Abstract:A new method for the synthesis of benzofurans or indoles via ligand-free CuBr-catalyzed coupling/cyclization of terminal alkynes with N-tosylhydrazones derived from o-hydroxy- or o-aminobenzaldehydes has been developed. A wide range of functional groups were found that are able to tolerate the reaction conditions.

Abstract:Ethyne is employed as coupling partner in copper-mediated cross-coupling reactions with N-tosylhydrazones and α-diazoacetate, leading to the development of a new synthetic method for terminal allenes. With this novel coupling method, the terminal allenes were obtained in good yields and with excellent functional group tolerance. Copper carbene migratory insertion is proposed as the key step in these transformations.

This method, which uses readily available starting materials and is operationally simple, offers 1,3-disubstituted allenes in moderate to good yields. Abstract:A highly efficient method for the synthesis of tri-aryl-substituted allenes has been developed through Cu(I)-catalyzed coupling of diaryldiazomethanes with terminal alkynes. Abstract:A mild three-component synthetic approach to versatile 2-amino-1,4-dihydropyridines from terminal alkynes, sulfonyl azides, and N-sulfonyl-1-aza-1,3-butadienes was successfully developed and relied on the in situ generation of metalated ynamide intermediates Ib to achieve a formal inverse electron-demand hetero-Diels-Alder reaction. Experimental results suggest that alkali metal cations (Li(+) and Cs(+) ions) might play a critical role to achieve the cycloaddition process.

Abstract:In this chapter, alkene synthesis based on the reaction of N-tosylhydrazones is described. The reactivity of tosylhydrazones is determined by either the acidity of α-proton and hydrazone proton or the electropositivity of the carbon of C=N bond. This leads to diverse reactivities and a series of N-tosylhydrazone-based olefination methodologies. Both non-catalytic and transition metal-catalyzed olefinations from N-tosylhydrazones are introduced in this chapter. Most of the transition metal-catalyzed reactions proceed via metal carbene transformations. The synthesis of alkenes through Pd-catalyzed cross-coupling reactions of N-tosylhydrazones is particularly attractive and will be discussed in detail.

Abstract:Significant attention has been given in the past few years to the selective transformations of N-tosylhydrazones to various useful compounds. However, the development of enantioselective versions poses considerable challenges. Herein we report a Pd-catalyzed enantioselective three-component coupling of N-tosylhydrazone, aryl halide, and terminal alkyne under mild conditions utilizing a novel chiral sulfinamide phosphine ligand (GF-Phos), which provides a facile access to chiral diarylmethyl alkynes, which are useful synthons in organic synthesis as well as exist as the skeleton in many bioactive molecules. A pair of enantiomers of the product could be easily prepared using the same chiral ligand by simply changing the aryl substituents of the N-tosylhydrazone and aryl halide. The salient features of this reaction include the readily available starting materials, general substrate scope, high enantioselectivity, ease of scale-up, mild reaction conditions, and versatile transformations.

Abstract:We report herein the copper- and palladium-catalyzed decarboxylative cross-coupling reactions of alkynyl carboxylic acids and N-tosylhydrazones, affording trisubstituted allenes and conjugated enynes, respectively. Abstract:A palladium-catalyzed reductive coupling between N-tosylhydrazones and aryl bromides has been developed. The reaction provides an efficient method for the synthesis of diarylmethanes and triarylmethanes via the formation of C(sp(2))-C(sp(3)) single bonds. This new methodology for the synthesis of diarylmethanes and triarylmethanes is featured by the ready availability of the starting materials, mild reaction conditions, and the tolerance of wide range of functional groups. The reaction follows a pathway including palladium carbene formation, migratory insertion, and reduction of the alkylpalladium(II) intermediate.

Abstract:C–C Bond formation and β-F elimination have been achieved in a Cu(I)-catalyzed cross-coupling reaction of terminal alkynes and trifluoromethyl ketone N-tosylhydrazones. The reaction represents an efficient synthesis of 1,1-difluoro-1,3-enyne derivatives. Mechanistically, the migratory insertion of the copper carbene intermediate leads to the C–C bond formation, which is followed by C–F bond cleavage.

Abstract:Herein, the use of triphenylmethylium tetrakis(pentafluorophenyl)borate as an efficient catalyst for the intermolecular dehydrative coupling of p‐hydroxymandelic acid hydrate with substituted aromatic amines and (thio)phenols was developed, which allows for the preparation of aryl acetic acid derivatives in a convenient manner. Notably, this method offers several advantages, including being metal‐free, prefunctionalization free, high atom efficiency, highc regioselective, and utilizing easily accessible starting materials. Additionally, it exhibits a broad substrate scope, enabling the synthesis of a range of aryl acetic acid derivatives. Furthermore, this method can also be applied to the etherification of arylthiol derivatives, facilitating the preparation of various thiolated products. The readily available starting materials and broad substrate scope enhance the applicability and potential utility of this method in organic synthesis.
