Abstract:The induction heating of magnetic materials implanted into malignant tumors used for magnetic induction hyperthermia is an emerging cancer treatment technology in recent years. It is shown that when a tumor tissue is heated to 40～70℃, the deposited thermal energy has a specific killing effect to cancerous cells and also a preventive effect against tumor formation, recurrence and metastasis. Besides, the hyperthermia treatment can also induce an anticancer immunological function in patients' body, thus further improve the curative effect. With the excellent performance in accuracy of targeting and special treatment effect, the magnetic induction hyperthermia for cancer treatment is drawing more and more attention in clinical antitumor researches. In this paper, the research progress of magnetic induction hyperthermia by using multiple magnetic materials in the laboratory of novel medical technology in Department of Engineering Physics, Tsinghua University is reviewed, including relevant aspects of magnetic field facility developments, magnetic material researches and hyperthermia-related biomedical researches. Further studies of magnetic induction hyperthermia treatment are to be carried out for a plan of Phase-I clinical trials.
Development of AC magnetic heating setup for cancer therapy with magnetic fluid hyperthermia

Abstract:The setup produces an AC (alternating current) magnetic field at the gap between the magnetic poles. Ferrite core in excitation winding enhances the magnetic field intensity, and bridge topology in circuit achieves high efficiency of power conversion. The frequency and magnetic field intensity are 25-120 kHz and 6-16 kA/m, respectively. Intensity measurement shows the uniformity of the magnetic field. The temperature increment of the magnetic nano-materials can be up to 28°C with magnetic heating under room temperature. The results suggest a way for clinical application in the magnetic fluid hyperthermia (MFH) cancer therapy.

Abstract:Targeted magnetic induction cancer therapy is expected to offer a new breakthrough in tumor treatment.Magnetic induction hyperthermia(MIH) for cancer treatment is a technique that has undergone extensive research and development.Recent studies in Germany,the United States and Japan have made significant progress and led to clinical trials.Furthermore,some research institutions in China have conducted in-depth research.In particular,with more than 10 years of relentless effort and exploration,Tsinghua University has many achievements in the research and development of MIH in China,particularly in terms of the hyperthermia system used for MIH in clinical trials.This paper summarizes the international and local research and development of MIH for cancer treatment.

Abstract:Magnetic induction hyperthermia (MIH), based on the Neer relaxation heat generation mechanism of the magnetic medium in an alternating magnetic field, is an adjuvant treatment technology for tumor lesions. A precise temperature control of the hyperthermia region is essential in order to ensure a relatively desired therapeutic. In this paper, an automatic shape optimization procedure is implemented to acquire tumor lesion optimal temperature distribution produced by a uniform magnetic field generating device using magnetic-thermal coupled finite element analysis and multi-objective optimization algorithms. The effect of multi-injection and blood perfusion is considered. The simulation result shows that an appropriate device shape and injection point location can produce a relatively ideal temperature field distribution in the tumor hyperthermia region.

Abstract:Magnetic nanoparticle-mediated hyperthermia (MHT) is a promising tumor theranostic technology due to its noninvasive nature and ability to penetrate deep tissues without greatly damaging normal tissues. To advance the clinical translation and application of MHT, we present a comprehensive overview of topics related to MHT, including the basic physical heating principles, magnetic nanoparticle design, biological effects and theranostic applications. First, the fundamental physical principles through which magnetic nanoparticles mediate hyperthermia are reviewed in detail. Subsequently, strategies to increase the magnetothermal effect and biological effects of MHT are highlighted. Then, MHT and multitechnology integration applications in precision diagnosis and treatment are introduced. Finally, the key challenges and outlooks of MHT for clinical purposes are discussed.

Abstract:The magnetic induction hyperthermia for malignant tumor is a kind of new techniques where the heat is generated to kill the tumor lesion tissues through the magnetic media by the application of alternating magnetic fields.Since the development of the inductive heat treatment in the 1960s,many attempts were made to improve the effect of the heat treatment,to develop magnetic mediums with multimodality function and to optimize the temperature field distribution,for achieving numerous excellent results.This paper reviews the experimental studies and clinical applications of several magnetic materials,such as the ferromagnetic thermal seed,the magnetic liquid,the magnetic liposomes,the magnetic glass ceramic and the magnetic bone cement.In the meantime,the challenges in improving the efficiency of therapy and optimizing the heat field are also discussed.

Abstract:Hyperthermia has become one of the most important means for tumor therapy in recent years,but there are some limitations.Magnetic fluid hyperthermia (MFH) as a newly developed means of hyperthermia has overcome the defects of conventional hyperthermia.It can be an assisted therapy,and even developed into an independent treatment.Therefore,the recent developments in the field of basic research and experimental research on magnetic fluid hyperthermia are reviewed in this article.The magnetic fluid characters,common magnetic fluids and alternating magnetic field devices are first introduced.Then,the research on in vitro trials,animal experiments and clinical studies about magnetic fluid tumor hyperthermia are emphatically presented.Although magnetic fluid hyperthermia has gradually entered into the clinical stage,it still has various problems and needs to further improve the treatment effect.

Abstract:Magnetic induction hyperthermia (MIH), is a technique that has developed rapidly in recent years in the field of tumor thermotherapy. It implants a magnetic heating medium (millimeter-sized heat seeds, micron-sized magnetic particles and nanometer-sized magnetic fluids, etc.) inside the tumor. The material heats up under the induction of an external alternating magnetic field (100–500 kHz), which causes a high temperature zone to rapidly form in the local biological tissues and induces apoptosis in tumor cells. Magnetic induction hyperthermia has the advantages of high safety, strong targeting, repeatable treatment, and the size of the incision during treatment is negligible compared to surgical resection, and is currently used in clinical treatment. However, the millimeter-scale heat seed heating that is typically used in treatments can result in uneven temperatures within the tissue. Common MIH heating devices are bulky and complex in design, and are not easy for medical staff to get their hands on, which are issues that limit the diffusion of MIH. In this view, this paper will discuss the basic theoretical research on MIH and the progress of MIH-related technologies, with a focus on the latest research and development results and research hotspots of nanoscale ferromagnetic media and magnetic heat therapy devices, as well as the validation results and therapeutic efficacy of the new MIH technology on animal experiments and clinical trials. In this paper, it is found that induction heating using magnetic nanoparticles improves the uniformity of the temperature field, and the magneto-thermal properties of nanoscale ferromagnetic materials are significantly improved. The heating device was miniaturized to simplify the operation steps, while the focusing of the magnetic field was locally enhanced. However, there are fewer studies on the biotoxicity aspects of nanomedicines, and the localized alternating magnetic field uniformity used for heating and the safety of the alternating magnetic field after irradiation of the human body have not been sufficiently discussed. Ultimately, the purpose of this paper is to advance research related to magnetic induction thermotherapy that can be applied in clinical treatment.

Abstract:Magnetic hyperthermia (MH) has been introduced clinically as an alternative approach for the focal treatment of tumors. MH utilizes the heat generated by the magnetic nanoparticles (MNPs) when subjected to an alternating magnetic field (AMF). It has become an important topic in the nanomedical field due to their multitudes of advantages towards effective antitumor therapy such as high biosafety, deep tissue penetration, and targeted selective tumor killing. However, in order for MH to progress and to realize its paramount potential as an alternative choice for cancer treatment, tremendous challenges have to be overcome. Thus, the efficiency of MH therapy needs enhancement. In its recent 60-year of history, the field of MH has focused primarily on heating using MNPs for therapeutic applications. Increasing the thermal conversion efficiency of MNPs is the fundamental strategy for improving therapeutic efficacy. Recently, emerging experimental evidence indicates that MNPs-MH produces nano-scale heat effects without macroscopic temperature rise. A deep understanding of the effect of this localized induction heat for the destruction of subcellular/cellular structures further supports the efficacy of MH in improving therapeutic therapy. In this review, the currently available strategies for improving the antitumor therapeutic efficacy of MNPs-MH will be discussed. Firstly, the recent advancements in engineering MNP size, composition, shape, and surface to significantly improve their energy dissipation rates will be explored. Secondly, the latest studies depicting the effect of local induction heat for selectively disrupting cells/intracellular structures will be examined. Thirdly, strategies to enhance the therapeutics by combining MH therapy with chemotherapy, radiotherapy, immunotherapy, photothermal/photodynamic therapy (PDT), and gene therapy will be reviewed. Lastly, the prospect and significant challenges in MH-based antitumor therapy will be discussed. This review is to provide a comprehensive understanding of MH for improving antitumor therapeutic efficacy, which would be of utmost benefit towards guiding the users and for the future development of MNPs-MH towards successful application in medicine.

Abstract:Magnetic induction hyperthermia, as a new heating technology in the treatment of malignant tumors, is to transfer the magnetic mediums into the target tumor tissues through various procedures. Magnetic mediums produce heat to kill tumor cells directly or induce tumor cell apoptosis under the extracorporeal alternating magnetic field. This paper discussed the influence of magnetic mediums, the key link in this technology, upon the effect of tumor hyperthermia. According to their physical size of different levels, respectively, we discussed the thermal effects and influential factors of mm-scale magnetic medium, micron-scale magnetic medium, and nano-scale magnetic medium. Mm-scale magnetic medium is used for heating deep tumors, and its materials and the distribution in vivo have great influence on magnetic induction of tumor therapy. Micron-scale magnetic medium is commonly used for arterial embolism hyperthermia. The thermal effect of nano-scale magnetic medium in alternative magnetic field has a significant effect on hyperthermia, mainly due to particle concentration, particle size and surface treatment and other factors. In addition, the curie point phenomenon of magnetic medium achieves self-regulating temperature in treatment, and the pattern of magnetic medium into tissues will also affect the hyperthermia targeting in vivo. To improve the histocompatibility of magnetic medium can ensure the clinical safety of the magnetic induction hyperthermia in tumor therapy.

Abstract:Magnetic inductive oncotherapy provides strong targeting, self-regulating, and good biocompatibility compared with ultrasound hyperthermia, microwave heating, radiofrequency capacitive heating method, etc.. In this method, ferromagnetic material was implanted into the tumor tissue. When applied alternating magnetic field, ferromagnetic materials produce heat, and kill the tumor tissue without damaging normal tissue. In this paper, the heating mechanism and characteristic of micrometer and nanometer ferromagnetic materials were discussed. The researching status of ferromagnetic materials used in magnetic inductive therapy was described, and the problems and prospect along the direction were proposed.

Abstract:Hyperthermia therapy for cancer has attracted much attention nowadays. The study on the heat transfer in the magnetic fluid and the tumor is crucial for the successful application of magnetic fluid hyperthermia (MFH). Water-based Fe3O4 magnetic fluid is expected to be a most appropriate candidate for MFH due to the good biocompatibility, high saturation magnetization, super-paramagnetization and high chemical stability. In this paper, we explore the heat generation and transfer in magnetic fluid which is placed under an AC magnetic field. It is found that the amplitude and the frequency of alternating magnetic field, particle size and volume fraction have a pronounce influence on maximum temperature of hyperthermia.

Abstract:Introduction: The temperature control of magnetic hyperthermia therapy mainly relies on circulating water cooling and regulating magnetic field intensity, which increases complexity in clinical applications. Using magnetic materials with appropriate Curie temperature has become an effective means to solve temperature monitoring and potentially achieve self-regulating temperature. Methods: A self-temperature-regulating Fe83Zr10B7 magnetic material was prepared. Based on this material, a simplified model of magnetic hyperthermia for arm tumors was established and verified using the finite- element method. The influence of magnetic field intensity and frequency on the heating power and temperature rise rate of different-sized and shaped magnetic media was studied. Additionally, factors such as the size, quantity, and spatial arrangement of the magnetic media were analyzed for their impact on the damage to tumors with different volumes and shapes. Results: Spherical shape is the most suitable for magnetic hyperthermia media, and the radius of the spherical magnetic media can be chosen according to the size of the tumor. For tumors with a radius below 10 mm, using magnetic media with a particle size of 3.5 mm is recommended. The optimal magnetic field conditions are H0 (10-12 kA/m) and f (110-120 kHz). Conclusion: Based on the good magnetic properties and heating performance of the Fe83Zr10B7 magnetic material, it is feasible to use it as a magnetic medium for magnetic hyperthermia. The results of this study provide references for the selection of thermal seed size and magnetic field parameters in magnetic hyperthermia.

Abstract:Magnetic hyperthermia (MH) utilizes magnetic iron oxide nanomaterials (MIONs) to generate nano-scale heat and boost reactive oxygen species production within cells exposed to an external alternating magnetic field. Unlike conventional thermal ablation therapies that produce heat on a macro-scale, MIONs act as point source of heat inside cells, which enables MIONs-mediated MH to modulate cellular functions and fate with precision in real-time. With key benefits such as deep tissue penetration and the ability to regulate processes in a temporal-spatial and quantifiable manner, MH is now emerging as a new cancer therapy. Most intriguing is the apparent ability for MH to alter specific biological pathways associated with an anti-tumor immune response. Research efforts are now accelerating to render MH applicable to the clinical setting, with the objective of supporting the treatment of common cancers such as hepatocellular carcinoma (HCC). In this perspective paper, we highlight the recent progress made in MH, with a particular focus on its ability to manipulate anti-tumor immune mechanisms and the therapeutic advantages demonstrated thus far for HCC. We explore the current challenges in this field, and provide our perspective on the outlook for MH and its role in cancer treatment.

Abstract:This paper proposes a medium frequency magnetothermal treatment method, which can be used for a certain depth of thermal hyperthermia of tumor tissue in vivo. Firstly, a magnetothermal treatment program using a vortex coil is designed with a medium frequency alternating magnetic field, which can better realize the external implementation. Next, the electromagnetic field of the vortex coil is simulated by finite element method, and the electromagnetic field distribution characteristics of the system are obtained for treatment reference. Then, the preparation and composition design of the gel suitable for the magnetothermal effect are studied, and the temperature rise experiment is carried out to verify the reliability of the temperature rise of the magnetothermal gel. Finally, the human thoracic cavity tissue model is established to calculate the magnetothermal effect of the gel on the lung tumor tissue. And the thermal effect and temperature distribution of different physical parameters on the tissue are obtained. The results show that the magnetothermal therapy can penetrate non-magnetic barriers and effectively generate a temperature rise on the magnetothermal gel injected into the body, so that the tumor tissue in a certain space is inhibited by the temperature rise in a short time. Therefore, the research results in this article provide important theoretical and clinical value for non-invasive hyperthermia of tumors in vivo.
