antibacterial mechanism of silkworms is still in its infancy. This review systematically summarized the routes of bacterial infection in silkworms, the antibacterial mechanism of silkworms after ingestion or wounding infection, and the intestinal bacteria and infection of silkworms. Finally, we will discuss silkworms as a model animal for studying bacterial infectious diseases and screening antibacterial drugs.

Abstract:Normally, silkworms, Bombyx mori, generate offspring by sexual activity. As we known, the hybrids of the first generation of the silkworm have higher cocoon production than pure lines. During the sericulture production, many processes are related with sex control. For example, sex sorting in the egg grainages, rearing of only male silkworm to save the mulberry leaf consumption and increase silk output and quality. Therefore it is very interested in understanding the sex control of the silkworm in theory and practice. Chinese sericultural scientists have been being engaged in the researches in the fields of artificifial parthogenesis, dispermic androgenesis, sex-limited varieties, sex linkage balanced lethal strain and high temperature sensitive male stocks for several decades and gained substantial achievement. Some of the achievements have been used in the commercial production. In this review, the authors introduced that the methods for control of the silkworm sex, and regulate the silkworm sex ratio according to different producing aim in the world and especially in China.
Silkworm-rearing Willingness,Sericulture Risk and Prevention Mea-sures——A Questionnaire Survey Based on 1782 Rural Households in 91 Counties of 14 Provinces

Abstract:A questionnaire survey to 1 782 sericultural households in 91 counties of 14 provinces revealed that 83.45% of the sericultural households did not quit growing mulberry and raising silkworm in the past and 83.89% of the sericultural households will continue to grow mulberry and raise silkworm.However,only 25.81% of the sericultural households will expand their production scale.Most of the sericultural households think that the income from sericultural industry is high but its risk is higher than other planting industries.Sericulture risks mainly come from price decline of silkworm cocoon,disease and pest outbreak on silkworm and mulberry,silkworm poisoning caused by waste gas emission and pesticide pollution,and output reduction of mulberry leaves and silkworm cocoons due to natural disasters.However,the primary reason that led to heavy loss between different provinces was obviously different.Nuclear polyhedrosis is the silkworm disease that occurs most frequently and draws most concern of the sericultural households.In 2010,silkworm diseases brought various degrees of loss to 75.48% of the total sericultural households.Major reasons leading to occurrence of mulberry diseases and pests and silkworm diseases include failure of mulberry field management and silkworm rearing practice not following technical requirements.Although sericultural households take active measures to cope with risk and reduce loss,they bear the major loss by themselves.Therefore,94.78% of the sericultural households hope the government to give subsidies to risk loss in mulberry planting and silkworm rearing.Meanwhile,it is expected to stabilize price of silkworm cocoons,accelerate renovation of mulberry and silkworm varieties,improve mechanization level in sericultural production,provide more technical training and guidance,enhance management of pesticide use during silkworm rearing period,control pollutant emissions of industrial enterprises during silkworm rearing period,supervise marketing of silkworm eggs and silkworm drugs,and include the potential risk loss in sericultural production into the scope of agricultural insurance.
Distribution of the Enterococci Isolated from Intestine of the Pebrine Infected Silkworm

Abstract:Two hundred of enterococci strains were isolated from the intestine of healthy, pebrine and bacterial flacherie diseased silkworms and classified by using API STREP(V.50) system based on numerical taxonomy so as to inquire into the ecological distribution of enterococci in the intestine of silkworm and relationship between Nosema bombycis and enterococci. The results showed that, in comparison with that of the healthy silkworms, the number of enterococci in silkworms infected with Nosema bombycis after third or forth moulting increased by 5.5 x 105 and 0.744 x 102 times respectively.The number of the species decreased from 6 to 3 and 4.The number of biochemical polymorphism types decreased from 27 to 13 and 14.The distribution frequency of Enterococcus avium decreased from 56% to 20% and 38% , the distribution frequency of Enterococcus faecium increased from 2% to 40% and 16% respectively. There are 59 biochemical polymorphism types within the 200 enterococci strains.

Abstract:Silkworms are an essential economic insect but are susceptible to diseases during rearing, leading to yearly losses in cocoon production. While chemical control is currently the primary method to reduce disease incidences, its frequent use can result in loss of susceptibility to pathogens and, ultimately, antibiotic resistance. To effectively prevent or control disease, growers must accurately, sensitively, and quickly detect causal pathogens to determine the best management strategies. Accurate recognition of diseased silkworms can prevent pathogen transmission and reduce cocoon loss. Different pathogen detection methods have been developed to achieve this objective, but they need more precision, specificity, consistency, and promptness and are generally unsuitable for in-situ analysis. Therefore, detecting silkworm diseases under rearing conditions is still an unsolved problem. As a consequence of this, there is an enormous interest in the development of biosensing systems for the early and precise identification of pathogens. There is also significant room for improvement in translating novel biosensor techniques to identify silkworm pathogens. This study explores the types of silkworm diseases, their symptoms, and their causal microorganisms. Moreover, we compare the traditional approaches used in silkworm disease diagnostics along with the latest sensing technologies, with a precise emphasis on lateral flow assay-based biosensors that can detect and manage silkworm pathogens.

Analysis on Sericulture Risk and Preventing Behavior of Sericulturists in Jiangsu and Zhejiang Province

Abstract:By questionnaire survey to 342 sericulturists from Jiangsu and Zhejiang province, we analysis their sericulture risk and preventing behavior, and come to a conclusion that sericulture has higher profits with higher risks. Sericulture risks mainly come from price declining of silkworm cocoon, pesticide poison, flaeherie, sewage and waste gas discharge, nature disasters and pest of mulberry trees. The loss of sericulturists from Jiangsu province mainly come from natural risks such as pesticide poison, flacherie, industrial waste gas etc., and the loss of sericuhurists from Zhejiang province mainly come from market risks of price declining of silkworm cocoon. Silkworm disease causes 78.65% of farmers" loss, nuclear polyhedrosis occurs most and sericulturists worried most. Mulberry plant diseases, insect pests and flacherie mainly caused by failure of mulberry field management and silkworm rearing against technical requirements. In order to reduce tile loss, famers take positive measures to keep away risks, but famers bear the loss mainly by themselves. So maximum of famers hope govermnent should give some subsidies, they also hope to keep the price of silkworm cocoon stable, accelerate renovation of variety, enhance pesticide use management, reduce pollution emissions and take silkworm production into agricultural insurance.

Abstract:Host-pathogen interactions are complex processes, which have been studied extensively in recent years. In insects, the midgut is a vital organ of digestion and nutrient absorption, and also serves as the first physiological and immune barrier against invading pathogenic microorganisms. Our focus is on Nosema bombycis, which is a pathogen of silkworm pebrine and causes great economic losses to the silk industry. A complete understanding of the host response to infection by N. bombycis and the interaction between them is necessary to prevent this disease. Silkworm midgut infected with N. bombycis is a good model to investigate the early host responses to microsporidia infection and the interaction between the silkworm and the microsporidium. Using Digital Gene Expression analysis, we investigated the midgut transcriptome profile of P50 silkworm larvae orally inoculated with N. bombycis. At 6, 12, 18, 24, 48, 72, and 96 h post-infection (hpi), 247, 95, 168, 450, 89, 80, and 773 DEGs were identified, respectively. KEGG pathway analysis showed the influence of N. bombycis infection on many biological processes including folate biosynthesis, spliceosome, nicotinate and nicotinamide metabolism, protein export, protein processing in endoplasmic reticulum, lysosome, biosynthesis of amino acids, ribosome, and RNA degradation. In addition, a number of differentially expressed genes involved in the immune response were identified. Overall, the results of this study provide an understanding of the strategy used by silkworm as a defense against the invasion by N. bombycis. Similar interactions between hosts and pathogens infection may exist in other species.
