The Extraction of Swine and Canine Spleen Transfer Factors and the Determination of Their Physical and Chemical Properties and Biological Characteristics

Abstract:Swine and canine spleen transfer factors(TFs) had been extracted by a general extracting method.And then based on the relational methods,the physical and chemical properties of two kinds of TFs—limpidity degree,pH,protein,ultraviolet spectrum,TF content,DNA content,nucleic acid content and amino acid varieties were determined and analyzed.At the same time,the biological characteristics of two kinds of TFs were also tested and examined according to relational rules including the asepsis test,the pyrogen test,the security examination,the hypersusceptibility examination and the antigen test.Finally,the activity of two kinds of TFs were determined and analyzed.The results showed that the extracted swine and canine spleen TFs in our laboratory contained DNA,nucleotide acid and so on,their physical and chemical properties were similar to that of cattle TF,sheep TF,chicken TF and other animal TFs,and no bacteria,no pyrogen,avirulence,no hypersusceptibility and no antigenicity,and A260nm/280nm value was also rather high index.Those results conformed with the reports on other animals TFs in China or other countries.According to OD value,the prepared TFs activity are about 2 units per mL and useful for experiment and clinic.
