Abstract:We report a rapid magnetic nanoparticle-mediated protocol to extract and detect DNA of E. coli O157:H7. Bare iron oxide nanoparticles (Fe 3 O 4 NPs) were used as solid phase support to extract high purity DNA from concentrated and spiked water samples. Electrochemical detection of the DNA is achieved by employing polyaniline-functionalized magnetic nanoparticles (PANI-MNPs) which serve as DNA pre-concentrator and transducer. The DNA-based nanobiosensor shows good selectivity and sensitivity (detection limit of 0.10 ng/μL DNA) and offers faster and comparable results relative to conventional methods. This DNA extraction and detection method is expected to be applicable in food, water, and environmental samples.
