Abstract:Three 9-wk feeding trials were performed to assess the effects of dietary protein and lipid levels on the growth, feed utilization, and body composition of turbot, Scophthalmus maximus Linnaeus, at three different growth stages with initial average weight 4.5 +/- 0.01, 59.1 +/- 0.24, and 209.1 +/- 0.21 g, respectively. Six practical test diets were formulated to contain three protein levels (45, 50, and 55%), each of which was supplemented with two lipid levels (12.0 and 16.0%), to produce a range of P : E ratios (from 87.4 to 110.0 mg protein/kcal). The results of three experiments suggested that growth performance generally improved with increasing dietary protein irrespective of dietary lipid. Fish fed diets with 55% protein (12 and 16% lipid, P : E ratio of 110.9 and 107.1 mg protein/kcal) had the highest specific growth rates (SGRs), feed efficiency ratios (FERs), and energy retention. At the same protein level, FER and protein efficiency ratio (PER) increased significantly with the increase of lipid except in large fish (initial average weight 209.1 +/- 0.21 g). SGR of medium turbot (initial average weight 59.1 +/- 0.24 g) was also improved by the lipid increase, indicating a protein-sparing effect of lipid.
Effects of A New Compound Biological Product on the Growth Performance of Turbot

Abstract:A home-made new compound biological product was added to the basic feed of turbot(Scophthalmus maximus L.) to study its effects on the growth performance of turbot.The feed groups including basic feed(group A,control),supplemented with 1.0%(group B),2.5%(group C) and 5.0%(group D) of compound biological product were used to make feed experiments,and the fish bodies were randomly selected from each group to determine their growth indexes and biochemical compositions of fish bodies.It is shown from the study results that the growth indexes including survival rate,body weight,specific growth rate,body length,total length and feed efficiency of experimental turbots from groups B,C and D were improved as compared with those from group A,the biochemical compositions of fish body such as moisture content and crude protein content were increased to a certain degree,and the contents of crude fat and ash were slightly decreased as compared with those from group A.It is shown from the experimental results that the new compound biological product added to the fish feed can effectively promote the fish growth and enhance the utilization ratio of fish feed,and can improve the nutritive components and muscle quality of fish body.It is shown from the experimental results that 2.5% of compound biological product added to the fish food is suitable.

Abstract:1. Five groups of juvenile turbot (Scophthalmus maximus) which had been given a diet free of fat for 12 weeks were given diets in which the lipid component (g/kg) was: oleic acid alone 50, oleic acid 40+linoleic acid 10, oleic acid 40+linolenic acid 10, oleic acid40+arachidonic acid 10 or oleic acid 40+cod-liver oil 10. These five experimental diets were given for 16 weeks. 2. Weight gains were highest in the group given the diet containing cod-liver oil and lowest in the groups given diets containing oleic acid alone or oleic acid+linoleic acid. Weight gains in the groups given oleic acid+arachidonic acid or linolenic acid were markedly inferior to those of the group given oleic acid+cod-liver oil. It is concluded that arachidonic acid is inferior to polyunsaturated fatty acids of the omega3 series in maintaining growth rate in turbot. 3. Fatty acid analyses of neutral lipids and phospholipids of liver and extrahepatic tissues did not suggest any evidence of desaturation of dietary oleic acid, linoleic acid or linolenic acid by the turbot. These experiments confirm previous isotopic evidence that turbot lack the necessary microsomal desaturases to perform this metabolic transformation.

Abstract:Three 56-day feeding trials were conducted to evaluate dietary lipid requirements and effects of dietary lipid levels on growth, digestive enzyme activities, antioxidant capacity, and lipid metabolism in turbot (Scophthalmus maximus L.) at three growth stages. The initial mean body weight of turbot was 9.13 ± 0.17 g, 50.10 ± 0.38 g, and 80.05 ± 0.76 g (small, medium, and large turbot), respectively. Five practical diets were formulated to contain 68.4, 93.9, 120.7, 147.8, and 171.2 g/kg lipid (L68.4, L93.9, L120.7, L147.8, and L171.2), respectively. Results of three trials showed that the weight gain rate and specific growth rate of turbot fed with L120.7, L147.8, and L171.2 diets were all significantly higher compared with turbot fed with L68.4 and L93.9 diets except for large turbot fed with the L147.8 diet. Activities of intestinal trypsin, lipase and hepatic catalase, superoxide dismutase, and total antioxidant capacity firstly increased and then decreased as dietary lipids increased. Meanwhile, malondialdehyde content decreased firstly but then increased with the elevation of dietary lipids in small and medium turbot, and it was significantly higher in the L171.2 group than the rest in large turbot. With increasing levels of dietary lipid, contents of whole-body lipid, liver lipid, serum total cholesterol, triglyceride, and low-density lipoprotein cholesterol were markedly increased at three stages. In addition, serum high-density lipoprotein cholesterol contents increased firstly and then decreased over the L120.7 group. Transcriptional levels of lipolysis-related genes lipin1 and lpl were significantly upregulated firstly and subsequently downregulated with increasing dietary lipid levels except lipin1 in medium fish. Meanwhile, a significant increase in lipogenesis-related genes lxr and pparγ expressions was detected in all fish. Based on the specific growth rate, the dietary lipid level of 130.1, 120.1, and 107.7 g/kg was optimal for the growth performance of turbot cultured at three phases, respectively. Additionally, the results indicated that high lipid diets may lead to abnormal lipid deposition and affect the physiological health of turbot.

Abstract:A 2×6 factorial experiment was conducted to evaluate the effects of chelated (Mintrex™ Zn, Zn–M) or inorganic (ZnSO4·7H2O, Zn–S) zinc as dietary zinc sources on growth, feed utilization, tissue zinc deposition and anti-oxidative responses of turbot (Scophthalmus maximus). Semi-purified diets were made to contain tricalcium phosphate and sodium phytate at levels of 2% and 0.5%, respectively, to resemble levels in practical diets. Ten experimental diets were made by adding either Zn–S or Zn–M to the basal diet to achieve five levels of dietary zinc (15, 45, 75, 105 and 135mg/kg diet) for each zinc source, respectively. The basal control diet and ten experimental diets were fed to groups (n=5) of juvenile turbot (initial mean weight: 4.78g) for 8weeks. Results showed that the specific growth rate (SGR), feed intake (FI), feed efficiency (FE), whole body and bone zinc concentration, whole body crude lipid content, serum superoxide dismutase (SOD) activity and glutathione peroxidase (GSH-PX) activity in serum or liver of turbot were significantly improved by zinc supplementation (P<0.05). There was no significant difference in the growth of turbot between the two zinc sources (P>0.05). On the basis of SGR, the dietary zinc requirement of juvenile turbot was estimated to be 60.2mg/kg, using broken-line regression analysis.

Abstract:Vitamin D (VD) plays a vital role in various physiological processes in addition to its classic functions on maintaining the balance of Ca and P metabolism. However, there still are gaps to understand in depth the issues on the precise requirement, metabolic processes and physiological functions of VD in fish. In this study, we investigated the effects of VD on the growth, intestinal health, host immunity and metabolism in turbot (Scophthalmus maximus L.), one important commercial carnivorous fish in aquaculture, through the supplementation of different doses of dietary VD3 (0, 200, 400, 800 and 1600 mu g VD3/kg diet). According to our results, the optimal VD3 level in the feed for turbot growth was estimated to be around 400 mu g/kg, whereas VD3 deficiency or overdose in diets induced the intestinal inflammation, lowered the diversity of gut microbiota and impaired the host resistance to bacterial infection in turbot. Moreover, the level of 1 alpha,25(OH)(2)D-3, the active metabolite of VD3, reached a peak value in the turbot serum in the 400 mu g group, although the concentrations of Ca and phosphate in the turbot were stable in all groups. Finally, the deficiency of dietary VD3 disturbed the nutritional metabolism in turbot, especially the metabolism of lipids and glucose. In conclusion, this study evaluated the optimal dose of dietary VD3 for turbot and provided the evidence that VD has a significant impact on intestinal health, host immunity and nutritional metabolism in fish, which deepened our understanding on the physiological functions and metabolism of VD3 in fish.
Effect of Premix in Different Levels on the Feeding and Growth of Turbot (Scophthalmus maximus)

Abstract:A feeding trial was conducted to investigate the different diets on the feeding and growth of turbot(Scophthalmus maximus).Three experimental diets included the same copmpositions except the levels of premix,diet1 16mg / g,diet2 20mg / g,diet3 24mg / g.The control diet was high quality imported feed EP4.Each diet was randomly assigned to 3samples of 100fish per aquarium(initial weight(51.00± 1.07) g).Fish were maintained at water temperature ranging from 13.0to 14.0℃,water salinity ranging from 25～28mg / g.Fish were fed twice daily for 30days.There is no significantly different on feeding rate,weight gain and specific growth rate among the different diets.The growth performance of experimental diets has reached the level of high quality imported feed.The feed conversion of D2is lower than D1and D3,the protein efficiency of D2is higher than D1and D3,but there are no significantly different.The results indicate the wide range of the turbot on premix.The 20mg / g levels of premix is appropriate to experimental feed of the turbot.
Interactive Effect of Dietary Taurine and Protein Sources on Feed Intake Growth Performance and Body Composition of Turbot (Scophthalmus maximus)

Abstract:A feeding trial was conducted to determine the dietary protein sources and taurine on feed intake,growth and metabolism of turbot (Scophthalmus maximus).Eight isonitrogenous and isolipidic diets containing fish meal (FM) or soy protein concentrate (SPC) as protein sources and 0,0.5 ％,1.0％ or 2.0％ taurine as supplementations were fed to juvenile fish for 8 weeks.Each diet was randomly assigned to triplicate groups of 18 juvenile fish (38.19±0.09 g).The results indicated that feed intake (FI),the specific growth rate (SGR),feed efficiency ratio (FE),protein efficiency ratio (PER),hepatosomatic index (HSI),serum total cholesterol concentration and serum triglycerides in fish fed FM-based diets were significantly higher than those of fish fed SPC-based diets (P＜0.05).In addition,carcass protein and lipid contents of fish fed SPC-based diets were significantly lower than those of fish fed FM-based diets (P＜0.001).SGR,FE,PER,serum total cholesterol concentration and carcass protein content were significantly improved with the increase of dietary taurine level (P＜0.05).Survival,FI,final weight,serum triglycerides,condition factor (CF) and viscerosomatic index (VSI) were not significantly different among fish fed the diets containing different levels of taurine (P＞0.05).In conclusion,the present study indicated that high SPC as protein source would reduce the growth performance of turbot.On the other hand,supplementing taurine in diets promoted the growth performance of juvenile turbot.
Feed Intake, Growth Performance and Cholesterol Metabolism in Juvenile Turbot (scophthalmus Maximus L.) Fed Defatted Fish Meal Diets with Graded Levels of Cholesterol

Abstract:A 10-week growth trial was conducted to examine the effects of dietary cholesterol on feed intake, growth performance and cholesterol metabolism in juvenile turbot (Scophthalmus maximus L.) (initial body weight 5.18±0.01g). Five isonitrogenous and isolipidic experimental diets were formulated with supplementation of 0.0, 0.5, 1.0, 1.5 and 2.0% cholesterol, and the final dietary cholesterol concentrations were 0.38, 0.76, 1.30, 1.80 and 2.22%, respectively. WGR of fish fed diets with 1.30, 1.80 and 2.22% cholesterol was significantly higher than that of fish fed the diet with 0.38% cholesterol, but no significant differences of WGR were found among fish fed diets with 1.30, 1.80 and 2.22% cholesterol. Also no significant differences were found in feed intake (FI) in all treatments. With dietary cholesterol increasing, a general upward trend of total cholesterol (TC), free cholesterol (FC), cholesterol ester (CE), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) in serum was exhibited. However, these kinds of cholesterol in serum were plateaued when fish fed diets with approximately over 1.30 to 1.80% cholesterol. The results of TC in liver and feces showed that, with increasing dietary cholesterol, only TC in feces exhibited an increasing trend. Quantitative PCR (qPCR) was used to assess the effects of dietary cholesterol on rate limiting enzyme 3-hydroxy-3-methylglutaryl-Coenzyme A (HMG-CoA) reductase in cholesterol synthesis and cholesterol 7α-hydroxylase (CYP7A1) in bile acid synthesis. HMG-CoA reductase expression in liver was significantly suppressed with increasing dietary cholesterol, while CYP7A1 expression in liver was significantly enhanced as dietary cholesterol increased. There were no significant differences of ghrelin expression in both gut and brain. It can be concluded that moderate dietary cholesterol was beneficial to growth performance, while feed intake promotion effect was limited in juvenile turbot in the present study. Feedback control might exist in the cholesterol metabolism of turbot to keep cholesterol in homeostasis.

Abstract:A ten-week feeding trial was conducted to evaluate the effect of replacing fishmeal with two differently processed cottonseed meals (CSM), namely XC and SC, separately in turbot (5.28 +/- 0.02g). Nine isonitrogenous and isoenergetic diets were formulated without fishmeal replacement (FM), 150 g/kg (XC15, SC15), 250 g/kg (XC25, SC25), 350 g/kg (XC35, SC35) and 450 g/kg (XC45, SC45) of fishmeal replaced by CSM. Fishmeal was successfully replaced by XC in turbot diets without growth reduction at 350 g/kg, but not by SC even at 150 g/kg. The apparent digestibility coefficients of SC-included diets were significantly lower than XC-included diets at same replacement level. The activities of aspartate aminotransferase and superoxide dismutase were significantly affected in XC45 and SC45 group. The XC45 and SC-included diets caused shortened distal intestine villi height and structural damage of liver compared with FM diet. This study indicated that different processing methods could affect the nutritional value of CSM and effect as a protein source for turbot. CSM with high quality could be an important alternative dietary protein source for juvenile turbot.
Effects of Silymarin on Growth Performance, Antioxidant Capacity and Immune Response in Turbot (scophthalmus Maximusl.)

Abstract:A 9-week feeding trial was conducted using triplicate groups of turbot (6.50 +/- 0.01 g) to explore the potential effects of silymarin. Three concentrations of silymarin (100, 200, and 400 mg/kg) were added to the plant protein-based diet. Fish were randomly distributed into fiberglass tanks (30 fish per tank). The results showed that adding 100 mg/kg silymarin significantly improved the growth performance, with no effects on feed utilization. The antioxidant capacity in the liver was significantly improved in the 100 mg/kg and 200 mg/kg silymarin groups by not only inducing the activities of superoxide dismutase (SOD) and catalase but also increasing the messager RNA (mRNA) expression levels ofSOD, glutathione peroxidase, and peroxiredoxin 6. Meanwhile, supplying 100 and 200 mg/kg of silymarin enhanced the heights of villi and enterocytes. Silymarin supplementation reduced the mRNA expression of interleukin-8 and tumor necrosis factor-alpha but induced the expression of transforming growth factor-beta (TGF-beta) in the intestine. These results indicated that silymarin was a potential nutraceutical that could enhance the growth performance and health status of turbot fed in a high plant protein diet. Adding 100 mg/kg silymarin to the plant protein diet achieved optimal performance in turbot.

Abstract:This study was designed to evaluate changes in the metabolic profile of liver and muscle of turbot (Scophthalmus maximus L.) fed fishmeal-based diet, diets containing size-fractionated fish protein hydrolysate and plant protein-based diet using H-1 NMR-based metabolomics approach combined with the growth. Fish protein hydrolysate (FPH) was obtained by enzymatic treatment, permeate fraction was obtained as UF by ultrafiltered step, and retentate fraction was retained as RF. FM diet contained fish meal used as a single protein source. Four other diets (PP, UF, FPH and RF) contained 180 g kg(-1) diet fish meal. 54, 55 and 55 g kg(-1) dry diet UF, FPH and RF were supplemented to UF, FPH and RF diets. All diets were formulated to be isolipidic and isonitrogenous fed to five triplicate groups of turbot (16.05 +/- 0.03 g) for 68 days. OPLS-DA in FM versus UF, FM versus FPH, FM versus RF and FM versus PP resulted in a reliable model for muscle and liver tissue, while OPLS-DA in UF versus FPH and UF versus RF only showed metabolites changes in liver tissue. Results indicated that metabolite changes among the different treatments were consistent with the growth tendency.
Effects of yeast extract supplemented in diet on growth performance, digestibility, intestinal histology, and the antioxidant capacity of the juvenile turbot (Scophthalmus maximus)

Abstract:An 8-week feeding experiment was conducted on the juvenile turbot (Scophthalmus maximus ) to evaluate the influence of yeast extract (YE) supplementation in the diet on growth performance, feed utilization, body composition, nutrient digestibility, intestinal histology, and antioxidant capacity. Four experimental diets were formulated with graded levels of yeast extract 0 (YE0), 1% (YE1), 3% (YE3), and 5% (YE5) and fed to turbots (initial body weight: 4.2 ± 0.1 g) with three replicates per diet and 200 fish in each replicate, respectively. The results showed that turbots fed with diets YE1 and YE3 displayed a significantly higher specific growth rate and protein efficiency rate than those fed with diets YE0 and YE5, while the feed conversion ratios in YE1 and YE3 groups were lower than those in YE0 and YE5. Fish fed with diets YE3 and YE5 showed higher body crude protein contents than those in groups YE0 and YE1. The highest apparent digestibility coefficients for dry matter and crude protein, digestive enzyme activities (trypsin, lipase, and amylase), and the height of the intestinal fold were observed in the YE3 group. YE3 treatment displayed a significantly higher superoxide dismutase (SOD) activity than the YE0 group, while the malondialdehyde (MDA) content in YE1 was significantly lower than those in YE0 and YE5. No significant difference was observed in serum physiological and biochemical parameters among all treatments. Overall, appropriate dietary supplementation of the yeast extract could improve the growth performance, digestibility, and antioxidant capacity of the juvenile turbot, and the recommended yeast extract level in the feed is 2.47%.

Abstract:Abstract In Europe, turbot aquaculture has a high potential for sustainable production, but the low tolerance to fishmeal replacement in the diet represents a big issue. Therefore, this study investigated the effects of more sustainable feed formulations on growth and feed performance, as well as nutritional status of juvenile turbot in recirculating aquaculture systems. In a 16-week feeding trial with 20 g juvenile turbot, one control diet containing traditional fishmeal, fish oil and soy products and two experimental diets where 20% of the fishmeal was replaced either with processed animal proteins (PAP) or with terrestrial plant proteins (PLANT) were tested. Irrespective of diets, growth performance was similar between groups, whereas the feed performance was significantly reduced in fish of the PAP group compared to the control. Comparing growth, feed utilisation and biochemical parameters, the results indicate that the fish fed on PAP diet had the lowest performance. Fish fed the PLANT diet had similar feed utilisation compared to the control, whereas parameters of the nutritional status, such as condition factor, hepato-somatic index and glycogen content showed reduced levels after 16 weeks. These effects in biochemical parameters are within the physiological range and therefore not the cause of negative performance. Since growth was unaffected, the lower feed performance of fish that were fed the PAP formulation might be balanced by the cost efficient formulation in comparison to the commercial and the PLANT formulations. Present study highlights the suitability of alternative food formulation for farmed fish.

Effects of Dietary Supplement of Schizochytrium Meal on Growth, Fatty Acid Profile and Activities of Digestive Enzymes in Turbot (scophthalmus Maximus L.) Larvae

Abstract:A feeding trial was conducted to investigate the effects of supplementation of Schizochytrium meal on growth, digestive enzymes activities and fatty acid composition of turbot (Scophthalmus maximus L.) larvae. Four isonitrogenous and isolipidic diets were formulated to contain 0 (S0, control diet), 50 (S5), 100 (S10) and 150 (S15) g kg(-1) Schizochytrium meal. Fish (initial body weight, 0.06 g) were randomly allotted to 12 square fiberglass tanks. Fish were fed 5 times daily (7:00, 10:00, 14:00, 17:00 and 21:00) for 28 days. No significant differences were observed in survival and intestinal morphology among fish fed various levels of algae meal (P>0.05). Fish fed diet S5 had significantly higher final body weight than that of fish fed diet S15, and no significant differences were observed among fish fed diets S0, S5 and S10 (P>0.05). Trypsin in intestinal segments, specific activities of alkaline phosphatase (AKP) in intestine and purified brush border membrane (BBM) of intestine were significantly higher in fish fed diet S5 than that of fish fed diet S15 (P<0.05). Specific activities of leucine-aminopeptidase (LAP) in intestine and purified BBM of intestine was significantly higher in fish fed diet S10 than that of fish fed diet S15 (P<0.05). Fish fed diets S5 and S10 had significantly higher docosahexaenoic (DHA, 22:6n-3), n-3 PUFAs content and n-3/n-6 ratio in muscle than fish fed diets S0 and S15 (P<0.05). Fish fed diets S5, S10 and S15 had significant lower C18:3n-3, eicosapentaenoic (EPA, 20:5n-3), C18:2n-6, n-6 polyunsaturated fatty acids (PUFAs) content in muscle than fish fed the control diets (P<0.05). No significant differences were observed in intestinal morphology (P>0.05). In conclusion, 50-100 g kg(-1) dry matter Schizochytrium meal in microdiets can improve growth performance and may be a valuable additive in microdiets of turbot larvae.

Abstract:Fishmeal could only be replaced by plant proteins at limited levels in aquafeeds, especially for carnivorous fish. In this study, an experiment was designed to evaluate the possibility of improving the utilization of plant proteins by maggot meal supplementation in turbot diet. Five diets were formulated: a reference diet (FM) containing 63% fishmeal and four experimental diets (35(0%), 35(3%), 40 (0%), 40(3%)) in which fishmeal was substituted at different levels by plant proteins with 0-3% maggot meal. Turbot (4.90 +/- 0.03 g) was fed with these diets for 9 weeks. Fishmeal was successfully replaced by plant proteins in turbot diet without growth reduction at 35% but not 40%. However, maggot supplementation (3%) at 40% plant protein replacement level achieved comparable growth performance with that of fishmeal. Maggot meal supplementation improved apparent digestibility coefficients, plasma hydroxyproline levels, intestine trypsin activities and activated target of rapamycin (TOR) signalling, all of which were decreased or down-regulated after high plant protein replacement. Therefore, this study demonstrated maggot meal, a potential valuable protein source for turbot.

Abstract:In this study,a feeding trial was conducted to evaluate the effects of adding different levels of sodium butyrate in high plant protein diets on growth performance,nutrient apparent digestibility coefficients and liver antioxidant function of juvenile turbot (Scophthalmus maximus L.),and to explore the feasible method for improving substitution of fish meal by plant protein.Using fish meal,soybean meal,corn gluten meal,vital gluten,peanut meal and beer yeast as protein sources,fish oil,coconut oil and soybean lecithin as lipid sources,and wheat meal as carbohydrate source,five isonitrogen and isocaloric experimental diets were formulated.Among them,the basal diet formulated with 60% fish meal was set as the positive control group (FM group),the substituted diet which used mixed plant protein sources to replace 50% fish meal was set as the negative control group (CON group),and the other three diets which were supplemented with 0.15% (D1 group),0.30% (D2 group) and 0.60% (D3 group) sodium butyrate into the substituted diet,respectively,were set as the experimental groups.Each diet was assigned to three replicates of 30 juvenile turbot with an initial body weight of (13.00±0.01) g for 58 days in an indoor aquaculture system.The results showed as follows: 1) with the increase of sodium butyrate supplemental level,the weight gain rate (WGR),specific growth rate (SGR) and feed efficiency (FE) were increased firstly and decreased subsequently.The WGR,SGR and FE in FM,D1 and D2 groups were significantly higher than those in CON group (P<0.05),while no significant difference in feed intake (FI) was observed among groups (P>0.05).For the viscerosomatic index (VSI),compared with FM group,the D2 group had no significant difference (P>0.05),but the other group were significantly decreased (P<0.05).No significant differences in whole fish moisture,crude protein and crude lipid contents were observed among groups (P>0.05).2) The apparent digestibility coefficient of dry matter in D1 group was significantly higher than that in CON group (P<0.05),while it showed no significant difference between the D1 and FM groups (P>0.05).For the apparent digestibility coefficient of protein,D1 group was significantly higher than that in CON,D2 and D3 groups (P<0.05),but no significant differences were showed between the D1 and FM groups (P>0.05).No significant difference in the apparent digestibility coefficients of dry matter and protein were observed between the D2 and D3 groups (P>0.05).3) Compared with the CON group,liver total antioxidant capacity (T-AOC) and catalase (CAT) activity in D2 group were significantly increased (P<0.05),but the liver malondialdehyde (MDA) content was significantly decreased (P<0.05).In conclusion,under this experimental condition,adding 0.15% sodium butyrate in the high plant protein diets can improve the growth performance,nutrient apparent digestibility coefficients and liver antioxidant function of juvenile turbot,but excessive adding sodium butyrate will decrease the growth performance of juvenile turbot.
Optimal Dietary Lipid Levels Alleviated Adverse Effects of High Temperature on Growth, Lipid Metabolism, Antioxidant and Immune Responses in Juvenile Turbot (scophthalmus Maximus L.).

Abstract:Fish physiological health is often negatively impacted by high-temperature environments and there are few studies on how dietary lipids affect fish growth and physiology when exposed to heat stress. The main objective of this research was to examine the impact of dietary lipid levels on growth and physiological status of juvenile turbot (Scophthalmus maximus L.) and determine if dietary lipid concentration could alleviate the possible adverse effects of heat stress. Five diets containing 6.81%, 9.35%, 12.03%, 14.74%, and 17.08% lipid, respectively, were formulated and fed to turbot (initial weight 5.13 +/- 0.02 g) under high-temperature conditions (24.0-25.0 C-degrees). Meanwhile, the diet with 12.03% lipid (considered by prior work to be an optimal dietary lipid level) was fed to turbot of the same size at normal temperature. Results suggested that, among the different dietary lipid levels under high-temperature conditions, fish fed the optimal lipid (12.03%) exhibited better growth compared to non-optimal lipid groups, as evidenced by higher weight gain and specific growth rate. Simultaneously, the optimal lipid diet may better maintain lipid homeostasis, as attested by lower liver and serum lipid, along with higher liver mRNA levels of lipolysis-related genes (pgc1 alpha, lipin1, ppar alpha, lpl and hl) and lower levels of synthesis-related genes (lxr, fas, scd1, ppar gamma, dgat1 and dgat2). Also, the optimal lipid diet might mitigate oxidative damage by improving antioxidant enzyme activity, decreasing malondialdehyde levels, and up-regulating oxidation-related genes (sod1, sod2, cat, gpx and ho-1). Furthermore, the optimal lipid may enhance fish immunity, as suggested by the decrease in serum glutamic-oxalacetic/pyruvic transaminase activities, downregulation of pro-inflammatory genes and up-regulation of anti-inflammation genes. Correspondingly, the optimal lipid level suppressed MAPK signaling pathway via decreased phosphorylation levels of p38, JNK and ERK proteins in liver. In summary, the optimal dietary lipid level facilitated better growth and physiological status in turbot under thermal stress.

Abstract:A nine-week feeding trial was conducted to comprehensively investigate the effects of different levels of dietary lipid on intestinal physiology of juvenile turbot. Three diets with different lipid levels (8%, 12% and 16%) were formulated, which were designated as the low-lipid group (LL), medium-lipid group (ML) and high-lipid group (HL), respectively. Each diet was fed to six replicate tanks, and each tank was stocked with 35 fish. The results revealed that medium dietary lipid (12%) increased the activities of intestinal digestive enzymes and brush border enzymes. Excessive dietary lipid (16%) decreased the intestinal antioxidative enzyme levels and increased the lipid peroxidation pressure. In addition, HL stimulated the occurrence of intestinal inflammation and significantly up-regulated the mRNA expression level of tumor necrosis factor-alpha (TNF-alpha), interleukin-1 beta (IL-1 beta), interferon-gamma (IFN-gamma) and transforming growth factor-beta (TGF-beta). Dietary LL and HL induced the apoptosis of intestinal epithelial cells. Sequencing of bacterial 16 s rRNA V4 region indicated that the abundance and diversity of intestinal microflora in fish fed with medium lipid diet (12%) were significantly higher than those in other groups, indicating the intestinal microflora ecology in group ML was more balanced. MetaStat analysis indicated that both low- and high-lipid diets significantly reduced the relative abundance of intestinal beneficial bacteria. In conclusion, results of this study demonstrated the sensitivity of intestinal health and microbiota to dietary lipid levels. From the perspective of microecological balance, medium dietary lipid (12%) was more conducive to maintaining the intestinal microflora stability of turbot.
