Abstract:Ethyne is employed as coupling partner in copper-mediated cross-coupling reactions with N-tosylhydrazones and α-diazoacetate, leading to the development of a new synthetic method for terminal allenes. With this novel coupling method, the terminal allenes were obtained in good yields and with excellent functional group tolerance. Copper carbene migratory insertion is proposed as the key step in these transformations.

Abstract:Methods toward the dimerization and homocoupling of terminal alkynes have been established that employ Co complexes along with tBuOK under different atmosphere. A variety of Z-1,3-enynes were furnished with a high Z:E (up to 93:7) ratio in moderate to very good yields under inert atmosphere. Alternatively, the Glaser homocoupling reaction of terminal alkynes was also accomplished with a Co catalyst, successfully affording various 1,3-diynes in excellent yields (up to 93%) under oxygen.

Abstract:Chiral allene and N -heteroaryl motifs are present in an ever-growing list of biologically active natural products and synthetic drugs. Although significant progress has been made in asymmetric syntheses of chiral allenes, general and practical protocols for enantioselective syntheses of chiral N -heteroaryl-substituted allenes from readily available starting materials still remain rare. Here we report a highly enantioselective synthesis of quinolinyl-substituted chiral allenes through a copper-catalyzed asymmetric allenylation of quinoline N -oxides with readily available 1,3-enynes. A variety of 1,3-enynes react with quinoline N -oxides, affording the corresponding quinolinyl-substituted allenes in high yields (up to 95%) and high enantioselectivities (up to 99% ee). This transformation tolerates a variety of functional groups, such as chloro, bromo, trifluoromethyl ether, tertiary amine, siloxy, carboxylic ester, imide, pyridine, and thiophene moieties. DFT calculations suggest a pathway involving an intramolecular nucleophilic addition of an allenyl copper intermediate with a coordinated quinoline N -oxide through a five-membered, rather than seven-membered, transition state.

Abstract:We report herein the copper- and palladium-catalyzed decarboxylative cross-coupling reactions of alkynyl carboxylic acids and N-tosylhydrazones, affording trisubstituted allenes and conjugated enynes, respectively. With readily available alkynyl carboxylic acids as the acetylene surrogates, the new strategies are operationally simple and show good functional group tolerance.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

A wide range of trisubstituted allenes may be synthesized efficiently via this method. Preliminary mechanistic studies revealed that CuI and Ti(OEt)4 are in charge of the formation of propargylic amine, while ZnBr2 is responsible for the transformation from propargylic amine to allene.

Abstract:The cadmium iodide‐mediated reaction between an amine and two molecules of terminal alkynes for the synthesis of allenes has been investigated by using DFT calculations. On the basis of these calculations, a plausible mechanism has been proposed. The results also reveal that pyrrolidine serves not only as a reactant but also as a shuttle in a proton relay.

Abstract:A Cu(I)-catalyzed cross-coupling reaction of terminal alkynes and bis(trimethylsilyl)diazomethane is reported. Mechanistically, the reaction is based on the recently developed cross-coupling reactions through metal-carbene migratory insertion. This reaction provides an efficient synthetic method for 1,1-disilyl allenes. Subsequent transformations of 1,1-disilyl allenes are investigated, which show diverse reactivities of these allenes.

Abstract:A copper-catalyzed three-component reaction of cyclobutanone oxime esters and 1,3-enynes in the presence of TMSCN or TMSCF3 has been developed. This mild protocol enjoys a broad substrate scope tolerating many functional groups, providing a facile access to 1,7-double-functionalized allenes, which are difficult to prepare. The allenyl nitrile products may be easily transformed into allenoic acid derivatives and stereodefined tetrasubstituted alkenes, demonstrating their potentials as platform molecules in synthesis. Abstract:Although the synthesis of allenes has made considerable progress, methods for tetrasubstituted allenes are still limited. In this paper, the CuBr2-catalyzed cross-coupling reaction of Grignard reagents and propargylic ammonium salts in tetrahydrofuran at 60 degrees C under argon delivered tetrasubstituted allenes in good to excellent yields. The reaction is thought to proceed by an S(N)2' pathway.

Abstract:Development of methods for the sp2 C-H transformations of allenes has received much attention, and it presents a powerful tool for the synthesis of complicated allene-containing bioactive molecules. With a copper-catalyzed radical relay, sp2 allenic C-H arylation and alkynylation were established herein, using various aryl boronic acids and trimethoxysilyl-substituted alkynes as carbon nucleophiles and using electrophilic N-F reagents as nitrogen-centered radical precursors. These methods featured excellent site selectivity to deliver fully substituted allenes efficiently. Moreover, with silyl-substituted allenes as substrates, a subsequent dual sp2 C-H functionalization process was established as well, which allowed for the divergent synthesis of multifunctionalized allenes, significantly expanding their chemical spaces.

Abstract:Cu(I)‐catalyzed reaction of diazo compounds generates a Cu(I)‐carbene intermediate that undergoes diverse transformations. In the past few years, the diazo compounds (or their precursor N ‐tosylhydrazones) have been established as cross‐coupling partners under transition‐metal catalysis, affording various organic compounds. Particularly the breakthrough has been made in allene synthesis by Cu(I)‐catalyzed carbene coupling with terminal alkynes. Moreover, the Cu(I)‐catalyzed coupling reaction of diazo compounds with terminal alkynes generates allene intermediate that undergoes tandem cyclization/coupling to afford cyclic compounds. This review article summarizes the most recent developments in allene synthesis based on the Cu(I)‐carbene coupling reactions and the utilization of allene intermediates in tandem reactions.

Abstract:Allenes are important building blocks in organic synthesis and have attracted much attention from researchers worldwide. So far, mono- and 1,3-disubstituted allenes can be prepared easily by applying the allenylation of terminal alkynes reaction of aldehydes. However, trisubstituted allenes have never been reported using this reaction due to the extremely low reactivity of ketones. Here, we report the one-pot synthesis of trisubstituted allenes from terminal alkynes and ketones utilizing cadmium iodide as the mediator. With this protocol, a series of different allenes, including 1,5-bisallenes and optically active allenols, which are especially important in synthetic chemistry, have been successfully prepared.
