The Influence of Intra-Articular Injection of Chitosan on Articular Cartilage of Knee Joint in Rabbits with Osteoarthritis

Abstract:Objective To investigate the influence of intra-articular injection of carboxymethylated chitosan (CMCTS) on cartilage degeneration and expression of matrix metalloproteinase (MMP)-1 and MMP-3 mRNA in cartilage in rabbit model of osteoarthritis (OA). Methods Twenty-four white rabbits underwent unilateral anterior cruciate ligament- transection (ACLT) and were divided into 3 groups randomly 5 weeks after surgery. Rabbits in group A received 0.3 ml of intra-articular 2% CMCTS injection into the operated knees once every two weeks. Rabbits in Group B were injected with 0.3 ml of 1% sodium hyaluronate (HA) once a week. Animals in group C were not injected. All animals were sacrificed 11 weeks after operation. Operated knees were harvested to observe gross morphological changes of the femoral condyles. Expression of MMP-1 and MMP-3 mRNA in cartilage from medial femoral condyles was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results Cartilage degeneration in untreated group was significantly more severe than that in CMCTS and HA injection groups. In CMCTS injection group, expression of MMP-1 and MMP-3 mRNA in cartilage was significantly suppressed compared with corresponding samples of HA injection group and untreated group. No significantly difference of MMP-1 and MMP-3 expression was found between HA injection group and untreated group. Conclusion CMCTS can significantly suppress the mRNA expression of MMP-1 and MMP-3 in cartilage of OA and reduce the severity of cartilage degeneration. CMCTS has a protective effect on articular cartilage of early stage OA.
