Abstract:Wild rice has adapted to weather and unfavorable environments under natural selection. It has been well recognized as a natural gene bank that conserves a lot of specific genes presently not available for extinct in the cultivated rice. There is an urgent need to explore these specific genes. The present paper summarized current researches in molecular mapping and cloning of useful genes from wild rice,and their potential application in breeding.

Abstract:Common wild rice (Oryza rufipogon Griff.), known as the ancestor of Asian cultivated rice (Oryza sativa L.), is the most important germplasm for rice improvement. The first male sterility gene was found in the wild rice, and introduced to the cultivated rice, which launched the fast development of the high-yielding hybrid rice. Other agronomically beneficial traits in the wild rice, such as rice tungro virus resistance, bacterial leaf blight (Xa21 gene) resistance and acid sulfate soil tolerance, have played important roles in rice breeding. China has the northernmost distribution area of wild rice possessing great genetic diversity. However, most of the populations of this species have disappeared in China over the last three decades, mainly caused by habitat loss, fragmentation and other human disturbances. Unfortunately, the decline of existing populations still continues. In the present study, we reviewed studies on genetic diversity and conservation of this wild rice in China, concentrating on population structure, pollen competition, pollen/gene flow from cultivated rice to wild rice, and ecological restoration in relation to in situ conservation. The relatively high genetic diversity of populations of O. rufipogon in China suggests that there is great value for conservation. Considerable gene flow from cultivated rice to wild rice may alter the genetic structure of natural populations of O. rufipogon and eventually lead to its genetic erosion. Pollen competition between wild and cultivated rice has caused a low rate of crop-to-wild gene flow, but it does not completely prevent gene flow from the crop. Effective isolation measures should be undertaken in the regions where in situ conservation of O. rufipogon is carried out. Reintroduction is an important alternative for the in situ conservation of wild rice species. As wild rice is an important genetic resource, both in situ and ex situ conservation strategies are needed.

Abstract:[Objective] To confirm genetic diversity distribution center of common wild rice,Oryza rufi-pogon Griff. , in Guangxi, and construct the core collection which will provide a reference for protecting and utilizing wild rice resources in Guangxi. [Method] A total number of 623 common wild rice re-sources from Yujiang River Basin,Hongshui River Basin,Nanliujiang River Basin and Guibei Mountain were used to study the genetic diversity with 24 SSR markers. The stepwise cluster analysis was used to construct 10% and 5% core collections of common wild rice in Guangxi. [Result]There were 114 alleles to be detected at 24 SSR loci. The number of alleles per locus (A) was 4. 75, the average number of ef-fective alleles (Ae) was 3. 000 1, the Shannon-weaver information index (I) was 1. 180 1 and the aver-age expected heterozygosity ( He ) was 0. 638 8. The order of genetic diversity of 9 regional populations was Yongning ( YN) > Lingui ( LG) > Fusui ( FS) > Rongxian ( RX) > Guigang ( GG) > Pingnan ( PN) > Gupeng ( GP) > Wulitang ( WLT) > Bobai ( BB) . The order of genetic diversity of four re-gions was Yujiang River Basin>Guibei Mountain > Nanliujiang River Basin > Hongshui River Basin. There were 31 copies belong to 5% core collection,including 14 copies of Yongning population and 12 copies of Fusui population which accounted for 5. 76% and 18. 75% of analyzed samples respectively.[Conclusion]The genetic diversity center of Guangxi common wild rice is Yujiang River Basin. Wild rice of Yongning is widely distributed and presents various types. Wild rice of Fusui shows great rich genetic diversity. Yongning and Fusui regional populations,which are main sources of the core collection sam-ples,are considered to be the most important populations of Guangxi common wild rice, and should be paid particular attention to their protection and exploitation.
[Genetic Diversity of Common Wild Rice (oryza Rufipogon Griff) by Using SSR Markers and Phenotypic Traits in Guangxi Province].

Abstract:In this paper, allelic variation and genetic diversity among geographies and growth habit types were studied by using 223 accessions of common wild rice primary core collection in Guangxi Province, with 34 SSR primers locating on 12 chromosomes of rice and 19 phenotypic traits. In the results, 24.91 alleles were detected per locus on average with a range from 7 to 48. Compared to the cultivated rice, the wild rice showed more allelic variations. The ratio of heterozygote of SSR locus was 32.01% on average, and it's range was 1.35% 81.31%. The frequency of heterozygote of SSR locus in Oryza rufipogon Griff was much higher than in Oryza sativa L. The geographical distribution of genetic diversity measured by SSR markers was not completely accordant with that by phenotypic traits. At DNA level, more wild rice individuals and higher genetic diversity were included within the area covering north latitude 22 degrees - 23 degrees and 23 degrees - 24 degrees (comprising Longan, Fusui, Yongning, Xiangzhou, Laibin, Xuanwu, Yulin and Guigang county), which formed the center of genetic diversity. But the center of genetic diversity at the phenotypic level located within north latitude 21 degrees - 22 degrees and 22 degrees - 23 degrees. Among the four growth habit types, the genetic diversity from high to low was found respectively in prostrate type, sloping type, slighting type, and erect type at both DNA and phenotypic levels.

Abstract:Using 36 SSR markers and 889 accessions of common wild rice in China, the genetic diversity and the divergence among different geographical populations are investigated. Guangdong Province has the largest number of alleles, which account for 84% of the total alleles detected in the study, followed by Guangxi Province. The Nei's gene diversity indices, from high to low, are in the sequence of Hainan, Guangdong, Guangxi, Fujian, Hunan, Jiangxi, and Yunnan provinces. Two genetic diversity centers of Chinese common wild rice are detected on the basis of geographic analysis, i.e., the region covering Boluo, Zijin, Lufeng, Haifeng, Huidong and Huiyang counties of Guangdong Province and the region covering Yongning, Longan, Laibin and Guigang counties of Guangxi Province. The common wild rice in Yunnan, Hunan, Jiangxi, and Fujian provinces are diverged into respectively independent populations with relatively large genetic distances, whereas, those in Hainan, Guangdong and Guangxi provinces have relatively low genetic divergence. Under the condition of geographic separation, natural selection is considered as one of the primary forces contributing to the divergence of common wild rice in China.

Abstract:Common wild rice (Oryza rufipogon Griff.), the progenitor of Asian cultivated rice (O. sativa L.), is endangered due to habitat loss. The objectives of this research were to evaluate the genetic diversity of wild rice species in isolated populations and to develop a core collection of representative genotypes for ex situ conservation. We collected 885 wild rice accessions from eight geographically distinct regions and transplanted these accessions in a protected conservation garden over a period of almost two decades. We evaluated these accessions for 13 morphological or phenological traits and genotyped them for 36 DNA markers evenly distributed on the 12 chromosomes. The coefficient of variation of quantitative traits was 0.56 and ranged from 0.37 to 1.06. SSR markers detected 206 different alleles with an average of 6 alleles per locus. The mean polymorphism information content (PIC) was 0.64 in all populations, indicating that the marker loci have a high level of polymorphism and genetic diversity in all populations. Phylogenetic analyses based on morphological and molecular data revealed remarkable differences in the genetic diversity of common wild rice populations. The results showed that the Zengcheng, Gaozhou, and Suixi populations possess higher levels of genetic diversity, whereas the Huilai and Boluo populations have lower levels of genetic diversity than do the other populations. Based on their genetic distance, 130 accessions were selected as a core collection that retained over 90% of the alleles at the 36 marker loci. This genetically diverse core collection will be a useful resource for genomic studies of rice and for initiatives aimed at developing rice with improved agronomic traits.

Abstract:Oryza rufipogon Griff. is a valuable germplasm resource for rice genetic improvement. However, natural habitat loss has led to the erosion of the genetic diversity of wild rice populations. Genetic diversity analysis of O. rufipogon accessions and development of the core collection are crucial for conserving natural genetic diversity and providing novel traits for rice breeding. In the present study, we developed 1,592 SNPs by multiplex PCR and next-generation sequencing (NGS) technology and used them to genotype 998 O. rufipogon accessions from 14 agroclimatic zones in Guangdong and Hainan Provinces, China. These SNPs were mapped onto 12 chromosomes, and the average MAF value was 0.128 with a minimum of 0.01 and a maximum of 0.499. The O. rufipogon accessions were classified into ten groups. The mean Nei's diversity index and Shannon–Wiener index (I) were 0.187 and 0.308, respectively, in all populations, indicating that O. rufipogon accessions had rich genetic diversity. There were also differences in the genetic diversity of O. rufipogon resources in the 14 regions. Hainan populations possessed higher levels of genetic diversity, whereas the Guangzhou population had lower levels of genetic diversity than did the other populations. Phylogenetic analysis revealed that the genetic relationship among the distribution sites of O. rufipogon was closely related to geographical location. Based on genetic distance, a core collection of 299 accessions captured more than 99% of the genetic variation in the germplasm. This study provides insights into O. rufipogon conservation, and the constructed core collection provides valuable resources for future research and genomics-assisted breeding of rice.

Abstract:The general aspects such as yield,quality and the resistance of hybrid rice in Guangxi have been studied using these hybrid rice combinations which have been certified and registered by Guangxi Committee for Crop Variety Registration from 2000 to 2005.It was pointed out that the yield,resistance and quality of hybrid rice could not fully meet with the real needs.It was put forward that elite germplasm of wild rice was essential for making breakthrough in hybrid rice breeding,and hybrid rice selective breeding should be strengthened.In order to enrich the diversity of hybrid rice type and further improve the yield,quality and resistance,super hybrid rice combinations should be selectively bred according to the ecological conditions in Guangxi by fully exploring the wild rice germplasm resources in Guangxi.

Abstract:Adapting to diversified natural environment in the long evolution process,wild rice has evolved rich genetic diversity and many superior characteristics including stresses tolerance,good quality and male sterility,which make wild rice the ideal genetic materials in rice breeding.The wild rice species in China mainly grow in ethical areas such as Yunnan,Guangxi and Guizhou.In fact,wild rice domestication and rice culturing have long been important components of ethical traditional culture.A lot of studies have been performed on evaluation of genetic resources,identification of excellent traits related genes and analysis of genetic diversity of wild rice in ethical areas.Some good characteristics related genes in wild rice have been introduced into the rice cultivars using the classic and modern breeding technologies,leading to elevated productivity and stress tolerance.With the advance of modern molecular biology,research and utilization of rice genetic resources in ethical area of China will be greatly deepened in future.

Abstract:Common wild rice ( Oryza rufipogon Griff.) is a valuable germplasm resource for rice genetic improvement. However, it is endangered due to habitat loss. Assessment of its genetic diversity is essential for germplasm characterization, utilization, and conservation. In the present study, we developed a 79,422-single-nucleotide polymorphism genome-wide array by specific-locus amplified fragment sequencing (SLAF-seq) and used it to genotype 110 common wild rice accessions from different agroclimatic zones in Guangdong Province, China. The number of SNPs on each chromosome was consistent with the physical length of the chromosome, and the average marker density was approximately 4.83 kb/SNP. The genome-wide linkage disequilibrium decayed to its half maximum within 0.52 kb for O. rufipogon . The common wild rice accessions were clustered into four subgroups, with some overlap in principal component analysis and a neighbor-joining tree that was also confirmed by model-based admixture. Analysis of molecular variance revealed significant genetic differentiation among individuals, accounting for 90.64% of the total molecular variance; however, the differences between the subgroups (Enping, Lufeng, Suixi and Fogang) were not significant. The significant genetic variations in resources collected in different years were observed in Enping and Suixi subgroups, but there was no significant genetic variation in Fogang subgroup. The pairwise fixation index ( F ST ) suggested that the populations from Enping and Lufeng were most differentiated while those from Enping and Fogang were least differentiated. Our results illustrated clear genetic relationships among 110 common wild rice accessions, showing a potential molecular marker-based strategy for preserving the genetic diversity and accelerating the systematic utilization of these important wild rice resources.

Abstract:The genetic diversity of rice germplasm is the basis for increases in rice yield and quality. The collection, assessment, and protection of the genetic diversity of rice germplasm is important for achieving sustainable agriculture and assuring food security. Many underdeveloped indigenous areas have abundant and valuable rice germplasm resources. However, in-depth assessments of the genetic diversity of rice germplasm from these areas and studies related to protecting these traditional cultures are not available. In this study, from 2005 to 2016, the authors have conducted in-depth evaluation of the genetic diversity of Kam fragrant glutinous rice germplasm resources in southeast Guizhou by using multidisciplinary comprehensive methods such as ethnobotany, cultural anthropology, and modern molecular markers. In total, 376 Kam fragrant glutinous rice samples from 42 villages in the Dong community in southeast Guizhou were collected. Agronomic traits of panicles were complex and exhibited diversity. Some varieties had good disease resistance and adaptation to cold and wet climates. The Dong people named the Kam fragrant glutinous rice varieties by using seven elements, including diverse traits, growth environment, and origin. Traditional folk classification, in addition to morphology and biological analysis using molecular markers, indicates that Kam fragrant glutinous rice includes 91 varieties. Kam fragrant glutinous rice comprises a very high number of varieties, most of which are japonica-type and exhibit a high level of genetic diversity. The traditional folk classification of Kam fragrant glutinous rice by the Dong community is consistent with the biological classification. The traditional naming of Kam fragrant glutinous rice provides an important reference for understanding its genetic diversity. The high level of genetic diversity in Kam fragrant glutinous rice is not only related to the natural environment of the area but also tightly linked with the abundant and diverse Dong ethnic traditional cultures, which has led to protection of Kam fragrant glutinous rice’s genetic diversity.

Abstract:The paper studied the wild rice population and phenotypic variation in which included qualitative traits and quantitative traits of a total of 23 phenotypic traits in the eastern region of Guangdong province,according to the survey data analysis,phenotypic variation,combined with DPS software SPSS and phenotypic correlation,clustering and analysis of variance.The results showed that:The eastern wild rice in a rich phenotypic variation exists,many important traits was significantly correlated,the cluster of seven eastern wild rice plants classes were significant differences between groups;the population material not only had high similarity,there is no variation of species,but also a degree of difference,with their own characteristics,eastern wild rice of the phenotypic variation with a certain regularity.

Abstract:Wild rice (Zizania latifolia) is an important genetic resource and it has important ecological functions in aquatic ecosystems as well. Here, we used landscape genetics to investigate how the landscape features, including rivers, mountains and habitat fragmentation, affect the genetic connectivity or create dispersal barriers for Z. latifolia. In this report, seventeen populations from theSanjiang Plain and its surrounding areas were genotyped by ten microsatellite markers. A high genetic differentiation and genetic discontinuity were found among populations within each river investigated, suggesting that the rivers be not acting as corridors for dispersal. Meanwhile, genetic discontinuity was detected from different sides of the Lesser Khingan and its branch, the Qinghei Mountains, demonstrating that gene flow was blocked by the complex topography of mountains. The results that historical gene flow was much higher than the contemporary gene flow might infer that recent habitat fragmentation resulted in decreased gene flow. For all sampled Z. latifolia populations, the result of low level of genetic variation (n(a) = 2.3, H-E = 0.328) and high genetic divergence (F-ST = 0.405, D-est = 0.414 and (empty set)(pt) = 0.424) was consistent with the decreased gene flow, an inbreeding systemand repeated genetic bottlenecks. A conservation strategy for protecting as many populations as possible to maximize genomic representation of the species is proposed. In addition, dredging the watercourses should be carried out to improve habitat stability and to facilitate connectivity among populations.
Diversity and Good Germplasm of Indigenous Rice Varieties in Yunnan Province

Abstract:Diversity and good germplasm were studied based on 5285 accessions in Yunnan indigenous rice varieties. The results showed that:(1) Yunnan Province is the most center of genetic diversity for Chinese cultivated rice and the central region and natural treasurehouse of good resources in China. There is great clear difference on diversity among characters, especially between prefectures,the diversity of Yunnan rice was classified into the center area, diffuse area and wasteland area.(2) Yunnan Province is the enrich center of good germplasm in Yunnan rice,Yunnan rice is the best material of researching the core collection.(3) The core collection established based on Yunnan rice resources could used to build the ideal core collection bank,but also definitude the phylogenesis law of good germplasm resources and establish the dynamic gene bank of core collection.

Abstract:Preserving many kinds of rice resources and rich variations, Guizhou Province is one of the districts with the highest genetic diversity of cultivated rice (Oryza sativa L.) in China. In the current research, genetic diversity and structure of 537 accessions of cultivated rice from Guizhou were studied using 36 microsatellite markers and 39 phenotypic characters. The results showed that the model-based genetic structure was the same as genetic-distance-based one using SSRs but somewhat different from the documented classification (mainly based on phenotype) of two subspecies. The accessions being classified into indica by phenotype but japonica by genetic structure were much more than that being classified into japonica by phenotype but indica by genetic structure. Like Ding Ying’s taxonomic system of cultivated rice, the subspecific differentiation was the most distinct differentiation within cultivated rice. But the differentiation within indica or japonica population was different: japonica presented clearer differentiation between soil-watery ecotypes than indica, and indica presented clearer differentiation between seasonal ecotypes than japonica. Cultivated rices in Guizhou revealed high genetic diversity at both DNA and phenotypic levels. Possessing the highest genetic diversity and all the necessary conditions as a center of genetic diversity, region Southwestern of Guizhou was suggested as the center of genetic diversity of O. sativa L. from Guizhou.

Abstract:As one of important original centers of Asian cultivated rice Oryza sativa L., China is abundant in genetic resources of rice and its wild relatives. They are not only useful for mountainous farmers to cope with heterogeneous microclimates, but also of extremely significance for rice genetic improvement programs in the future. Unfortunately, the combination of new conditions, including rapid population growth, new agricultural technology and swift economic & cultural changes produces a new environment. As a result, most of wild populations have disappeared, and others are at the edge of the extinction. Moreover, widespread adoption of high-yielding rice varieties (HYVs) have led to biological impoverty of rice germplasms, as local rice varieties are abandoned for modern varieties. These processes, known as “genetic erosion”, are generally summarized as two types in Chinese rice genetic resources. Based on our recent studies on the conservation of Chinese rice biodiversity, this paper reviews the biodiversity, ethnobotany and genetic erosion of rice and its wild relatives in China. Finally, some suggestions for the ways to enhance efforts to conserve rice genetic resources, including investigation of genetic erosion, studies on population genetics and conservation genetics using molecular approaches, settings of in situ & ex situ conservation priorities, significance of ethnobotanical knowledge for taking in situ conservation, and necessities of extensive participation in the conservation activities, are given.

Abstract:The widespread adoption of improved high2yield rice varieties , which in a large scale replaced tradi2 tional and local rice varieties , has resulted in a great genetic erosion of t he cultivated rice genepool.This pro2 cess has narrowed genetic background of t he cultivated rice.As a result , modern rice varieties become vulnera2 ble and can not stand t he attack of new disease and insect pests , and t he unfavorable changes of environments.Meanwhile , t he long2term use of chemical pesticides and herbicides significantly deteriorated agricultural e2 cosystem.In order to change t his vicious cycle of t he rice ecosystem , it is very important and necessary to ex2 plore and utilize t he genetic biodiversity in t he rice genepool to broaden t he genetic background of t he cultivated rice.The rice genepool encompasses varieties of Asia rice and African rice , weedy rice , wild species of rice in t he genus O ryz a , and species in t he related genera of t he tribe Oryzeae.These are t he essential germplasm re2 sources for t he furt her improvement of cultivated rices.However , due to t he change of agricultural patterns , rapid development of social economy , industrialization and urbanization , biodiversity of t he rice genepool , in2 cluding wild species of rice , is under t hreat .Many populations of wild O ryz a species have been extinct from t heir original sites or are diminishing.Therefore , it is absolutely necessary and essential to strategically safe2 guard and conserve biodiversity of t he rice genepool and to effectively utilize t he elite rice germplasm resources in rice breeding programs.This is t he only way to guarantee t he long2term and sustainable production of rice varieties.
