Pathological changes of aorta,liver in type 2 diabetes rat and the intervention study of rosiglitazone

Abstract:Objective Atheroclerosisc is closely related to glucose and lipid metabolism.Diabetic patients are more easily comlicated with fatty liver and atherosclerosis than nondiabetic subjects.The aim of this study is to observe the pathological change of aorta and liver in type 2 diabetes rats at 6 week and 12 week,and to observe the intervention effect of rosiglitazone.Methods 80 male Wistar rats were randomized to control group,high fat diet group,diabetes group,and rosiglitazone treatment group(diabetes plus rosiglitazone treatment).Type 2 diabetes models were developed and rosiglization group was treated with rosiglitazone.Animal was sacrificed respectively at 6 week and 12 week.Liver and aorta tissues were collected.All of the animal tissues were carried on HE stain.Pathological changes of these tissues in different groups were observed under the light microscope.Results 1.At 6 week,there was no evident pathological changes of aorta observed in control group,high fat diet group,diabetes group and rosiglitazone group.At 12 week,different degree pathological changes of aorta were observed in diabetes group,high fat diet group and rosiglitazone group.2.There were no evident liver pathological changes observed in control group and high fat diet group at 6 week.But adipose degeneration of liver were developed in diabetes group,high fat groupand rosiglitazone group at 6 week and at 12 week.Conclusion 1.At 6 week,there are no histologic changes of aorta in high fat diet group and diabetes group.Pathological changes of liver in diabetes group and rosiglitazone group have been observed under light microscope.2.There are pathological changes in aorta and liver in high fat diet group,diabetes group and rosiglitazona group at 12 week.3.All of these changes can be relieved by rosiglitazone intervention.
