Abstract:Background The genes encoding adiponectin receptor 1 (ADIPOR1) and small ubiquitin-like modifier 4 (SUMO4) have been linked to anti-atherogenic effects, but little is known about whether polymorphisms in the two genes, acting separately or interacting, affect risk of coronary artery disease (CAD) without diabetes. Methods We genotyped 200 CAD patients without diabetes and 200 controls without CAD or diabetes at three single-nucleotide polymorphisms (SNPs) in ADIPOR1 and one SNP in SUMO4, which were chosen based on previous studies. Potential associations were also explored between these SNPs and clinical characteristics of CAD without diabetes. Results Risk alleles at three SNPs in ADIPOR1 (rs7539542-G, rs7514221-C and rs3737884-G) and the G allele at SNP rs237025 in SUMO4 significantly increased risk of CAD without diabetes, with ORs ranging from 1.79 to 4.44. Carriers of any of these four risk alleles showed similar adverse clinical characteristics. Compared with individuals with a CC or GC genotype, those with a GG genotype at rs3737884 were at significantly higher risk of CAD that affected the left anterior descending coronary artery (OR: 6.77, P = 0.009), the right coronary artery (OR: 4.81, P = 0.028) or a relatively large number of vessels (P = 0.04). Individuals carrying a risk allele at one or more of the three SNPs in ADIPOR1 as well as a risk allele at the SNP in SUMO4 were at significantly higher risk of CAD without diabetes than individuals not carrying any risk alleles (OR: 5.82, 95% CI: 1.23. 27.7, P = 0.013). Conclusions SNPs in ADIPOR1 and SUMO4 are associated with elevated risk of CAD without diabetes, and SNPs in the two genes may interact to jointly affect disease risk.
Association of Single Nucleotide Polymorphisms in ADIPOQ Gene with Risk of Hypertension: a Systematic Review and Meta-Analysis.

Abstract:BACKGROUND:Hypertension has been continuing to be a major contributor to the global burden of disease and to the global mortality, leading to over 10 million deaths each year. The purpose of this study was to investigate the association between Adiponectin gene polymorphism with Essential hypertension (EH).METHODS:PubMed, EMbase, the Cochrane Library, and China National Knowledge Infrastructure (CNKI) were searched independently by two investigators. Pooled odds ratios and 95% confidence intervals were calculated to estimate the associations of Adiponectin polymorphism with EH.RESULTS:Thirteen studies with 3198 cases and 3076 controls for meta-analysis (MA) were included in present study. Pooled results showed that rs2241766 polymorphism is associated with the risk of EH in the allelic model (G vs. T: OR=1.10; 95% CI, 1.01-1.21). In the <40 years subgroup, rs2241766 polymorphism is associated with the risk of EH in allele model (G vs. T: OR=1.43; 95% CI, 1.06-1.94), recessive model (GG vs. GT + TT: OR=5.26, 95% CI=1.47-18.76), homozygous model of GG (GG vs.TT: OR=5.27, 95% CI=1.47-18.95), and rs266729 in recessive model (GG vs. GT + TT: OR=2.33, 95% CI=1.33-4.08).CONCLUSIONS:Our meta-analysis results show that the rs2241766 polymorphism is associated with the risk of hypertension. There still need a larger sample with better design to verify.
Genetic Variability in Adiponectin Receptor 1 (adipor1) is Associated with the Risk of Coronary Artery Disease in Type 2 Diabetes in Chinese Population

Abstract:Objective To investigate whether adiponectin receptor 1 gene ( AdipoR1 ) polymorphisms were associated with the risk of coronary artery disease (CAD) in type 2 diabetic patients in China.Methods Of total 307 individuals with type 2 diabetes,205 with CAD as cases and 102 without CAD as controls were recruited.The PCR-RFLP assay or directly DNA sequencing was adopted to genotype all the subjects for a total of 10 haplotype-tagging single nucleotide polymorphisms (SNPs) in AdipoRl gene.The frequencies of genotypes and alleles at each SNP were compared between the case and control groups.Results (1) At SNP rs1342387,GG homozygotes had a higher risk for CAD compared to AA homozygotes in type 2 diabetics (OR= 2.245,95 % CI = 1.113-4.530,P = 0.024;OR' = 2.491, 95%CI 1.354-6.885,P' = 0.031,after adjusted for other known CAD risk factors),and the carriers of genotype AG had the increased risk of CAD compared to those of genotype AA(adjusted OR' = 3.053, 95%CI 1.085-5.718,P' = 0.007).(2) At SNP rs12045862,CC homozygotes had a higher risk for CAD compared to TT homozygotes (adjusted OR' = 2.751,95%' 1.063-7.115,P' = 0.037).(3) Genotypic combinations of SNPs rs1342387 and rs12045862 were associated with the risk for CAD in type 2 diabetic patients.GX/CC was associated with the increased risk for CAD(adjusted OR' = 3.646,95%CI 1.253- 10.608,P'=0.018).AA/TX provided protection from CAD in type 2 diabetic patients (adjusted OR' = 0.260,95%CI 0.113-0.601,P' = 0.002).(4) In overweight or obese (BMI≥24) type 2 diabetics, rs1342387 and rs12045862 showed significant interaction with obesity in relation to CAD risk (P for interaction0.05); the associated rsl342387 and rsl2045862 found in the general type 2 diabetics were consistent in association with CAD risk in the subgroup(P0.05) ;genotype CC at rs7539542 was found to associate with increased CAD risk compared to genotype GG(OP =4.714,P =0.036)in the subgroup. Conclusions Genetic variations in AdipoR1 are associated with the risk of coronary artery disease,and interact with obesity on CAD risk in type 2 diabetes in Chinese population.

Abstract:Background Adiponectin, an adipokine facilitating insulin action, has antiatherogenic effects. This study investigated whether common single nucleotide polymorphisms (SNPs) in the adiponectin gene influenced plasma adiponectin level and whether they were associated with the risk of coronary artery disease (CAD) and its angiographical severity in type 2 diabetes in Chinese population. Methods 11 tagging SNPs were genotyped in 1110 subjects with or without CAD in type 2 diabetes. Variants of adiponectin gene were determined by Taqman polymerase chain reaction method. The plasma adiponectin concentrations were measured by sandwich enzyme-linked immunosorbent assay. The severity and extent of coronary atherosclerosis were assessed using the angiographic Gensini score and Sullivan Extent score. Results Among the 11 SNPs, the minor G allele of SNP rs266729 was significantly associated with higher odds of CAD (odds ratio (95% CI) = 1.49 (1.10 - 2.16), P = 0.022) after adjusting for covariates. In stepwise multivariate logistic regression, SNP rs266729 was a significant independent factor of CAD. Multivariate linear regression analysis revealed that rs266729 (β = −0.101, P < 0.0001), rs182052 (β = −0.044, P = 0.0035), and rs1501299 (β = 0.073, P < 0.0001) were significantly associated with adiponectin level, and also indicated that the minor G allele of SNP rs266729 had higher Gensini score (β = 0.139, P < 0.001) and Sullivan Extent score (β = 0.107, P < 0.001). Haplotypes analysis revealed different haplotype distributions in case and control subjects (P = 0.0003), with two common haplotypes GGG and GAG of the rs266729, rs182052, and rs1501299 being associated in heterozygotes with a greater than threefold increase in cardiovascular risk (odds ratio (95% CI)=3.39 (1.83 - 6.30), P = 0.0001). Conclusions In our population, genetic variants in the adiponectin gene influence plasma adiponectin levels, and one of them is a strong determinant of CAD susceptibility and its angiographical severity in type 2 diabetes. This study has provided further evidence for a role of adiponectin in the development of CAD.

Abstract:OBJECTIVE:Adiponectin receptor 1 (encoded by ADIPOR1) is one of the major adiponectin receptors, and plays an important role in glucose and lipid metabolism. However, few studies have reported simultaneous associations between ADIPOR1 variants and type 2 diabetes (T2D), coronary artery disease (CAD) and T2D with CAD. Based on the "common soil" hypothesis, we investigated whether ADIPOR1 polymorphisms contributed to the etiology of T2D, CAD, or T2D with CAD in a Northern Han Chinese population.METHODS:Our multi-disease comparison study enrolled 657 subjects, including 165 with T2D, 173 with CAD, 174 with both T2D and CAD (T2D+CAD), and 145 local healthy controls. Six ADIPOR1 single nucleotide polymorphisms (SNPs) were genotyped and their association with disease risk was analyzed.RESULTS:Multi-case-control comparison identified two ADIPOR1 variants: rs3737884-G, which was simultaneously associated with an increased risk of T2D, CAD, and T2D+CAD (P-value range, 9.80×10(-5)-6.30×10(-4); odds ratio (OR) range: 1.96-2.42) and 16850797-C, which was separately associated with T2D and T2D+CAD (P-value range: 0.007-0.014; OR range: 1.71-1.77). The risk genotypes of both rs3737884 and 16850797 were consistently associated with common metabolic phenotypes in all three diseases (P-value range: 4.81×10(-42)-0.001). We observed an increase in the genetic dose-dependent cumulative risk with increasing risk allele numbers in T2D, CAD and T2D+CAD (P trend from 1.35×10(-5)-0.002).CONCLUSIONS:Our results suggest that ADIPOR1 risk polymorphisms are a strong candidate for the "common soil" hypothesis and could partially contribute to disease susceptibility to T2D, CAD, and T2D with CAD in the Northern Han Chinese population.

Abstract:Objective: Many studies have suggested that adiponectin gene might be involved in the development of coronary artery disease (CAD). However, the results have been inconsistent. In this study, the authors performed a meta-analysis to assess the associations of +45T/G, +276G/T and -11377C/G polymorphisms in adiponectin gene with CAD susceptibility. Methods: Published literature from PubMed and EMBASE databases were searched. Pooled odds ratio (OR) and corresponding 95% confidence interval (CI) were calculated using fixed-or random-effects model. Results: Sixteen studies (4394 cases/8187 controls) for +45T/G polymorphism, fifteen studies (3569 cases/7463 controls) for +276G/T polymorphism, and thirteen studies (3531 cases/7072 controls) for -11377C/G polymorphism were included in the meta-analysis. The overall results showed that there was a statistically significant association between -11377C/G polymorphism and CAD (G vs. C: OR=1.15, 95% CI 1.07-1.24). Similar results were observed among European (G vs. C: OR=1.11, 95% CI 1.02-1.20) and East Asian populations (G vs. C: OR=1.27, 95% CI 1.11-1.45). However, no significant association was found for +45T/G or +276G/T polymorphism with CAD susceptibility. Conclusions: The meta-analysis indicated the significant association of -11377C/G polymorphism, but not +45T/G or +276G/T polymorphism, with CAD susceptibility. However, large-scale studies with the consideration of gene-gene and gene-environment interactions should be conducted to investigate the associations in future. (C) 2012 Elsevier Ltd. Abstract:Single-nucleotide polymorphisms (SNPs) of ADIPOQ, ADIPOR1, and ADIPOR2 have been associated with type 2 diabetes mellitus (T2DM), but there are many conflicting results especially in Chinese populations. To investigate the contribution of the adiponectin genes and their receptors to T2DM, a case-control study was performed and 11 SNPs of ADIPOQ, ADIPOR1, and ADIPOR2 were genotyped in 985 T2DM and 1,050 control subjects. rs16861194 (−11426 A>G) in the putative promoter of ADIPOQ was associated with T2DM (P = 0.007; OR = 1.29, 95% CI 1.08–1.55). None of the other 10 SNPs were associated with T2DM in this study, although rs2241766 and rs1501299 were reported to be associated with T2DM in previous Chinese studies. There was also no significant difference found from the ADIPOQ haplotype analysis, which contains rs16861194. In addition, we also assessed potential gene-gene interactions in three genes and no interactions were found. In conclusion, our results supported the ADIPOQ gene as a possible risk factor for type 2 diabetes in Han Chinese population.
Genetically Driven Adiposity Traits Increase the Risk of Coronary Artery Disease Independent of Blood Pressure, Dyslipidaemia, Glycaemic Traits

Abstract:Adiposity has been associated with the risk of coronary artery disease (CAD) in observational studies, but their association may differ according to specific characteristics of studies. In Mendelian randomization (MR) analyses, genetic variants are used as instrumental variables (IVs) of exposures to examine causal effects to overcome confounding factors and reverse causation. We performed MR analyses for adiposity (n = 322,154) on risk of CAD (60,801 cases and 123,504 controls) based on the currently largest genome-wide association studies. The estimated associations between adiposity traits and CAD were calculated by an inverse-variance weighted method with and without excluding the IVs, which are associated with the well-known risk factors of CAD. Genetic variants are identified to be associated with the well-known risk factors of CAD by a cross-phenotype meta-analysis method. Our results furnished strong evidence for a causal role of adiposity in risk of CAD, with the odds ratios (ORs) for CAD being 1.53 (95% CI 1.36–1.72) for body mass index (BMI), 1.48 (1.20–1.96) for waist–hip ratio (WHR), and 1.34 (1.07–1.59) for WHR adjusted for BMI (WHRadjBMI), respectively. After excluding mediators-associated IVs from the MR analyses, the corresponding ORs were 1.46 (1.28–1.67) for BMI, 1.39 (1.01–1.93) for WHR, and 1.38 (1.04–1.84) for WHRadjBMI, respectively. Furthermore, our results suggested that central adiposity and general adiposity might pose a similar risk for CAD. In summary, our data supported that genetically driven adiposity traits imposed the risk of CAD independent of blood pressure, dyslipidaemia, glycaemic traits, and type 2 diabetes.
Association of ADIPOQ Gene Polymorphisms and Coronary Artery Disease Risk: a Meta-Analysis Based on 12 465 Subjects.

Abstract:Introduction: Coronary artery disease (CAD) is one of the most common cardiovascular diseases and is a major cause of morbidity and mortality worldwide. Various researchers have investigated the role of ADIPOQ gene in the risk of CAD, yet their results have been inconsistent.Methods: To evaluate the association between ADIPOQ genetic polymorphisms and CAD risk, relevant studies published before October 2011 were identified by searching PubMed and EMBASE. Studies were selected using previously defined criteria. The strength of the relationship between the four single nucleotide polymorphisms (SNPs) of the ADIPOQ gene and CAD risk was assessed using odds ratios (ORs).Results: A total of 12 465 subjects from 17 case-control studies were identified in the present study. Based on the relevant studies, it was determined that the risk of CAD was not associated with rs2241766 in any genetic model. Increased risk of CAD was associated with rs266729 in allele contrast (1.11, [1.03, 1.20]) and dominant genetic model (1.15, 95% CI: [1.05, 1.27]); increased risk of CAD was also associated with rs822395 in additive (1.63, 95% CI: [1.19, 2.22]) and recessive genetic model (1.71, 95% CI: [1.27, 2.30]). It was further determined that the rs1501299 polymorphism reduced the risk of CAD in the additive (0.80, 95% CI: [0.67, 0.94]) and recessive genetic model (0.81, 95% CI: [0.68, 0.95]). In the stratified analysis, significant associations were found in Asian subjects for rs266729 and in Caucasian subjects for rs1501299.Conclusion: There is an association between ADIPOQ gene polymorphisms and CAD risk. Different SNPs of the ADIPOQ gene have different associations with CAD risk, and appear to increase risk in individuals of Asian ethnicity while decrease the CAD risk in Caucasians. Abstract:BACKGROUND:Adaptor proteins containing PH domain, PTB domain, and leucine zipper motif 1 and 2 (APPL1/2) play a key role in cell proliferation in many tissues. APPL1 or APPL2 as an adaptor for adiponectin receptors mediates the signaling pathway of adiponectin which acts as an anti-atherosclerotic adipokine. This study aimed to investigate whether genetic variations in the APPL1/2 genes affect the risk of coronary artery disease (CAD) in Chinese patients with type 2 diabetes mellitus (T2DM).METHODS:Seven haplotype-tagging single nucleotide polymorphisms (tag-SNPs) were selected from CHB HapMap database (Phase II) and total 203 CAD-positive cases and 106 CAD-negative controls with T2DM were genotyped for the 7 tag-SNPs by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay.RESULTS:The minor allele G of rs4640525 at APPL1 locus was protective from CAD in patients with T2DM, with the carriers of genotype CC at higher risk of CAD compared with non-carriers (OR = 2.830, 95%CI 1.285 - 6.230, P = 0.010; OR' = 4.992, 95%CI = 1.758 - 14.173, P' = 0.003, after adjustment for the other known CAD risk factors); the homozygotes of AA at rs11112412 in APPL2 gene had higher risk of CAD compared with those of GG (adjusted OR' = 5.697, 95%CI 1.006 - 32.257, P' = 0.049).CONCLUSION:Genetic variation(s) in APPL1/2 may be associated with CAD risk in T2DM in Chinese population.
Association of the Adiponectin Gene Rs1501299 G>T Variant, Serum Adiponectin Levels, and the Risk of Coronary Artery Disease in a Chinese Population

Abstract:BACKGROUND:The present study aimed to investigate whether the single nucleotide polymorphism (SNP) 276G>T (rs1501299) of the adiponectin (ADIPOQ) gene was associated with the risk of coronary artery disease (CAD) and serum adiponectin levels in a Chinese population.METHODS:The rs1501299 polymorphism of the ADIPOQ gene was genotyped in 438 subjects with angiographically diagnosed CAD and 443 controls. Levels of serum adiponectin were determined in 152 CAD subjects and 155 controls.RESULTS:The CAD subjects had GT and TT genotypes more frequently, and had GG genotype less frequently than the controls. The OR increased and was significant after adjustment for known CAD risk factors. Significant difference was also observed with T allele being more frequent among the CAD subjects. The T allele at the rs1501299 polymorphism was associated with a higher risk of CAD. The mean adiponectin levels of CAD patients were lower than control subjects. No significant correlation was seen of different genotypes with serum adiponectin levels.CONCLUSIONS:The adiponectin rs1501299 G>T variant was positively related with an increased risk of CAD, and the CAD patients had lower adiponectin levels which were not affected by the different genotypes of rs1501299 in the present study.

Abstract:Objective: The study aimed to investigate the association between adiponutrin gene (ADPN) SNP rs20729707 and overweight/obesity or type 2 diabetes in Chinese population. Methods: We genotyped SNP rs2072907 using multiplex ligase detection reaction (LDR) assay in 500 type 2 diabetes patients and 331 nondiabetic subjects who were divided into groups as non-obese (n=155), and overweight/obese (n=176). Body height, weight, waist and hip circumferences were measured. WHR and BMI were determined. Total cholesterol, triglyceride and high density lipoprotein (HDL) were also measured. Insulin resistance was assessed by the homeostasis model assessment (HMA) method. Allele and genotype frequencies were compared between type 2 diabetes patients and nondiabetic controls, as well as between nondiabetic overweight/obese and nonobese subjects. Correlations between genotypes and obesity-related phenotypes were also analyzed in whole nondiabetic subjects, nondiabetic men and women respectively. Results: No significant correlation was found between SNP rs2072907 and obesity. C allele frequency tended to higher in male type 2 diabetic patients than that in nondiabetic controls. Significantly greater fasting insulin, 2 h insulin, the integrated area under the curve of plasma insulin levels and HOMA-IR were found in men with CC genotype compared with GG+GC genotype (P＜0.01~0.05). In the MANOVA analysis, SNP rs2072907 C allele was showed to be correlated with HOMA-IR in male. Conclusion: In conclusion, ADPN is not a major susceptable gene for obesity or type 2 diabetes in Chinese population. ADPN SNP rs2072907 might confer an increased risk of hyperinsulinaemia in nondiabetic male Chinese.

Abstract:BACKGROUND:Metabolic syndrome traits play an important role in the development of colorectal cancer. Adipokines, key metabolic syndrome cellular mediators, when abnormal, may induce carcinogenesis.METHODOLOGY/PRINCIPAL FINDINGS:To investigate whether polymorphisms of important adipokines, adiponectin (ADIPOQ) and its receptors, either alone or in combination with environmental factors, are implicated in colorectal cancer, a two-stage case-control study was conducted. In the first stage, we evaluated 24 tag single nucleotide polymorphisms (tag SNPs) across ADIPOQ ligand and two ADIPOQ receptors (ADIPOR1 and ADIPOR2) among 470 cases and 458 controls. One SNP with promising association was then analyzed in stage 2 among 314 cases and 355 controls. In our study, ADIPOQ rs1063538 was consistently associated with increased colorectal cancer risk, with an odds ratio (OR) of 1.94 (95%CI: 1.48-2.54) for CC genotype compared with TT genotype. In two-factor gene-environment interaction analyses, rs1063538 presented significant interactions with smoking status, family history of cancer and alcohol use, with ORs of 4.52 (95%CI: 2.78-7.34), 3.18 (95%CI: 1.73-5.82) and 1.97 (95%CI: 1.27-3.04) for smokers, individuals with family history of cancer or drinkers with CC genotype compared with non-smokers, individuals without family history of cancer or non-drinkers with TT genotype, respectively. Multifactor gene-environment interactions analysis revealed significant interactions between ADIPOQ rs1063538, ADIPOR1 rs1539355, smoking status and BMI. Individuals carrying one, two and at least three risk factors presented 1.18-fold (95%CI:0.89-fold to 1.58-fold), 1.87-fold (95%CI: 1.38-fold to 2.54-fold) and 4.39-fold (95%CI: 2.75-fold to 7.01-fold) increased colorectal cancer risk compared with those who without risk factor, respectively (P(trend) <0.0001).CONCLUSIONS/SIGNIFICANCE:Our results suggest that variants in ADIPOQ may contribute to increased colorectal cancer risk in Chinese and this contribution may be modified by environmental factors, such as smoking status, family history of cancer and BMI.

Relation Between ADIPOQ Gene Polymorphisms and Type 2 Diabetes in a Chinese Population.

Abstract:Objective: We aimed to investigate the relation between adiponectin gene (ADIPOQ) polymorphisms and type 2 diabetes mellitus (T2DM) in a Chinese population. Methods: The present study included 510 subjects with normal glucose tolerant (NGT) and 510 patients with type 2 diabetic. Five SNPs (rs2241767, rs3821799, rs182052, rs1501299 and rs7627128) were genotyped by TaqMan methods. Results: Of these 5 SNPs, three SNPs (rs1501299, rs182052, and rs7627128) were found to be significantly associated with T2DM. The haplotypes AAT (Construction of rs1501299, rs182052, and rs7627128) was frequent in T2DM patients (OR=2.051, 95% CI: 1.439 similar to 2.923, P<0.001), but GAT (Construction of rs1501299, rs182052, and rs7627128) was frequent in the control group (OR=0.65, 95% CI: 0.540 similar to 0.805, P<0.001). Conclusion: The ADIPOQ gene variants and haplotype were associated with the risk for development of type 2 diabetes.

No association was found between SNPrs2072907 and obesity or T2DM. ADPN was not an obesity/T2DM susceptibility gene in Chinese population.

Abstract:Objective To investigate the relationship between adiponectin rs266729 and rs1501299 gene polymorphism and the risk of coronary heart disease(CHD).Methods The adiponectin rs266729 and rs1501299 single nucleotide polymorphism(SNP) were geno typed in CHD patients(n =105) and non-CHD patients(n =75) using matrix-assisted laser desorption/ionization-time of flight(MALDI-TOF).The genotype and the frequency of each genotype and allele were determined.Results There was statistical difference in rs1501299 genotypes and allele gene between CHD group and control group(both P ＜ 0.05).The frequency distribution of each genotype and allele gene of rs266729 showed no statistical difference between CHD group and control group (both P ＞ 0.05).Logistic regression analysis revealed that adiponectin rs1501299 locus with G allele was independently associated with CHD(OR =1.731,95％ CI 1.091-2.747,P ＜ 0.05).Conclusion The rs1501299 site may be associated with the incidence of CHD,and the G allele may be the genetic susceptibility gene for CHD.
Genetic Variations In Dipeptidyl Peptidase 4 Gene Have A Synergetic Effect With Body Mass Index On The Risk Of Coronary Artery Disease In A Chinese Han Population With Type 2 Diabetes

Abstract:Objective: Dipeptidyl peptidase 4 (DPP4) not only plays an important role in pancreatic beta cell regulation, but also shows possible protective functions in cardiovascular system. The study aimed to determine whether genetic variations in DPP4 were related to the susceptibility of coronary artery disease (CAD) in Chinese patients with type 2 diabetes (T2D). Methods: In total, 745 patients with T2D of Chinese Han population were enrolled in the case-control study, and 501 of them were diagnosed as CAD. Six haplotype-tagging SNPs in DPP4 gene were selected and genotyped using polymerase chain reaction and restriction fragment length polymorphism. The association between gene polymorphisms and the risk of CAD was analyzed using logistic regression analyses. Results: Among all six genotyped SNPs, the variation of the SNP rs3788979 was significantly related to the risk of CAD in dominant inheritance mode after adjusting for confounders (OR' = 1.663, 95% CI' = 1.051-2.630, p' = 0.030). In subgroup analysis, the carriers of genotype GG or GA had a significantly higher risk of CAD, compared with genotype AA carriers in overweight and obesity subgroup (OR' = 2.070, 95% CI' = 1.256-3.416, p' = 0.004). The SNP rs3788979 showed an interactive effect with BMI in terms of CAD risk (beta = 0.191, p' for interaction = 0.048). In non-smoking subgroup, the carriers of genotype GG or GA had a raised risk of CAD than carriers of AA (OR' = 2.041, 95% CI' = 1.196-3.484, p' = 0.009). The smokers with GG or GA genotype had an increased CAD risk compared to non-smokers with AA genotype (OR' = 3.351, 95% CI' = 1.589-7.064, p' = 0.001). Conclusions: The SNP rs3788979 in the DPP4 gene is associated with the risk of CAD, and has a synergetic effect with BMI on the risk of CADin Chinese Han patients with T2D.

Abstract:OBJECTIVE:systemically evaluate the association between +276G/T single nucleotide polymorphisms of adiponectin gene and coronary atherosclerotic heart disease in Chinese population.METHODS:8 case-control studies about +276G/T polymorphism that were based on our inclusion criterion and available in the literature were reviewed. Meta analysis was carried out by Review Manager 4.2 software.RESULTS:Results from Metaanalysis demonstrated no significant heterogeneity among the studies about +276G/T polymorphisms and showed significant association between the +276G/T polymorphism and coronary disease in the Chinese population, and the fixed effects OR of the allele +276G to susceptibility of the disease was (OR = 1.93, 95% CI 1.32-2.82, P = 0.0007).CONCLUSION:The current paper on Meta-analysis demonstrated a correlation between the +276G/T single nucleotide polymorphism and the cononary disease in Chinese population.

Abstract:Background. Glucagon-like peptide 1 (GLP-1) bestows protective effects upon the cardiovascular system through direct cardiovascular interactions or by improvements to metabolic function. Both these effects are thought to be at least partly mediated by the GLP-1 receptor (GLP-1R). This case-controlled study investigated whether polymorphisms in the GLP-1R gene affect the risk of cardiovascular disease in type 2 diabetic patients in the Chinese Han population. Methods. Eleven haplotype-tagging single nucleotide polymorphisms (SNPs), distributed across 22 kb of the 39 kb GLP-1R gene, were selected and genotyped in diabetic patients from a Chinese Han population. Patients were classified based on the severity of coronary artery stenosis. Coronary artery stenosis was ≥50% in 394 patients (coronary artery disease- (CAD-) positive group), and coronary artery stenosis was <50% in 217 patients (control group). Allele and genotype frequencies were compared between the two groups at all 11 SNPs. Results. When considered in recessive inheritance mode, patients with the GG genotype at rs4714210 had a lower CAD risk than patients with other genotypes (OR = 0.442, 95% CI = 0.258–0.757, p=0.002), even when other known CAD risk factors were taken into account (ORa = 0.440, 95% CIa = 0.225–0.863, pa=0.017). In additive inheritance mode, GG genotype carriers at rs4714210 exhibited a lower risk of CAD than AA carriers (ORa = 0.475, CIa = 0.232–0.970, pa=0.041). Conclusion. In type 2 diabetic patients from a Han Chinese population, some variations in the GLP-1R gene were associated with a lower risk of developing CAD.
