Abstract:Batch-to-batch variability of traditional Chinese medicine (TCM) has already heavily limited its industrial development. By utilizing direct analysis in real time mass spectrometry (DART-MS) and multivariate statistical analysis, the present study introduced a novel and rapid methodology for TCM manufacturing process monitoring. Danshen alkaline precipitation was employed as an example operating unit to demonstrate the effectiveness of this new technique. A total of 15 batches run under normal operating conditions were used to develop a reference principal component analysis model and then enabled the establishment of multivariate control charts. Hotteling T2 and DModX charts were applied to examine batch-to-batch reproducibility of 12 test batches. Artificial variations including starting material change and process fault were identified, which was in a good agreement with conventional analysis result. Further understanding of the deviating process behavior was achieved by investigating the contribution plot of abnormal batches. The results showed that DART-MS, in conjunction with multivariate analysis, provided valuable information on model process and gave us a new perspective for TCM manufacturing process monitoring.

Abstract:Spectral domain optical coherence tomography (SD-OCT) has recently attracted a lot of interest in the pharmaceutical industry as a fast and non-destructive modality for direct quantification of thin film coatings that cannot easily be resolved with other techniques. While previous studies with SD-OCT have estimated the intra-tablet coating uniformity, the estimates were based on limited number of B-scans. In order to obtain a more accurate estimate, a greater number of B-scans are required that can quickly lead to an overwhelming amount of data. To better manage the data so as to generate a more accurate representation of the intra-tablet coating uniformity without compromising on the achievable axial resolution and imaging depth, we comprehensively examine an algebraic reconstruction technique with OCT to significantly reduce the data required. Specifically, a set of coated pharmaceutical tablets with film coating thickness in the range of 60-100 μm is investigated. Results obtained from performing the reconstruction reveal that only 30% of the acquired data are actually required leading to a faster convergence time and a generally good agreement with benchmark data against the intra-tablet coating uniformity measured with terahertz pulsed imaging technology.

Abstract:One unusual and challenging scientific field that has received only cursory attention to date is the three-dimensional (3D) microstructure and spatial distribution of drug(s) and formulation materials in solid dosage forms. This study aims to provide deeper insight into the relationships between the microstructure of multiple-unit pellet system (MUPS) tablets and the spatial distribution of the active pharmaceutical ingredient (API) and excipients to facilitate the design of quantitative models for drug delivery systems. Synchrotron radiation X-ray microcomputed tomography (SR-μCT) was established as a 3D structure elucidation technique, which, in conjunction with liquid chromatography coupled to mass spectrometry (LC-MS) or liquid chromatography with evaporative light-scattering detector (LC-ELSD) enables chemical analysis of tablets. On the basis of the specific interior construction of theophylline MUPS tablets, the spatial distribution of materials was acquired by quantifying microregion samples that had been validated by SR-μCT for their locations in the MUPS tablets. The 3D structure of the MUPS tablets was catalogued as three structural domains: a matrix layer (ML), a protective cushion layer (PCL), and pellets (PL). Compared with the components in the ML, components in the PL had a larger proportion of theophylline, sucrose, and diethyl phthalate and a smaller proportion of lactose and sodium lauryl sulfate, whereas glyceryl monostearate was found to account for a large portion of the PCL. Microstructural characterization-guided zonal chemical determination represents a new approach for quality assessment and the development of drug delivery systems with in-depth insight into their constituent layers on a new scale.
Characterization of Herbal Powder Blends Homogeneity Using Near-Infrared Spectroscopy

Abstract:Homogeneity of powder blend is essential to obtain uniform contents for the tablets and capsules. Near-infrared (NIR) spectroscopy with fiber-optic probe was used as an on-line technique for monitoring the homogeneity of pharmaceutical blend during the blending process instead of the traditional techniques, such as high performance liquid chromatograph (HPLC) method. In this paper NIRS with a SabIR diffuse reflectance fiber-optic probe was used to monitor the blending process of coptis powder and lactose (excipient) with different contents, and further qualitative methods, like similarity, moving block of standard deviation and mean square were used for calculation purposes with the collected spectra after the pretreatment of multiplicative signal correction (MSC) and second derivative. Correlation spectrum was used for the wavelength selection. Four different coptis were blended with lactose separately to validate the proposed method, and the blending process of "liu wei di huang" pill was also simulated in bottles to verify this method on multiple herbal blends. The overall results suggest that NIRS is a simple, effective and noninvasive technique can be successfully applied to the determination of homogeneity in the herbal blend.

Abstract:This exploratory study investigated the minimum required Raman mapping area for predicting sustained-release tablet dissolution profiles based on intra-tablet homogeneity. The aim was to minimize scanning time while achieving reliable dissolution profile predictions. To construct the sample set, we controlled the blending time to introduce variability in the homogeneity of the tablets. The dissolution prediction models were established using the partial least squares regression under different Raman mapping area. The accuracies of the prediction results were evaluated according to the difference factor f 1 and Intersection–Union two one-sided t-tests (IU TOST) methods, and the implications conveyed by the results were discussed. The results showed that the homogeneity of sustained-release tablet affects the minimum required mapping area, and the tablets with higher homogeneity show higher prediction accuracy when using the same mapping area to model the dissolution profiles of tablets.

Abstract:Tablets are usually spray-coated in the pharmaceutical industry, and coating uniformity is important for the coating process. In this study, discrete element method-discrete droplet method (DEM-DDM) is applied for the investigation of the coating process, tablets are described by different particle shape models, and the influence of different particle shape models on inter-tablet coating uniformity is investigated. Multi-sphere approach with two accuracies, multi-super-ellipsoid approach, and polyhedron approach are applied to describe the biconvex cylinder tablet. The proposed model is verified by a lab-scale coating experiment within a coating pan first. And then, the coating process in a coating drum with baffles is investigated. It can be found that inter-tablet coating uniformity is almost the same for the polyhedron and multi-super-ellipsoid approaches, and the inter-tablet coating uniformity obtained by the multi-sphere approach is lower than both of them. The influence of different parameters on inter-tablet coating uniformity is also studied, including the droplet diameter, the drum rotation speed, the spray range angle, and the nozzle number, which can provide guidance for the tablet coating process.

Abstract:Coating uniformity is important for the quality, safety, and efficacy of tablets in the pharmaceutical industry. Among the various research methods, discrete element method (DEM) is a powerful tool for numerically investigating the coating process in particle level, increasing the understanding of coating uniformity and improving related processes. In this study, the combination of DEM and discrete droplet method (DDM) is applied for the simulation of tablet coating process, including the intra-tablet coating uniformity of differently shaped tablets. The biconvex tablets are described by multi-super-ellipsoid approach in this study. In order to obtain the coating thickness distribution on each tablet, two particle surface-based meshing methods are proposed. The proposed intra-tablet DEM-DDM coating model is subsequently verified by the qualitative comparison with both theoretical models and experimental results from other literature. Finally, the influence of tablet shape on the intra-tablet coating uniformity is investigated, including tablets with different aspect ratios and height ratios.

Abstract:Process analysis can effectively stabilize pharmaceutical quality and optimize the control of production process. For the sustained-release digestion-aid tablets, the coating film thickness is an important indicator to measure the quality of products. Traditional method mainly spot-checks tablets and measures with visual microscopy, which is time-consuming and laborious. This study attempted to use a portable near-infrared spectroscopy for rapid detection of a Chinese medicine tablets from production line. First, PLS regression models were established for coating film at twelve different locations of the tablet section, and the results showed that the correlation coefficients of training and validation sets were all over 0.80. Subsequently, the twelve locations were divided into six groups to further establish regressions. After chemometrics optimization, the optimal of six group models were generally better than single location models, with Rc2 and Rv2 all above 0.85, and RMSEV values all below 2.0. The mean relative error of prediction of the optimal model was 9.49%. The pharmaceutical process detection based on the portable NIR spectroscopy met the demand of managing digestion-aid tablet coating data conveniently. The proposed approach can successfully realize on-site and online pharmaceutical monitoring and has a promising practical value.

Abstract:Objective:To establish a rapid analysis method of determining the contents of Danshensu and protocatechuic aldehyde in the dripping solution of Fufangdanshen dripping pills,and on-line monitoring the homogeneity of the blending process.Method:Near-infrared(NIR) transflectance spectroscopy was first used to collect NIR spectra of the dripping solution.With HPLC analysis values as reference,the partial least square regression(PLSR) calibration models were built after wavelength selection and multiplicative scatter correction(MSC) + 1st derivative pretreatment,which were applied to on-line determine the contents of ingredients in the dripping solution of Fufangdanshen dripping pills,and on-line monitor the homogeneity and end-point of the blending process.Result:The PLSR calibration models of NIR spectral have high correlation coefficients(R) and low root mean square error of prediction(RMSEP).Danshensu:r=0.9929,RMSEP=0.368;Protocatechuic aldehyde:r=0.9937,RMSEP=0.159.The proposed method was successfully used in on-line monitoring the blending process of dripping solution.Conclusion:NIR transflectance spectroscopy is accurate,robust and can be used routinely to determine the contents of ingredients and the uniformity of distribution in the solution of dripping pills.

Abstract:Assessment of particle distribution has a wide range of applications in scientific researches and industrial processes. The knowledge of uniformity, density and agglomeration of distribution is essential for material, medical and many other fields. In this paper, we propose a set of parameters for evaluating the uniformity, density and agglomeration of particle distribution which is based on image processing. Distribution uniformity is evaluated by the value of coefficient of variationcv based on voronoi diagram, and by the eccentricity and deviation angle of particle distribution. The density of distribution is investigated via numerical density and area density which illustrate the area distribution at certain density levels. The assessment of agglomeration is carried out through the separation of particle distribution and the counting of different agglomeration situations. These three aspects of assessment will be demonstrated to fully describe the particle distribution. Microscopic images taken from chip on glassCOG are used to show the assessment process, and the characteristics of distribution can be obtained clearly and comprehensively.
In-line monitoring of blending process of Zhongsheng pill powder using miniature near-infrared analyzer

Abstract:Objective: To establish a simple and rapid method based on diffuse reflectance NIR spectra to in-line monitor the blending process of Zhongsheng pill powder. Method: Zhongsheng pill powder consisted of Zhongsheng spray powder, artificial cowbezoar powder and bile arisaema powder. By means of a revolutionary miniature near-infrared analyzer mounted onto the blender, the powder homogeneity was determined in real time with noninterference. The moving block standard deviation (MBSD) method was used to identify the end point of blending process, providing real-time data and instant feedback about the blending course. Evaluation of blend uniformity was carried out by stratified in-process sampling, and the content of baicalin in samples was determined by high performance liquid chromatography (HPLC). Result: The results calculated through MBSD method were generally coincided with HPLC analysis. Blend homogeneity was assumed to be reached when the standard deviations between spectra approached 0.002, and RSD values of baicalin content in samples from six positions were lower than 5%. Moreover, stratified in-process sampling has significant influence on blending process when the powder loading volume was small. Conclusion; Based on this study, NIR spectroscopy can be possibly extended to be applied in blending process of Zhongsheng pill powder, ensuring a remarkable saving of time and manpower employed and creating a reliable and efficient process measurement technology.

Abstract:Very few efforts have been directed toward interpreting the differences between chromatographic and spectroscopic fingerprints for the purpose of quality control, although similarity analysis based on fingerprints is commonly applied to quality consistency evaluation of herbal medicines (HM). In the current study, comparative evaluation of high-performance liquid chromatography (HPLC), ultraviolet (UV), and near-infrared (NIR) fingerprint techniques combined with chemometric methods has been conducted to control the quality of Chinese herbal injections. A set of Danshen injections (DSI) are chosen as an example of the relevance to contemporary HM. Similarity analysis by principal component analysis, identification using linear discriminant analysis and k nearest neighbor classifiers, and control charts based on Hotelling's T-2 and DModX statistics are employed to evaluate the quality consistency of the DSI samples from three different manufacturers. The results in this study demonstrate that the UV spectroscopy technique combined with chemometric methods is useful for the identity and consistency evaluation of DSI. Such an approach is believed to be equally applicable to other HM.
An Application for the Quantitative Analysis of Pharmaceutical Tablets Using a Rapid Switching System Between a Near-Infrared Spectrometer and a Portable Near-Infrared Imaging System Equipped with Fiber Optics

Abstract:We present a rapid switching system between a newly developed near-infrared (NIR) spectrometer and its imaging system to select the spot size of a diffuse reflectance (DR) probe. In a previous study, we developed a portable NIR imaging system, known as D-NIRs, which has significant advantages over other systems. Its high speed, high spectral resolution, and portability are particularly useful in the process of monitoring pharmaceutical tablets. However, the spectral accuracies relating to the changes in the formulation of the pharmaceutical tablets have not been fully discussed. Therefore, we improved the rapid optical switching system and present a new model of D-NIRs (ND-NIRs) here. This system can automatically switch the optical paths of the DR and NIR imaging probes, greatly contributing to the simultaneous measurement of both the imaging and spot. The NIR spectra of the model tablets, including 0-10% ascorbic acid, were measured and simultaneous NIR images of the tablets were obtained. The predicted results using spot sizes for the DR probe of 1 and 5 mm diameter, resulted in concentrations of R2 = 0.79 and 0.94, with root mean square errors (RMSE) of 1.78 and 0.89, respectively. For tablets with a high concentration of ascorbic acid, the NIR imaging results showed inhomogeneity in concentration. However, the predicted values for the low concentration samples appeared higher than the known concentration of the tablets, although the homogeneity of the concentration was confirmed. In addition, the optimal spot size using NIR imaging data was estimated to be 5-7 mm. The results obtained in this study show that the spot size of the fiber probe, attached to a spectrometer, is important in developing a highly reliable model to determine the component concentration of a tablet.

Abstract:Monitoring of tablet quality attributes in direct vicinity of the production process requires analytical techniques that allow fast, non-destructive, and accurate tablet characterization. The overall objective of this study was to investigate the applicability of multispectral UV imaging as a reliable, rapid technique for estimation of the tablet API content and tablet hardness, as well as determination of tablet intactness and the tablet surface density profile. One of the aims was to establish an image analysis approach based on multivariate image analysis and pattern recognition to evaluate the potential of UV imaging for automatized quality control of tablets with respect to their intactness and surface density profile. Various tablets of different composition and different quality regarding their API content, radial tensile strength, intactness, and surface density profile were prepared using an eccentric as well as a rotary tablet press at compression pressures from 20MPa up to 410MPa. It was found, that UV imaging can provide both, relevant information on chemical and physical tablet attributes. The tablet API content and radial tensile strength could be estimated by UV imaging combined with partial least squares analysis. Furthermore, an image analysis routine was developed and successfully applied to the UV images that provided qualitative information on physical tablet surface properties such as intactness and surface density profiles, as well as quantitative information on variations in the surface density. In conclusion, this study demonstrates that UV imaging combined with image analysis is an effective and non-destructive method to determine chemical and physical quality attributes of tablets and is a promising approach for (near) real-time monitoring of the tablet compaction process and formulation optimization purposes.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:In this study, the application of near-infrared (NIR) spectroscopy for online monitoring of the Moluodan extraction process was investigated. Paeoniflorin, the main active component of Moluodan, was chosen as the quality index. Samples were partitioned into calibration and validation sets by random select and set partitioning based on joint X-Y distances algorithm (SPXY), respectively. Wavelengths for modeling were selected by manual method and competitive adaptive reweighted sampling (Cars) algorithm. Particle swarm optimizationbased least square support vector machines (PSO-LS-SVM) and partial least squares models were both established for quantitative analysis to determine the content of paeoniflorin online. At last, 8 models were obtained according to the combination of these algorithms and the SPXY-Cars-PSO-LS-SVM model had the best quantitative analysis performance compared with the other 7 models. Specifically, in the developed SPXY-Cars-PSO-LS-SVM model, the determination coefficients of the calibration R2c and validation R2v sets were 0.99 and 0.95, respectively, and the root mean square errors of the calibration and validation sets were 0.012 and 0.024 mg/ mL, respectively, and the relative standard errors of the calibration and validation sets were 2.84% and 6.34%, respectively. These results suggested that the appropriate sample partition, wavelength selection, and regression analysis methods in this study, namely, the SPXY-Cars-PSO-LS-SVM algorithm combined with NIR spectroscopy, could be an effective and real-time approach for online monitoring of the extraction process of Moluodan.

Abstract:Raman and near-infrared (NIR) chemical mapping are widely used methods in the pharmaceutical industry to understand the distribution of components within a drug product. Recent advancements in instrumentation have enabled the rapid acquisition of high-resolution images. The comparison of these techniques for the analysis of pharmaceutical tablets has not recently been explored and thus the relative performance of each technique is not currently well defined. Here, the differences in the chemical images obtained by each method are assessed and compared with scanning electron microscopy with energy dispersive X-ray microanalysis (SEM-EDX), as an alternative surface imaging technique to understand the ability of each technique to acquire a chemical image representative of the sample surface. It was found that the Raman data showed the best agreement with the spatial distribution of components observed in the SEM-EDX images. Quantitative and qualitative comparison of the Raman and NIR images revealed a very different spatial distribution of components with regards to domain size and shape. The Raman image exhibited sharper and better discriminated domains of each component, whereas the NIR image was heavily dominated by large pixelated domains. This study demonstrated the superiority of using Raman chemical mapping compared with NIR chemical mapping to produce a chemical image representative of the sample surface using routinely available instrumentation to obtain a better approximation of domain size and shape. This is fundamental for understanding knowledge gaps in current manufacturing processes; particularly relating the relationship between components in the formulation, processing condition, and final characteristics. By providing a means to more accurately visualize the components within a tablet matrix, these areas can all be further understood.
Batch-to-Batch Quality Consistency Evaluation of Botanical Drug Products Using Multivariate Statistical Analysis of the Chromatographic Fingerprint

Abstract:Botanical drug products have batch-to-batch quality variability due to botanical raw materials and the current manufacturing process. The rational evaluation and control of product quality consistency are essential to ensure the efficacy and safety. Chromatographic fingerprinting is an important and widely used tool to characterize the chemical composition of botanical drug products. Multivariate statistical analysis has showed its efficacy and applicability in the quality evaluation of many kinds of industrial products. In this paper, the combined use of multivariate statistical analysis and chromatographic fingerprinting is presented here to evaluate batch-to-batch quality consistency of botanical drug products. A typical botanical drug product in China, Shenmai injection, was selected as the example to demonstrate the feasibility of this approach. The high-performance liquid chromatographic fingerprint data of historical batches were collected from a traditional Chinese medicine manufacturing factory. Characteristic peaks were weighted by their variability among production batches. A principal component analysis model was established after outliers were modified or removed. Multivariate (Hotelling T 2 and DModX) control charts were finally successfully applied to evaluate the quality consistency. The results suggest useful applications for a combination of multivariate statistical analysis with chromatographic fingerprinting in batch-to-batch quality consistency evaluation for the manufacture of botanical drug products.

Abstract:Traditional Chinese medicine (TCM) products are usually manufactured through batch processes. To improve batch-to-batch reproducibility, the feasible approaches for real-time monitoring of batch evolution need to be developed. In-line near-infrared (NIR) spectroscopy combined with multivariate data analysis as an efficient process analytical technology (PAT) tool, is presented in this study for real-time batch process monitoring. Liquid–liquid extraction is a widely used purification technology in the TCM manufacture, and selected as the example to demonstrate the effectiveness of this PAT tool. Multi-way partial least squares (MPLS) model was developed based on in-line measured NIR spectral data of ten normal operation condition (NOC) batches. Three kinds of multivariate control charts (scores, Hotelling T2 and DModX) were used to monitor the evolution of six test batches with artificial batch variations, including the change of starting material quality attributes and abnormal operation conditions. The approach was found very effective for real-time monitoring of process deviations from NOC batches. It is an alternative promising tool for monitoring batch reproducibility of the unit operations during the manufacture of TCM.
