Nanosecond Pulsed Electric Fields Enhance Chondrogenesis of Mesenchymal Stem Cells (mscs) Partially Via Phosphorylation of P38-Mapk

Abstract:Purpose: Biophysical signals play critical roles in chondrogenesis of mesenchymal stem cells (MSCs), whereas, however, unknown target effects and varied dose effects limit their wide application. In previous study, we found that nanosecond pulsed electric fields (nsPEFs) regulated phenotype of primary chondrocytes via activation of Wnt/β-catenin signaling pathways, as well as enhanced chondrogenesis of MSCs through P38-MAPK signaling pathways. In consideration of their dose effects and multiple target effects, hereafter, we comprehensively evaluate effects of nsPEFs with varied voltage and duration with an aim to further optimize chondrogenesis. Methods: Primary Porcine bone marrow-derived stem cells (bMSCs) were subjected to nsPEFs with 16 parameters (durations:10ns, 60ns, 100ns, 300ns) plus (voltages: 5kv/cm, 10kv/cm, 20kv/cm, 30kv/cm). Proliferation of the cells was evaluated by cck8 (Cell Counting Kit-8) test at day1 and day3. Gene expression of COL I/II/X, aggrecan (AGG) and Sox9 were evaluated at day 3 and 14 respectively while histology and quantification of glycosaminoglycan (GAG) were done at day 21 after bMSCs were subjected to 5 pulses of electric field (10 kV/cm or 20 kV/cm, 100ns duration). Protein expressions of P38-MAPK and phosphorylation of P38-MAPK were tested with western blotting at 1 hour of nsPEFs treatment. Results: nsPEFs with relatively short duration and low voltages enhanced proliferation (less than two-folds), while they gradually inhibited proliferation (up to 10 percent of the controls) of the cells with increased duration and voltages. nsPEFs with 100ns of both 10Kv/cm and 20Kv/cm were used in later experiment, as the former one mildly enhanced 10 percent of proliferation and the latter one significantly hindered 70% of proliferation. Gene expression of COL II and Sox9 increased significantly at day 3 (One-way anova, p=0.01), while gene expression of COL I/X remained stable, when the cells were subjected to both 10 kV/cm and 20 kV/cm of nsPEFs. 10 kV/cm of nsPEFs increased COL II at day 3.(1.9-fold, p=0.01). Gene expression of aggrecan (AGG) increased with 2.5-fold at day 3 (p=0.001) when subjected to 10Kv/cm, while there was no obvious changes in 20kv/cm group. Quantification of accumulated GAG showed that nsPEFs at 10 kV/cm increased to 1.11-fold while 20 kV/cm decreased to 0.94-fold. However, alcian blue staining showed there was no difference between 10 kV/cm nsPEFs and 20 kV/cm nsPEFs groups. Expression of p-P38 protein was increased at 1 hour after 10 kV/cm and 20kV/cm. nsPEFs treatment. Conclusions: nsPEFs with optimized parameters could potentially promote chondrogenesis with well-defined mechanism, while phosphorylation of P38-MAPK could be involved.
