Intraventricular Injection of Liposome-Mediated VEGF Plasmids Enhances Brain Repair Following Ischemia-Reperfusion Injury

Abstract:Objective To elucidate whether the over-expression of vascular endothelial growth factor (VEGF) in the brain has neuroprotection against cerebral ischemic injury and its neuroprotective mechanism. Methods Focal cerebral ischemia was made by middle cerebral artery occlusion (MCAo) for 30 min in adult rat. Neurological deficits were analyzed by six-grade system developed by Longa. Infarct was measured by cresyl violet (CV) staining and infarct volume was analyzed by an image processing and analysis system. The expression of von Willebrand factor (vWF) and CRMP-4 was analyzed with immunohistochemical technique. Results Intraventricular injection of VEGF plasmid mixed with liposome boosted angiogenesis in ischemic striatum of adult rat, expedited the recovery process of body weight and neuronal scores, significantly reduced the infarct volume to (17.38±1.56)% compared with control plasmid treatment \[(22.35± 2.04)%, n = 5, P 0.05\] . It enhanced CRMP-4 positive cells in ischemic striatum from 13.75 4.52 in control group to 42 8.41 in VEGF treatment group (P 0.05). Conclusion Liposome-mediated transfer of VEGF plasmid into rat brain plays neuroprotective roles in pathophysiological process following cerebral ischemic injury in adults.
