Detection and Susceptibility of Moraxella Catarrhalis

Abstract:To establish the polymerase chain reaction(PCR) methed for detecting Moraxella catarrhalis with DNase testas reference and determine minimum inhibitory concentration(MIC) of antimicrobial agents against M. catarrhalis. Spe-cific fragment of M. catarrhalis DNA were amplified by PCR. DNase of M. catarrhalis was detected by DNase test. MICs of an-timicrobial agents against Moraxella catarrhalis were determined by agar dilution method and β-lactamases were detected by nitro-cefin test. The range of MIC50 and MIC90 of seven antibiotics on 46 β-1actamases-producing strains of M. catarrhalis were ≤0.06 -2mg/L, 0.25-4mg/L respectively. DNase test and PCR were both opsitive in 48 of 68 c1inica isolates and the other 20strains were all negative, the results of PCR were all the same as DNase test in detecting M.catarrhalis. The incidence ofβ-lacta-mases in M. cutarrhalis was 95.8 % (46/48). Two β-lactamases-negative M. catarrhalis strains were susceptible to seven antibi-otics tested and the MICs were both lower than 0.06mg/L.PCR method can be used to identify M. catarrhalis withadvantages of rapidity, specificity and easy to perform.β-1actamases-producing M. catarrhalis strains were susceptible toβ-lactam/β-lactamase inhibitor combination, the third-generation cephaloaporins, quinolones and minocycline.
