Abstract:An optical setup that can generate a focused light spot, with its shape tunable from a very sharp peak-centered beam to a donut-shaped dark-centered beam, is always desired in optical tweezing. This setup is now realized by incorporating an image inverting interferometer (III) and a helical phase element into a radially polarized beam generator, which enables the trapping and control of a large variety of particles with a single optical tweezing system. The performance of this system is investigated theoretically.

Abstract:Recent advances in the field of optical tweezer technology have shown intriguing potential for applications in cardiovascular medicine, bringing this laboratory nanomechanical instrument into the spotlight of translational medicine. This article summarizes cardiovascular system findings generated using optical tweezers, including not only rigorous nanomechanical measurements but also multifunctional manipulation of biologically active molecules such as myosin and actin, of cells such as red blood cells and cardiomyocytes, of subcellular organelles, and of microvessels in vivo . The implications of these findings in the diagnosis and treatment of diseases, as well as potential perspectives that could also benefit from this tool, are also discussed.

Abstract:Manipulating very small objects has been a big challenging task in many areas. As the most basic tool to manipulate, fabricate, characterize, assemble and test the micro/nano scale devices and biological samples, micromanipulation keeps receiving attentions over the area of micro/ nano technology especially micro automation. Research along this direction is becoming a major stream in the world. This paper summarized and reviewed the latest advancement in developing the micro/nano manipulation method. According to the working principles, classifications on the currently available micro/nano tweezers were made. Particularly, those enabled from using the mechanical, hydraulic, electrical, magnetic, acoustic, optical, thermal effects or their combinations were reviewed. Some of their typical applications in manipulating the micro/nano objects were introduced. It indicates that advancement of the micro/nano tweezer technologies will create more powerful tools in studying and implementing the micro world.

Abstract:Near-field optical tweezers is a new technology based on the near-field optical theory, which can stably trap particles. Compared with conventional far-field optical tweezers based on the single-beam gradient force, near-field optical tweezers which overcomes the diffraction limitation of near-field light optical resolution, thermal effects and many other disadvantages, can achieve the capture on particles in nano order, and have aroused much attention in the field of physics, cell engineering, biomedicine, and so on. Firstly, the model of near-field optical tweezers based on evanescent field and the basic principles of trapping were described. Secondly, the development of total reflection prism optical tweezers, probe type optical tweezers, nano-pore optical tweezers, focused evanescent field optical tweezers, micro-nano fiber optical tweezers, and micro-resonator coupling structure type near-field optical tweezers was introduced. In the end, the applications of optical tweezers in the field of biomedicine were mainly introduced.

Abstract:Optical tweezers (OTs) can transfer light momentum to particles, achieving the precise manipulation of particles through optical forces. Due to the properties of non-contact and precise control, OTs have provided a gateway for exploring the mysteries behind nonlinear optics, soft-condensed-matter physics, molecular biology, and analytical chemistry. In recent years, OTs have been combined with microfluidic chips to overcome their limitations in, for instance, speed and efficiency, creating a technology known as "optofluidic tweezers." This paper describes static OTs briefly first. Next, we overview recent developments in optofluidic tweezers, summarizing advancements in capture, manipulation, sorting, and measurement based on different technologies. The focus is on various kinds of optofluidic tweezers, such as holographic optical tweezers, photonic-crystal optical tweezers, and waveguide optical tweezers. Moreover, there is a continuing trend of combining optofluidic tweezers with other techniques to achieve greater functionality, such as antigen-antibody interactions and Raman tweezers. We conclude by summarizing the main challenges and future directions in this research field.

Abstract:As a versatile tool for trapping and manipulating neutral particles, optical tweezers have been studied in a broad range of fields such as molecular biology, nanotechnology, and experimentally physics since Arthur Ashkin pioneered the field in the early 1970s. By levitating the sensor with a laser beam instead of adhering it to solid components, excellent environmental decoupling is achieved. Furthermore, unlike levitating particles in liquid or air, optical tweezers operating in vacuum are isolated from environmental thermal noise, thus eliminating the primary source of dissipation present for most inertial sensors. This attracted great attention in both fundamental and applied physics. In this paper we review the history and the basic concepts of optical tweezers in vacuum and provide an overall understanding of the field.

Abstract:As noninvasive tools of high-resolution micromanipulation and force measurement, optical tweezers have been widely applied to researches in life science. Holographic optical tweezers show higher flexibility than the conventional single-trap optical tweezers in producing arbitrarily patterned trap arrays with the help of the spatial light modulator, which has significant potential in application to the biomedical research. In this paper, we review the basic principle of holographic optical tweezers, hologram algorithm, and the progress in the applications of holographic optical tweezers in biological research. We expect that this review will provide a helpful reference to the community that will apply holographic optical tweezers to the biological research.

Abstract:Optical tweezers have been widely used for optical trapping and manipulating particles at the microscopic scale with a tightly focused laser beam. Over the recent years, optical fiber tweezers have attracted extensive attention due to their simple fabrication process, compact structure, strong anti-interference ability, convenient utilization, compatibility with chip devices, and flexible operation. In this review, the principle of optical force-based trapping is described, and the different fabrication methods of optical fiber tweezers are comprehensively discussed, including heating and pulling, chemical etching, grinding and polishing, coaxial splicing, photopolymer manufacturing, focused ion beam milling, and other emerging super-surface processing techniques. Then, the applications of such tweezers in optical manipulation, optical drive, optical transportation, optical imaging, and optical measurement are summarized. Finally, the future development prospects of optical fiber tweezers are presented.

Since their introduction in 1986, the optical tweezer has become an important tool for research in the fields of biology, physical chemistry and soft condensed matter physics. Recent advances promise to take optical tweezers out of the laboratory and into the mainstream of manufacturing and diagnostics; they may even become consumer products. The next generation of single-beam optical traps offers revolutionary new opportunities for fundamental and applied research.

Abstract:Since their invention in 1986 by Arthur Ashkin and colleagues, optical tweezers have become an essential tool in several fields of physics, spectroscopy, biology, nanotechnology, and thermodynamics. In this Tutorial, we provide a primer on how to calibrate optical tweezers and how to use them for advanced applications. After a brief general introduction on optical tweezers, we focus on describing and comparing the various available calibration techniques. Then, we discuss some cutting-edge applications of optical tweezers in a liquid medium, namely to study single-molecule and single-cell mechanics, microrheology, colloidal interactions, statistical physics, and transport phenomena. Finally, we consider optical tweezers in vacuum, where the absence of a viscous medium offers vastly different dynamics and presents new challenges. We conclude with some perspectives for the field and the future application of optical tweezers. This Tutorial provides both a step-by-step guide ideal for non-specialists entering the field and a comprehensive manual of advanced techniques useful for expert practitioners. All the examples are complemented by the sample data and software necessary to reproduce them.

Abstract:In the past three decades, the ability to optically manipulate biomolecules has spurred a new era of medical and biophysical research. Optical tweezers (OT) have enabled experimenters to trap, sort, and probe cells, as well as discern the structural dynamics of proteins and nucleic acids at single molecule level. The steady improvement in OT’s resolving power has progressively pushed the envelope of their applications; there are, however, some inherent limitations that are prompting researchers to look for alternatives to the conventional techniques. To begin with, OT are restricted by their one-dimensional approach, which makes it difficult to conjure an exhaustive three-dimensional picture of biological systems. The high-intensity trapping laser can damage biological samples, a fact that restricts the feasibility of in vivo applications. Finally, direct manipulation of biological matter at nanometer scale remains a significant challenge for conventional OT. A significant amount of literature has been dedicated in the last 10 years to address the aforementioned shortcomings. Innovations in laser technology and advances in various other spheres of applied physics have been capitalized upon to evolve the next generation OT systems. In this review, we elucidate a few of these developments, with particular focus on their biological applications. The manipulation of nanoscopic objects has been achieved by means of plasmonic optical tweezers (POT), which utilize localized surface plasmons to generate optical traps with enhanced trapping potential, and photonic crystal optical tweezers (PhC OT), which attain the same goal by employing different photonic crystal geometries. Femtosecond optical tweezers (fs OT), constructed by replacing the continuous wave (cw) laser source with a femtosecond laser, promise to greatly reduce the damage to living samples. Finally, one way to transcend the one-dimensional nature of the data gained by OT is to couple them to the other large family of single molecule tools, i.e., fluorescence-based imaging techniques. We discuss the distinct advantages of the aforementioned techniques as well as the alternative experimental perspective they provide in comparison to conventional OT.

Abstract:Optical tweezers have become the method of choice in single-molecule manipulation studies. In this Primer, we first review the physical principles of optical tweezers and the characteristics that make them a powerful tool to investigate single molecules. We then introduce the modifications of the method to extend the measurement of forces and displacements to torques and angles, and to develop optical tweezers with single-molecule fluorescence detection capabilities. We discuss force and torque calibration of these instruments, their various modes of operation and most common experimental geometries. We describe the type of data obtained in each experimental design and their analyses. This description is followed by a survey of applications of these methods to the studies of protein–nucleic acid interactions, protein/RNA folding and molecular motors. We also discuss data reproducibility, the factors that lead to the data variability among different laboratories and the need to develop field standards. We cover the current limitations of the methods and possible ways to optimize instrument operation, data extraction and analysis, before suggesting likely areas of future growth.

Abstract:Abstract The possibility for the manipulation of many different samples using only the light from a laser beam opened the way to a variety of experiments. The technique, known as Optical Tweezers, is nowadays employed in a multitude of applications demonstrating its relevance. Since the pioneering work of Arthur Ashkin, where he used a single strongly focused laser beam, ever more complex experimental set-ups are required in order to perform novel and challenging experiments. Here we provide a comprehensive review of the theoretical background and experimental techniques. We start by giving an overview of the theory of optical forces: first, we consider optical forces in approximated regimes when the particles are much larger (ray optics) or much smaller (dipole approximation) than the light wavelength; then, we discuss the full electromagnetic theory of optical forces with a focus on T-matrix methods. Then, we describe the important aspect of Brownian motion in optical traps and its implementation in optical tweezers simulations. Finally, we provide a general description of typical experimental setups of optical tweezers and calibration techniques with particular emphasis on holographic optical tweezers.
Microrobots and micromachines based on optoelectronic tweezers

Abstract:Optoelectronic tweezers (OET) are a state-of-art technique that combines light stimuli with electric field together by leveraging the photoconductive effect of semiconductor materials. Herein, the behavior of microobjects can be directly controlled by inducing the change of electric fields on demand in an optical manner. Relying on this light-induced electrokinetic effect, OET offer tremendous advantages in micromanipulation such as programmability, flexibility, versatility, high-throughput, and ease of integration with other characterization systems, thus showing impressive performance compared to those of many other manipulation techniques. A lot of research on OET has been reported in recent years and the technology has developed rapidly in various fields of science and engineering. In this chapter, we first introduce the working mechanism and typical structures of OET devices. Then, applications of OET in cell manipulation, cell analysis and cell electroporation are discussed. In the following part, OET-actuated microrobots and micromachines are introduced, including their working principles and applications. Finally, we overview the recent progress in the commercialization of OET technology for biopharmaceutical applications and end the chapter with challenges and future perspectives.

Abstract:Since the pioneering work of Arthur Ashkin, optical tweezers have become an indispensable tool for contactless manipulation of micro- and nanoparticles. Nowadays optical tweezers are employed in a myriad of applications demonstrating the importance of these tools. While the basic principle of optical tweezers is the use of a strongly focused laser beam to trap and manipulate particles, ever more complex experimental set-ups are required in order to perform novel and challenging experiments. With this article, we provide a detailed step- by-step guide for the construction of advanced optical manipulation systems. First, we explain how to build a single-beam optical tweezers on a home-made microscope and how to calibrate it. Improving on this design, we realize a holographic optical tweezers, which can manipulate independently multiple particles and generate more sophisticated wavefronts such as Laguerre-Gaussian beams. Finally, we explain how to implement a speckle optical tweezers, which permit one to employ random speckle light fields for deterministic optical manipulation.

Abstract:Abstract Optical tweezers and associated manipulation tools in the far field have had a major impact on scientific and engineering research by offering precise manipulation of small objects. More recently, the possibility of performing manipulation with surface plasmons has opened opportunities not feasible with conventional far-field optical methods. The use of surface plasmon techniques enables excitation of hotspots much smaller than the free-space wavelength; with this confinement, the plasmonic field facilitates trapping of various nanostructures and materials with higher precision. The successful manipulation of small particles has fostered numerous and expanding applications. In this paper, we review the principles of and developments in plasmonic tweezers techniques, including both nanostructure-assisted platforms and structureless systems. Construction methods and evaluation criteria of the techniques are presented, aiming to provide a guide for the design and optimization of the systems. The most common novel applications of plasmonic tweezers, namely, sorting and transport, sensing and imaging, and especially those in a biological context, are critically discussed. Finally, we consider the future of the development and new potential applications of this technique and discuss prospects for its impact on science.

Abstract:Magnetic tweezers,as one of micro-manipulation techniques,was developed in the field of life science research in recent years.The magnetic tweezers built by some assembly required,and the micromanipulation experiment accomplished by the 2.8-diameter beads,were introduced.The recorded video was analyzed by search algorithm,thus the motion trail and velocity of the beads can be calculated,that obtained the same result as the theoretical analysis.The magnetic tweezers is proved to be practical.
