Intranasal Administration of VEGF with Effect on Angiogenesis in Infarction Boundary Zones in Rats with Cerebral Ischemia

Abstract:Objective To study the effect of intranasal administration of vascular endothelial growth factor (VEGF) on angiogenesis in infarction boundary zones following cerebral infarction in rats. Methods Adult male Sprague-Dawley rats were divided into sham operation group (n = 6). VEGF group (n=12) and control group (n=12). The VEGF and control groups were intranasally administrated with VEGF or saline daily starting 3 rd after the middle cerebral artery occlusion (MCAO), as well as BrdU (50 mg/kg) was intraperitoneally injected daily for 13 days. Modified Neurological Severity Scores (mNSS) were performed at 1. 7 and 14 days after MCAO. The BrdU~+ /vWF~+ and microvcssels were detected by immunohistochemistry and three-dimensional image respectively 14 days after MCAO. Results At day 7 and day 14 after MCAO, the mNSS of VEGF group were significantly lower than that in control group (P0. 01). Compared with control rats, the number of BrdU~+/vWF~+ cells in infarction boundary significantly increased in VEGF group (P0. 01). VEGF increased the numbers of FITC-dextran perfused vessels in infarction boundary regions. Conclusions Intranasal administration of VEGF could improve angiogenesis in the infarction boundary following cerebral infarction in rats, and improve neurological recovery functionally.
