Abstract:Solanesol, a main polyprenol occurred in tobacco, has gained the attention of biochemists because of its value as a source of isoprene units for the synthesis of metabolically active quinones and vitamin K analogs. This article deals with extraction and purifyication of solanesol. Ground tobacco leaf was extracted with petroleum ether in 50°C water bath under reflux for 2h, and then filtrated, the powder leaf was re-extracted successively with petroleum ether and the extracts were pooled and concentrated to get pasty residue. The residue was saponified and subsequently loaded onto silica gel column chromatography. The chemical was eluted using a binary solvent mixture of petroleum ether–acetone (90:10, v/v) and the eluent was collected by fraction size of 5ml. Tentative identification was carried out using TLC. The fractions mainly contained solanesol were pooled and dried by rotary evaporation. The yield of solenosol was 0.38% of the dry weight of tobacco leaf, and the purity was 83.04%.

Abstract:The method of solid-phase extraction (SPE) for the concentration and clean-up of tobacco extract samples during solanesol analysis was proposed in this work. A column (200 mm × 4 mm i.d.) packed with 0.10 g silica gel (with particle size of 70 μm, porosity of 0.5 and surface area of 400 m2 g−1) was used as SPE cartridge. Several extraction parameters, such as sample loading flow (0.3–7 mL min−1), sample volume (5–50 mL), the column adsorption capacity and elution were evaluated to provide a fast, quantitative and reproducible SPE method. A mean solanesol recovery was 97.5 ± 1.6% (mean ± sd) and mean intra- and inter-day reproducibility was higher than 97%. It can be used in the analysis of solanesol in tobacco extracts.

Abstract:Beta-carotene was isolated from discarded tobacco leaves by column chromatography using petroleum ether as eluent. Its structure was characterized by 1H-NMR and 13C-NMR spectra. The purity of the beta-carotene was determined to be 98% by a high performance liquid chromatographic (HPLC) method which was demonstrated to be accurate and reproducible. Infra-red spectrum and melting point of the isolated sample were also characterized in agreement with those reported in literature. The overall yield of 80%, the high purity and the easiness to recycle as the results of only a single solvent employed, would make the method an important reference for future research of beta-carotene and a favorable technique for future development.
The Study of Refining Solanesol by Molecular Distillation

Abstract:Molecular distillation is a kind of high vacuum distillation method,which is suitable for the separation of products with high boiling point,high viscidity,heat sensitivity and bioactivity.From the raw solanesol,the solanesol with high purity was produced by twice molecular distillation.The effects of evaporation temperature,feed flow rate and temperature,operating pressure and agitated speed on the refine of solanesol were studied.The optimal operation conditions for molecular distillation of solanesol were found as follows;the operating pressure of 10 Pa,evaporation temperature of 200～220℃,feed rate of 350～400 mL·h-1,feed temperature of 100℃and agitated speed of 300～400 r·min-1.Under the above conditions,the concentration and recovery of the solanesol obtained by the experiments were about 97.6%and 77.1%,respectively.Those data satisfy the requirement of separating solanesol,and it also shows that the molecular distillation process is feasible for the task of refining solanesol.

Abstract:The effect of different parameters such as extraction time,resolution temperature and extraction pressure on the supercritical carbon dioxide extraction of solanesol was investigated.With pretreatment such as saponification,the optimal extraction conditions were determined according to orthogonal experiments as entrainer 90% ethanol(volume fraction),extraction pressure 35 MPa,extraction temperature 50 ℃ and resolution temperature 40 ℃.After extraction for 3 h,mass fraction of the solanesol was 40.14%,and extraction rate was as high as 98.53%.

Abstract:The optimum extraction technology of tobacco absolute oil from discarded tobacco leaves by the mixture solvent n-hexane and acetone is investigated.The effects of mass ratio of n-hexane to acetone,ratio of liquid to solid,extraction temperature and extraction time are studied by single factor test.Based on the single factor test,orthogonal experiments are performed to optimize the above conditions.The results show that the optimum extraction technology is as follows: mass ratio of n-hexane to acetone is 6∶4,ratio of liquid to solid is 12∶1,extraction temperature is 50 ℃,extraction time is 4 h.Based on the optimum extraction conditions,the yield of tobacco absolute oil reaches 6.95%.

Abstract:The extraction of an essential oil from discarded tobacco leaves using a combination of solvent extraction and steam distillation (SE–SD) was investigated. The effect of different parameters, such as temperature, extraction time and liquid–solid ratio, on the extraction yield was investigated using the Response Surface Method (RSM) with the Box–Behnken Design (BBD) method. The temperature and liquid–solid ratio were found to have a significant effect on the yield of extract (oleoresin). A yield of 3.38±0.03% of the oleoresin (final essential oil yield was 0.89%) could be extracted under the following optimized conditions: a temperature of 41°C, a liquid–solid ratio of 6.27mL/g and a time of 5h. Composition and characteristics of the discarded tobacco essential oils and oleoresins obtained by SE–SD, hydrodistillation and solvent extraction were analyzed by gas chromatography/mass spectrometric detection, to allow the extraction methods to be compared. The results showed that the same compounds were largely present in the different tobacco essential oils, but their relative concentrations differed. General characteristics of the extracts obtained from the three extraction methods showed that the SE–SD essential oil was of better quality and therefore of higher value. Thus, SE–SD is the optimum process for the extraction of high quality essential oil from discarded tobacco.

Abstract:The central composite design combined with response surface methodology was used to optimal operating parameters of ethanol-modified supercritical CO2 extraction and evaluate the effects of operating parameters on crude-solanesol yield and extract anti-free-radical activity (DPPH radical scavenging activity). The results show that pressure, temperature and time appeared independent and interactive effects on the crude solanesol yield as well as the extract anti-free-radical activity. The optimal conditions for crude-solanesol yield were 45°C, 38MPa and 2.6h, while that for the extract anti-free-radical activity 47°C, 36MPa and 2.5h. Under the optimal conditions, the highest solanesol extracted could arrived at 3.74wt% with 52.3wt% purity (96.8% extraction ratio), and the maximal extract anti-free-radical activity 57.49%. Meanwhile, it was found that the extract anti-free-radical activity was correlated very well with the crude solanesol yield, resulting in model with high coefficient of determination (R2=0.946). The research demonstrates that ethanol-modified supercritical CO2 extraction conducted by the central composite design combined with response surface methodology, which could produce high crude solanesol yield with high quality as well high level of bioactivity, is a prospective alternative to the traditional heat-reflux extraction.

Abstract:Solanesol, an aliphatic terpene alcohol composed of nine isoprene units, is mainly found in solanaceous plants. Particularly, tobacco (Nicotiana tabacum), belonging to the Solanaceae family, is the richest plant source of solanesol, and its leaves have been regarded as the ideal material for solanesol extraction. Since the discovery of solanesol in tobacco, significant progress has been achieved in research on solanesol’s bioactivities, medicinal value, accumulation, extraction technology, and determination methods. Solanesol possesses strong free radical absorption ability and antioxidant activity owing to the presence of several non-conjugated double bonds. Notably, solanesol’s anti-inflammatory, neuroprotective, and antimicrobial activities have been previously demonstrated. Solanesol is a key intermediate in the synthesis of coenzyme Q10, vitamin K2, and the anticancer agent synergiser N-solanesyl-N,N′-bis(3,4-dimethoxybenzyl) ethylenediamine. Other applications of solanesol include solanesol derivative micelles for hydrophobic drug delivery, solanesol-derived scaffolds for bioactive peptide multimerization, and solanesol-anchored DNA for mediating vesicle fusion. Solanesol accumulation in plants is influenced by genetic and environmental factors, including biotic stresses caused by pathogen infections, temperature, illumination, and agronomic measures. Seven extraction technologies and seven determination methods of solanesol are also systematically summarized in the present review. This review can serve as a reference for solanesol’s comprehensive application.

Abstract:Objective:To isolate and purify 10-deacetyl baccatinⅢ(10-DABⅢ)from leaves and branches of Taxus mairei.Methods:10-DABⅢwas extracted and purified by liquid-liquid and back ex- traction,column chromatography and recrystallization,and elucidated by UV and HPLC.Results:The total yield of 10-DABⅢ,a precursor of anti-cancer drug taxol and docetaxel was 7.9% and the purity of 10-DASⅢwas 91%.Conclusion:The novel isolation process for 10-DABⅢwas warranted for the pilot manufacture.
Comparison on Bioactivities of Solanesol Extracted from Tobacco Leaves by Different Methods

Abstract:[Objective] The aim was to select suitable method for extracting solanesol from tobacco leaves.[Method] DPPH method and agar plate diffusion were used to investigate the antioxidant and antibacterial activities of solanesol extracted from tobacco leaves by 3 methods.[Result] The rates of semi-scavenging(IC50) of solanesol extracted by soxhlet,ultrasonic and supercritical extractions from saponated tobacco leaf powder on DPPH were 56.43,54.77 and 49.48 mg/L resp.,that of solanesol extracted by soxhlet and ultrasonic extractions from non saponated tobacco leaves on DPPH were 47.22 and 44.45 mg/L resp.and that of Vc(CK) was 13.89 mg/L.The curve of DPPH scavenge rate of mixture of solanesol and Vc with equal volumes was between theirs.The antibacterial activities of solanesol extracted by different methods in order were supercritical extraction soxhlet extraction ultrasonic extraction.Saponification had negative effects on both antioxidant and antibacterial activities of extracts.[Conclusion] The bioactivities of solanesol extracted from tobacco leaves by different methods in order were supercritical extraction soxhlet extraction ultrasonic extraction.

Abstract:Solanesol, the precursor for the synthesis of coenzyme Q10, is currently recovered from tobacco leaves by conventional extraction techniques that require multiple purification steps and a large amount of organic solvents. We recently identified tomato leaves as an alternative source of solanesol and hypothesized that a high-pressure CO2 extraction could be used as a clean extraction process. The effect of CO2 pressure and temperature on the extraction of solanesol was determined to achieve high yield and purity. It was found that solanesol could be extracted efficiently by subcritical CO2 at 25 °C from tomato leaves. The extract contained 40% solanesol and other active compounds such as vitamin K1. A higher level of purity of 93% was achieved using a secondary purification step. Different conventional methods for solanesol extraction was compared to determine the most efficient technique for production of solanesol from tomato leaf. The highest yield of solanesol was achieved at nearly 1% dry weight with using subcritical CO2, which was superior to conventional methods.

Abstract:New method for the production of nicotine from waste tobacco by saponification,distillation,(acidification),inspissation,extraction had been investigated.Thus,1 000 g waste tobacco was saponified by 3 L of 0.1 mol/L sodium hydroxide solution at 60 ℃ for 4 h.Nicotine solution was obtained from saponification solution by distillation,after acidification,inspissation and extraction with chloroform,nicotine was obtained from chloroform solution by distillation with purity of 99.23% and yield of 82.7%.
RP-HPLC Determination of Solanesol in Tobacco Leaves

Abstract:Objective:A reversed phase high performance liquid chromatography method for the determination of solanesol in the extract of tobacco leaves was developed.Method:The analysis was performed on KNAUER-C_(18) re- versed-phase analytical column(4 mm×250 mm,5 μm)at the temperature of 30 ℃.The mobile phase consist of 100% methanol,which was pumped at 1.0 mL·min~(-1).The detecting wavelength was set at 215 nm.Results:The standard curve was linear ranged from 0.05 μg to 1.43 μg for solanesol.Intra-day RSD and inter-day RSD of this method was 1.3% ,2.8%,respectively.The average recovery(n=9)was 108.5%.Conclusion:Solanesol in the extract of tobacco leaves could be determined with this method for the advantage of simplicity,conveniency,ac- curacy and reproducibility.

Abstract:BACKGROUND: Aromatic waste water is the main by-product of industrial essential oil distillation. To recover the dissolved essential oil from this distillation waste water, an approach combining column adsorption (CA) and supercritical fluid extraction (SFE) was proposed. Yulania liliiflora was selected as a case study. Activated carbon of mixed porosity was employed and quantitative desorption of the adsorbed oil was achieved using supercritical carbon dioxide (SC-CO2).RESULTS: The optimized condition of CA-SFE was 18 MPa, 308 K, dynamic extraction for 60 min and CO2 flow rate of 3.6 g min(-1). A recovery of 0.668 +/- 0.050 g kg(-1) (n=3) was achieved for the dissolved oil from distillation waste water, significantly higher (P < 0.05) than that obtained by liquid-liquid extraction (LLE) (0.405 +/- 0.032 g kg(-1), n =3). Yulania liliiflora recovered oil was rich in eucalyptol (52.6-55.2%), alpha-terpineol (15.2-18.1%) and terpinen-4-ol (8.1-8.7%).CONCLUSION: The CA-SFE approach is highly efficient and especially suitable for large-scale application. The recovered essential oil is valued for being rich in organoleptically important and biologically active compounds. (C) 2015 Society of Chemical Industry

Abstract:Sucrose esters (SEs) were successfully extracted from Oriental tobacco leaves using a new methodology based on accelerated solvent extraction followed by hydrophilic-lipophilic balanced cartridge cleanup step. The SEs were detected by HPLC with ion-trap MS detection using an electrospray interface operated in the positive ion mode. This methodology combines the high efficiency of extraction provided by a pressurized fluid and the highly sensitive characterization offered by ion-trap MS. Under the optimized conditions, 14 SEs were first identified among a total of 23 SEs found in Oriental tobacco leaves. Under the same conditions, only four new SEs were extracted by using traditional ultrasound-assisted extraction and liquid-solid extraction methods. The present method might be potentially useful in high-efficiency extraction and sensitive characterization of SEs from complex matrices such as tobacco leaves.

Abstract:The objective of this work is to study the feasibility of vacuum membrane distillation (VMD) by hydrophilic membranes to recover petroleum ether from the extracting solution of solanesol. Being different from the traditional membrane distillation (MD) process, hydrophilic membranes were used to avoid the membrane wetting for the nonaqueous solutions. The VMD performances of polyvinylidene fluoride (PVDF) membrane and two kinds of polyacrylonitrile (PAN) membranes with different structures were compared in the process of recovering petroleum ether. The results showed that good solvent flux (> 15 kg/m(2) h) and solanesol rejection (> 98%) for the PAN membranes were obtained, indicating that PAN membranes had considerable potential use in this area, while the PVDF membrane was not appropriate for the nonaqueous solution system because of membrane wetting. The effects of operation conditions on VMD performances of the two PAN membranes were investigated, and it was found that high-feed temperatures, low-downstream pressures enhanced the permeate flux for both membranes, and the flux of PAN1 membrane decreased more obviously than that of PAN2 membrane under high-feed concentration because of its larger pore size. General models including Knudsen and viscous flows were proposed, and a good agreement between the experimental and the theoretical fluxes was obtained. Additionally, temperature and concentration polarizations were proved to have an obvious influence on mass transport of PAN1 membrane. It was advisable to work at the downstream pressure of 20 kPa, the feed temperature of 30 degrees C, and the feed flow rate of 27.5 mL/s to obtain favorable results for PAN2 membrane.

Abstract:Currently, in the ongoing development of the tobacco industry, a large amount of tobacco rhizomes is discarded as waste. These wastes are usually disposed of through incineration or burial. However, these tobacco wastes still have some economic value. High-purity nicotine has a promising market outlook as the primary raw material for electronic cigarette liquid. Nicotine is not only found in tobacco leaves but also in the rhizomes of tobacco plants. This study presents a method for treating tobacco waste and extracting high-purity nicotine from it. After mixing the raw material powder and entrainer in specific ratios, as much of the nicotine in tobacco roots can be extracted as possible using supercritical carbon dioxide extraction. The effects of temperature, the ratio of the entrainer, and the volume fraction of ethanol in the entrainer on the nicotine yield in supercritical fluid extraction (SFE) at 25 MPa for 120 min were discussed. By using 90% ethanol (a raw material mass-to-volume ratio of 1:5) as the entrainer, we obtained the highest nicotine yield of 0.49% at 65 °C. Meanwhile, the purity of the crude extract was 61.71%, and after purification, it increased to 97.57%. In this way, we can not only obtain nicotine with market value but also further reduce the harm to the environment caused by tobacco waste disposal.
