Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:Herein,an organocatalytic enantioselective desymmetrizing hydroalkoxylationof 1,4- and 1,8-diynes is disclosed, which represents a unique chiralBronsted acid-catalyzed desymmetrization of diynes. This protocolprovides a facile and practical access to enantioenriched 1,3-diaminopropanolderivatives and & gamma;-butyrolactones with a wide substrate scopeand generally high enantioselectivities. Besides, the backbones ofthese products constitute the structural core of numerous bioactivemolecules, and they can serve as valuable precursors for the expeditiousassembly of versatile N- and O-heterocycles. Moreover, control experimentsand theoretical calculations are employed to confirm the mechanisticrationale and elucidate the origin of enantioinduction.

Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:A novel asymmetric Pd-catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates and alpha,beta-unsaturated pyrazolones has been developed to afford highly functionalized tetrahydrofuran-fused spiro-heterocyclic skeletons with excellent stereoselectivity. A diverse range of enantioenriched spiro-tetrahydrofuran-pyrazolones, possessing three contiguous stereogenic centers including one all-carbon quaternary, were obtained in high yields and excellent enantioselectivities (up to 90% yield and 98% ee). In addition, the practical application of this protocol was also demonstrated by gram-scale synthesis and further transformations into various compounds. Preliminary mechanistic experiments indicated that the allylic 1, 3-strain was crucial to the stereoselective control. We developed an asymmetric Pd-catalyzed [3 + 2] cycloaddition of alpha,beta-unsaturated pyrazolones with vinylethylene carbonates. An array of spiro-tetrahydrofuran-pyrazolones were obtained in high yields with excellent enantioselectivities.

Abstract:A series of novel chiral bifunctional tertiary amine–thioureas based on spirobiindane were designed and synthesized as organocatalysts. One of these catalysts was shown to promote the asymmetric Michael addition reaction of 1,3-diphenylpropane-1,3-dione to nitroolefins, affording the desired products in good yields (up to 95%) and enantioselectivities (up to 98% ee).

Abstract:In this paper, the organocatalytic asymmetric 1,4-Michael addition reaction of azadienes and α-thiocyanoindanones was investigated. A series of chiral benzofuran compounds containing thiocyano group and quaternary carbon center were synthesized in moderate yields with good enantioselectivities (up to 90:10 er) and high diastereoselectivities (up to >95:5 dr). This is the first case of 1,4-Michael addition reaction using α-thiocyanoindanones to obtain a series of chiral thiocyano compounds and further broaden the scope of application of azadiene substrates. In addition, a possible reaction mechanism is also described in the article.

Abstract:A novel type of highly efficient chiral C2-symmetric bipyridine-N,N'-dioxides ligands prepared from L-octahydroindole-2-carboxylic acid and successfully applied in the synthesis of optically pure 1,2,3-trisubstituted cyclopropanes employing sulfoxonium ylides with α,β-unsaturated 2-acyl imidazoles has been developed, affording the corresponding adducts in 88-98% yields with up to 99% enantioselectivity and >20:1 dr under mild conditions with broad scope. In addition, a scale-up experiment and the synthetic transformations of the cycloadducts further highlighted the synthetic utility.

Abstract:The catalytic enantioselective diorganozinc additions to cyclic diketones including pyrazolin-4,5-diones and isatins have been developed. In the presence of morpholine-containing chiral amino alcohol ligand, the corresponding chiral cyclic tertiary alcohols were produced in good to excellent yields (up to 97 %) and enantioselectivities (up to 95 % ee). The notable feature of this protocol includes its mild reaction conditions, Lewis acid additives free and broad functional group tolerance.

Abstract:The bifunctional organocatalyst C3 N,N'-dioxide has been successfully applied to the asymmetric cascade Michael/hemiacetalization reaction of α-substituted cyano ketones and β,γ-unsaturated α-ketoesters for the synthesis of multifunctionalized chiral dihydropyrans. The corresponding products were obtained in excellent yields (up to 99%) with high to excellent enantioselectivities (up to 99% ee).

Abstract:A novel asymmetric [4 + 2] cycloaddition of 2-methylidenetrimethylene carbonate with pyrrolidone-derived enones has been achieved to produce the functionalized tetrahydropyran-fused spirocyclic scaffolds via palladium-catalysis. An array of enantioenriched spiro-pyrrolidine-2,3-diones bearing adjacent quaternary and tertiary stereocenters are obtained in high yields with excellent enantioselectivities (up to 96% yield and 99% ee). The further transformation of the product has been accomplished for the construction of medical interesting β2,2-amino acids and β-lactams. Preliminary mechanistic research was well conducted.

Abstract:A highly efficient copper(I)-catalyzed asymmetric 1,3-dipolar cycloaddition of azomethine ylides with benzo[b] thiophene 1,1-dioxides as the dipolarophiles was developed, providing elegant access to novel chiral tricyclic pyrrolidine-fused benzo[b] thiophene 1,1-dioxide derivatives in good to excellent yields (up to 99%) with excellent diastereo- and enantioselectivities (up to > 25 : 1 dr, up to 99% ee). Furthermore, the cycloadducts could be conveniently converted to synthetically important tricyclic pyrrolidine-fused benzo[b] thiophene and tetracyclic oxazolidin-2-one derivatives via simple transformations.

Abstract:A novel chiral spirocyclic amide (SPA)-derived triazolium organocatalyst has been designed and demonstrated to effect asymmetric homo- and heterodialkylations of various bisoxindoles, enabling enantioselective construction of vicinal all-carbon quaternary stereocenters. These reactions feature excellent enantioand diastereoselectivities (up to 99% ee and >20:1 dr) as well as good to high yields (up to 89% over two steps). As an application of this methodology, the first asymmetric total synthesis of (-)-chimonanthidine has been achieved.

Abstract:Enantioselective transformations of olefins are among the most important strategies for the asymmetric synthesis of organic compounds. Chemo-, diastereo-, and stereoselective control of reactions with internal acyclic alkenes for the construction of functionalized acyclic alkanes still remain a persistent challenge. Here, we report a palladium- catalyzed asymmetric regiodivergent Heck-type diarylation of internal acyclic alkenes. The 1,2-diarylation of two accessible acyclic alkenes, cinnamyl carbamates and enamides with diazonium salts and aromatic boronic acids, furnishes products containing vicinal stereogenic centers via the stereospecific formation of carbonyl coordination-assisted transient palladacycles. Moreover, the asymmetric migratory diarylation of enamides enables the formation of incontiguous stereocenters by an interrupted diastereoselective 1,3-chain-walking process. This protocol streamlines access to highly functionalized multisubstituted enantioenriched carbamates and amine derivatives which are embedded in the key biologically active motifs.
