Intra-articular injection of transforming growth factor beta1 and sodium hyaluronate on rabbit osteoarthritis

Abstract:AIM: To investigate the effects of intra-articular injection of transforming growth factor (TGF) β1 and sodium hyaluronate (SH) on experimental osteoarthritis in rabbits, and observe the expression of matrix metalloproteinase-3 (MMP-3) in articular cartilage.METHODS: The experiment was accomplished in the Central Laboratory, Clinical Medical College of Southeast University between March and August 2006. In this investigation, 32 mature New Zealand white rabbits were randomly divided into 4 groups: model group, SH group, TGF β1 group and mixture group, with 8 rabbits in each. Then models of osteoarthritis in knee of rabbits were established by Hulth's technique and then injected intra-articularly with normal saline, SH, TGF β1 (50 ng) and mixture of the last two, respectively, 0.3 mL once a week, for five weeks. At the sixteenth week, the rabbits were anesthetized and executed. Then the degeneration of articular cartilage was observed. Meanwhile, the expression of MMP-3 in articular cartilage was detected.RESULTS: All 32 rabbits were involved in the result analysis.①Histomorphology of articular cartilage: Mankin score was used to rank the cartilage sample in 8 cases of each group (10.38, 10.13, 6.50, 12.38). Intra-articular injection of TGF β1 or SH alleviated the degeneration of cartilage and decreased the Mankin's marks (P < 0.01). Combined injections of TGF β1 and SH had more obvious effect than their sole injection (P < 0.05).②The expression of MMP-3 in articular cartilage: TGF β1 injected solely or combined with SH remarkably decreased the expression of MMP-3 in chondrocyte, compared with that of model group (44.50±4.92)%, (26.38±4.78)%, (62.13±4.52)%, t =7.457, 15.375, P < 0.05, but the expression of MMP-3 in chondrocyte was not decreased significantly by SH injection solely (61.00±5.10)%, P > 0.05.CONCLUSION: Intra-articular injection of TGF β1 and/or SH could decrease the number of chondrocytes that express MMP-3 positively, so as to delay the process of osteoarthritis. The combined use of them might greatly improve the effects.
