Expressions of stearoyl-CoA desturase-1 on rats fed with fat-rich diet and the effects of rosiglitazone

Abstract:Objective To study the expressions of stearoy1 CoA desturase 1 (SCD 1) on nonalcoholic fatty liver induced by fat rich diet and the effect of rosiglitazone.Methods Thirty five SD rats were divided into control, experimental and therapeutic groups. Experimental rats were fed with fat rich diet for 4,8 and 24 weeks, and the control rats were fed with normal diet simutanously. The therapeutic rats were fed with fat rich diet for 24 weeks and from 9th week rats were treated with rosiglitazone for 16 weeks. The SCD 1 mRNA and β actin mRNA of rat liver were analysed by RT real time fluorescence PCR.Results The body weight of two experimental groups increased progressively. The hepatocytes, stained with H E, of fat rich group were filled with fat under microscopy and met the diagnosis criteria of fatty liver at 8th week. The SCD 1 mRNA/β actin mRNA of control and experimental rats at 8th and 24th week were 0.83 ±0.28 vs 0.39 ±0.18 ( P 0.05 )and 0.88 ±0.22 vs 0.47 ±0.23 ( P 0.05 ),respectively. The SCD 1 mRNA/β actin mRNA of experimental rats and rosiglitazone treated rats were 0.47 ±0.23 vs 1.10 ±0.30 ( P 0.05 ).Conclusions The rat model of nonalcoholic fatty liver can be produced successfully by feeding with fat rich diet. Fat rich diet could decrease the level of liver SCD 1 mRNA. Low level of SCD 1 mRNA may result in excessive fat deposit in liver and forming nonalcoholic fatty liver. Rosiglitazone therapy could increase the level of SCD 1 mRNA and reduce fat deposit in rat liver. SCD 1 might be involved in the pathogenesis of nonalcoholic fatty liver induced by fat rich died.
