Abstract:Cell-surface functional groups (amino, carboxyl, hydroxyl, as well as phosphate) were chemically modified in various ways to enhance the demulsification capability of the demulsifying bacteria Alcaligenes sp. S-XJ-1. Results demonstrated that the demulsifying activity was significantly inhibited by amino enrichment with cetyl trimethyl ammonium bromide, amino methylation, hydroxyl acetylation, and phosphate esterification, but was gradually promoted by carboxyl blocking with increasing the extents of esterification. Compared with the raw biomass, an optimal esterification of carboxyl moieties enhanced the demulsification ratio by 26.5% and shortened the emulsion half-life from 24 to 8.8 h. The demulsification boost was found to be dominated by strengthened hydrophobicity (from 53° to 74°) and weakened electronegativity (from −34.6 to −4.3 mV at pH 7.0) of the cell surface, allowing the rapid dispersion and adsorption of cells onto the oil-water interface. The chemical modification of the functional groups on the biomass surface is a promising tool for the creation of a high-performance bacterial demulsifier.
Effect of Saponins on N-Hexane Removal in Biotrickling Filters.

Abstract:Saponins was applied to enhance the removal of n-hexane in a biotrickling filter (BTF) in this study. Comparison experiments were carried out to examine the effect of saponins on n-hexane removal in two BTFs at various saponins concentrations, n-hexane loading rates (LRs) and gas empty bed contact times (EBCTs). Results show that the optimum concentration of saponins in nutrient feed was 50.0mgL(-1). When organic LR of n-hexane increased from 47.8 to 120.0gm(-3)h(-1), the removal efficiency (RE) for BTF1 (with saponins) and BTF2 (without saponins) decreased from 91.3% to 83.3% and from 62.8% to 56.8%, respectively. As gas EBCT decreased from 30.0 to 7.5s, the RE declined from 88.4% to 64.5% for BTF1 and from 61.4% to 38.3% for BTF2. Saponins could also decrease the biomass accumulation rate within the medium bed. These results could be referred in the design and operation of BTFs for hydrophobic VOC removal.
Optimization of Biodemulsifier Production from Alcaligenes Sp. S-XJ-1 and Its Application in Breaking Crude Oil Emulsion.

Abstract:A biodemulsifier-producing strain of Alcaligenes sp. S-XJ-1, isolated from petroleum-contaminated soil of the Karamay Oilfield, exhibited excellent demulsifying ability. The application of this biodemulsifier significantly improved the quality of separated water compared with the chemical demulsifier, polyether, which clearly indicates that it has potential applications in the crude oil extraction industry. To optimize its biosynthesis, the impacts of carbon sources, nitrogen sources and pH were studied in detail. Paraffin, a hydrophobic carbon source, favored the synthesis of this cell wall associated biodemulsifier. The nitrogen source ammonium citrate stimulated the production and demulsifying performance of the biodemulsifier. An alkaline environment (pH 9.5) of the initial culture medium favored the strain's growth and improved its demulsifying ability. The results showed paraffin, ammonium citrate and pH had significant effects on the production of the biodemulsifier. These three variables were further investigated using a response surface methodology based on a central composite design to optimize the biodemulsifier yield. The optimal yield conditions were found at a paraffin concentration of 4.01%, an ammonium citrate concentration of 8.08 g/L and a pH of 9.35. Under optimal conditions, the biodemulsifier yield from Alcaligenes sp. S-XJ-1 was increased to 3.42 g/L.

Abstract:Biodemulsifier is a new type of demulsifiers for breaking oil-water emulsion. One demulsifier-producing strain, Alcaligenes sp. S-XJ-1 could grow on waste diesel oil (WDO), dry weight of the strain was up to 2.0 g/L after being cultivated for 7 d, 10 g/L S-XJ-1 cell suspension reduced surface tension of water from 72.0 mN/m to 29.7 mN/m. Biodemulsifier produced by S-XJ-1 was 0.3 g/L, its critical micelle concentration (CMC) was 150 mg/L, showing a better surface activity than chemical surfactant SDS. Furthermore, it showed an demulsifying efficiency over 70% for W/O model emulsion. It was indicated S-XJ-1 could utilize C14-C20 n-alkanes composing waste diesel oil by GC-MS, C20 n-alkane was almost completely comsumed by S-XJ-1 with utilization ratio of 99%. In addition, n-alkanes utilization ratio and demulsifying capability increased when length of carbon chain increased. Both utilization ratio and demulsifying capability of biodemulsifier produced by S-XJ-1 using C20 n-alkane as the carbon source were superior to other n-alkanes, and similar to waste diesel oil. It was identified the biodemulsifier produced by S-XJ-1 using waste diesel oil was lipopeptide by TLC and FTIR.

Abstract:Biodemulsifiers are environmentally friendly agents used in recycling oil or purifying water from emulsion, yet the demulsifying feature of cell-surface composition remains unclear. In this study, potentiometric titration, attenuated total reflectance-Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry were combined to characterize cell-surface chemical composition of demulsifying strain Alcaligenes sp. S-XJ-1 cultivated with different carbon sources. Cells cultivated with alkane contained abundant elemental nitrogen and basic functional groups, indicating that their surface was rich in proteins or peptides, which contributed to their highest demulsifying efficiency. For cells cultivated with fatty acid ester, the relatively abundant surface lipid contributed to a 50% demulsification ratio owing to the presence of more acidic functional group. The cells cultivated with glucose exhibited a high oxygen concentration (O/C similar to 0.28), which indicated the presence of more polysaccharides on the cell surface. This induced the lowest demulsification ratio of 30%. It can be concluded that cell surface-associated proteins or lipids other than the polysaccharide of the demulsifying strain played a positive role in the demulsification activity. In addition, the cell-surface oligoglutamate compounds identified in situ were crucial to the demulsifying capability.
Effective biodemulsifier components secreted by Bacillus mojavensis XH-1 and analysis of the demulsification process

Abstract:The purpose of the present study was to investigate the effective components of the demulsifying bacterial strain Bacillus mojavensis XH-1 and its demulsification process. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and the shotgun LC-MS/MS method were used to separate and identify proteins with efficient demulsification activity. The zeta potential changes of the emulsion before and after addition of the biodemulsifier were tested, and the relationships between oil-in-water interfacial tension, the demulsification efficiency and the biodemulsifier structure were examined. The study results indicate that the effective biodemulsifier components were extracellular proteins attached to the cells or secreted into the culture solution that presented as a 50-80 kDa band observed by SDS-PAGE. Six of the proteins were unknown or unnamed, and the demulsifying functions of another 14 proteins had not been previously reported. The main demulsification mechanisms were determined to be solubilization and replacement. When the concentration of the biodemulsifier was low, the replacement mechanism dominated, and the demulsification ratio increased with the biodemulsifier concentration. Solubilization dominated when a high concentration of biodemulsifier was provided, and the demulsification ratio decreased as the biodemulsifier concentration increased.

Abstract:Environment-friendly demulsifying strains could be applied in breakup of emulsion produced in the petroleum industry. The demulsifying strain Alcaligenes sp. S-XJ-1 was found to be capable of utilizing paraffin or certain pure n-alkane as its sole carbon source, and exhibited a strong cell surface hydrophobicity above 70% and demulsifying ability above 80%. A maximum biomass of 6.27 g/L was obtained when the demulsifying strain was cultured using eicosane as carbon source. A positive correlation appeared to exist between the resulting biomass and alkane carbon chain length, indicating that waste mineral oils rich in long chain alkane could be potential substrates for producing biodemulsifiers. The highest emulsion breaking ratio of 96% was obtained when the demulsifying strain was cultured using hexadecane as carbon source. Its biological demulsification process was analyzed by using Turbiscan Lab (R) Expert as following: began with the flocculation and coalescence of dispersed droplets, followed by the settling of dispersed droplets, which finally induced separation of the oil and water phases. The critical point of phase separation in the emulsion by the strain produced using hexadecane appeared in the first hour of the demulsification process, which exhibited faster emulsion-breaking speed than that produced by paraffin.
Extraction of surface active substance and analysis of demulsifying characteristics for the demulsifying strain Alcaligenes sp. S-XJ-1

Abstract:Extraction and identification of surface active substance of Alcaligenes sp. S-XJ-1, as well as description of its emulsion breaking process were conducted to reveal the demulsifying characteristics of this demulsifying strain. Alkali solvent was adopted in the extraction process with conditions optimized as 35 degrees C, 0.08 mol x L(-1) of alkali concentration, 12 g x L(-1) of sample to solution ratio, and 4 h of extraction time by launching both single-factor and orthogonal tests. Under this optimal condition, the extracted surface active substance (the extraction ratio was 36.1%) achieved 77% emulsion breaking ratio for 500 mg x L(-1) within 48 h. FT-IR showed the existence of glycolipids, lipids and proteins in the surface active substance, the molecular weight of which mainly scattered between 55 and 61 256. Saccharides, lipids and proteins were identified as the three chief components in surface active substance with the content of 22.2%, 7.5% and 13.4%, respectively. The proteins were further proved to take the most responsibility for the emulsion breaking ability. Moreover, obvious difference in the emulsion breaking process was demonstrated between the original demulsifying strain S-XJ-1 and the extracted surface active substance by real time observation of Turbiscan Lab Expert. The results suggested that the demulsifying efficiency of the strain was jointly contributed by its surface active substance and demulsifying cell morphology, and the former possessed higher functional priority than the latter.
Study on Effective Biodemulsifier Components Secreted by Demulsifying Bacterial Strain XH-1

Abstract:The demulsifying effective substances of bio-demulsifier secreted by Bacillus mojavensis XH-1 with high demulsifying efficiency were researched and their physiochemical properties were analyzed. The study results indicate that for O /W model emulsions,more than 85% efficiency of demulsification was obtained for the strain within 24h and 100% efficiency was acquired within 48h. The effective components of biodemulsifier were extracellular substances,that did not possess thermostability,but cool resistance,and their activity were inhibited by pepsin,trypsin and urea. The active demulsifying substances were extracted using different protein extraction methods,and all of the extracts exhibited demulsification activities,indicating that the active substance possessed protein characteristics. The results of ultraviolet spectrophotometry in combination with infrared spectrum scanning demonstrate that the active components of the biodemulsifier were extracellular proteins.
Characterization of the Extracellular Biodemulsifier of Bacillus Mojavensis XH1 and the Enhancement of Demulsifying Efficiency by Optimization of the Production Medium Composition

Abstract:The demulsifying bacterium XH1 was identified as a Bacillus mojavensis by the 16S rDNA gene. The extracellular biodemulsifier produced by this species was purified by ethanol extraction and column chromatography through a sephadex and silicon gel column. Preliminary investigation using UV-vis and TLC indicated that the biodemulsifier had two components a protein and a lipopeptide. All major components of the medium, including the sources of soluble and insoluble carbon, nitrogen, phosphate, and metal ions were investigated to improve the biosynthesis and efficiency of the biodemulsifier. The optimal carbon sources were glucose and liquid paraffin. Glucose participated in the biosynthesis of the demulsifier, while liquid paraffin promoted the lipophilicity and secretion of biosurfactants. The absence of yeast extract, ammonium chloride or phosphate (K2HPO4/KH2PO4) had a negative effect on the production of the biodemulsifier and significantly inhibited its activity. To further enhance the biodemulsifier efficiency, the optimal medium composition was determined using the response surface methodology (RSM) based on the central composite rotation design (CCRD). Using the optimized biodemulsifier production medium: 8.5 g/l glucose: 3% (v/v) liquid paraffin; 1.5 g/l yeast extract; 3.36 g/l NH4Cl and15 g/l phosphate, the demulsifying ratio increased 35.5% and biodemulsifier yield increased to 2.07 g/l. Evaluation of screening methods for demulsifying bacteria and characterization of lipopeptide bio-demulsifier produced by Alcaligenes sp.

Abstract:In this paper, surface tension measurement, oil-spreading test and blood-plate hemolysis test were attempted in the screening of demulsifying bacteria. After the comparison to the screening results obtained in demulsification test, 50mN/m of surface tension of culture was proposed as a preliminary screening standard for potential demulsifying bacteria. For the identification of efficient demulsifying strains, surface tension level was set at 40mN/m. The detected strains were further verified in demulsification test. Compared to using demulsification test alone as screening method, the proposed screening protocol would be more efficient. From the screening, a highly efficient demulsifying stain, S-XJ-1, was isolated from petroleum-contaminated soil and identified as Alcaligenes sp. by 16S rRNA gene and physiological test. It achieved 96.5% and 49.8% of emulsion breaking ratio in W/O and O/W kerosene emulsion within 24h, respectively, and also showed 95% of water separation ratio in oilfield petroleum emulsion within 2h. The bio-demulsifier was found to be cell-wall combined. After soxhlet extraction and purification through silicon-gel column, the bio-demulsifier was then identified as lipopeptide biosurfactant by TLC and FT–IR.
Analysis of Biological Demulsification Process of Water-in-oil Emulsion by Alcaligenes Sp. S-XJ-1.

Abstract:A demulsifying strain (S-XJ-1) was isolated from petroleum-polluted soil and identified as Alcaligenes sp. It showed emulsion breaking ratio of 81.3% for W/O emulsion within 24h when the cell concentration was 500mg/L. Evolution of water droplets during the biological demulsification process was investigated using a Turbiscan stability analyzer and microphotography. Further investigation focused on cell surface hydrophobicity and oil–water interfacial properties. The biological demulsification process began with rapid dispersal of the cells into the oil phase and adsorption onto the oil–water interface. This occurred due to high cell surface hydrophobicity and the presence of amphiphilic compounds in the cell walls. The cells had higher interfacial activity than the emulsifier molecules, and they displaced some of the emulsifier molecules, which effectively reduced the interfacial tension gradient. As a result, the interfacial film strength decreased, the water droplets coalesced and eventually phase separation occurred.
Enhanced Solubilization of Pyrene by Saponin,a Plant-Derived Biosurfactant

Abstract:Biosurfactants have great advantages in the remediation of organic contaminated soils because of their environmentally friendly nature. The solubilization of pyrene by saponin,a plant-derived biosurfactant,was studied and the results indicated that saponin can greatly enhance the solubilization of pyrene in solution and the weight solubilization ratio (MSR) of saponin to pyrene was greater than those of some representative nonionic surfactants. The enhanced solubilization of pyrene by saponin showed strong dependence on the solution pH and ionic strength. It decreased as the solution pH increased from 4.0 to 8.0,but increased as the NaCl concentration increased from 0.01 mol·L-1 to 1.0 mol·L-1,which can be attributed to the increased or decreased critical micelle concentration (CMC) of saponin solution with the changing pH or ionic strength,respectively. Saponin was more effective in enhancing pyrene solubilization at low pH and high ionic strength. In addition,heavy metal ions (Zn2+ and Cd2+) can also enhance the solubilization of pyrene by saponin.
Characterization and Phylogenetic Analysis of Biodemulsifier-Producing Bacteria

Abstract:Based on demulsification performance, twenty biodemulsifier-producing strains were isolated from various environmental sources. Five of them achieved nearly or over 90% of emulsion breaking ratio within 24 h. With the aid of biochemical and physiological tests and 16S rDNA analysis, these isolates were classified into eleven genera, in which six genera (Brevibacillus sp., Dietzia sp., Ochrobactrum sp., Pusillimonas sp., Sphingopyxis sp. and Achromobacter sp.) were firstly reported as demulsifying strains. Moreover, with data in this study and other literatures, a phylogenic tree was constructed, showing a rich diversity of demulsifying bacteria. Half of these bacteria belong to Actinobacterale order, which is famous for hydrocarbon degradation and biosurfactant biosynthesis. However, some strains in the same genera differed remarkably in demulsifying capability, surface properties and biochemical and physiological characteristics. This implied the biosynthesis and composition of biodemulsifier were more complicated than expected.
Separation and Purification of Alcaligenes Biodemulsifier by Dichloromethane-Water Extraction and Silica Gel Column Chromatograph

Abstract:Biodemulsifier is one of the green demulsifier which could be hopeful used in crude oil industry. In this paper, pretreatment with dichloromethane and water extraction was used to extract active demulsification substance from the demulsifying bacteria Alcaligenes sp. S-XJ-1. The crude extract were consist of four amino compounds and one composition of saccharide by thin layer chromatography. It was established experiment condition of silica gel column chromatograph for crude extract purification. Demulsification ratio of the purified sample G4 was 70.8% after 48h. The purified active substance was a peptide compound by further analysed with infrared spectrum and amino acid analysis.
Separation and Characterization of Effective Demulsifying Substances from Surface of Alcaligenes Sp S-Xj-1 and Its Application in Water-In-Kerosene Emulsion

Abstract:The main goal of this work was to analyze the effect of surface substances on demulsifying capability of the demulsifying strain Alcaligenes sp. S-XJ-1. The demulsifying substances were successfully separated from the cell surface with dichloromethane-alkali treatment, and exhibited 67.5% of the demulsification ratio for water-in-kerosene emulsions at a dosage of 356 mg/L. FT-IR, TLC and ESI-MS analysis confirmed the presence of a carbohydrate-protein-lipid complex in the demulsifying substances with the major molecular ions from mass-to-charge ratio (m/z) 165 to 814. After the substances separated, the cell morphology changed from aggregated to dispersed, and the concentration of cell surface functional groups decreased. Cell surface hydrophobicity and the ability of cell adhesion to hydrophobic surface of the treated cells was also reduced compared with original cell. It was proved that the demulsifying substances had a significant effect on cell surface properties and accordingly with demulsifying capability of Alcaligenes sp. S-XJ-1. (C) 2013 Elsevier Ltd. Simultaneous Removal of Polycyclic Aromatic Hydrocarbons and Heavy Metal from Co-Contaminated Soils by Saponin, a Plant-Derived Biosurfactant

Abstract:Biosurfactants have great advantages in the remediation of organic contaminated soils as their environmental friendly nature. The performance of saponin,a plant-derived biosurfactant,for the removal of polycyclic aromatic hydrocatbons(PAHs) and heavy metal from co-contaminated soils were studied. The results indicated that saponin can greatly enhance the solubilization of phenanthrene and pyrene in solution by the partition of them into saponin micelle and then the removal of phenanthrene and pyrene from PAHs-contaminated soil. The maximum removal rates of 84.1% and 81.4% were obtained for phenanthrene and pyrene from PAHs-contaminated soil,respectively. Saponin could form complex compound with heavy metals by the external carboxyl groups,which greatly enhanced the removal of zinc and cadmium from heavy metal contaminated soil and the removal rates of 93.0% and 79.6% were obtained for zinc and cadmium with a sapoin concentration of 0.4%,respectively. For the co-contaminated soil with PAHs and heavy metals,87.6% of phenanthrene,83.5% of pyrene,92.3% of zinc and 78.6% of cadmium were simultaneously removed by saponin at a concentration of 0.4%. The removal rates of zinc and cadmium from co-contaminated soil did not show obvious difference with the presence of PAHs,but the addition of heavy metals slightly enhanced the removal of phenanthrne and pyrene from the co-contaminated soil. Thus,saponin had the potential capability to simultaneously remove PAHs and heavy metal from the co-contaminated soils.

Abstract:As a new member of demulsifier family, biodemulsifier is applied in oil-water emulsion breaking. A strain, XJ-T-1, was isolated from petroleum-contaminated soil and identified as Alcaligenes sp.. Its physicochemical properties, demulsification capability and utilization of waste oil were further investigated. With waste frying oil (WFO) as carbon source, the demulsifier produced by Alcaligenes sp. showed high demulsifying capability. Moreover, the production of demulsifier was 4.6 times of that generated with paraffine as the carbon source. The CMC(-1) of biodemulsifier produced by paraffine and waste frying oil was achieved at 10 and 20, respectively. The biodemulsifier cultured with paraffine as the carbon source achieved 96% and 50% of emulsion breaking ratio in W/O (water in oil) and O/W (oil in water) model emulsion, while the demulsifier cultured on waste frying oil II as carbon source exhibited 97.8% and 65% demulsification ratio in the two model emulsions correspondingly. From the dynamic change of kerosene breaking ratio, emulsion breaking ratio and water breaking ratio during demulsification process, it was found that this biodemulsifier reacted with the continuous phase of the emulsion prior to its reaction with the dispersed phase. It was identified the valid part of biodemulsifier produced by WFO II was lipopeptide by TLC and IR.

Abstract:The inability to demulsify oil-in-water emulsions via green and efficient processes is a challenging problem in many industrial processes. As a novel biodemulsifier, protein demulsifiers display excellent dispersibility and stability, but their demulsification mechanisms are not clear, which severely restricts their large-scale production and application. In this study, the demulsification mechanism of the high-efficiency protein biodemulsifier oxalate decarboxylase (Bacm OxdC), which is secreted by the Bacillus mojavensis XH1 strain, for an oil-in-water emulsion was analyzed. The results showed that Bacm OxdC was spontaneously adsorbed at the oil-water interface and turned its hydrophobic amino acids outward to increase its hydrophobicity and break the emulsified system. Furthermore, it effectively reduced the oil-water interfacial tension and interfacial film strength, thereby reducing the oil-water interfacial energy and finally enabling demulsification. To further improve the demulsification efficiency and reusability, Fe3O4@SiO2@OxdC-DDSA was prepared. This method provided a magnetic response for Bacm OxdC and enabled efficient demulsification. The demulsification rate of Fe3O4@SiO2@OxdC-DDSA reached 98.1% at 24 h, which was 30.7% higher than that of the original Bacm OxdC. After three cycles, the demulsification rate still reached 89.3%, proving it has excellent recyclability. This work is the first study on the demulsification mechanism of protein biodemulsifiers and provides useful insights into the demulsification mechanism of biodemulsifiers for oil-in-water emulsions. In addition, a promising high-efficiency modification technique for protein biodemulsifiers was proposed, which provided information for the development of biodemulsifiers for oil-water separation.

Abstract:A highly efficient demulsifying strain, LH-6, was isolated from petroleum-contaminated soil and identified as Bacillus cereus by 16S rDNA gene analysis. It achieved 95.61 and 95.40 % demulsifying ratios within 12 h for water-in-oil (W/O) and oil-in-water (O/W) model emulsions, respectively. Fourier transform infrared spectroscopy (FT-IR) and thin-layer chromatography (TLC) detections indicated that the LH-6’s extracellular biodemulsifiers were different types of lipopeptides for the W/O and O/W emulsions. Optimization of the culture medium composition was conducted to improve the biosynthesis and demulsifying efficiency of the biodemulsifier. The optimal carbon source was liquid paraffin, while waste frying oil could also be an alternative carbon source. The optimal nitrogen sources were ammonium sulfate and yeast extract. To further enhance the biodemulsifier efficiency, the optimal cultivation conditions were determined using response surface methodology (RSM) based on central composite rotation design (CCRD). Using the optimized cultivation conditions, the demulsifying ratios increased to 98.23 and 97.65 % for the W/O and O/W model emulsions, respectively.
