Abstract:Hydrogenolysis has emerged as one of the most effective means of converting polymeric lignin into monoaromatic fragments of value. Reported yields may be higher than for other methods and can exceed the theoretical yields estimated from measures of the content of lignin's most readily cleaved alkyl-aryl ether bonds in beta-ether units. The high yields suggest that other units in lignin are being cleaved. Diaryl ether units are important units in lignin, and their cleavage has been examined previously using simple model compounds, such as diphenyl ether. Herein, the hydrogenolysis of model compounds that closely resemble the native lignin 4-O-5 diaryl ether units was analyzed. The results provided unexpected insights into the reactivity and partial cleavage of these compounds. The models and lignin polymer produced not only monomers, but also unusual 1,3,5-meta-substituted aromatics that appear to be diagnostic for the presence and the cleavage of the 4-O-5 diaryl ether unit in lignin.

Abstract:The effective cleavage of C-O bonds in linkages of lignin was one of the significant strategies promoting lignin valorization. Herein, the strategy of C-O bonds cleavage of lignin using metal triflate as the catalyst was developed. The carboxylic acid or alcohol could be used as the nucleophile to stabilize the reactive intermediates formed during the depolymerization of lignin, and the corresponding ester/ether compounds could be obtained. This catalytic system was suitable for the C-O bond cleavage in α-O-4 and β-O-4 linkages with excellent efficiency. Additionally, reaction conditions were optimized. The reaction mixture was detected by 1 H NMR, and no other byproducts were found. As for treated lignin samples, the cleavage of C-O bonds in linkages was determined by 2D HSQC NMR, the increased content of the phenol hydroxyl group was proved by FT-IR, and the reduced molecular weight was investigated by GPC. Furthermore, multiple phenolic compounds were detected by GC-MS in the reaction mixtures.

Abstract:Lignin, the planet's most abundant renewable source of aromatic compounds, is difficult to degrade efficiently to welldefined aromatics. We developed a microwave-assisted catalytic Swern oxidation system using an easily prepared catalyst, MoO2Cl2(DMSO)(2), and DMSO as the solvent and oxidant. It demonstrated high efficiency in transforming lignin model compounds containing the units and functional groups found in native lignins. The aromatic ring substituents strongly influenced the selectivity of -ether phenolic dimer cleavage to generate sinapaldehyde and coniferaldehyde, monomers not usually produced by oxidative methods. Time-course studies on two key intermediates provided insight into the reaction pathway. Owing to the broad scope of this oxidation system and the insight gleaned with regard to its mechanism, this strategy could be adapted and applied in a general sense to the production of useful aromatic chemicals from phenolics and lignin.

Abstract:In order to better understand the specific substituent effects on the electrochemical oxidation process of β-O-4 bond, a series of methoxyphenyl type β-O-4 dimer model compounds with different localized methoxyl groups, including 2-(2-methoxyphenoxy)-1-phenylethanone, 2-(2-methoxyphenoxy)-1-phenylethanol, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanol, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanol have been selected and their electrochemical properties have been studied experimentally by cyclic voltammetry, and FT-IR spectroelectrochemistry. Combining with electrolysis products distribution analysis and density functional theory calculations, oxidation mechanisms of all six model dimers have been explored. In particular, a total effect from substituents of both para-methoxy (on the aryl ring closing to Cα) and Cα-OH on the oxidation mechanisms has been clearly observed, showing a significant selectivity on the Cα-Cβ bond cleavage induced by electrochemical oxidations.

Abstract:While the depolymerization of lignin to chemicals catalyzed by ionic liquids has attracted significant attention, the relevant molecular mechanism, especially the cleavage of specific bonds related to efficient depolymerization, still needs to be deeply understood for the complexity of this natural aromatic polymer. This work presents a detailed understanding of the cleavage of the most abundant β-O-4 bond in the model system, guaiacylglycerol β-guaiacyl ether, by a Brønsted acidic IL (1-methyl-3-(propyl-3-sulfonate) imidazolium bisulfate ([C3SO3Hmim][HSO4]) using density functional theory calculation and molecular dynamics simulation. It has been found that [C3SO3Hmim][HSO4] generates zwitterion/H2SO4 via proton transfer with an energy barrier of 0.38 kcal/mol, which plays a dominant role in the lignin depolymerization process. Subsequently, the reaction can be carried out via three potential pathways, including (1) the dehydration of α-C-OH, (2) dehydration of γ-C-OH, and (3) the protonation of β-O. The electrophilic attack of H2SO4 and the hydrogen-bonding interaction between GG and zwitterion are the two most important factors to promote the depolymerization reaction. In all steps, the dehydration of α-C-OH route is computed to be favored for the experiment. The relatively higher energy barrier for β-O-4 bond dissociation among these reaction steps is attributed to the hindrance of the self-assembled clusters of GG in the mixed system. Further, the dense distribution of H13([C3SO3Hmim]) surrounding O21(GG), indicated by sharp peaks in RDFs, reveals that -SO3H in cations plays a substantial role in solvating lignin. Hopefully, this work will demonstrate new insights into lignin depolymerization by functionalized ILs in biomass conversion chemistry.

Abstract:Lignin in lignocellulosic biomass is the only renewable source for aromatic compounds, and effective valorization of lignin remains a significant challenge in biomass conversion processes. We have performed density functional theory calculations and experiments to investigate the cleavage mechanism of the C–O ether bond in the lignin model compound 2-phenoxy-1-phenylethanol with a β-O-4 linkage over a Pd(111) catalyst surface model. We propose the favorable reaction pathway to proceed as follows: the dilignol reactant gets dehydrogenated first on the α-carbon and then on the −OH group to generate its corresponding ketone 2-phenoxy-1-phenylethanone; the ketone continues to get dehydrogenated on the β-carbon by first a equilibrated keto–enol tautomerization to its enol form and then −OH dehydrogenation; the C–O ether bond cleavage happens afterward, leading to one-aromatic-ring surface intermediates followed by hydrogenation to yield acetophenone and phenol.

Abstract:Hydrothermal-controlled selective cleavage of CβO and CαCβ bonds was well realized via modification the side-chain structure of lignin. To elucidate the reaction pathway and the origin of selectivity, the sites of the benzylic alcohol (Cα–OH) and benzylic ketone (CαO) on the cleavage mechanisms of βO4 lignin dimers were investigated. Results clearly demonstrated the hydrothermolysis process could be divided into three stages: (1) intramolecular elimination reactions, (2) the cleavage of CαCβ or CβO bonds and (3) secondary reactions of intermediates. Reaction pathways were highly susceptible to the side-chain structure. The CαCβ and CβO bonds of 1-(4-hydroxy-3-methoxyphenyl)-2-(2-methoxyphenoxy)-ethanone tended to directly cleave and form aromatic monomers. However, many different species dimers were produced via the intramolecular elimination reactions when the hydroxyl group was introduced at the Cα or Cγ position, which participated in a variety of reaction mechanisms, resulting in more complex products distribution. In addition, the results also indicated that dehydration of Cα-OH group significantly facilitated the cleavage of CβO bonds, but oxidation of the Cα-OH into Cα=O group weakened the CαCβ bond. Based on the compositions and structures of products, several cleavage pathways of dimers were proposed, explaining well the origin of selectivity.
A DFT Mechanistic Study on Base-Catalyzed Cleavage of the β-O-4 Ether Linkage in Lignin: Implications for Selective Lignin Depolymerization

Abstract:The detailed mechanism of the base-catalyzed C-C and C-O bond cleavage of a model compound representing the β -O-4 linkage in lignin is elucidated using DFT calculations at the M06/6-31G* level of theory. Two types of this linkage have been studied, a C2 type which contains no γ -carbinol group and a C3 type which contains a γ -carbinol. Cleavage of the C2 substrate is seen to proceed via a 6-membered transition structure involving the cation of the base, the hydroxide ion and the α-carbon adjacent to the ether bond. The reaction with KOH has the lowest activation barrier of 6.1 kcal mol −1 with a calculated rate constant of 2.1 × 10 8 s −1 . Cleavage of the C3 substrate is found to proceed via two pathways: an enol-formation pathway and an epoxide-formation pathway. The first path is the thermodynamically favored pathway which is similar to the pathway for the C2 substrate and is the preferred pathway for the isolation of an enol-containing monomer. The second path is the kinetically favored pathway, which proceeds via an 8-membered transition state involving a hydrogen hopping event, and is the preferred pathway for the isolation of an epoxide-containing monomer. The KOH-catalyzed reaction also has the lowest activation barrier of 10.1 kcal mol −1 along the first path and 3.9 kcal mol −1 along the second path, with calculated rate constants of 2.4 × 10 5 s −1 and 8.6 × 10 9 s −1 respectively. Overall, the results provide clarity on the mechanism for the base-catalyzed depolymerization of lignin to phenolic monomers. The results also suggest both NaOH and KOH to be the preferred catalysts for the cleavage of the β -O-4 linkage in lignin.

Abstract:Electrochemical oxidation is a promising and effective method for lignin depolymerization owing to its selective oxidation capacity and environmental friendliness. Herein, the electrooxidation of non-phenolic alkyl aryl ether monomers and β-O-4 dimers was experimentally (by cyclic voltammetry, in situ spectroelectrochemistry, and gas chromatography-mass spectroscopy) and theoretically (by DFT calculations) explored in detail. Compared to the reported literature (T. Shiraishi, T. Takano, H. Kamitakahara and F. Nakatsubo, Holzforschung, 2012, 66(3), 303-309), 1-(4-ethoxyphenyl)ethanol showed a distinguishable oxidation pathway, where the resulting carbonyl product surprisingly underwent a bond cleavage on alkyl-aryl ether to ultimately produce a quinoid like compound. In contrast, β-O-4 dimers, like 2-phenoxy-1-phenethanol and 2-phenoxyacetophenone also demonstrated electrochemical oxidation induced by Cβ-O and Cα-Cβ bond cleavages. For the oxidation products, the presence of the Cα-hydroxyl group in dimers was the key to selectively generate aldehyde-containing species under mild electrochemical conditions, otherwise it produces alcohol-containing products following a different mechanism compared to the Cα[double bond, length as m-dash]O containing dimers.

Abstract:Cleaving the C-C bonds in lignin to afford value-added low-molecular-weight aromatic products is promising but a challenging project. In this contribution, a mild and efficient strategy for one-step selective C-C bond cleavage in beta-O-4 and beta-1 lignin models has been developed over pyridinium-based photocatalysts. The strong electron-accepting ability of the pyridinium core unit and the neutral nitrogen atom of the pyridazine substituent group provide a proton coupled electron transfer (PCET) channel to promote hydrogen dissociation from the OH or CH groups to form the corresponding free radical intermediates, thus directly triggering the cleavage of C-C bonds via a beta-scission process. Benzaldehyde and phenyl formate are eventually formed as the main products. This work not only enriches the application of the pyridinium family as a low-cost, eco-friendly, and sustainable alternative for producing value-added chemicals from renewable biomass resources, but also provides a new idea for designing novel organic catalysts with PCET features to decrease the consumption of precious metals.
Highly selective cleavage C–O ether bond of lignin model compounds over Ni/CaO–H-ZSM-5 in ethanol

Abstract:Herein, 2-(2-methoxyphenoxy)-1-phenylethanol (β-O-4), 2-methoxyphenyl anisole (α-O-4) and 4-phenoxyphenol (4-O-5) were selected as typical lignin model compounds. Given the effectiveness of traditional acid–base catalysts for lignin depolymerisation, a novel Ni/CaO–H-ZSM-5(60) catalyst was prepared to investigate the difficulty level of C–O bond of three model compounds cleavage in ethanol. It was observed that Ni/CaO–H-ZSM-5(60) had prominent performance on the C–O bond cleavage at very mild conditions (140 °C, 1 MPa H2). Among them, the C–O bond of α-O-4 and β-O-4 could be completely cleaved within 60 min. Although the C–O bond of 4-O-5 had high bond energy, 41.2% of conversion was occurred in 60 min. The introduction of CaO could regulate the acidity of H-ZSM-5 to enhance the ability to break C–O bonds. Moreover, the possible pathways of C–O ether bonds in three lignin model compounds cleavage were proposed in order to selectively obtain target products from the raw lignin degradation.

Abstract:In this study, a β-O-4 dimer with abundant oxygen substituents was successfully synthesized, and was used as the model compound for lignin to study the pyrolysis mechanism. Py-GC/MS (micro pyrolyzer coupled with gas chromatography/mass spectrometry) was employed to identify the distribution of pyrolytic products under temperatures of 150–850°C. It was found that the yields of methoxylated monoaromatics underwent tendencies of first increase and then decrease at high temperature. Polyaromatics and benzofuran were only detected at high temperature. Based on experimental analysis, a detailed pyrolysis kinetic model was developed by combining the density functional theory (DFT) and the transition state theory (TST). The homolysis of Cβ-O was the most favorable route for the initial depolymerization of dimer rather than the concerted retro-ene fragmentation. For the evolution of intermediate products, the breakage of ether bond and the keto-enol tautomerization of enols were the most favorable routes, while intramolecular cyclization and group dissociation exhibited high energy barriers and low reaction rates. The methoxyl in guaiacol preferred to undergo demethylation leading to the formation of catechol.

Abstract:Lignin is an alternative renewable feedstock for the production of low molecular weight aromatics. The cleavage of phenyl side-chain CαCβ bond is a valid depolymerization pathway to obtain aromatic monomers. The selective cleavage of the CαCβ bonds in two lignin model dimers and lignin with ferric sulfate as the catalyst was investigated to narrow the distribution of aromatic monomers. The CαCβ bonds in phenolic and non-phenolic dimers were selectively cleaved under microwave irradiation. The acidity promoted the etherification of Cα-hydroxyl groups with methanol solvent. Both the etherification and the decomposition of alkenyl products occurred in fast reaction steps, but the formation of alkenyl products was a decomposition-determining step. Density functional theory (DFT) calculations indicated that the phenolic dimer was more thermodynamically favorable to form alkenyl product than non-phenolic dimer, which resulted in the CαCβ bond cleavage reactivity of phenolic dimer being higher than that of non-phenolic dimer. Microwave heating contributed to 96.3% of CαCβ bond cleavage of organosolv lignin (OL), and a narrow distribution of aromatic monomers which mainly contained vanillin, syringaldehyde, methyl vanillate, and methyl syringate. This work highlights the prospect of microwave-assisted selective cleavage of the CαCβ bond to achieve the selective production of aromatic monomers from lignin.

Abstract:A phenolic β - O -4-type lignin model compound with a C 6 -C 3 -type side-chain and two guaiacyl nuclei and its derivatives were individually acidolyzed in aqueous 82 vol% 1,4-dioxane containing 0.2 mol/L HBr, HCl, or H 2 SO 4 at 85 °C in this study. The obtained results, combined with those in the series of this study in which phenolic and non-phenolic C 6 -C 3 - and C 6 -C 2 -type analogues were used, generated the following results and proposals. The H 2 O addition reproducing the original compound is preponderant as a reaction mode of the benzyl cation intermediate (BC) over the other modes leading to the β - O -4 bond cleavage, especially in the acidolyses using H 2 SO 4 and those of C 6 -C 3 -type compounds. A well-known phenomenon whether formaldehyde is liberated from C 6 -C 3 -type compounds is regulated by acid types appears less clearly in phenolic than in non-phenolic compounds. Br ̄ as well as Cl ̄ nucleophilically assists the H ̄ transfer from the β - to α -carbon of BC, especially in C 6 -C 3 -types, which accelerates and regulates the acidolyses using HBr as well as HCl. Furthermore, Br ̄ is suggested to nucleophilically attack the β -carbon of non-phenolic C 6 -C 3 -type BC to cleave the β - O -4 bond, which explains the characteristically peculiar observation in the acidolysis of the non-phenolic C 6 -C 3 -type compound using HBr described in our previous report.

Abstract:It remains challenging to achieve the selective cleavage of C-C bonds in lignin or lignin model compounds to produce aromatic products in high yield and selectivity. We have developed a redox-neutral photocatalytic strategy to accomplish this goal in both beta-O-4 and beta-1 lignin models at room temperature (RT) via proton-coupled electron transfer (PCET) process without any pretreatments of substrate, by adjusting the alkalinity of base to obtain a lignin models/base PCET pair with a bond dissociation free energy close to 102 kcal/mol. Without breaking down C-beta-C-gamma bond and any C-O bonds, this PCET method is 100% atom economy and produces exclusive C-alpha-C-beta bond cleavage products, such as benzaldehydes (up to 97%) and phenyl ethers (up to 96%), in high to excellent yields and selectivities. Preliminary studies indicated that the PCET strategy is also effective for the depolymerization of native lignin at RT, thus providing significantly important foundation to the depolymerization of lignin. Abstract:Lignin valorization to aromatic fine chemicals requires selective cleavage of a high-energy C-C bond under mild conditions. Herein, we report the efficient and selective C-C bond cleavage of lignin model substrates through a chlorine (Cl) atom as the hydrogen atom transfer agent generated by visible-light excitation of the metal-free photocatalyst perylene diimide. With the assistance of the Cl atom, the overall yield of C-C bond cleavage products in model substrate beta-O-4 linkages reached 82.5% under visible-light excitation. This work makes use of metal-free photocatalysts and ubiquitous chloride as an alternative sustainable method for lignin depolymerization.
