Mass spectrometry (MS) and chromatography have been extensively developed in the past few decades and hold a distinguished position in qualification and separation science. Abstract:Today, chromatography is the backbone of separation science and is being used in all research laboratories and pharmaceutical industries of the world. In these chromatography techniques, HPLC is one of the chromatographic techniques, which is mostly used analytical technique. Recent developments in chromatographic supports and instrumentation for liquid chromatography (LC) are enabling rapid and highly efficient separations. This article represents a brief review of HPLC along with its principle and instrumentation. It describes about new trends in HPLC such as RRLC, UPLC, UFLC and Nano LC. In this article mainly focus on detailed comparison of new developments of HPLC in terms of instrumental operating conditions, applications and advantages over HPLC of each technique. Abstract:The liquid chromatography-tandem mass spectrometry was an important technology of modern separation and analysis because of its high separating power,high sensitivity,high specificity and wide range of application.Compared with single mass spectrometry,the tandem mass spectrometry was more sensitive and selective,and it can provide more abundant information about molecular structure.Firstly,this paper explained the fundamental principle of tandem mass spectrometry,common mass analyzer and its characteristic,different tandem mode of tandem mass spectrometry and data acquisition of the liquid chromatography-tandem mass spectrometry.Secondly,the author introduced its latest progressive application in the drug metabolism,such as determination of the pharmacokinetics parameter of the drug using tandem mass spectrometry,identification of the drug metabolites using various tandem mass mode,sum of the fit and unfit of each mode,application strategy of the identification of the drug metabolites,and prospects for the application in the drug metabolism in the future.

Abstract:Biological toxin is a kind of important life phenomenon in nature, an important tool and the material basis to explore the secrets of life science and development of new drugs, and is also a key preventive treatment object in the field of food safety. Mycotoxin and marine biotoxins which are two kinds of natural toxic substances associated with food safety, have the characteristics of strong toxicity and pollute widely and a great harm to human health. Among the detection means of mycotoxin and marine biotoxin, the technique of liquid chromatography coupled with mass spectrometry (LC-MS) is widely used for its sensitive and efficient characteristics. Based on the LC-MS technique as a breakthrough point, its application in biological toxins related to food safety testing analysis in recent years were reviewed, as well as the related sample processing technology progress.

Abstract:Microchip-mass spectrometry (chip-MS) has been emerging as an excellent analytical tool in the analysis of complex biological samples. The microchip can play an important role, as in cell culture and sample-preparation steps prior to mass spectral identification, which benefit from its ability to handle small sample quantities with the potential for high-throughput parallel analysis. We describe recent progress in chip-MS, including approaches that combined microchip devices with electrospray ionization and matrix-assisted laser desorption/ionization. We then review the main applications of chip-MS in proteomics and cell analysis in the past three years. We also look at the implications for the future of the field. (C) 2013 Elsevier Ltd. Abstract:Laser desorption ionization mass spectrometry (LDI-MS) has been developed to be an essential tool for the analysis and identification of a large variety of species, ranging from small metabolites, drugs and pollutants to peptides and proteins and even to bacteria. Matrix assisted LDI-MS has shown a great power for macromolecules, but its analytical capability to low mass compounds is limited due to inherent drawbacks, including abundant interference in low mass range (<800 Da) caused by matrix self-dissociation and poor reproducibility due to inhomogeneous crystallization. Several matrix-free methods have been developed to solve these issues, mainly through altering organic matrix with inorganic nanostructured surfaces or nanomaterials as LDI medium. In the past five years, improvements on conventional silicon-based, metal-based, metal oxide-based and carbon-based LDI media as well as emerging novel materials such as 2D nanomaterials and organic frameworks have gained important progresses. Meanwhile, with the deep research in light-matter interaction, LDI mechanism studies begin to grow and become to attach great importance. Advances in both medium and mechanism promote to expand the applicable potential of nanostructure/nanomaterial-assisted LDI-MS in many aspects, including trace analysis, structural analysis and MS imaging. In this review, we survey the major progresses in this area in recent years. We also sketch remaining challenges and directions for future development.

Abstract:Since the end of 20th century, chromatographic science has experienced a rapid development in China. Along with the current developments of life science, material science, energy science and environmental science, especially the rise of researches in functional genomics, proteomics, metabolomics, glycomics etc, chromatography has got more close attention and extensive applications. During this period, the chromatography researchers of China have made significant achievements, published a considerable number of papers in the world-class academic journals and made some impacts on the international research. This review summarizes some of the representative studies of chromatographic research in China about the basic theories of chromatography, new separation and analytical methods and techniques, new columns and enrichment materials, new equipments and applications of chromatographic analysis in the last decade.

Abstract:Instrument miniaturization is one of the critical issues to improve sensitivity, speed, throughput, and to reduce the cost of analysis. Microfluidics possesses the ability to handle small sample amounts, with minimal concerns related to sample loss and cross-contamination, problems typical for standard fluidic manipulations. Moreover, the native properties of microfluidics provide the potential for high-density, parallel sample processing, and high-throughput analysis. Recently, the coupling of microfluidic devices to mass spectrometry, especially electrospray ionization (ESI) and matrix-assisted laser desorption/ionization (MALDI), has attracted an increasing interest and produced tremendous achievements. The interfaces between microfluidics and mass spectrometry are one of the primary focused problems. In this review, we summarize the latest achievements since 2008 in the field of the technologies and applications in the combining of microfluidics with ESI-MS and MALDI-MS. The integration of several analytical functions on a microfluidic device such as sample pretreatment and separations before sample introduction into the mass spectrometer is also discussed.

Abstract:The combination techniques of microfluidic devices with mass spectrometry provide a promising platform for cell analysis. As mass spectrometry owns dominant advantages of high sensitivity and high chemical specificity to screen and identify trace targets from complex matrix, it is an ideal detection method for cellular samples. This review described the recent advance in cell analysis improved by the new strategies of chip-mass spectrometry. We discussed the coupling technologies of chip to electrospray ionization mass spectrometry (ESI-MS), chip-integrated sample pretreatment methods, cellular drug metabolism, cell-cell interactions and the application of matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) in cell analysis. Future possible uses of paper or probe based ambient ionization techniques for cell analysis were presented.

Abstract:Hyphenation of electrochemistry (EC) and mass spectrometry has become a powerful tool to study redox processes. Approaches that can achieve this hyphenation include integrating chromatography/electrophoresis between electroinduced redox reactions and detection of products, coupling an EC flow cell to a mass spectrometer, and performing electrochemical reactions inside the ion source of a mass spectrometer. The first two approaches have been well reviewed elsewhere. This Minireview highlights the inherent electrochemical properties of many mass spectrometry ion sources and their roles in the coupling of electrochemistry and mass spectrometric analysis. Development of modified ion sources that allow the compatibility of electrochemistry with ionization processes is also surveyed. Applications of different in-source electrochemical devices are provided including intermediate capturing, bioanalytical studies, nanoparticle formation, electrosynthesis, and electrode imaging.
