Abstract:Chiral phosphine-catalyzed asymmetric [3 + 2] annulation reaction of various Boc-amino-substituted chalcones with allenoates has been developed, leading to efficient formation of 1,4,5-trisubstituted cyclopentenes with high yields, excellent diastereoselectivities and enantioselectivities (up to 99% ee and up to 98% yield).

Abstract:A catalyst system, [Rh(COD)Cl]2-BINAP-AgSbF6, has been developed as a second-generation catalyst for the cycloisomerization of 1,6-enynes tethered by carbon chains. Cyclopentanes and cyclopentanones, which can contain functional groups, such as the 1,4-dienes, vinyl ether, aldehyde etc., were obtained from readily available starting materials in high yields and selectivities. Both regioselectivities and enantioselectivities are excellent: only 1,4-dienes were observed and in over 99% ee.

Abstract:Highly enantioselective [4 + 2] annulation of activated alkenes with α-substituted allenoates catalyzed by amino acid-based bifunctional phosphines has been developed for the first time, which provides an easy access to optically enriched functionalized cyclohexenes. Abstract:Asymmetric cycloaddition reactions are the most powerful tool to the expeditious construction of enantioenriched cyclic motifs in organic chemistry. In sharp contrast to well-developed cycloaddition reactions via the palladium-trimethylenemethane (Pd-TMM) intermediate, hetero (3 + 2) cycloadditions of the heteroallyl cations remain rare, largely due to their thermally forbidden nature. To the best of our knowledge, there is no example of asymmetric version leading to enantioenriched heterocycles reported so far. Herein we enabled the first example of catalytic asymmetric (3 + 2) cycloaddition of electrophilic palladium-heteroallyl zwitterion intermediate (Pd-OTMM or Pd-NTMM) with cyclic or acyclic 1,3-dienes via a pathway terminated with C-N or C-O bond formation, delivering the highly substituted or fused pyrrolidine and tetrahydrofuran rings in high yields with excellent regio-, diastereo-, and enantioselectivity. Engineering the PC-Phos, one of the chiral sulfinamide phosphine (Sadphos) type ligands, by introducing the di-tert-butyl or/and 3,5-difluorophenyl group is a vital component in achieving excellent catalytic reactivity and enantioselectivity.

Abstract:By employing readily available cyclopropylacetylene and its derivatives as the pentadienylation reagent, an asymmetric regioselective asymmetric α-pentadienylation reaction of aldehydes is developed by cooperative catalysis of a chiral Pd(0) catalyst and a chiral Brønsted acid in the presence of a subschoichmetric amount of an achiral amine. α-Pentadienylated aldehydes are afforded with high yields and enantioselectivities as well as excellent E/Z ratios.

Abstract:An enantio- and diastereodivergent [2+2+1] annulation reaction of allyl ketones, acidic methylene compounds, and nitroalkenes to assemble highly functionalized cyclopentanes from readily available substances enabled by asymmetric relay catalysis of chiral bifunctional squaramide and palladium complex has been established. This method showcases that allyl ketones can serve as latent 1,2-dication synthons via a linear-selective allylic C–H functionalization and sequential 1,4-conjugated addition, enabling the rapid assembly of cyclopentane skeleton with a broad scope of methylene nucleophiles and nitroalkenes. Notably, chiral bifunctional squaramide catalyst engages in both the intermolecular and intramolecular Michael addition reactions accompanying with a kinetic resolution behavior to amplify the enantioselection. In addition, the stereodivergent synthesis of diastereomers from the resultant chiral cyclopentane derivatives is also accessible by a simple manipulation with base.

Abstract:AbstractDue to the synergistic “push–pull” effect of vicinal electron‐donating and electron‐withdrawing groups, donor–acceptor (D‐A) cyclopropanes have been recognized as one of the most powerful building blocks to generate polyfunctional reactive intermediates after a strain‐driven ring cleavage. Enantioselective reactions of D‐A cyclopropanes provide an efficient approach to enantioenriched acyclic and cyclic compounds. A number of chiral Lewis/Brønsted acids, transition metals, and organocatalysts have been designed for such transformations, including ring‐openings, annulations, and rearrangements. This minireview highlights the developments and new advances in this field and describes new synthetic opportunities offered by these interesting methodologies.

Abstract:With the use of a bifunctional chiral phosphine as the catalyst, the asymmetric [3+2] annulation of Morita-Baylis-Hillman carbonates with cyclic 1-azadienes proceeded smoothly under mild conditions to give various enantiomerically enriched cyclopentene derivatives bearing three consecutive tertiary stereocenters and a sulfamate moiety in moderate to excellent yields with moderate to excellent enantioselectivities and excellent diastereoselectivities.

The methodology performs well over a broad scope of substrates, which provides facile access to a series of highly functionalized bicyclic pyrrolidine/cyclopentane derivatives in good to high yields with excellent stereoselectivities.

Abstract:Herein we report a catalytic enantioselective (2 + 3) cycloaddition in which a vinyl phosphonium intermediate serves as the 2-carbon component. The reaction involves an umpolung umpolung coupling sequence enabled by haloacrylates and chiral enantioenriched phosphepine catalysts. The reaction shows good generality providing access to an array of cyclopentenes, with mechanistic studies supporting stereospecific formation of the vinyl phosphonium intermediate which then undergoes annulation with turn over limiting catalyst elimination. Beyond defining a new approach to cyclopentenes these studies demonstrate that haloacrylates can replace ynoates in reaction designs that require exclusive umpolung coupling at both carbons

Abstract:The first Lewis acid and chiral Brønsted acid cooperatively catalyzed asymmetric cascade ring opening/aza-Piancatelli rearrangement reaction of furyl-substituted donor-acceptor cyclopropanes is achieved, enabling the construction of functionalized aminocyclopentenones bearing α-quaternary carbon stereocenters in high yields with excellent enantio- and diastereoselectivities under remarkably low catalyst loading of 0.2-1.2 mol %.

Abstract:An efficient double catalytic system, combining chiral amine and 2-mercaptobenzoic acid, is applied for α',β-regioselective [4+2] annulations of 2-cyclopentenone with a diversity of activated alkenes, constructing multifunctional chiral bicycle[2,2,1]heptane scaffolds in good to excellent yields and enantioselectivities. In comparison with the traditional cross-dienamine species between 2-cyclopentenone and chiral amine, the interrupted enamine intermediate containing a covalently linked thiol catalyst shows significantly improved reactivity.

Abstract:Herein, we report the enantioselective phosphine-catalyzed β,γ-annulation of electron-poor allenes with bifunctional malonates. The reaction exploits a 2C phosphonium synthon that when accessed using (R)-SITCP gives 23 cyclopentanes with high stereoselectivity (most >95:5 er and >9:1 dr) and yield. In addition to the (3+2) annulation, a one-pot three-component variant to give the same cyclopentanes and a (3+2) annulation/Dieckmann cyclization cascade, along with mechanistic studies, are reported.

Abstract:A highly enantio- and diastereoselective organocatalytic formation of cyclopropanes embedded in a complex bridged polycyclic architecture is disclosed. In the presence of a chiral phosphoric acid catalyst, this reaction generates four new stereogenic centers and three new C–C bonds efficiently from isochromene acetals and vinylboronic acids under mild conditions. Different from conventional asymmetric cyclopropanation strategies, this process does not involve carbenes or carbenoids. The complex products can serve as precursors to useful homoenolate equivalents. Mechanistically, DFT studies provided insights into the key transition states of the enantiodetermining [4+2] cycloaddition, in which the enantioselectivity is induced by the chiral phosphate counter anion of the isobenzopyrylium intermediate.

Abstract:An asymmetric intermolecular [4 + 2] cycloaddition of 1,3-dienes with dialkyl acetylenedicarboxylates, which was catalyzed by a rhodium(i)-chiral phosphoramidite complex, was developed. This protocol provided a highly enantioselective access to prepare carbonyl substituted cyclohexa-1,4-dienes with up to 96% yield and >99% ee. Notably, a cycloaddition on the 10 g scale gave the product in 92% yield and with 99% ee, which showed great potential for the scale-up synthesis of carbonyl substituted cyclohexa-1,4-dienes. In addition, oxidative aromatizations and hydrolysis of the products were also investigated.
