Abstract:Halloysite is a nanostructured clay mineral with hollow tubular structure, which has recently found an important role as delivery system for drugs or other active molecules. One of these is curcumin, main constituent in the rhizome of the plant Curcuma Longa, with a series of useful pharmacological activities, hindered by its poor bioavalaibility and solubility in water. In this study, Halloysite clay nanotubes (HNTs) were characterized in terms of both structure and biocompatibility and they were used for curcumin delivery to cancer cells. The performed 3-(4, 5-dimethythiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) assay showed that HNTs have a high biocompatibility, also when coated with polymers, while curcumin is highly toxic for cancer cells. The release kinetics of curcumin from HNTs was investigated by the dialysis bag method, showing a slow and constant release of the drug, which can be further controlled by adding layers of polyelectrolytes to the external surface of the tubes. Successful polymer coating was followed by Zeta potential. The Trypan Blue assay showed a cytotoxic effect of loaded HNTs, proportional to the concentration of tubes and the incubation time. Successful HNTs uptake by breast cancer cells was demonstrated by Confocal Laser Scanning Microscopy images. All results indicate that HNTs are a promising carriers for polyphenol delivery and release.

Abstract:Nanoscale drug delivery systems have emerged as promising alternatives to overcome the problems associated with by conventional chemotherapy for cancer treatment such as poor drug stability and bio-distribution. Herein, we report a single walled carbon nanotubes (SWCNTs) based drug delivery system functionalized with polysaccharides such as alginate (ALG) and chitosan (CHI), which can be loaded with an anticancer drug curcumin (CUR). Modification of SWCNTs renders high drug loading efficiency and sustained drug release, imperative for drug activity. These were characterized through various tools viz, microscopic (transmission electron microscopy, scanning electron microscopy and atomic force microscopy) and zeta potential analysis. Incorporation of CUR inside the modified SWCNTs was studied through Fourier transform infrared spectroscopy, fluorescence and UV-visible spectroscopy. In vitro release studies were conducted to gain an insight into the pH-dependent release behavior of the entrapped CUR from modified SWCNTs. The anti-cancer potential was further demonstrated using human lung adenocarcinoma (A549) cells as a model system. Various cell culture based assays were performed to study the ability of released CUR from modified SWCNTs for inhibiting the cancer cell proliferation by inducing apoptosis.
Enhancement of curcumin antitumor efficacy and further photothermal ablation of tumor growth by single-walled carbon nanotubes delivery system in vivo

Abstract:Curcumin, a commonly used natural product for antitumor therapy, is unable to achieve full potential due to poor bioavailability. Based on our previous report of a novel delivery system for curcumin using functionalized single-walled carbon nanotubes by phosphatidylcholine and polyvinylpyrrolidone (SWCNT-Cur), we further evaluated SWCNT-Cur's performance in vivo and characteristics in vitro. SWCNT-Cur significantly increased the blood concentration of curcumin, up to 18-fold, in mice. And in a murine S180 tumor model, SWCNT-Cur exhibited significantly higher inhibition efficacy on tumor growth and no obvious toxicity in main organs. Moreover, photothermal therapy induced by SWCNT under near-infrared radiation further facilitated SWCNT-Cur to inhibit the tumor growth in vivo. In addition, solvent residue is negligible in SWCNT-Cur formulation, and hydrogen bonding was formed between void carriers and curcumin, as demonstrated by GC chromatograph and IR spectra. Furthermore, experiments of confocal microscopy and spectrofluorometer showed that SWCNT-Cur gave a six-fold higher uptake for curcumin compared to native curcumin in human prostate cancer PC-3 cells. In conclusion, curcumin delivery with functionalized SWCNT is a promising strategy to enhance anticancer activity in vivo by enhancing cell uptake and blood concentration, changing physicochemical properties of curcumin and combining phototherapeutic with chemotherapeutic effects.

Abstract:Curcumin (Cur) and Melittin (Mel) are two natural extracts that have been shown anti-tumor effects. However, their applications are limited due to poor oral bioavailability and the lack of tumor-targeting property. Here, we developed a novel nanocomposite that enabled the co-delivery of Cur and Mel, which consists of α-lactalbumin protein nanotubes (NTs), positively charged N,N,N-trimethyl chitosan (TMC), and a tumor-targeting cyclic peptide iRGD. The results showed that NTs/Cur-TMC-Mel-iRGD incorporated the advantages of each component, for instance, effective compounds loading by NTs, improved cellular uptake by TMC, prolonged accumulation in tumors by iRGD as well as synergistic anti-tumor effects of Cur and Mel. In the tumor-bearing mice, NTs/Cur-TMC-Mel-iRGD treatment remarkably induced cancer cell apoptosis while inhibiting cell proliferation, leading to suppressed tumor growth. Besides, no obvious adverse effects were observed in the blood physiology and tissue histology. Overall, our study provided an effective strategy for co-delivering Cur and Mel, which has a potential for translational clinical research aiming to treat solid tumors.

Abstract:Low water solubility, extensive metabolism, and drug resistance are the existing unavoidable disadvantages of the insoluble drug curcumin in biomedical applications. Herein, we employed d-α-tocopherol polyethylene glycol 1000 succinate (TPGS)-functionalized near-infrared (NIR)-triggered photothermal mesoporous nanocarriers with bamboo charcoal nanoparticles (TPGS-BCNPs) to load and deliver curcumin for improving its bioavailability. This system could considerably increase the accumulation of curcumin in cancer cells for enhanced curcumin bioavailability via simultaneously promoting the cellular internalization of the as-synthesized composite (TPGS-BCNPs@curcumin) by the size effect of NPs and considerably triggering controlled curcumin release from TPGS-BCNPs@curcumin by NIR stimulation and reducing efflux of curcumin by the P-glycoprotein (P-gp) inhibition of TPGS, so as to enhance the therapeutic effect of curcumin and realize a better chemo-photothermal synergetic therapy in vitro and in vivo. Besides cancer therapy, studies indicated that curcumin and some carbon materials could be used as radical scavengers that play an important role in the radioprotection of normal cells. Hence, we also investigated the free-radical-scavenging ability of the TPGS-BCNPs@curcumin composite in vitro to preliminarily evaluate its radioprotection ability for healthy tissues. Therefore, our work provides a multifunctional delivery system for curcumin bioavailability enhancement, chemo-photothermal synergetic therapy of cancer, and radioprotection of healthy tissues.
Development of curcumin-loaded galactosylated chitosan-coated nanoparticles for targeted delivery of hepatocellular carcinoma

Abstract:Curcumin (CUR) has good antitumor effects, but its poor aqueous solubility severely limits its clinical application and the systemic nonspecific distribution of the free drug in tumor patients is a key therapeutic challenge. In order to overcome the limitations of free drugs and improve the therapeutic efficacy, we developed novel galactosylated chitosan (GC)-modified nanoparticles (GC@NPs) based on poly (ethylene glycol) methyl ether- block -poly (lactide- co -glycolide) (PEG-PLGA), which can target asialoglycoprotein receptor (ASGPR) expressed on hepatocellular carcinoma cells and have excellent biocompatibility. The results showed that the drug loading (DL) of CUR was approximately 4.56 %. A favorable biosafety profile was maintained up to concentrations of 500 μg/mL. Furthermore, in vitro cellular assays showed that GC@NPs could be efficiently internalized by HepG2 cells via ASGPR-mediated endocytosis and successfully released CUR for chemotherapy. More importantly, in vivo anti-tumor experiments revealed that GC@NPs were able to accumulate effectively within tumor sites through EPR effect and ASGPR-mediated endocytosis, leading to superior inhibition of tumor growth compared to free CUR. Overall, GC@NPs are a promising CUR nanocarrier for enhanced tumor therapy with a good biosafety profile.

Abstract:Zeolitic imidazolate framework-8 (ZIF-8) is a promising material for drug delivery, but its poor stability limits its wide application. In this work, homogeneous dodecahedral ZIF-8 was prepared by the room-temperature solution stirring method, and to improve the stability of ZIF-8, ZIF-8@mSiO 2 and DZIF-8 (ZIF-8 after calcination) @PMO composites with the core-shell structure were obtained using mesoporous silica (mSiO 2 ) and periodic mesoporous organic silica (PMO) coatings. Folic acid-coupled chitosan (CS-FA) was modified on nanoparticles, and curcumin (CUR) was used as a model anti-tumor drug to obtain the drug delivery systems with targeting and pH stimulation response: ZIF-8@mSiO 2 @CUR@CS-FA and DZIF-8@PMO@CUR@CS-FA, which exhibited good dispersion and high drug loading capability. In addition, we used the MTT assay to assess the cytotoxicity of the drug-carrying systems and selected human cervical cancer cells (Hela) as model cells. The results of the in vitro experiments suggested that the final CUR-loaded drug delivery systems had significantly increased anticancer efficiency compared with the free drug CUR, which improved the therapeutic effect of the hydrophobic CUR on tumors. In conclusion, the pH-responsive nanocarriers provided potential for the targeted delivery of anticancer medication, resulting in a platform for drug-controlled release and cancer therapy.

Highly Soluble and Stable, High Release Rate Nanocellulose Codrug Delivery System of Curcumin and AuNPs for Dual Chemo-Photothermal Therapy

Abstract:As a natural antitumor drug, curcumin (CUR) has received increasing attention from researchers and patients due to its various medicinal properties. However, currently CUR is still restricted due to its low and stand-alone therapeutic effects that seriously limit its clinical application. Here, by using cellulose nanocrystals (CNCs) as a nanocarrier to load CUR and AuNPs simultaneously, we developed a hybrid nanoparticle as a codrug delivery system to enhance the low and stand-alone therapeutic effects of CUR. Aided with the encapsulation of beta-cydodextrin (mu CD), both the solubility and the stability of CUR are greatly enhanced (solubility increased from 0.89 to 131.7 mu g/mL). Owing to the unique rod-like morphology of CNCs, the system exhibits an outstanding loading capacity of 31.4 mu g/mg. Under the heat effects of coloaded AuNPs, the system demonstrates a high release rate of 77.63%. Finally, with CNC as a bridge nanocarrier, all aforementioned functions were integrated into one hybrid nanoparticle. The all-in-one integration ensures CUR to have enhanced therapeutic effects and enables the delivery system to exhibit combined chemo-photothermal therapy outcomes. This work presents a significant step toward CUR's clinical application and provides a new strategy for effective and integrative treatment of tumor disease.
Efficient Delivery of Curcumin by Alginate Oligosaccharide Coated Aminated Mesoporous Silica Nanoparticles and In Vitro Anticancer Activity against Colon Cancer Cells

Abstract:We designed and synthesized aminated mesoporous silica (MSN-NH2), and functionally grafted alginate oligosaccharides (AOS) on its surface to get MSN-NH2-AOS nanoparticles as a delivery vehicle for the fat-soluble model drug curcumin (Cur). Dynamic light scattering, thermogravimetric analysis, and X-ray photoelectron spectroscopy were used to characterize the structure and performance of MSN-NH2-AOS. The nano-MSN-NH2-AOS preparation process was optimized, and the drug loading and encapsulation efficiencies of nano-MSN-NH2-AOS were investigated. The encapsulation efficiency of the MSN-NH2-Cur-AOS nanoparticles was up to 91.24 ± 1.23%. The pH-sensitive AOS coating made the total release rate of Cur only 28.9 ± 1.6% under neutral conditions and 67.5 ± 1% under acidic conditions. According to the results of in vitro anti-tumor studies conducted by MTT and cellular uptake assays, the MSN-NH2-Cur-AOS nanoparticles were more easily absorbed by colon cancer cells than free Cur, achieving a high tumor cell targeting efficiency. Moreover, when the concentration of Cur reached 50 μg/mL, MSN-NH2-Cur-AOS nanoparticles showed strong cytotoxicity against tumor cells, indicating that MSN-NH2-AOS might be a promising tool as a novel fat-soluble anticancer drug carrier.

Abstract:Owing to great anticancer activity and excellent biosafety, curcumin (Cur) has potential development in cancer therapy. Nevertheless, the anticancer activity of Cur is limited by the poor selectivity toward tumors and low bioavailability. Inspired by coordination-driven self-assembly, it is a promising strategy for controlling drug delivery due to its rich structure and properties, easy composition integration, and biodegradation. Here, a fucoidan-based Cur nanoparticle constructed from natural components synthesized via a green one-pot reaction of Cur, fucoidan (FU), and Zn2+ based on coordination-driven self-assembly, namely, FU/Zn2+/Cur-nanoparticles (FZC-NPs), was reported for improving cancer therapy. The obtained FZC-NPs displayed a small hydrodynamic diameter, high stability, high drug loading efficiency, glutathione (GSH)-triggered drug accurate release behaviors, fast intracellular uptake, and good penetrability ability into the tumor spheres. Compared to free Cur, FZC-NPs had a stronger ability to induce mitochondrial damage and inhibit tumor cell invasion and migration. The in vivo experiments showed that FZC-NPs had effective accumulation in tumor sites, excellent antitumor activity, and good biocompatibility. These results confirmed that the developed FZC-NPs is a safe and anticancer performance therapeutic agent for cancer-targeted therapy.

Abstract:<p>In order to achieve high loading and efficient delivery of curcumin, phenylboronic acid-conjugated chitosan nanoparticles were prepared by a simple desolvation method. These nanoparticles exhibited a regular spherical shape with the average size about 200−230 nm and narrow size distribution, which were kinetically stable under physiological condition. Due to boronate ester formation between curcumin and phenylboronic acid groups in the nanoparticles, and the hydrogen bonding interactions between curcumin and nanocarriers, curcumin was successfully loaded into the nanoparticles with high drug loading content. These curcumin-loaded nanoparticles showed pH and reactive oxygen species (ROS)-triggered drug release behavior. <em>In vitro</em> cell experiments revealed that the blank nanoparticles were completely nontoxic to cultured cells, and the curcumin-loaded nanoparticles exhibited efficient antitumor efficiency against cancer cells. Moreover, the drug-loaded nanoparticles performed an enhanced growth inhibition in three-dimensional multicellular tumor spheroids. Thus, these nanocarriers would be a promising candidate for curcumin delivery in tumor treatment.</p>

Abstract:A multiwalled carbon nanotube (MWNT)-based drug delivery system was developed by covalently combining carbon nanotubes with the antitumor agent 10-hydroxycamptothecin (HCPT) using hydrophilic diaminotriethylene glycol as the spacer between nanotube and drug moieties. The surface functionalizations of the nanotube were carried out by enrichment of carboxylic groups with optimized oxidization treatment, followed by covalently linking hydrophilic diaminotriethylene glycol via amidation reaction, and then HCPT was chemically attached to carbon nanotubes through a cleavable ester linkage. It is demonstrated that the obtained MWNT-HCPT conjugates are superior in antitumor activity both in vitro and in vivo to clinical HCPT formulation. In vivo single photon emission computed tomography (SPECT) imaging and ex vivo gamma scintillation counting analyses reveal that MWNT-HCPT conjugates have relatively long blood circulation and high drug accumulation in the tumor site. These properties together with the enhanced cell uptake and multivalent presentation of HCPT on a single nanotube benefit substantially the antitumor effects and would boost significantly the applications of carbon nanotubes in the biomedicine field.

Abstract:Chitosan is a biotechnological derivative of chitin receiving a widespread pharmaceutical and biomedical applications. It can be used to encapsulate and deliver cancer therapeutics with inherent pH-dependent solubility to confer drug targeting at tumour microenvironment and anti-cancer activity synergizing cancer cytotoxic drug actions. To further reduce the off-target and by-stander adverse effects of drugs, a high targeted drug delivery efficiency at the lowest possible drug doses is clinically required. The chitosan has been functionalized with covalent conjugates or complexes and processed into nanoparticles to encapsulate and control drug release, to avoid premature drug clearance, to deliver drugs passively and actively to cancer site at tissue, cell or subcellular levels, and to promote cancer cell uptake of nanoparticles through membrane permeabilization at higher specificity and scale. Nanomedicine developed using functionalized chitosan translates to significant preclinical improvements. Future challenges related to nanotoxicity, manufacturability, selection precision of conjugates and complexes as a function of cancer omics and their biological responses from administration site to cancer target need critical assessments.
Drug delivery potential of carbon and boron nitride nanotubes: A DFT-D3 analysis of curcumin binding interactions

Abstract:Recent experimental research has focused on loading the anticancer molecule curcumin (CurcH) into both carbon and boron nitride nanotubes (CNTs and BNNTs) to assess their efficiency as drug delivery carriers, addressing CurcH bioavailability issues. This study investigates CurcH adsorption on the inner and outer surfaces of zigzag (13,0) and (16,0) CNTs and BNNTs using the Grimme Dispersion-corrected Density Functional Theory (DFTD3) method which more accurately describes weak interaction forces. Geometry optimization, adsorption energies, and Mulliken charges were performed for each nanotube@CurcH complex, revealing that the interaction occurs through a physisorption process. Among the various CurcH positions explored, the one closest to the inner sidewall of the (16,0) CNT exhibited the strongest binding affinity compared to the other nanostructures. Analysis of the predicted electronic properties, including band structure and total Density of States, revealed a considerable increase in electrical conductivity within BNNTs upon CurcH adsorption, in contrast to CNTs. Moreover, the Partial Density of States analysis and HOMO-LUMO isosurfaces were further examined to elucidate the electronic contributions of nanotube-CurcH entities around the Fermi level. These theoretical findings suggest that both CNTs and BNNTs exhibit favorable adsorption properties for CurcH, indicating their potential as promising nanocarriers for enhancing CurcH's therapeutic efficacy while minimizing its systemic toxicity.

Abstract:Abstract New drug-delivery systems have remained a challenge for pharmaceutical scientists due to the use of expensive polymers and the low loading capacity of prepared nanoparticles. There is pressure to develop formulations that contain not only cheaper materials but also have controlled-release properties. Halloysite nanotubes (HNTs) are a naturally occurring clay mineral similar to kaolin, possessing a special particle shape in the form of an ultramicroscopic multilayered hollow cylinder. Its uses encompass a wide range in anticancer therapy, sustained- and controlled-release drug-delivery systems, cosmetics, delivery of proteins, vaccines and genes. These advantages are due to its biocompatibility, significant mechanical strength and natural availability. The surfaces of the tubules can be modified by coating different polymers for application in the drug-delivery system. This review is focused on the various aspects of HNTs such as structure, properties, loading methods, applications and characterizations.

Abstract:As one of carbon-based nanomaterials, single-walled carbon nanotubes (SWCNTs) are widely regarded as potentially potent drug delivery carriers on account of their prominent properties. Nevertheless, their biomedical application, particularly in the drug delivery field has been seriously restricted by some inherent defects. In this study, SWCNTs materials were functionalized by covalent and non-covalent approaches, respectively. In short, the pristine SWCNTs were first purified with strong oxidizing acids (H 2 SO 4 /HNO 3 ), and the resulting carboxylated ones (CNTs) were attached sequentially by different modification agents, including polyethylene glycol (PEG), polyethyleneimine (PEI), folic acid (FA) and chitosan (CS). Various nanocarriers were then systematically characterized and comparatively evaluated. The results illustrated that all CNTs samples could act as drug delivery carriers since they had high drug loading efficiency, good biocompatibility and responsive drug release. In comparison with other CNTs, multi-functionalized ones (CNTs-PPFC) exhibited more excellent performance, such as rapid drug release at low pH condition, higher cell internalization efficiency, and enhanced antitumor activity toward MCF-7 cells. These advantages should be attributed to their better dispersion state and comparably higher affinity with tumor cells, which favor the more efficient selective cellular uptake and subsequent drug accumulation. Moreover, further pharmacological mechanism analysis also revealed that CNTs-PPFC/DOX could induce the apoptosis of MCF-7 cells most effectively, by triggering ROS overproduction and affecting cell cycle distribution. In conclusion, the multi-functionalized CNTs-PPFC can be utilized as promising nanocarriers, and the findings will contribute to the rational design of novel delivery vehicles for anticancer drugs.

Abstract:This paper presents the first phase shifter based on sol-gel Barium Calcium Zirconate Titanate (BCTZ) thin ferroelectric film. It is a coplanar waveguide distributed phase shifter with BCTZ varactors used as tuning elements. It presents a continuous phase shift of 285° at 12 GHz (180° at 8 GHz) with a 20 V bias voltage. Its figure of merit is 26°/dB and its return loss is better than 12 dB for the 8 to 12 GHz frequency bandwidth. The presented phase shifter is the only phase shifter based on low cost sol-gel deposition to compete with the best reported X band ferroelectric phase shifters.

Abstract:The research for alternative administration methods for anticancer drugs, towards enhanced effectiveness and selectivity, represents a major challenge for the scientific community. In the last decade, polymeric nanostructured delivery systems represented a promising alternative to conventional drug administration since they ensure secure transport to the selected target, providing active compounds protection against elimination, while minimizing drug toxicity to non-target cells. In the present research, poly(glycerol sebacate), a biocompatible polymer, was synthesized and then nanostructured to allow curcumin encapsulation, a naturally occurring polyphenolic phytochemical isolated from the powdered rhizome of Curcuma longa L. Curcumin was selected as an anticancer agent in virtue of its strong chemotherapeutic activity against different cancer types combined with good cytocompatibility within healthy cells. Despite its strong and fascinating biological activity, its possible exploitation as a novel chemotherapeutic has been hampered by its low water solubility, which results in poor absorption and low bioavailability upon oral administration. Hence, its encapsulation within nanoparticles may overcome such issues. Nanoparticles obtained through nanoprecipitation, an easy and scalable technique, were characterized in terms of size and stability over time using dynamic light scattering and transmission electron microscopy, confirming their nanosized dimensions and spherical shape. Finally, biological investigation demonstrated an enhanced cytotoxic effect of curcumin-loaded PGS-NPs on human cervical cancer cells compared to free curcumin.

Abstract:Biomolecule-based coordination complex nanoassemblies are a new type of functional materials that are attracting increasing attention. They could possess functionalities that are not readily attainable with other materials, and represent a promising research area that can be exploited in coordination chemistry and materials science. Using bioactive folic acid molecule as a linker, Ni-folate-hydrazine coordination complex nanotubes (CCNTs) have been effectively constructed using the solvothermal method. This is not only the first example of the CCNTs being formed using a nonpyridyl-based molecule as a linker, but also the first report on biomolecule-based CCNTs (BMB-CCNTs) with anticancer activity. It does not require any post treatment to achieve targeted delivery and biocompatible performance. The BMB-CCNTs are sufficiently stable at normal pH of 7.4 until it enters a tumor cell, subsequently it breaks open to release drug in the tumor cell. Furthermore, it overcomes the major limitations of antibody-drug and folate-drug conjugates and is a potential smart multi-functional nanomedicine system. The results of in vitro cytotoxicity assay reveal that the antitumor ability of BMB-CCNTs is similar to cisplatin (CDDP), while their cytotoxicity for normal cells is lower than the latter. Furthermore, BMB-CCNTs exhibit excellent performance as drug carriers and target agents for delivering drugs into tumor cells. Bio-TEM and confocal laser scanning microscope images trace the uptake process of CDDP-CCNTs by a tumor cell. CDDP-CCNTs exhibit dual anti-cancer effect.
