The Measurement of HBV DNA in peripheral blood mononuclear cells and its clinical applicability

Abstract:Objective To quantify HBVDNA in peripheral blood mononuclear cells(PBMCs) using fluorescence quantitative polymerase chain reaction(FQ-PCR) based on TaqMan chemistry and its clinical applicability. Methods The blood was collected from patients with acute or chronic hepatitis B. The PBMCs were isolated from the blood by Ficoll and the genomic DNA was extracted. The number of HBV DNA in PBMCs and plasma were detected by FQ-PCR. Results ①The positive rates of HBV DNA in PBMCs and plasm were 80 3% and 72 9% respectively( P 0 05),their results were related( P 0 01). ②The positive rates and viral load of HBV in PBMCs and plasm were reduced from HBsAg(+)HBeAg(+)HBcAb(+) to HBsAg(+)HbcAb(+) group and there was a good correlation between them. ③The copies of HBV DNA in PBMCs and plasm were reduced with the severity of hepatitis related liver injury from the mild chronic hepatitis to the serve chronic hepatitis and hepatocirrhosis and the positive rates were obviously reduced from 92 3% to 43 8% in plasma and significantly increased from 56 9% to 93 8% in PBMCs. The copies of HBV DNA were correlated between in the PBMCs and plasma in the chronic hepatitis groups( P 0 05). Conclusion The results supported that HBV-infected PBMCs might participate in persistence of HBV and interfered with functions of the mononuclear cells during the immune response against the virus and might be responsible for chronicity.
