The Clinical Significance of Balance Between Type Ⅰ and Type Ⅱ Immune Responses of Lymphocyte Subpopulations in Myelodysplastic Syndrome

Abstract:Objective: To evaluate the clinical significance of the subsets of CD4+ , CDS+ cells and the relationship of the activated T cells in peripheral blood from patients with myelodysplastic syndrome (MDS). Methods: We investigated the intracytoplasmic expression on CD4+ and CD8+ T cells after in vitro cultivation with PMA,Ionomycin and Monensin in 14 patients with RA and 1 patient with RAEB, Flow cytometry was used for the counts of CD3+ CDS IFN-(+ cells (Th1), CD3+ CD8- IL-4+ cells (Th2), CD3+ CD8+ IFN-(+ cells (Tc 1), CD3+ CD8+ IL-4+ cells (Tc2). Furthermore, we studied the relationship between the Th cells, Tc cells and the activated T cells. Results: The percentages of Thl cells in peripheral blood from patients with MDS were higher than those of control group. Th cells were deviated to the Thl cells in the body of MDS. The increased production of cytokines in MDS which inhibit hematopoiesis such as IFN-γ,TNF-β might reflect deviations of Th distributions. There were no significant differences in the proportion of Th2, Tc1 and Tc2 cells. The relationship between the Th1 cells and CD4+ CD45RO+ cells was positive (r=0. 573,P0.05) .while Th1 cells versus CD4+ CD45RO+ cells was negative (r=-0. 509,P0.05). There was no relationship between the expression rates of Th2, Tc1, Tc2 cells and CD4+ CD45RO+ cells(P 0.05). Conclusion:There was the imbalanced development of Th cells and the overproduction of cytokines inhibitory to hematopoiesis in MDS. T lymphocytes played an important role in producing these cytokines.
