Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:A novel palladium-catalyzed cascade reaction of o-iodostyrenes, internal alkynes and formates to access various indene-1-acetates was developed. The method enables the construction of three C-C and one C-O bonds through sequential double carbopalladation and aryloxycarbonylation under a palladium/air system. This protocol exhibited high regioselectivity and broad functional group tolerance. In addition, the synthetic utility of this protocol has been successfully demonstrated by the gram-scale synthesis and the late-stage modification of a series of complex bioactive molecules.

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:A practical and effective palladium-catalyzed C–H activation/alkene insertion/annulation has been reported for the synthesis of furans and cyclopropanes from cyclic 1,3-diketones or 1,3-indandione and diverse alkenes, resulting in moderate to good yields. This protocol demonstrates excellent selectivity and is well-compatible with a wide range of alkene substrates, exhibiting exceptional regioselectivities, high efficiency, and good functional group tolerance.

Abstract:A photoinduced palladium‐catalyzed ring‐opening hydroarylation of furans to form functionalized indenes was achieved via an intramolecular reductive coupling of aryl bromide with furan using silane as the reductant. Deuterium tracking and DFT analysis revealed a reasonable reaction pathway involving: 1) the homolytic cleavage of the furan C−O bond induced by the α‐allyl radical; 2) a 1,6‐H atom transfer from carbon to oxygen.

Abstract:A tandem reaction initiated by Au(I)-catalyzed C1-C5 cyclization of enediynes bearing an internal carboxy nucleophile has been developed, providing a distinctive methodology for the assembly of fused indenes including indeno[1,2-c]isochromen-5(11H)-ones and indeno[1,2-b]pyran-2(5H)-ones. A mechanism involving the concerted formation of the five- and six-membered rings was proposed.

Abstract:Herein, a new iodide-mediated three-component annulation reaction of secondary anilines, cyclohexanones and elemental sulfur is demonstrated, which allows access to various phenothiazines with the merits of formation of multiple chemical bonds in one single operation, high step and atom efficiency, readily available feedstocks and catalyst system, good substrate and functional group compability. The developed chemistry capable of constructing novel phenothiazines with structural diversity offers a significant basis for further applications.

Abstract:A highly efficient heteroannulation between [60]fullerene (C60) and N-methoxy-1H-indole-1-carboxamides has been successfully achieved via the palladium-catalyzed C-H activation and subsequent cyclization. This protocol has remarkable functional group tolerance for the synthesis of C60-fused 3,4-dihydropyrimido[1,6-a]indol-1(2H)-ones. Furthermore, the electrochemically generated dianionic C60-fused 3,4-dihydropyrimido[1,6-a]indol-1(2H)-ones can be regioselectively transformed into 1,2,3,4-, 1,2,3,16-, and 1,4,9,25-adducts of C60.

Abstract:The asymmetric C–H annulation of O-pivaloyl 1-indolehydroxamic acid with donor/acceptor diazo compounds has been achieved for the first time, to the best of our knowledge, by using a rhodium catalyst embedded in a chiral binaphthyl backbone. This protocol constitutes a straightforward route for the synthesis of a new family of 1,2-dihydro-3H-imidazo[1,5-a]indol-3-one derivatives having a quaternary carbon stereocenter in high yields and excellent enantioselectivity (up to 98:2 er).

Abstract:An efficient and versatile method was developed for the access of multisubstituted 1,8-naphthyridine derivatives through a one-pot, three-component protocol from heterocyclic ketene animals, malononitrile dimer, and aryl aldehydes in high yields. The 1,8-naphthyridines were formed via Knoevenagel condensation, aza-ene reaction, imine-enamine tautomerization, and intramolecular cyclization.

Abstract:The enantioselective [3 + 2] annulation of α-hydroxy-1-indanones with yne-enones was accomplished by the dinuclear zinc catalysts via a Brønsted base and Lewis acid cooperative activation model. A wide range of enantioenriched spiro[2,3-dihydrofuran-2,2'-inden-1-one] derivatives were accommodated in good yields with excellent levels of diastereoselectivities and enatiostereoselectivities (up to 84% yield, 20:1 dr, 99% ee). This transformation features broad substrate scope, gram-scale synthesis, and the late-stage functionalization of complex molecule.

Abstract:Indole-containing polyaromatic scaffolds are widely found in natural products, pharmaceutical agents, and π-conjugated functional materials. Often, the synthesis of these highly valuable molecules requires a multistep sequence. Therefore, a simple, one-step protocol to access libraries of polyaromatic indole scaffolds is highly desirable. Herein we describe the direct synthesis of polysubstituted indolo[2,1-a]isoquinoline analogues via a double C-H annulation cascade using triazene as an internally cleavable directing group. Evidence from HRMS and theoretical calculations suggests that an unprecedented 1,2-alkyl migration might be responsible for the in situ cleavage of the directing group. Both kinetic isotope effects and DFT calculations suggested that the alkyne insertion step is rate-limiting for the second C,N annulation reaction.

Abstract:An efficient synthesis of N-fused polycyclic indoles by a palladium-catalyzed annulation/acyl migration cascade reaction is described. The reaction is ligand-free, scalable, and provides access to a diverse range of useful indole scaffolds from readily available starting materials. Supporting mechanistic studies indicate that the reaction likely proceeds via an intramolecular α-arylation mechanism. The synthetic utility of this protocol is demonstrated by a gram-scale reaction and syntheses toward indole alkaloids and a HSP90 inhibitor.

Abstract:An efficient one-pot synthesis of 1,2-dihydro-2-oxo-3-pyridinecarboxylate derivatives starting from enones and ethyl cyanoacetate in moderate to good yields is described. This novel tandem [3+3] annulation method mediatedby FeCl 3 involves Michael addition and ketone-nitrile annulation followed by aromatization-dehydrogenation. A plausible mechanism is also proposed.
